Abstract
The invention belongs to the technical field of cosmetics, and particularly relates to an anti-wrinkle composition and application thereof. The anti-wrinkle composition provided by the invention is prepared from beta-nicotinamide mononucleotide, echinacea purpurea extract, acetyl chitosamine, coffee extract, pullulanase polysaccharide and alpine edelweiss extract, wherein the weight ratio of beta-nicotinamide mononucleotide to echinacea purpurea extract is 0.01-5: 0.05 to 0.5:0.5 to 5:0.1 to 5:1 to 5:0.5 to 2. The anti-wrinkle composition provided by the invention has the advantages of high active ingredients and good anti-aging and anti-wrinkle effects due to the synergistic effect of the components, and can be applied to preparation of anti-oxidation, anti-aging and anti-wrinkle skin care products. The mask solution in the anti-wrinkle freeze-dried powder mask prepared from the anti-wrinkle composition provided by the invention is used together with the freeze-dried powder containing beta-nicotinamide mononucleotide, so that the skin water content and the skin elasticity value of a tester can be effectively improved, and the anti-wrinkle freeze-dried powder mask has a relatively obvious improvement effect on dry lines and wrinkles of the face.
Description
Technical Field
The invention belongs to the technical field of cosmetics, and particularly relates to an anti-wrinkle composition and application thereof.
Background
Skin aging refers to the deterioration of the skin's ability to protect and regulate the body, the inability of the skin to adapt to the changes of the internal and external environments, and the appearance of the skin to change its overall conditions such as color, luster, shape and texture. Skin aging is a natural phenomenon of the life process, and although not a deterrent, the pace of aging can be slowed by some means.
In recent years, in the research on various cosmetic raw materials for skin aging resistance, NMN is one of the important raw materials for anti-aging and anti-wrinkle which are newly discovered. NMN (β -nicotinamide mononucleotide), a precursor substance of the cofactor NAD + of the long-life protein in the human body. NAD + is also called coenzyme i, is known as nicotinamide adenine dinucleotide, also called nicotin diphosphate, and is involved in thousands of reactions in cellular respiration and energy metabolism in every cell. NAD + is an important coenzyme in the tricarboxylic acid cycle, promotes the metabolism of sugars, fats, and amino acids, and participates in the synthesis of energy. NAD + is in turn the sole substrate for coenzyme I consuming enzymes (the sole substrate for DNA repair enzyme PARP, the sole substrate for longevity protein Sirtuins, the sole substrate for cyclic ADP ribose synthase CD 38/157). Therefore, NAD + is very important for the human body and plays an important role in human body aging resistance. However, with aging, NAD + levels in the human body are gradually decreased, resulting in decreased mitochondrial activity, accelerated aging of mitochondria, cells, and even the whole body, and gradually enter the vicious circle, and the human body gradually ages. Therefore, NMN supplementation can help human self repair DNA damage, thereby achieving the purpose of delaying aging.
NMN is a single substance in body cells, has higher safety when being used as a skin care product, and can prevent skin from being rough and aged, reduce skin wrinkles and keep the skin bright and elastic when being used as the skin care product. However, NMN is easily decomposed in aqueous solution, which greatly affects the performance of its efficacy. In the current product formula, the NMN is applied to anti-aging and skin-care, so that the anti-aging and anti-wrinkle effects are greatly weakened in practical use, but the stability of the product in a dry environment is very good.
NMN is a single substance in body cells, and has high safety when being used as a skin care product, and the skin care product can prevent skin from being rough and aged, reduce skin wrinkles and keep the skin bright and elastic. At present, most of the products on the market are anti-aging products with single formula, and the stability of NMN in the formula of an aqueous solution is also a problem which is difficult to solve.
Therefore, how to synergistically improve the anti-wrinkle effect of the NMN and effectively improve the stability of the NMN in the formula of the facial mask is a technical problem which needs to be solved urgently.
Disclosure of Invention
In order to solve the problems of the prior art, it is an object of the present invention to provide an anti-wrinkle composition having excellent anti-wrinkle, anti-oxidation and anti-aging effects, and use thereof. The second purpose of the invention is to provide an anti-wrinkle freeze-dried powder mask composition which has the advantages of high active ingredients, oxidation resistance, aging resistance and water replenishing and relieving. The third purpose of the invention is to provide a preparation method of the anti-wrinkle freeze-dried powder mask composition, which has the advantage of maximally retaining bioactive substances and thermosensitive substances in materials so as to improve the efficacy of the mask.
The technical scheme of the invention is as follows:
an anti-wrinkle composition comprises beta-nicotinamide mononucleotide, echinacea purpurea extract, acetyl chitosamine, coffee extract, pullulanase polysaccharide and edelweiss extract.
Further, the weight ratio of the beta-nicotinamide mononucleotide, the echinacea purpurea extract, the acetyl chitosamine, the coffee extract, the pullulanase polysaccharide and the edelweiss extract is 0.01-5: 0.05 to 0.5:0.5 to 5:0.1 to 5:1 to 5:0.5 to 2.
Furthermore, the weight ratio of the beta-nicotinamide mononucleotide, the echinacea purpurea extract, the acetyl chitosamine, the coffee extract, the pullulanase and the edelweiss extract is 3:0.2:2:2:2:1.
in the above composition: the echinacea purpurea extract is rich in chicoric acid, can separate 27 triterpenoids with anti-inflammatory effects, can inhibit induced allergy of skin, and can treat dermatitis; can inhibit the permeability of capillary vessel to achieve anti-inflammatory effect; contains polyphenols and polysaccharides, and can scavenge free radicals in vivo and protect organism from damage of excessive free radicals generated by internal or external factors; the compound has obvious absorption peaks in UVA and UVB wave band ranges, can effectively inhibit inflammation caused by UVB irradiation, reduces redness, and improves the restoring force of cells under pressure conditions.
The acetyl chitosamine is small molecular polysaccharide, is the basic composition unit of many important polysaccharides in biological cells, and is an important precursor for synthesizing bifidus factors. The keratin metabolism is normalized by the glycoprotein on the surface of the keratinocyte, thereby removing the cutin and maintaining the normal metabolism function of the surface keratinocyte. And can promote skin hydration, thereby effectively increasing the transdermal absorbability of other effective components. The acetyl chitosamine can inhibit tyrosinase activity by inhibiting the glycosylation of tyrosinase, reduce melanin synthesis, reduce free radical damage to skin, resist wrinkle and aging, and enhance skin tissue repair ability.
The coffee extract is an alkaloid and can effectively resist free radicals threatening the health of people; contains high-content polyphenol compounds, is a powerful antioxidant; the coffee extract can also enhance vasoconstriction, stimulate cell metabolism, and promote blood circulation.
The pullulanase is obtained by fermenting aureobasidium pullulans, so that the pullulanase is called pullulanase. The pullulan polysaccharide is a straight chain homoglycan glucose consisting of glucose residues and is connected into maltotriose by alpha-1,4-glycosidic bond, the maltotriose is connected into macromolecule pullulan by alpha- (1 → 6) glycosidic bond, and the pullulan polysaccharide is widely applied to cosmetics to promote cell regeneration and belongs to a natural healthy cosmetic raw material.
The extract of the alpine edelweiss has the effects of inhibiting the activity of the elastin and promoting the generation of the elastin, can slow down the degradation of the elastin, can maintain the elasticity of the skin, and can be used as an anti-wrinkle cosmetic.
The components are reasonably mixed according to a specific proportion to obtain an effective anti-wrinkle composition, and experiments prove that the composition has quick, effective and continuous anti-wrinkle effect through the synergistic effect of the components, and can be applied to skin care products, such as anti-wrinkle essence, cream, facial masks and the like.
The invention also provides an anti-wrinkle lyophilized powder mask which comprises the anti-wrinkle composition;
the anti-wrinkle lyophilized powder mask consists of lyophilized powder, mask solution and mask cloth; the weight ratio of the freeze-dried powder to the facial mask solution is 1:4.
Further, the freeze-dried powder is prepared from the following raw materials in percentage by weight: 2 to 6 percent of mannitol, 1 to 3 percent of trehalose, 0.01 to 5 percent of beta-nicotinamide mononucleotide and the balance of water to 100 percent.
Further, the mask solution is prepared from the following raw materials in percentage by weight: 0.1 to 20 percent of humectant, 0.4 to 1 percent of preservative, 0.1 to 0.5 percent of thickening agent, 0.05 to 0.5 percent of echinacea purpurea extract, 0.5 to 5 percent of acetyl chitosamine, 0.1 to 5 percent of coffee extract, 1 to 5 percent of pullulanase, 0.5 to 2 percent of alpine edelweiss extract and the balance of water to 100 percent.
Further, the humectant is selected from two or more of glycerol, pentanediol, propylene glycol and sorbitol.
Further, the preservative is selected from one or more of 1,2-hexanediol, p-hydroxyacetophenone, phenoxyethanol, and ethylhexyl glycerin.
Further, the thickening agent is selected from one or more of xanthan gum, carbomer, cellulose, acryloyl dimethyl taurate/VP copolymer, acrylic acid (ester)/C10-30 alkanol acrylate cross-linked polymer.
Furthermore, the preparation method of the freeze-dried powder comprises the following steps:
s1, heating water to 80-90 ℃, cooling the water to 40-50 ℃, adding mannitol, trehalose and beta-nicotinamide mononucleotide, and uniformly stirring and mixing to obtain a colorless and transparent freeze-dried powder solution;
and S2, cooling the anti-wrinkle freeze-dried powder solution obtained in the step S1 to 38 ℃, adjusting the pH value to 5.0-7.0, and then carrying out vacuum freeze-drying to obtain the freeze-dried powder.
Further, the preparation method of the mask solution comprises the following steps:
(1) Heating water to 80-90 ℃, adding a thickening agent, keeping the temperature for 20min, and uniformly stirring to obtain a mixed solution;
(2) Cooling the mixed solution obtained in the step (1) to 40-50 ℃, adding a humectant, a preservative, an echinacea purpurea extract, acetyl chitosamine, a coffee extract, pullulanase polysaccharide and a edelweiss extract, uniformly stirring, cooling to 38 ℃, and adjusting the pH to 5.5-6.5 to obtain the colorless and transparent mask solution.
Compared with the prior art, the anti-wrinkle composition and the anti-wrinkle freeze-dried powder mask provided by the invention have the following advantages:
(1) The anti-wrinkle composition provided by the invention has the advantages of high active ingredients and good anti-aging and anti-wrinkle effects through the synergistic effect of the components such as the beta-nicotinamide mononucleotide, the echinacea purpurea extract, the acetyl chitosamine, the coffee extract, the pullulanase, the alpine edelweiss extract and the like, and can be applied to the preparation of anti-oxidation, anti-aging and anti-wrinkle skin care products, anti-wrinkle essence, cream, facial masks and the like.
(2) The mask solution in the anti-wrinkle freeze-dried powder mask prepared from the anti-wrinkle composition provided by the invention is used together with the freeze-dried powder containing beta-nicotinamide mononucleotide, so that the skin water content and the skin elasticity value of a tester can be effectively improved, and the anti-wrinkle freeze-dried powder mask has a relatively obvious improvement effect on dry lines and wrinkles of the face.

In the following examples and comparative examples, reagents not specifically described were conventional reagents and were purchased from conventional reagent manufacturers, and some of the raw material manufacturers were as follows:
echinacea purpurea extract was purchased from Denko jasmine fragrance (Nantong) Inc.; acetochitosamine was purchased from Huaxi Biotech Ltd; coffee extracts were purchased from i.r. aistitato Ricerche application srl; pullulanase polysaccharide was purchased from Hayashibara Co, ltd.; the extract of Hibiscus hirsutus is obtained from Luojie Biotech, inc., guangzhou; the beta-nicotinamide mononucleotide is purchased from VilQi medicine research and development Limited company in Shenzhen city.
Embodiment 1-5 an anti-wrinkle lyophilized powder mask and preparation method thereof
As shown in table 1 and table 2 below, a weight ratio table (in weight percentage) of 5 examples of the anti-wrinkle lyophilized powder mask of the present invention, including lyophilized powder and mask solution is provided.

The preparation method of the freeze-dried powder comprises the following steps:
s1, heating water to 90 ℃, cooling the water to 45 ℃, adding mannitol, trehalose and beta-nicotinamide mononucleotide, and uniformly stirring and mixing to obtain a colorless and transparent freeze-dried powder solution;
and S2, cooling the anti-wrinkle freeze-dried powder solution obtained in the step S1 to 38 ℃, adjusting the pH value to 5.0-7.0, and then carrying out vacuum freeze-drying to obtain the freeze-dried powder.
The preparation method of the mask solution comprises the following steps:
(1) Heating water to 90 ℃, adding a thickening agent, keeping the temperature for 20min, and uniformly stirring to obtain a mixed solution;
(2) Cooling the mixed solution obtained in the step (1) to 45 ℃, adding a humectant, a preservative, an echinacea purpurea extract, acetyl chitosamine, a coffee extract, pullulanase polysaccharide and a edelweiss extract, uniformly stirring, cooling to 38 ℃, and adjusting the pH to 5.5-6.5 to obtain the colorless and transparent mask solution. Test example I evaluation of anti-wrinkle Effect
1. Test materials: the anti-wrinkle freeze-dried powder mask prepared in the embodiment 1-5 and the comparative example 1-5 is prepared in the comparative example.
2. Test objects: healthy men and women of 30-55 years old have aging problems such as dry lines, fine lines, wrinkles, statute lines and the like on the faces; no allergic diseases, no allergic history of cosmetics and other external preparations; no history of photosensitive diseases exists in the past, and medicines influencing the photosensitivity are not used in the near term; the skin of the tested part has no phenomena of birthmarks, inflammations, scars, hirsutism and the like; it is possible to understand the course of the trial, voluntarily taking the trial, for a total of 110, divided into 11 groups of 10 persons each.
3. The test method comprises the following steps:
(1) Testing instrument and principle: skin analysis tester VISIA-CR: pictures were taken before and after the subjects used the product by VISIA-CR to compare facial/ocular wrinkles and texture changes.
Skin elasticity tester MPA580: the test principle is based on the principle of suction and stretching, where a negative pressure is generated on the skin surface to be tested to suck the skin into a specific test probe, and the depth of the skin sucked into the test probe is measured by a non-contact optical test system. The test probe includes a transmitter and receiver of light, the ratio of which (the ratio of transmitted light to received light is proportional to the depth of the skin being inhaled) and then analyzed by the MPA software to determine the elastic properties of the skin.
Skin moisture content tester CM825: the principle of moisture measurement is based on the fact that the dielectric constants (< 7) of water and other substances vary considerably, and that, depending on the moisture content, a suitably shaped measuring capacitor changes with the change in the capacitance of the skin, which is within the measurement range, so that the moisture content of the skin can be measured. A greater test value indicates greater skin moisture.
(2) The application method of the facial mask comprises the following steps:
the usage amount is as follows: the facial mask liquid and the lyophilized powder are mixed, and the mixture is administered 1 time each day in the morning and evening. The use of skin care cosmetics other than the test samples, oral preparations for anti-aging, etc. is avoided during the test.
(3) The test method and indexes are as follows:
subjects underwent VISIA photograph, skin elasticity, skin moisture test. The trial part is the whole face of the face, the samples are continuously used for 1 week, 2 weeks, 4 weeks and 8 weeks before the test respectively, the skin of the test part is tested by the same person, and the difference value before and after the test is compared. Before testing, the test can be carried out after the face is washed and then the room is kept at constant temperature and very wet for 15 min. And (3) testing environmental conditions: the test environment temperature is 20-22 ℃, the humidity is 40-60%, and real-time dynamic monitoring is carried out.
Testing indexes are as follows: surface texture, skin elasticity, skin moisture content, and calculating average surface wrinkle reduction, skin elasticity increase, and skin moisture increase for different time periods of the subject.
4. Test results
The experimental data of the skin moisture content values are shown in table 3 and fig. 1, the experimental data of the skin elasticity values are shown in table 4 and fig. 2, and the skin eye dry wrinkle change pictures are shown in fig. 3 and fig. 4.

As can be seen from the results of table 3 above and fig. 1, the anti-wrinkle composition of the present invention has a relatively significant effect of increasing the moisture content of the skin through the synergistic effect between the efficacy components, wherein example 3 is most significant; meanwhile, the results of comparative examples 1 to 5 show that the lack of any anti-wrinkle component is somewhat less effective than example 3, but also has a tendency to increase the moisture content of the skin compared to the blank control; the components achieve the effect mainly through the synergistic effect, and when one component is absent, the synergistic relationship among the components is destroyed, so that the overall effect is deteriorated.

As can be seen from the results of table 4 above and fig. 2, the anti-wrinkle composition of the present invention has a relatively significant effect of improving the skin elasticity value through the synergistic effect between the efficacy components, of which example 3 is most significant; meanwhile, the results of comparative examples 1 to 5 show that the lack of any anti-wrinkle component is less effective than example 3, but there is also a tendency to improve skin elasticity compared to the blank control, which is the least effective.
Further, fig. 3 and 4 are graphs of eye dry wrinkles after 8 weeks using the mask prepared in example 3 for two testers.
From the results of fig. 3 and 4, it can be seen that the anti-wrinkle composition of the present invention has a significant effect of improving dry lines and wrinkles of the face.

2. The anti-wrinkle composition according to claim 1, wherein the weight ratio of β -nicotinamide mononucleotide, echinacea purpurea extract, acetyl chitosamine, coffee extract, pullulanase polysaccharide, edelweiss alpine extract is 0.01 to 5:0.05 to 0.5:0.5 to 5:0.1 to 5:1 to 5:0.5 to 2.
3. The anti-wrinkle composition according to claim 1, wherein the weight ratio of β -nicotinamide mononucleotide, echinacea purpurea extract, acetyl chitosamine, coffee extract, pullulanase polysaccharide, edelweiss alpine extract is 3:0.2:2:2:2:1.
4. use of an anti-wrinkle composition according to any one of claims 1 to 3 for the preparation of an anti-wrinkle, anti-oxidant, anti-aging skin care product.
5. An anti-wrinkle lyophilized powder mask, characterized by comprising the anti-wrinkle composition according to any one of claims 1 to 3.
6. The anti-wrinkle lyophilized powder mask according to claim 5, which consists of lyophilized powder, mask solution and mask cloth; the weight ratio of the freeze-dried powder to the mask solution is 1:4.

The anti-wrinkle lyophilized powder mask as claimed in claim 5, wherein the lyophilized powder is prepared from the following raw materials in percentage by weight: 2 to 6 percent of mannitol, 1 to 3 percent of trehalose, 0.01 to 5 percent of beta-nicotinamide mononucleotide and the balance of water to 100 percent;
the mask solution is prepared from the following raw materials in percentage by weight: 0.1 to 20 percent of humectant, 0.4 to 1 percent of preservative, 0.1 to 0.5 percent of thickening agent, 0.05 to 0.5 percent of echinacea purpurea extract, 0.5 to 5 percent of acetyl chitosamine, 0.1 to 5 percent of coffee extract, 1 to 5 percent of pullulanase, 0.5 to 2 percent of alpine edelweiss extract and the balance of water to 100 percent.
8. The anti-wrinkle lyophilized powder mask according to claim 5, wherein the moisturizer is selected from two or more of glycerin, pentanediol, propylene glycol, and sorbitol;
the preservative is selected from one or more of 1,2-hexanediol, p-hydroxyacetophenone, phenoxyethanol and ethylhexyl glycerol;
the thickening agent is selected from one or more of xanthan gum, carbomer, cellulose, acryloyl dimethyl ammonium taurate/VP copolymer, acrylic acid (ester)/C10-30 alkanol acrylate cross-linked polymer.
9. The anti-wrinkle lyophilized powder mask as claimed in claim 5, wherein the preparation method of the lyophilized powder comprises the following steps:
s1, heating water to 80-90 ℃, cooling the water to 40-50 ℃, adding mannitol, trehalose and beta-nicotinamide mononucleotide, and uniformly stirring and mixing to obtain a colorless and transparent freeze-dried powder solution;
