Verification of the essence described in examples 1-3 and comparative examples 1-10 with reduced skin itching and erythema
1. Test instrument:
1. the invention uses a percutaneous moisture loss measuring instrument of Finland Delfin VapoMeter SWL5201, and the VapoMeter uses a closed cavity measuring principle, so that the measurement is not influenced by indoor air flow. The measurement time is short, ensuring the minimization of blocking normal evaporation.
2. Using finnish Delfin antra 3D photographs, antra 3D uses innovative optical methods and complex mathematical algorithms to acquire three-dimensional images. This way, data can be extracted from images relating to the three-dimensional shape of the skin, which will enable quantification of the treatment effect and monitoring of the condition over time. For example, the texture, smoothness, and depth and width of fine lines and wrinkles of the skin are measured. With its multispectral analysis, the antra software can also measure pigmentation and redness, which will provide meaningful information about sun damage, veins, etc.
2. Experimental principle:
TEWL values, transdermal water loss (Transepidermal water loss, TEWL) measures the change in vapor pressure of moisture adjacent to the skin surface. TEWL value indicates the loss of water from the stratum corneum, and is an important criterion for evaluating barrier function of the stratum corneum. The lower the TEWL value of the skin, the better the barrier function of the skin, and vice versa. The efficacy of the moisturizing and repairing cosmetic can be evaluated by testing the value of the TEWL of the skin moisture loss, and the moisturizing and repairing cosmetic can also be applied to the monitoring of allergic patch tests, contact dermatitis, physiotherapy, burn and new tissues, and whether the protection function of the skin is destroyed can be found in time. Wherein, the unit of TEWL is: g/hm 3 。
3. Test population:
40 persons in each group, 3 groups of volunteers aged 18-60 years to meet the experimental requirements.
4. Test conditions:
the subjects were rested for 30 minutes at room temperature of 20.+ -. 2 ℃ and relative humidity of 50.+ -. 10% and were monitored dynamically in real time and the relevant data was retested.
5. The experimental process comprises the following steps:
1. establishment of damaged skin barrier condition
200 mu L + -10 mu L of sodium dodecyl sulfate solution 1% (w/w) is injected into the small chamber of the patch tester, the filter paper sheet is soaked, the filter paper sheet is adhered to the curved sides of the left and right forearms of a subject by using hyposensitization tapes, the distance between the edges of the small chamber of the adjacent patch tester is not less than 3cm, and the filter paper sheet is uniformly adhered to the skin by using palm light pressure for 24 hours.
After removing the plaque tester, the test person sits still for 30min in a test environment which accords with the standard (after the indentation disappears), the test person measures the TEWL value for the center position of each plaque test area of the test person by using a Delfin vapoMeter (skin water shunt loss instrument), each area is measured once, and the measurement result is expressed by the average value of the TEWL values of three plaque test areas and is taken as an initial value (TEWL 0); photographing each plaque area of the subject by using a Delfin Antera3D camera, analyzing by using an instrument, and setting a measurement result to RED0 by taking the measurement average value of the three plaque areas as a redness initial value;
the left arm and the right arm are smeared with samples randomly, so that the equilibrium is ensured to be reached statistically;
the sample is dispensed to the subject and the subject is allowed to smear according to instructions, the smear of the sample being required to completely cover the entire plaque area. Measuring the sample using amount by an analytical balance, and using for 48 hours;
before each test, cleaning the tested part with clear water, sucking the tested part with a chipless water-absorbing dry paper towel, and resting for 30min under a constant temperature and humidity environment (the temperature is 20-22 ℃ and the relative humidity is 40-60%), so that the tested part cannot drink water and beverages, and the tested part keeps relaxed to avoid touching the patch area;
the test subject is subjected to a return visit at 24h and 48h after using the sample according to the use requirement, the volunteer fills out a questionnaire, the skin condition of the tested area of the volunteer is recorded, the test subject enters a test room to rest 30 minutes in advance, and the test step of T0 is repeated;
test part: experiments were performed with arm sections as shown in FIG. 1
The first set of zones Z1 used example 1, zone Z2 used comparative example 3, zone Z3 used comparative example 1, zone Z4 used comparative example 2, zone Z5 was blank;
the second set of zones Z1 used example 1, zone Z2 used example 3, zone Z3 used comparative example 10, zone Z4 used example 2, zone Z5 was blank, zone Z6 used comparative example 9;
the third group of zones Z1 used example 1, zone Z2 used comparative example 5, zone Z3 used comparative example 7, zone Z4 used comparative example 4, zone Z5 used comparative example 6, zone Z6 used comparative example 8.
6. Experimental results:
1. skin Transcutaneous Water loss (TEWL) test
The test sets TEWL at day 0 as TEWL0 measured in the subject, TEWL as TEWL1 after 24 hours, TEWL as TEWL2 after 48 hours, TEWL change values as Δtewl, Δtewl=tewl2-TEWL 0, TEWL change rate = Δtewl/tewl0 x 100%, TEWL change rate is negative, and the lower the value, the less moisture loss of skin after 48 hours, i.e. the better the skin barrier function is repaired, the healthier the skin is, table 1 shows the average Δtewl values for each group, as shown in table 2 below:
TABLE 2
2. Pi Fufan Red average result analysis
The average RED value RED of the skin on day 0 measured by a subject is set as RED0, RED is set as RED1 after 24 hours, RED is set as RED2 after 48 hours, RED change value is Δred, Δred=red 2-RED 0, RED change rate= Δred/RED0 is 100%, RED change rate is negative, and the lower the value is, the smaller the RED degree of the skin after 48 hours is, namely, the less inflammatory reaction in the skin is, the allergic symptoms are reduced, and the better the effects of relieving allergy and reducing erythema of the product are. Table 2 shows the average Δred values for each group, as shown in table 3 below;
TABLE 3 Table 3
3. Volunteer questionnaire
According to volunteer questionnaire statistics, 100% of volunteers using the examples considered the product to have reduced itching, tingling, and no subjective intention to scratch the area under test; the use of the volunteers of the comparative example was 50% of the volunteers considered to reduce itching, tingling, and 50% of the volunteers had the sensation of wanting to scratch the test area.
Experimental results illustrate:
from the above experimental findings, it can be seen that:
the first group of results, TEWL change rate and RED change rate, comparative examples 1-3> example 1, demonstrate that the effects of better repairing skin barrier, reducing skin moisture loss, reducing skin erythema and reducing itching feeling can not be achieved by arbitrarily changing the active ingredients in the essence of the invention into the commonly used ingredients with good anti-allergy and repairing effects.
The second set of results, TEWL rate of change and RED rate of change, comparative example 10> example 1> comparative example 9> example 2> example 3, show that the higher the amount of active ingredient added in the essence of the present invention, the smaller the TEWL and RED rate of change, i.e., the less the skin moisture loss, the less the reddish skin symptoms, the more complete the barrier function of the stratum corneum of the skin, the less the skin moisture loss, and the better the skin erythema reducing effect. In comparative example 9, the active ingredients of the essence of the invention are further improved, but the experimental results prove that the content of each ingredient is blindly improved, and the skin barrier can not be better repaired, and the skin redness symptom is relieved, so that the content interval of the essence of the invention is the interval with the best effect. The essence of comparative example 10 is inferior to the essence of the present invention in repairing skin barrier and reducing redness, so that it is shown that the essence of the present invention is a very excellent product for reducing skin sensitive symptoms and reducing skin redness and itching.
The third group of results, TEWL change rate and RED change rate, comparative examples 4-8> example 1, show that the effects of repairing skin barrier, reducing erythema and reducing skin itching feeling are not as good as those of the essence of the invention when only a certain active ingredient in the essence of the invention is added and the instant addition amount reaches the maximum value.
Comparison of test pictures
The front-back comparison of the arm redness images of a part of volunteers using the examples 1-3 of the present invention is shown in fig. 2-4, respectively, and it can be intuitively seen from the figures that the skin redness reduction is obvious by using the examples 1 and 2 as shown in fig. 2 and 3, and the skin redness reduction is most obvious by using the example 3 as shown in fig. 3, which indicates that the skin redness reduction capability of the essence of the present invention is very excellent.
