Verification of essence solutions described in examples 1 to 3 and comparative examples 1 to 10 for reducing skin itch and erythema
1. Testing the instrument:
1. the invention uses a finland Delfin vapoMeter SWL5201 percutaneous water loss measuring instrument, and the vapoMeter uses a closed cavity measuring principle, so that the measurement is not influenced by indoor air flow. The measurement time is short, ensuring the minimum of the blockage of normal evaporation.
2. Photographs were taken using Delfin antra 3D, finland, the antra 3D using innovative optical methods and complex mathematical algorithms to acquire three-dimensional images. This allows data to be extracted from the image relating to the three-dimensional shape of the skin, which will enable quantification of the effect of the treatment and monitoring of the condition over time. For example, the texture, smoothness of the skin, and depth and width of fine lines and wrinkles are measured. With its multispectral analysis, the Antera software can also measure pigmentation and redness, which will provide meaningful information about sun damage, veins, etc.
2. The experimental principle is as follows:
TEWL value, transdermal water loss (TEWL), measures the change in the vapor pressure of moisture near the surface of the skin. TEWL values represent the loss of water from the stratum corneum and are an important criterion for evaluating the barrier function of the stratum corneum. A lower TEWL value of the skin indicates a better barrier function of the skin, whereas the opposite is the worse. The efficacy of the moisturizing and repairing cosmetic can be evaluated by testing the TEWL value of the skin moisture loss, and the method can also be applied to allergic patch test, contact dermatitis, physical therapy, burn and monitoring of new tissues, so as to find out whether the protective function of the skin is damaged or not in time. Wherein the unit of TEWL is: g/hm 3 . 3. Testing the population:
40 persons in each group, 3 groups in total, and volunteers with the age of 18-60 years according with the experimental requirements.
4. And (3) testing conditions are as follows:
subjects rested for 30 minutes at room temperature 20 ± 2 ℃ and relative humidity 50 ± 10% and were monitored dynamically in real time and were retested for relevant data.
5. The experimental process comprises the following steps:
1. establishment of impaired skin barrier
Injecting 200 μ L + -10 μ L sodium dodecyl sulfate solution 1% (w/w) into the cell, infiltrating the filter paper sheet, applying to the left and right forearm of the subject with hypoallergenic tape, wherein the edge distance between adjacent cells is not less than 3cm, and applying to skin by pressing with palm for 24 hr.
2. After removing the spot tester, the testee sits still for 30min in a standard test environment (after the indentation disappears), a tester uses a Delfin Vapometer to measure TEWL values at the central position of each spot test area of the testee, each area is measured, and the measurement result is expressed by the average value of the TEWL values of three spot test areas and is used as an initial value (TEWL 0); photographing each spot test area of the subject by using a Delfin Antera3D camera, analyzing by using an instrument, and setting the average value of the measurement of the three spot test areas as a redness initial value as RED0;
3. randomly smearing samples on the left and right arms to ensure that the balance is achieved statistically;
4. samples were distributed to subjects and allowed to smear according to the instructions, the sample application required complete coverage of the entire plaque area. Measuring the using amount of the sample by using an analytical balance for 48 hours;
5. before each test, cleaning the tested part with clear water, drying with a non-scrap absorbent dry paper towel, and resting in a constant-temperature and constant-humidity environment (temperature: 20-22 ℃, relative humidity 40-60%) for 30min, so that water and beverage can not be drunk, the tested person keeps relaxed, and the spot sticking area is prevented from being touched;
6. according to the use requirements, the test subject makes a return visit at 24h and 48h after using the sample, the volunteers fill in questionnaires, the skin condition of the tested area of the volunteers is recorded, the test room is entered 30 minutes in advance for rest, and the test step of T0 is repeated;
7. testing parts: experiments were performed with arm zones as shown in FIG. 1
The first set of zone Z1 used example 1, zone Z2 used comparative example 3, zone Z3 used comparative example 1, zone Z4 used comparative example 2, and zone Z5 blank;
the second set of zone Z1 used example 1, zone Z2 used example 3, zone Z3 used comparative example 10, zone Z4 used example 2, zone Z5 blank, zone Z6 used comparative example 9;
the third set of zone Z1 used example 1, zone Z2 used comparative example 5, zone Z3 used comparative example 7, zone Z4 used comparative example 4, zone Z5 used comparative example 6, and zone Z6 used comparative example 8.
6. The experimental results are as follows:
1. skin transdermal Water loss Rate (TEWL) test
The test sets the subject's measured TEWL on day 0 as TEWL0, TEWL after 24 hours as TEWL1, TEWL after 48 hours as TEWL2, TEWL variance as Δ TEWL,. DELTA.tewl = TEWL2-TEWL0, TEWL variance =Δtewl/TEWL0 100%, TEWL variance is negative and the lower the number, the less water lost from the skin after 48 hours, the better the skin barrier function repaired and the healthier the skin, table 1 shows the average Δ TEWL values for each group as shown in table 2 below:
TABLE 2
First group | TEWL Change Rate (%) | Second group | TEWL Change Rate (%) | Third group | TEWL Change Rate (%) |
Example 1 | -15.350 | Example 1 | -17.516 | Example 1 | -16.427 |
Comparative example 1 | -9.851 | Example 2 | -31.084 | Comparative example 4 | -12.184 |
Comparative example 2 | -8.273 | Example 3 | -43.118 | Comparative example 5 | -10.837 |

2. Average skin redness results analysis
The test set the average RED-burning value RED of the skin on the 0 th day measured by the subject as RED0, RED1 after 24 hours, RED2 after 48 hours, RED change value as Δ RED = RED2-RED0, RED change rate =Δred/RED0 x 100%, RED change rate is negative, and the lower the value, the smaller the RED-burning degree of the skin after 48 hours, i.e. the less the inflammatory reaction inside the skin, the lower the allergic symptoms, the better the product relieves the allergy, and reduces the erythema. Table 2 shows the average Δ RED values for each set, as shown in table 3 below;
TABLE 3
3. Volunteer questionnaire
According to the statistics of the questionnaire of the volunteers, 100% of the volunteers using the examples considered that the product had reduced itching and tingling sensation, and wanted to scratch the test area without subjective will; the use of comparative example in 50% of volunteers was considered to reduce the itching, tingling sensation, and 50% of volunteers had the sensation of trying to scratch the test area.
The experimental results show that:
according to the above experimental conclusion, it can be seen that:
1. the first group of results, namely the TEWL change rate and the RED change rate, are comparative examples 1-3 and example 1, and show that the active ingredients in the essence provided by the invention are arbitrarily changed into the common ingredients with good anti-allergy and repairing effects, so that the effects of better repairing skin barriers, reducing skin moisture loss, reducing skin erythema and reducing itching feeling cannot be achieved.
2. The second group of results, the TEWL change rate and the RED change rate, the comparative example 10, the example 1, the comparative example 9, the example 2 and the example 3 show that the TEWL change rate and the RED change rate become smaller correspondingly as the addition amount of the active ingredients of the essence liquid is higher, namely, the skin moisture loss is smaller, the skin redness symptom is smaller, the stratum corneum barrier function of the skin is more complete, the skin is not easy to lose moisture, and the effect of reducing the skin erythema is better. The comparative example 9 is to improve the active ingredients of the essence of the invention, but the experimental results prove that the content of each ingredient is improved blindly, so that the skin barrier can not be repaired better and the skin redness symptom can not be relieved, and therefore, the content interval of the essence of the invention is the interval with the best effect. The essence of comparative example 10 is inferior to the essence of the present invention in terms of repairing skin barrier and reducing redness, thus indicating that the essence of the present invention is a very excellent product for reducing skin sensitivity symptoms, and reducing skin redness and itching.
3. The third group of results, the TEWL change rate and the RED change rate, comparative examples 4-8 and example 1 show that the effect of repairing skin barrier, reducing erythema and reducing skin pruritus is not as good as that of the essence of the invention when only one active ingredient in the essence of the invention is added and the instant addition amount reaches the maximum value.
4. Test Picture comparison
Before and after selecting partial arm redness images of volunteers using examples 1-3 of the present invention, detailed images are shown in fig. 2-4 respectively, and it can be seen from the figures that the reduction of skin redness is more obvious using examples 1 and 2 as shown in fig. 2 and 3, and the reduction of skin redness is most obvious using example 3 as shown in fig. 3, which shows that the ability of the essence of the present invention to reduce skin redness is very excellent.
