Abstract

The mass percentage distribution ratio and the preparation parameters of the components in the moisturizing, repairing and anti-aging composition containing ceramide are shown in the following table 1:

The moisturizing, repairing and anti-aging composition containing ceramide of example 1 is prepared by the following steps:
(1) Proportionally mixing phospholipid, emulsifier, ceramide NP, cholesterol and antioxidant, heating to 80 deg.C, and stirring to obtain oil phase; wherein the emulsifier is PEG-20 sorbitan cocoate, and the antioxidant is vitamin E.
(2) Mixing water, glycerol, and butanediol at a certain proportion, heating to 80 deg.C, and making into water phase.
(3) Adding the heated water phase into the oil phase, rapidly stirring, homogenizing for 3min to obtain emulsion;
(4) Circulating the emulsion for 3 times by a high-pressure homogenizer under the pressure of 800bar to obtain a transparent ceramide liposome solution;
(5) Cooling to 45 deg.C, adding antiseptic composed of caprylyl hydroximic acid and ethylhexyl glycerol, and mixing to obtain the final product.
The moisturizing, repairing and anti-aging composition containing ceramide in example 2 is prepared by the following steps:
(1) Proportionally mixing phospholipid, emulsifier, ceramide NP, cholesterol and antioxidant, heating to 85 deg.C, and stirring to obtain oil phase; wherein the emulsifier is PEG-40 hydrogenated castor oil, and the antioxidant is coenzyme Q10.
(2) Mixing water, glycerol, and butanediol at a certain proportion, heating to 85 deg.C, and making into water phase.
(3) Adding the heated water phase into the oil phase, rapidly stirring, homogenizing for 5min, and making into emulsion;
(4) Circulating the emulsion for 3 times by a high-pressure homogenizer under the pressure of 1000bar to obtain a transparent ceramide liposome solution;
(5) Cooling to 45 deg.C, adding antiseptic composed of caprylhydroxamic acid and ethylhexyl glycerol, and mixing well to obtain the final product.
The moisturizing, repairing and anti-aging composition containing ceramide in example 3 is prepared by the following steps:
(1) Proportionally mixing phospholipid, emulsifier, ceramide NP, cholesterol and antioxidant, heating to 85 deg.C, and stirring to obtain oil phase; wherein the emulsifier is a mixture of PEG-20 sorbitan cocoate and PEG-40 hydrogenated castor oil in a ratio of 1.
(2) Mixing water, glycerol and butanediol at a certain proportion, heating to 85 deg.C, and making into water phase.
(3) Adding the heated water phase into the oil phase, rapidly stirring, homogenizing for 5min, and making into emulsion;
(4) Circulating the emulsion for 3 times by a high-pressure homogenizer under the pressure of 1200bar to obtain a transparent ceramide liposome solution;
(5) Cooling to 45 deg.C, adding antiseptic composed of caprylhydroxamic acid and ethylhexyl glycerol, and mixing well to obtain the final product.
The moisturizing, repairing and anti-aging composition containing ceramide of example 4 is prepared by the following steps:
(1) Proportionally mixing phospholipid, emulsifier, ceramide NP, cholesterol and antioxidant, heating to 85 deg.C, and stirring to obtain oil phase; wherein the emulsifier is a mixture of laureth-23, steareth-21 and tween 80, and the antioxidant is a mixture of vitamin E and coenzyme Q10.
(2) Mixing water, glycerol and butanediol at a certain proportion, heating to 85 deg.C, and making into water phase.
(3) Adding the heated water phase into the oil phase, rapidly stirring, homogenizing for 5min, and making into emulsion;
(4) Circulating the emulsion for 3 times by a high-pressure homogenizer under the pressure of 1200bar to obtain a transparent ceramide liposome solution;
(5) Cooling to 45 deg.C, adding capryl hydroximic acid antiseptic, and mixing to obtain the final product.
The moisturizing, repairing and anti-aging composition containing ceramide in example 5 is prepared by the following steps:
(1) Proportionally mixing phospholipid, emulsifier, ceramide NP, cholesterol and antioxidant, heating to 85 deg.C, and stirring to obtain oil phase; wherein the emulsifier is polyglycerol 10 myristate, and the antioxidant is idebene.
(2) Mixing water, glycerol, and butanediol at a certain proportion, heating to 85 deg.C, and making into water phase.
(3) Adding the heated water phase into the oil phase, rapidly stirring, homogenizing for 5min, and making into emulsion;
(4) Circulating the emulsion for 3 times by a high-pressure homogenizer under 1500bar pressure to obtain transparent ceramide liposome solution;
(5) Cooling to 45 deg.C, adding ethylhexyl glycerol as antiseptic, and mixing to obtain the final product.
Particle size measurement
The size and the distribution of the particle size are very important to the stability of the system, so that the stability of the moisturizing and repairing anti-aging composition system containing ceramide can be intuitively reflected by continuously detecting the particle size in different time periods.
Table 2 shows the average particle size data of different moisturizing and rejuvenating anti-aging compositions containing ceramide (samples of examples 1 to 5) stored at room temperature for 1 to 28 days.

As can be seen from table 2, the particle size of the moisturizing, repairing and anti-aging composition containing ceramide prepared in examples 1 to 5 is about 200nm, the average particle size increases with the time, the increase of the homogenization pressure is beneficial to delaying the aggregation of phospholipid ceramide liposome molecules, and when the homogenization pressure is 1500bar, the particle size of the moisturizing, repairing and anti-aging composition containing ceramide according to the present invention changes minimally, so a high homogenization pressure should be selected during homogenization.
Moisturizing repair test
The moisturizing, repairing and anti-aging compositions containing ceramide prepared in examples 1 to 5 were dispersed in serum at an addition amount of 10%, 50 patients with mild hormone-dependent disease history (red and swollen skin, itching and pain) were randomly divided into 5 groups of 10 patients each, sprayed once in the morning and evening, and tested for TEWL values before and after use after 7 consecutive days; among them, TEWL refers to transdermal water loss, and is one of the indexes for evaluating the skin barrier function. When barrier function is impaired, the TEWL value increases; conversely, when the TEWL value decreases, barrier repair is suggested.
Wherein the rate of change = (after use-before use)/100% before use, and a smaller rate of change indicates a slower water loss and a better barrier function of the skin.

From table 3, it can be seen that: the skin of a patient can be obviously improved by adding the essence water of the moisturizing, repairing and anti-aging composition of 10 percent, and the moisturizing, repairing and repairing effects of the composition are proved to be in line with expectations.

3. the moisturizing, repairing and antiaging composition containing ceramide as claimed in claim 1 or 2, wherein the emulsifier is one or more selected from PEG-20 sorbitan cocoate, PEG-40 hydrogenated castor oil, polyglycerol 10 myristate, laureth-23, steareth-21, and Tween 80.
4. The moisturizing, repairing and anti-aging composition containing ceramide as claimed in claim 1 or 2, further comprising 0.1-1% by mass of an antioxidant, wherein the antioxidant is one or more of vitamin C, vitamin E, coenzyme Q10 and idebene.
5. A method for preparing the moisturizing, repairing and anti-aging ceramide-containing composition as claimed in any one of claims 1 to 4, comprising the steps of:
(1) Proportionally mixing phospholipid, emulsifier, ceramide NP, cholesterol and antioxidant, heating, stirring, and making into oil phase;
(2) Mixing water, glycerol, and butanediol at a certain proportion, and heating to obtain water phase;
(3) Adding the heated water phase into the oil phase, quickly stirring uniformly, and homogenizing for 3-5 min to prepare emulsion;
(4) Circulating the emulsion for 3 times by a high-pressure homogenizer under the pressure of 800-1500 bar to obtain a transparent ceramide liposome solution;
(5) Cooling, adding appropriate amount of antiseptic, and mixing to obtain the final product.
6. The preparation method of moisturizing, repairing and anti-aging ceramide-containing composition according to claim 5, wherein: heating the oil phase in the step (1) and the water phase in the step (2) to 80-85 ℃.
7. The method for preparing the moisturizing, repairing and anti-aging ceramide composition as claimed in claim 5, wherein the composition comprises: the preservative in the step (5) is a mixture of caprylhydroxamic acid and ethylhexyl glycerin.
