The gentian root extract, the yeast/zinc fermentation product, the wenzhou mandarin orange peel extract, the aframomum melegueta seed extract and the tetrahydro-methylpyrimidine carboxylic acid are combined, and the anti-inflammatory repair effect is improved through the mutual synergistic effect of the components; the anti-inflammatory repairing composition can remove free radicals of the skin and inhibit the damage effect of the free radicals on skin cells; the anti-inflammatory repairing composition can repair contact dermatitis and allergic dermatitis caused by external stimulation, and quickly recover the homeostasis of skin.
Description
Technical Field
The invention relates to an anti-inflammatory repairing composition and a preparation method and application thereof, belonging to the field of cosmetics.
Background
The skin is used as a barrier for the human body to contact with the outside, mainly consists of three layers, namely epidermis, dermis and subcutaneous tissue, and plays an important role in protecting the health of the human body. Skin is the tissue that wraps the surface of the body outside the muscles and is the largest organ of the human body. The skin is composed of sebum membrane, horny layer keratin, lipid, etc., and has effects in resisting harmful substances, irritants, ultraviolet rays, etc., keeping moisture, regulating and resisting inflammation. Environmental pollution, over-cleaning of skin, hormones, antibiotics, etc. cause increasingly sensitive and problematic skin. In order to accelerate the repair effect of some existing repair skin care products, an inflammatory factor inhibitor such as an antihistamine anti-allergy agent is added into the skin care product, and the inflammatory factor inhibitor can only simply relieve anaphylactic reaction and cannot repair skin barriers. Furthermore, some of the inflammation factor inhibitors are dependent on the skin even after long-term use.
Disclosure of Invention
The anti-inflammatory repairing composition can solve various skin inflammation problems, repair contact dermatitis and allergic skin caused by stimulation and enable the skin to quickly recover homeostasis.
In order to achieve the purpose, the invention adopts the technical scheme that: an anti-inflammatory remedial composition comprising active efficacy ingredients comprising gentian root extract, yeast/zinc fermentation product, citrus unshiu peel extract, aframomum melegueta seed extract and tetrahydro-methyl pyrimidine-carboxylic acid; the active functional component accounts for 0.5-10.5% of the composition by mass.
Radix Gentianae belongs to Gentiana (Gentiana) family, and has effects of clearing heat, eliminating dampness, clearing liver fire, and treating jaundice due to damp-heat, cholecystitis, anorexia, conjunctival congestion, urinary tract infection, herpes zoster and hyperthyroidism. Modern pharmacological research shows that the plant has the effects of resisting bacteria, resisting tumor, invigorating stomach, slowing down heart rate, etc.; the yeast/zinc fermentation product is also called as zinc-rich yeast, is rich in zinc element necessary for skin metabolism, and zinc, amino acid, polypeptide and other high-activity small molecular substances exist in a chelating form, so that the unique skin irritation of inorganic zinc is reduced, the zinc-rich yeast/zinc fermentation product is more suitable for being used in skin care products, has the effects of yeast essence, and has various skin benefiting effects of diminishing inflammation, removing acne, controlling oil, reducing dandruff and the like; the wenzhou mandarin orange peel extract has the function of promoting the generation of the cerebral amide, and the cerebral amide has very important physiological functions in organisms, so the extract has the function of activating skin, has good anti-aging effect by combining the proliferation capacity of the extract on cells, and has good anti-aging performance by combining the proliferation capacity of the extract on the cells; aframomum melegueta seed extract is obtained from Aframomum melegueta seed of West Feddedy, from a herb of the family of the same genus as ginger (Zingiberaceae) and perennial, by opening white to pink flowers to form the future fruit, a bright red capsule, containing many conical seeds, due to its powerful ginger-like active molecule gingerol has anti-inflammatory effect and flavonoid has antioxidant effect; the tetrahydro-methyl pyrimidine carboxylic acid exists in halotolerant bacteria as osmotic pressure compensating solute, plays a chemical transmitter-like role in cells, has a stable protection effect on the cells in adverse environment, and has the functions of activating skin and resisting aging. Through a large number of experiments, the inventor of the application discovers that the combination of the gentian root extract, the yeast/zinc fermentation product, the wenzhou mandarin orange peel extract, the aframomum melegueta seed extract and the tetrahydro-methyl pyrimidine carboxylic acid has a synergistic effect and can obviously improve the anti-inflammatory repair effect of the composition.
As a preferred embodiment of the anti-inflammatory rejuvenating composition according to the present invention, in the active efficacy ingredients, the mass ratio of the gentian root extract, the yeast/zinc fermentation product, the satsuma mandarin peel extract, the aframomum melegueta seed extract and the tetrahydro methylpyrimidine carboxylic acid is gentian root extract: yeast/zinc fermentation product: citrus unshiu peel extract: aframomum melegueta seed extract: tetrahydro-methyl-pyrimidine-carboxylic acid =0.1 to 3:0.15 to 3:0.1 to 2:0.1 to 2:0.05 to 0.5.
As a preferred embodiment of the anti-inflammatory repair composition according to the present invention, in the active efficacy ingredients, the mass ratio of the gentian root extract, the yeast/zinc fermentation product, the satsuma mandarin peel extract, the aframomum melegueta seed extract and the tetrahydro-methyl pyrimidine carboxylic acid is gentian root extract: yeast/zinc fermentation product: citrus unshiu peel extract: aframomum melegueta seed extract: tetrahydro-methyl-pyrimidine carboxylic acid =1.5:1:0.5:1:0.2.
as a preferred embodiment of the anti-inflammatory rejuvenating composition according to the invention, the composition further comprises a humectant, an antioxidant, a chelating agent, a thickener, a pH adjusting agent, a solubilizer, a preservative and a fragrance.
As a preferred embodiment of the anti-inflammatory rejuvenating composition according to the invention, the moisturizer comprises butylene glycol and 1, 3-propylene glycol; the antioxidant comprises allantoin; the chelating agent comprises disodium EDTA; the thickener comprises acrylic acid (ester)/C10-30 alkanol acrylate cross-linked polymer; the pH regulator comprises aminomethyl propanol; the solubilizer comprises PEG-40 hydrogenated castor oil; the preservative comprises phenoxyethanol and ethylhexyl glycerin; the fragrance includes a perfume.
As a preferred embodiment of the anti-inflammatory repairing composition, the composition comprises 3 to 10 percent of humectant, 0.01 to 0.2 percent of antioxidant, 0.01 to 0.05 percent of chelating agent, 0.1 to 0.45 percent of thickening agent, 0.07 to 0.42 percent of pH regulator, 0.02 to 0.048 percent of solubilizer, 0.15 to 0.8 percent of preservative and 0.005 to 0.012 percent of aromatic agent by mass percent.
As a preferred embodiment of the anti-inflammatory remedial composition of the present invention, the composition further comprises a solvent; the solvent is camellia extract. The camellia extract is used as a solvent, so that the anti-inflammatory repair effect can be obviously enhanced, and the skin can quickly recover the homeostasis.
The invention also provides a preparation method of the anti-inflammatory repairing composition, which comprises the following steps:
(1) Uniformly mixing a humectant, an antioxidant, a chelating agent, a thickener and a solvent for emulsification, stirring and dispersing at 85-88 ℃ under the condition of 20-30r/min, and homogenizing under the condition of 1500r/min for 5-8 min until complete dissolution to obtain a phase A;
(2) Stirring the phase A at 25-30r/min, cooling to 50-55 ℃, adding a pH regulator, and stirring for 15-20 min to a transparent state to obtain a phase B;
(3) Stirring the phase B at 25-30r/min, cooling to 42-45 ℃, adding active functional components, and stirring for 8-10 min to obtain phase C;
(4) And stirring the phase C at the speed of 25-30r/min, cooling to 38-40 ℃, adding an aromatic, a solubilizer and a preservative, and stirring for 15-20 min until the mixture is completely uniform to obtain the anti-inflammatory repairing composition.
The invention also provides application of the anti-inflammatory repairing composition in preparing skin care products with anti-inflammatory and repairing effects.
As a preferred embodiment of the use according to the invention, the skin care product comprises a solution, emulsion, cream, paste, gel, foundation or spray.
Compared with the prior art, the invention has the beneficial effects that: the invention provides an anti-inflammatory repairing composition, which combines gentian extract, yeast/zinc fermentation product, wenzhou mandarin orange peel extract, aframomum melegueta seed extract and tetrahydro-methyl pyrimidine carboxylic acid, and the anti-inflammatory repairing effect is improved through the mutual synergistic effect of the components; the anti-inflammatory repairing composition can remove free radicals of the skin and inhibit the damage effect of the free radicals on skin cells; the anti-inflammatory repairing composition can repair contact dermatitis and allergic dermatitis caused by external stimulation, and enables skin to quickly recover homeostasis.

Detailed Description
To better illustrate the objects, aspects and advantages of the present invention, the present invention will be further described with reference to specific examples.
Examples 1 to 5
An anti-inflammatory rejuvenating composition, the components and their amounts are shown in table 1.

The method of preparation of the anti-inflammatory remedial composition of examples 1-5 includes the steps of:
(1) Uniformly mixing a humectant, an antioxidant, a chelating agent, a thickening agent and a solvent for emulsification, stirring and dispersing at 85-88 ℃ under the condition of 20-30r/min, and homogenizing under the condition of 1500r/min for 5-8 min until complete dissolution to obtain phase A;
(2) Stirring the phase A at the speed of 25-30r/min, cooling to 50-55 ℃, adding a pH regulator, and stirring for 15-20 min to a transparent state to obtain a phase B;
(3) Stirring the phase B at 25-30r/min, cooling to 42-45 ℃, adding active functional ingredients, and stirring for 8-10 min to obtain phase C;
(4) And stirring the phase C at the speed of 25-30r/min, cooling to 38-40 ℃, adding an aromatic, a solubilizer and a preservative, and stirring for 15-20 min until the mixture is completely uniform to obtain the anti-inflammatory repairing composition.
Comparative examples 1 to 5
The components and their contents of the anti-inflammatory repair compositions of comparative examples 1 to 5 are shown in table 2.

The anti-inflammatory remedial compositions of comparative examples 1-5 were prepared in the same manner as in example 2.
Examples of effects
1. Evaluation of safety
The anti-inflammatory repair compositions of examples 1 to 5 and comparative examples 1 to 5 were evaluated for cosmetic irritation.
The tested population is female, 100 people and the age is between 16 and 65 years. The skin of the subject is healthy, has no allergic history of the skin disease, and meets the volunteer selection standard of the subject. Randomly grouped into 10 groups of 10 persons each.
The test method is a human body patch test. Selecting a qualified spot tester, placing 0.020-0.025g of a tested object in the spot tester, sticking the spot tester with the tested object on the back or the bent side of the forearm of the tested object by using a non-irritating adhesive tape, and lightly pressing with a palm to uniformly stick the spot tester on the skin for 24 hours. And (5) removing the tested spot tester for 30min, and observing skin reaction after the indentation disappears. If the result is negative, the test is observed once more at 24h and 48h after the patch test. The results were recorded according to the reference standards of "standards for hygiene of cosmetics" in 2007 and "standards for safety of cosmetics" in 2015.
Evaluation criteria: level 0: negative reaction; stage 1: suspicious reactions, only faint erythema; and 2, stage: weak positive reaction, erythema, infiltration, edema, and pimple; and 3, stage: strong positive reaction, erythema, infiltration, edema, pimple and reaction beyond the tested area; and 4, stage 4: very strong positive reaction, obvious erythema, severe infiltration, edema, herpes fusiformis and reaction beyond the tested area.
The results of the patch test on the skin of a human body show that: the skin reactions of the subjects were observed 24h and 48h after the test by the patch test of the skin-brightening anti-wrinkle compositions of examples 1 to 5 and comparative examples 1 to 5, and the skin reactions of all the subjects were negative, which indicates that the anti-inflammatory repairing composition prepared by the invention is safe to use.
2. Skin redness reduction test
(1) Subject: 60 healthy male and female subjects, age range: 21-70 years old;
(2) Sample receiving: the anti-inflammatory remedial compositions of examples 1-3.
(3) The experimental method comprises the following steps: the inner arm of the test was patched with SDS (1%) and the relevant test was performed 24h after the patch had been applied before the test. The test areas were 3 × 3CM in size, respectively, SDS was smeared only to a size of 1CM squared. During the test period, six groups were applied with the anti-inflammatory repair compositions of examples 1 to 3 in the morning and evening, respectively, and the observation time nodes were before application (day 0) and day 5, respectively. Photographs of the effect of forearm redness at day 0 and day 5 of the subjects were taken as shown in fig. 1-3. As can be seen from FIGS. 1 to 3, the anti-inflammatory and skin-repairing compositions of examples 1 to 3 of the present invention are effective in reducing skin redness.
2. Anti-inflammatory repair test
(1) The test method comprises the following steps: 100 women with reddish faces, pruritus, dryness and erythema at the age of 30-65 years are selected as trial objects and divided into 10 groups of 10 persons, and the anti-inflammatory repairing compositions of examples 1-5 and comparative examples 1-5 are respectively tried in each group. Before test, after cleaning face, the tester is in constant temperature and humidity environment (temperature 21-23 deg.C, humidity 55-65%) for 30min, and then takes facial photograph by using VISA-CR as primary data, the subject applies each sample on face, uses for 1 time every day for 20 days, cleans face on 0, 10, and 20 days, and then takes skin condition test by using VISA-CR, and respectively records as D 0 、D 10 And D 20 . It is noted that each group D 0 、D 10 And D 20 Values are the average of these values in the group, and the facial skin improvement before and after use is calculated by instrument software, with lower scores indicating more severe skin inflammation.
(2) The test results are shown in Table 3.

As can be seen from table 3, the anti-inflammatory repair effect is more remarkable with the increase in the addition amount of the active efficacy ingredient in the anti-inflammatory repair compositions of examples 1 to 5, but when the addition amount of the active efficacy ingredient exceeds 4.2%, the increase in the anti-inflammatory repair effect is not remarkable, that is, the anti-inflammatory repair effect tends to be saturated. The anti-inflammatory repair effect of the composition of example 2 is better than that of comparative examples 1-5, which shows that the active functional ingredients of the invention, namely the gentian root extract, the yeast/zinc fermentation product, the wenzhou mandarin orange peel extract, the aframomum melegueta seed extract and the tetrahydro-methyl pyrimidine carboxylic acid, have good anti-inflammatory repair effect through synergistic effect.
Finally, it should be noted that the above embodiments are only used for illustrating the technical solutions of the present invention and not for limiting the protection scope of the present invention, and although the present invention is described in detail with reference to the preferred embodiments, it should be understood by those skilled in the art that modifications or equivalent substitutions can be made to the technical solutions of the present invention without departing from the spirit and scope of the technical solutions of the present invention.
Claims (10)
1. An anti-inflammatory remedial composition comprising an active efficacy ingredient comprising gentian root extract, yeast/zinc fermentation product, satsuma mandarin peel extract, aframomum melegueta seed extract, and tetrahydromethylpyrimidine carboxylic acid; the active functional component accounts for 0.5-10.5% of the composition by mass.
2. An anti-inflammatory remedial composition according to claim 1 wherein the active efficacy ingredient is a combination of gentian root extract, yeast/zinc fermentation product, citrus unshiu peel extract, aframomum melegueta seed extract and tetrahydro methyl pyrimidine carboxylic acid in a mass ratio of gentian root extract: yeast/zinc fermentation product: citrus unshiu peel extract: aframomum melegueta seed extract: tetrahydro-methyl-pyrimidine carboxylic acid = 0.1-3: 0.15 to 3:0.1 to 2:0.1 to 2:0.05 to 0.5.
3. An anti-inflammatory remedial composition according to claim 2 wherein the active efficacy ingredients are gentian root extract, yeast/zinc fermentation product, satsuma mandarin peel extract, aframomum melegueta seed extract and tetrahydro methyl pyrimidine carboxylic acid in a mass ratio of gentian root extract: yeast/zinc fermentation product: the extract of the citrus unshiu peel: aframomum melegueta seed extract: tetrahydro-methyl-pyrimidine carboxylic acid =1.5:1:0.5:1:0.2.
4. an anti-inflammatory repair composition according to claim 1 further comprising a humectant, an antioxidant, a chelating agent, a thickener, a pH adjusting agent, a solubilizer, a preservative and a fragrance.
5. An anti-inflammatory rejuvenating composition as claimed in claim 4 wherein said humectant comprises butylene glycol and 1, 3-propylene glycol; the antioxidant comprises allantoin; the chelating agent comprises disodium EDTA; the thickening agent comprises acrylic acid (ester)/C10-30 alkanol acrylate cross-linked polymer; the pH regulator comprises aminomethyl propanol; the solubilizer comprises PEG-40 hydrogenated castor oil; the preservative comprises phenoxyethanol and ethylhexyl glycerin; the fragrance includes a perfume.
6. An anti-inflammatory repair composition according to claim 4, wherein said composition comprises, in mass percent, from 3 to 10% of a moisturizing agent, from 0.01 to 0.2% of an antioxidant, from 0.01 to 0.05% of a chelating agent, from 0.1 to 0.45% of a thickening agent, from 0.07 to 0.42% of a pH adjusting agent, from 0.02 to 0.048% of a solubilizing agent, from 0.15 to 0.8% of a preservative and from 0.005 to 0.012% of a fragrance.
7. An anti-inflammatory remedial composition according to claim 1 further comprising a solvent; the solvent is camellia extract.
8. A method of preparing an anti-inflammatory rejuvenating composition as claimed in any one of claims 1 to 7, comprising the steps of:
(1) Uniformly mixing a humectant, an antioxidant, a chelating agent, a thickener and a solvent for emulsification, stirring and dispersing at 85-88 ℃ under the condition of 20-30r/min, and homogenizing under the condition of 1500r/min for 5-8 min until complete dissolution to obtain a phase A;
(2) Stirring the phase A at 25-30r/min, cooling to 50-55 ℃, adding a pH regulator, and stirring for 15-20 min to a transparent state to obtain a phase B;
(3) Stirring the phase B at 25-30r/min, cooling to 42-45 ℃, adding active functional components, and stirring for 8-10 min to obtain phase C;
(4) And stirring the phase C at the speed of 25-30r/min, cooling to 38-40 ℃, adding an aromatic, a solubilizer and a preservative, and stirring for 15-20 min until the mixture is completely uniform to obtain the anti-inflammatory repairing composition.
9. Use of an anti-inflammatory rejuvenating composition according to any one of claims 1 to 7 for the preparation of a skin care product with anti-inflammatory, rejuvenating effect.
10. The use of claim 9, wherein the skin care product comprises a solution, emulsion, cream, paste, gel, foundation or spray.
