Description
技术领域
本发明属于检测技术领域,涉及卵巢功能检测,具体地说是一种试纸条样品垫处理液、其制备方法及其应用。
背景技术
在免疫层析中,抗体作为最常使用的检测试剂,常用来标记有色、荧光乳胶颗粒、胶体金颗粒或磁珠等形成抗体标记复合物。免疫层析中常用结合垫吸收待检测试剂,而待检测试剂在流经结合垫时,抗体标记复合物能够被有效释放,并处于一种稳定状态;同时,它还需要有良好的流动属性以保证样品能均一可靠的转移和结合到硝酸纤维素膜(NC膜)上。
样品垫位于检测装置的末端,主要功能是接收样品,并以一种均匀一致的方式将它转运至结合物释放区域或分析膜,具体的转运位置根据检测装置构造不同而定。除此之外,样品垫还有许多其他的功能:a.过滤样品中的颗粒物质和细胞,保证检测时不会发生样品浸渍过重的情况;b.通过化学物质浸润修饰样品,减小样品样品到达结合物释放和检测区时的差异;c.消除假阳:添加封闭物质以减少反应膜与分析物或检测制剂之间的非特异结合;d.提高检测的灵敏度:添加使样品变粘稠的物质降低爬速,增加反应时间。
评价卵巢储备功能的指标包含抗缪勒氏管激素、抑制素B、卵泡刺激素和促黄体生成素等激素分子,这些激素分子在免疫层析中,荧光免疫分析试纸的样品垫吸收的主要是被PBS稀释的血清溶液,标本在层析的过程中需要将标记物从标记垫中快速洗脱出来,并使之能很好地溶解在标本液中;同时,标本中的蛋白质类物质的亲水性往往并不是很好,容易在纤维介质中的被吸附。
发明内容
本发明的目的,是要提供一种试纸条样品垫处理液,以实现增加样品垫的亲水性、助于快速润湿;同时,保护样品中抗原,提高检测的特异性、灵敏度、降低交叉感染率的目的;
本发明的另一个目的,是要提供上述试纸条样品垫处理液的制备方法,以实现制备方法简便易行目的;
本发明还有一个目的,是要提供上述试纸条样品垫处理液的应用。
为实现上述目的,本发明所采用的技术方案是:
一种试纸条样品垫处理液,制成它的有效成分的原料,以重量份数计包括硼砂3.5~4份,表面活性剂20~35份,聚乙二醇15~25份,海藻糖8~12份,聚乙烯吡咯烷酮30~40份,酪蛋白1~3份和Tris缓冲液980~990份。
作为一种限定,所述表面活性剂,包括两性表面活性剂和非离子型表面活性剂;
所述聚乙二醇的分子量为4000~8000;
所述Tris缓冲液浓度为0.15~0.25mM。
作为进一步限定,所述两性表面活性剂,为S9溶液;
所述非离子型表面活性剂,为S14溶液、S17溶液和S19溶液中的至少一种;
表面活性剂中溶质的浓度为5~10g/L;
所述两性表面活性剂和非表面活性剂的重量比为10:15~18。
作为另一种限定,所述试纸条样品垫处理液的pH值为7.5~9。
本发明还提供了上述试纸条样品垫处理液的制备方法,该方法为将硼砂、表面活性剂、聚乙二醇、海藻糖、聚乙烯吡咯烷酮和酪蛋白混合均匀后,加入Tris缓冲液,即得所述试纸条样品垫处理液。
本发明还提供了上述试纸条样品垫处理液的的应用,它用作检测卵巢功能的荧光试纸条样品垫处理液。
由于采用了上述的技术方案,本发明与现有技术相比所取得的有益效果是:
①本发明提供的试纸条样品垫处理液中,添加了合理配比的两性表面活性剂S17和海藻糖,海藻糖封闭液可有效减少抗体的非特异性吸附,从而达到提高检测体系信噪比的目的,极大提高了试纸条检测的灵敏性,使得评估卵巢储备功能的四项标志物可以在同一试纸条上进行联合检验;
②本发明提供的试纸条样品垫处理液中,酪蛋白具有很好的憎水性,能够很好的在溶胶中使用,不会随溶液流动而溶于水,牢固固定在NC膜上,降低了由于使用环境而造成的背景升高;大分子PVP可以增加样品垫的亲水性,同时其水溶液有悬浮分散作用,可用于保护样品中的微生物抗原;非表面活性剂可提高亲水性和润湿性,降低液体的表面张力,有利于检测样品中的抗原和试纸条中的金标抗体快速充分的反应;本发明提供的试纸条样品垫处理液之所以具有良好湿润性效果,是配方中各个组分之间的配伍所决定的;
③现有技术中处理液为酸性,而样品在酸性条件下变化较大,其在流动过程中捕捉抗体表面产生正电荷,这将引起与带有负电荷的金标粒子发生非特异性反应,如果样品中含有大量的酸性物质或者含有带有大量正电荷的蛋白质,它们在到达捕捉抗体区之前很可能与金标颗粒非特异性结合,从而使得试纸条检测的灵敏性降低,产生不良的交叉反应。本发明提供的试纸条样品垫处理液中添加了合理配比的硼砂,使样品垫处理液为弱碱性,在检测酸性样品时可有效增加试纸条检测的灵敏性,降低产生不良的交叉反应风险;PEG6000具有优良的润滑性、保湿性、分散性,用做基质,起调节粘稠性的作用,有利于检测样品中的抗原和试纸条中的抗体快速反应;
④经本发明提供的试纸条样品垫处理液处理后的试纸条,能够快速检测出样品中与卵巢功能相关的激素的含量,检测成本低,操作简单、快速,检测结果直观;
⑤本发明提供的试纸条样品垫处理液,容易储存,稳定性好,在2~30℃条件下有效期长达18个月;
⑥本发明提供的试纸条样品垫处理液适用范围较广,可以采用人血清、血浆、全血作为样本,用于检测卵巢储备功能的指标包含抗缪勒氏管激素、抑制素B、卵泡刺激素、促黄体生成素等激素的样品垫处理液。
综上所述,本发明提供的样品垫处理液,可以增加样品垫的亲水性,有助于样品垫快速润湿,促进层析作用的进行,大分子类物质形成的水溶液具有悬浮分散作用,可保护样品中抗原,提高检测的特异性、灵敏度,降低交叉感染率,提高了抗体的亲的水性,能够更好的使标记物释放并且使得样本溶液和荧光标记物不分离。
本发明适用于制备试纸条样品垫处理液,制备出的处理液进一步用作检测卵巢功能的荧光试纸条样品垫处理液。

实施例1一种试纸条样品垫处理液的制备方法
本实施例为一种试纸条样品垫处理液的制备方法,本实施例包括以下步骤:
取3.85g硼砂,10g浓度为5g/L两性表面活性剂S9,16.5ml浓度为10g/L的非表面活性剂tritonX-100(S14)溶液,20gPEG6000,10g海藻糖,35g聚乙烯吡咯烷酮(PVP),2g酪蛋白,混合均匀后,加入983.5ml浓度为0.2mM的Tris缓冲液定容至1L,即得试纸条样品垫处理液α1,经检测试纸条样品垫处理液α1的pH值为8.5。
实施例2~6试纸条样品垫处理液的制备方法
实施例2-6分别为一种试纸条样品垫处理液的制备方法,它们的步骤与实施例1基本相同,不同之处仅在于原料用量及工艺参数的不同,具体详见表1:

本实施例为试纸条样品垫处理液的性能试验,主要内容如下:
S1.配置现有技术中的样品垫处理液
现有技术中的样品垫处理液β1:0.05wt%Tris缓冲液、1wt%的tritonX-100、0.5wt%的PEG6000、0.4wt%牛血清蛋白和0.02wt%鼠抗人红细胞;
S2.分别取4.5mL(150μL/cm)样品垫处理液α1和样品垫处理液β1,分别均匀涂抹在20*300mm的样品垫上,置于架子上晾干过夜后,用于检测卵巢功能的免疫层析过程中;
结果显示:样品垫处理液β1处理过的试剂盒判读结果:FSH的检测灵敏度为5mIU/mL,最大检测范围达150mIU/mL;LH的检测灵敏度为5mIU/mL,最大检测范围达150mIU/mL;AMH的检测灵敏度为0.1ng/mL,最大检测范围达10ng/mL;INHB的检测灵敏度为10pg/mL,最大检测范围达2000pg/mL;上述四种检测指标的线性相关系数R2均大于0.99,表明检测结果可靠;
样品垫处理液α1处理过的试剂盒判读结果:FSH的检测灵敏度为1mIU/mL,最大检测范围达200mIU/mL;LH的检测灵敏度为2mIU/mL,最大检测范围达200mIU/mL;AMH的检测灵敏度为0.05ng/mL,最大检测范围达25ng/mL;INHB的检测灵敏度为8pg/mL,最大检测范围达2000pg/mL;上述四种检测指标的线性相关系数R2均大于0.99,表明检测结果更可靠,本发明能更好的提高检测灵敏度和准确度。
可知,相较于现有技术中的处理液,本发明制备的样品垫处理液检测灵敏度较高,检测范围较大。
Claims (2)
1.一种荧光试纸条样品垫处理液,其特征在于:制成它的有效成分的原料,以重量份数计包括硼砂3.5~4份,表面活性剂20~35份,聚乙二醇15~25份,海藻糖8~12份,聚乙烯吡咯烷酮30~40份,酪蛋白1~3份和 Tris缓冲液980~990份;
所述表面活性剂,包括两性表面活性剂和非离子型表面活性剂;
所述聚乙二醇的分子量为4000~8000;
所述Tris缓冲液浓度为0.15~0.25mM;
所述两性表面活性剂,为S9溶液;
所述非离子型表面活性剂,为S14溶液、S17溶液和S19溶液中的至少一种;
表面活性剂中溶质的浓度为5~10g/L;
所述两性表面活性剂和非离子型表面活性剂的重量比为10:15~10:18;
所述荧光试纸条样品垫处理液用作检测卵巢功能的荧光试纸条样品垫处理液;
所述卵巢功能的指标为抗缪勒氏管激素、抑制素B、卵泡刺激素和促黄体生成素;
所述荧光试纸条样品垫处理液的pH值为7.5~9。
2.根据权利要求1所述的荧光试纸条样品垫处理液的制备方法,其特征在于:该方法为将硼砂、表面活性剂、聚乙二醇、海藻糖、聚乙烯吡咯烷酮和酪蛋白混合均匀后,加入Tris缓冲液,即得所述荧光试纸条样品垫处理液。
