本发明公开了一种芦荟软胶囊及其制备方法和应用,芦荟软胶囊由芦荟提取物和椰枣籽提取物制备获得,重量比为1:0.6到1:1;通过乙醇萃取芦荟花获得第一芦荟提取物并浓缩;通过乙醇萃取芦荟叶获得第二芦荟提取物并浓缩;通过甲醇萃取椰枣籽得到第一椰枣籽提取物;通过己烷萃取椰枣籽得到第二椰枣籽提取物,混合后蒸发浓缩获得椰枣籽浓缩液;利用第一、第二芦荟提取物和椰枣籽浓缩液制备获得芦荟软胶囊。本发明,通过芦荟提取物和椰枣籽提取物的配伍,利用芦荟抗炎、抗病毒活性的作用,配合椰枣籽抗病毒、抗氧化、抗菌的作用,对治疗消化性溃疡有一定的良好疗效,并且价格低、使用安全。

本发明涉及抗溃疡药物技术领域,具体涉及一种芦荟软胶囊及其制备方法和应用。
背景技术
目前,芦荟产品主要有芦荟保健品和芦荟化妆品,包括芦荟洗浴、芦荟洗涤产品,如牙膏、香皂、洗发护发、沐浴液、保湿霜、清洁剂、洗手液、芦荟凝胶汁、芦荟浓缩液。其中,芦荟保健品通常制备成芦荟软胶囊,在提供美白皮肤,排除体内毒素以及保护肠胃的功能的同时,使用更加方便。
例如,中国发明专利CN 105521178 B公开了一种芦荟软胶囊及其制备方法和应用,所述芦荟软胶囊的填充物由以下组分按照质量比制成:植物油的质量为芦荟发酵液质量的1.1~1.2倍,助悬剂的质量为芦荟发酵液质量的0.1~0.5倍;所述芦荟发酵液由以下组分按照质量比制成:红缘拟层孔菌发酵液的质量为芦荟全叶冻干粉质量的0.5~1倍、金耳菌株发酵液的质量为芦荟全叶冻干粉质量的1~2.5倍。药理实验证明,所述芦荟软胶囊能减轻血栓性湿重,能明显延长小鼠出血与凝血时间,具有改善微循环、抗凝血作用,具有抗血栓的作用,有助于抑制血栓形成的作用。
然而,现有的芦荟软胶囊,主要用于美白皮肤,排除体内毒素。
有鉴于此,需要对现有的芦荟软胶囊进行改进,在美白排毒的同时,提高抗溃疡功效。
发明内容
针对上述缺陷,本发明所要解决的技术问题在于提供一种芦荟软胶囊及其制备方法和应用,以解决现有的芦荟软胶囊抗溃疡功效不强的问题。
为此,本发明提供的一种芦荟软胶囊,由由芦荟提取物和椰枣籽提取物制备获得,其中,芦荟提取物与椰枣籽提取物的重量比为1:0.6到1:1。
在上述芦荟软胶囊中,所述芦荟提取物包括芦荟花提取的第一芦荟提取物和芦荟叶提取的第二芦荟提取物,第一芦荟提取物和第二芦荟提取物所比例为1:1。
本发明还提供了一种芦荟软胶囊的制备方法,包括以下步骤:
通过乙醇萃取粉末状的芦荟花,获得第一芦荟提取物并浓缩;
通过乙醇萃取粉末状的芦荟叶,获得第二芦荟提取物并浓缩;
通过甲醇萃取椰枣籽粉末得到第一椰枣籽提取物;
通过己烷萃取椰枣籽粉末得到第二椰枣籽提取物;
混合第一椰枣籽提取物和第二椰枣籽提取物,并蒸发浓缩获得椰枣籽浓缩液;
利用第一、第二芦荟提取物和椰枣籽浓缩液制备获得芦荟软胶囊。
在上述方法中,使用旋转蒸发器浓缩所述第一、第二芦荟提取物,真空度为95~98kPa。
在上述方法中,将椰枣籽从椰枣果实中分离出来,然后浸泡在热水中,并洗涤除去粘附的枣果肉,获得干净的椰枣籽。
在上述方法中,将芦荟鲜花和芦荟绿叶分别在阴凉处干燥后粉化,并储存在密封容器中。

具体实施方式
下面将结合本发明实施例附图,对本发明实施例的技术方案进行清楚、完整地描述,显然,以下所描述的实施例,仅仅是本发明的一部分实施例,而不是全部的实施例。基于本发明的实施例,本领域普通技术人员在没有做出创造性劳动的前提下,所获得的所有其他实施例,都属于本发明保护的范围。
为了对本发明的技术方案和实现方式做出更清楚地解释和说明,以下介绍实现本发明技术方案的几个优选的具体实施例。
需要说明的是,本文中“内、外”、“前、后”及“左、右”等方位词是以产品使用状态为基准对象进行的表述,显然,相应方位词的使用对本方案的保护范围并非构成限制。
本发明提供的一种芦荟软胶囊,由芦荟提取物和椰枣籽提取物制备获得,其中,芦荟提取物与椰枣籽提取物的重量比为1:0.6到1:1。
其中,具体配比可以为:芦荟提取物与椰枣籽提取物1:0.6,1:0.7,1:0.8,1:0.9和1:1。
芦荟富含许多活性药理化合物,如矿物质,凝集素,维生素,糖蛋白,生物碱,氨基酸,蒽醌苷,具有抗菌,镇痛,抗炎,抗肿瘤,抗病毒活性等功效。
枣椰籽是各种重要化合物的良好来源,如饱和和不饱和脂肪酸、酚类化合物、生育酚和甾醇,具有抗病毒、抗氧化、抗癌、抗菌和降血糖等特性。采用上述芦荟和枣椰籽提取物制备的芦荟软胶囊,不仅具有美白皮肤,排除体内毒素等保健功效,还具有一定的治疗消化性溃疡的疗效。
对于上述芦荟软胶囊,本发明提供了如下的一种制备方法,包括以下步骤:
第一步:制备芦荟提取物。
本发明中的芦荟提取物,不仅使用芦荟花,还会使用芦荟叶。具体地,
将芦荟鲜花和芦荟绿叶分别在阴凉处干燥后粉化,并储存在密封容器中。
取500克粉末状的芦荟花用30升乙醇萃取6-10小时,然后利用旋转蒸发器将萃取液浓缩成红棕色半固体状,获得第一芦荟提取物。其中,浓缩时旋转蒸发器的真空度为95~98kPa。
取500克粉末状的芦荟叶用30升乙醇萃取6-10小时,然后利用旋转蒸发器将萃取液浓缩成深绿色胶质团,获得第二芦荟提取物。其中,浓缩时旋转蒸发器的真空度为95~98kPa。
萃取芦荟花粉末和芦荟叶粉末时,采用冷浸渍法在室温下进行。
第二步:制备椰枣籽提取物。
将椰枣籽从椰枣果实中分离出来,并在50-60℃的热水中浸泡2小时,然后洗涤除去椰枣籽上粘附的椰枣果肉,将干净的椰枣籽在60℃下烘烤干燥一天,然后研磨成椰枣籽粉末。
取500克椰枣籽粉末用5升甲醇萃取,得到红色的第一椰枣籽提取物。
取300克椰枣籽粉末用3升己烷萃取,得到黄色的第二椰枣籽提取物。
将第一椰枣籽提取物和第二椰枣籽提取物混合,并使用旋转蒸发器蒸发浓缩获得椰枣籽浓缩液。
第一步和第二步中,萃取和使用旋转蒸发器浓缩的工艺均为本领域的常规方法,在此不再赘述。
利用上述的第一、第二芦荟提取物和椰枣籽浓缩液制备获得芦荟软胶囊。其中。芦荟提取物与椰枣籽提取物的重量比为1:0.6到1:1,第一芦荟提取物和第二芦荟提取物所比例为1:1。
芦荟软胶囊的制备可采用下述方法实现。
步骤110,取4.275g氧化铁红和17.1g的纯化水置于不锈钢桶中搅拌30分钟,使适当分散,然后过胶体磨研磨均匀,得到氧化铁红料。
步骤120,取357.9纯化水和150g甘油置化胶罐中,加热至65~75℃后,再加入375g明胶和步骤110制备的氧化铁红料,保温75℃,使明胶溶胶,搅拌约60分钟,直至明胶全部溶解。
步骤130,将化胶罐抽真空,使真空度达到-0.08MPa,至胶液无气泡。
步骤140,胶液经80目滤袋过滤储存于保温桶中,在50~60℃保温备用。
步骤150,将300g大豆油和25g蜂蜡加入配料罐中,加热至65-75℃,搅拌30分钟,使蜂蜡完全熔融,降温至40℃±5℃,得油蜡液。
步骤160,加入芦荟提取物和椰枣籽提取物,搅拌60分钟,置胶体磨中研磨,得均质料液,过80目滤网,置于料液桶中静置4小时左右备用。
步骤170,将步骤140配制好的胶液和步骤160研制好的均质料液投入软胶囊压丸机压制成丸,软胶囊压丸机转速为2.5~3.0转/分钟,喷体温度为38~48℃,胶皮厚度为0.7mm~0.8mm,调节装量为0.5g/粒,压丸期间保持室温18~26℃,相对湿度45~65%。
步骤180,通过转笼预干机定型,转笼中放上洁净的擦丸布,擦除软胶囊表面的残余石蜡油,第一节和最后一节转笼不放擦丸布;定型完毕的软胶囊送入干燥间干燥,定型时每一节转笼的定型时间不低于15分钟,室温18~26℃,相对湿度45~65%。
步骤190,将定型后的软胶囊均匀撒布于洁净的晾丸盘中,每盘厚度以铺平晾丸盘为宜,在干燥间中干燥,干燥温度为21℃~25℃,相对湿度≤45%,干燥40小时左右,使软胶囊胶皮水分含量为9%-13%。
当软胶囊漏油时,需要将无法擦干净的部分软胶囊经洗丸机,用95%的食用乙醇洗涤,直至软胶囊表面干净,在洗丸过程中不得破坏软胶囊的形状。
本发明提供的芦荟软胶囊,可应用于制备抗溃疡药物和保健品。
大鼠胃溃疡的抗溃疡活性试验如下:
1、材料和方法
将45只体重150-200克(8-10周龄)的大鼠随机分为三组(每组15只)。
第1组:对照组,不使用任何药物。
第2组:雷尼替丁组,以剂量水平50mg/kg口服,每日早晚各一次,连续服用5天。
第3组:芦荟软胶囊组,以剂量水平250mg/kg和500mg/kg口服,,每日早晚各一次,连续服用5天。
大鼠溃疡采用幽门结扎术诱导。术前在单独的笼子中禁食24小时,在麻醉醚的帮助下进行麻醉,腹部切口打开,胃的幽门部分被稍微抬出并结扎,5天后打开腹部,解剖胃,并将内容物排入玻璃管中。测量胃液的体积并以2000rpm离心10分钟,从上清液中,取等分试样(每种1ml)测定pH值以及总酸度和游离酸度。
2、pH测量结果
使用1ml蒸馏水稀释1ml胃液等分试样,然后使用pH计测量pH。
3、总酸度和游离酸度的测定
使用1ml蒸馏水稀释1ml胃液等分试样,然后转移到50ml锥形烧瓶中,采用滴定法测量总酸度和游离酸度。
4、胃溃疡的宏观评价
打开胃并用盐水冲洗以消除任何胃内容物或血块后,使用10倍放大镜评估溃疡结果如下:
综合以上具体实施例的描述,本发明提供的芦荟软胶囊及其制备方法和应用,对治疗消化性溃疡具有一定的疗效,丰富了芦荟软胶囊的功能。

Claims (8)
1.一种芦荟软胶囊,其特征在于,由芦荟提取物和椰枣籽提取物制备获得,其中,芦荟提取物与椰枣籽提取物的重量比为1:0.6到1:1。
2.根据权利要求1所述的芦荟软胶囊,其特征在于,所述芦荟提取物包括芦荟花提取的第一芦荟提取物和芦荟叶提取的第二芦荟提取物,第一芦荟提取物和第二芦荟提取物所比例为1:1。
3.一种芦荟软胶囊的制备方法,其特征在于,包括以下步骤:
通过乙醇萃取粉末状的芦荟花,获得第一芦荟提取物并浓缩;
通过乙醇萃取粉末状的芦荟叶,获得第二芦荟提取物并浓缩;
通过甲醇萃取椰枣籽粉末得到第一椰枣籽提取物;
通过己烷萃取椰枣籽粉末得到第二椰枣籽提取物;
混合第一椰枣籽提取物和第二椰枣籽提取物,并蒸发浓缩获得椰枣籽浓缩液;
利用第一、第二芦荟提取物和椰枣籽浓缩液制备获得芦荟软胶囊。
4.根据权利要求3所述的方法,其特征在于,使用旋转蒸发器浓缩所述第一、第二芦荟提取物,真空度为95~98kPa。
5.根据权利要求3所述的方法,其特征在于,将椰枣籽从椰枣果实中分离出来,然后浸泡在热水中,并洗涤除去粘附的枣果肉,获得干净的椰枣籽。
6.根据权利要求3所述的方法,其特征在于,将芦荟鲜花和芦荟绿叶分别在阴凉处干燥后粉化,并储存在密封容器中。
7.根据权利要求3所述的方法,其特征在于,采用冷浸渍法在室温下用乙醇萃取芦荟花和芦荟叶粉末。
8.如权利要求1所述的芦荟软胶囊在制备抗溃疡药物或保健品中的应用。
