Abstract
本发明提供了一种具有美白作用的组合物及其制备方法和应用,属于食品保健品技术领域。本发明公开的组合物中添加了酵母抽提物、SOD酵母粉、N‑乙酰神经氨酸、橄榄粉、米糠油粉、烟酰胺,通过控制酵母抽提物、SOD酵母粉和橄榄粉的质量比为1:1‑1.5:1‑3明显提高了组合物的美白效果;还在组合物中添加了抗性糊精、低聚木糖和壳寡糖作为膳食纤维和益生元成分,使得到的组合物在改善肠道健康的基础上具有更好的稳定性。

技术领域
本发明属于食品保健品技术领域,具体涉及一种具有美白作用的组合物及其制备方法和应用。
背景技术
随着人们生活质量的提高以及女性对于美白的追求,在化妆品领域,美白产品层出不穷,几乎是每个成年女性的必备品,但是随着人们认知水平不断提高,以及内调外养的健康理念的普及,将最天然来源的物质用于美白产品已然成为一种趋势,口服美容产品已经成为新一代炙手可热的美白单品,如何研究出美白效果显著、安全可靠且稳定性好的可食用美白产品,已经成为口服美容领域的热点和重点。
经研究发现,影响皮肤白皙程度的主要原因分为内在因素和外在因素,内在因素包括遗传因素、年龄因素、内分泌因素;外在因素包括环境因素、炎症反应、生活习惯等。其中最直接的因素便是环境因素,当人体受到紫外线照射等外界刺激时,黑色素细胞中的麦拉宁色素激活了酪氨酸酶活性,刺激酪氨酸转换成为多巴,从而在基底层中形成更多的黑色素,黑色素转移至表皮层形成色素沉着。目前,市场上大多数口服美白产品成分单一,作用机理不明确,效果不佳。
如中国专利申请202010515673.6中公开了一种美白祛斑的果味片及其制备方法,其中包含蓝莓果粉5-13份、酵母抽提物5-16份、葡萄粉6-16份、桑葚粉6-10 份、针叶樱桃果粉8-16份、圣女果粉10-16份、维生素C 4-8份等。该发明将含有抗氧化成分的蓝莓果粉、葡萄粉、桑葚粉、针叶樱桃果粉和圣女果粉搭配使用,使其果味片中富含维生素C和维生素E和其他维生素等,从而具有美白、延缓人的衰老、减少皱纹祛斑、排毒,保护肝脏、抗过敏等作用。但是该申请仅从理论上推断其有美白效果,但是美白效果并没有验证,并且该产品中添加了维生素C,本领域技术人员公知维生素C常态下不稳定,容易氧化,从而使得到的产品稳定性低。
再如中国专利申请202110514011.1中公开了一种刺梨保湿美白口服液的制备方法。该方法,包括以下步骤:(1)中药活性成分提取、(2)富硒发酵物制取、 (3)制备浓缩刺梨汁、(4)制备SOD酵母抽提物、(5)调配。该发明出刺梨等原料中的多糖、蛋白质等水溶性成分,使得制得的中药活性成分有耐高温组分和常温活性成分,在提高刺梨汁的营养成分的同时有效增强刺梨汁中成分的稳定性,但是该申请得到的口服液的美白效果有待提高。
因此,亟需开发一种美白效果显著、安全可靠且稳定性好的美白作用组合物及其制备方法和应用。
发明内容
基于现有技术中存在的问题,本发明旨在提供一种美白效果显著、安全可靠且稳定性好的美白作用组合物及其制备方法和应用,本发明通过控制饮料中的具体组分以及各组分之间的配比,明显提高了组合物的美白效果和稳定性。
为了实现上述目的,本发明采用以下技术方案:
一种具有美白作用的组合物,按重量份数计包括以下组分:酵母抽提物2-9 份、SOD酵母粉2-9份、N-乙酰神经氨酸1-5份、橄榄粉4-10份、米糠油粉0.5-2 份、烟酰胺0.01-0.05份;
优选地,所述的组合物按重量份数计包括以下组分:酵母抽提物3-7份、SOD 酵母粉4-7份、N-乙酰神经氨酸2-4份、橄榄粉6-9份、米糠油粉1-2份、烟酰胺0.02-0.04份;
在一些优选实施方案中,所述的酵母抽提物、SOD酵母粉和橄榄粉三者比例为1:1-1.5:1-3;
再优选地,所述的酵母抽提物、SOD酵母粉和橄榄粉三者比例为3:3:4。
作为一个优选地技术方案,所述的组合物按重量份数计包括以下组分:酵母抽提物6份、SOD酵母粉6份、N-乙酰神经氨酸3份、橄榄粉8份、米糠油粉 1.2份、烟酰胺0.02份。

作为一个优选的技术方案,所述的组合物按重量份数计包括如下组分:酵母抽提物6份、SOD酵母粉6份、N-乙酰神经氨酸3份、橄榄粉8份、米糠油粉 1.2份、烟酰胺0.02份、抗性糊精3份、低聚木糖2份和壳寡糖1.5份。
本发明在实施过程中,在组合物中添加了酵母抽提物、SOD酵母粉、N-乙酰神经氨酸、橄榄粉、米糠油粉和烟酰胺,通过控制酵母抽提物、SOD酵母粉和橄榄粉三者比例在1:1-1.5:1-3,使得到的组合物具有更好的美白效果。
其中,酵母抽提物由酿酒酵母经发酵、酶解、分离、浓缩、干燥等工艺制备得到。其中含有的谷胱甘肽(GSH)是一种含γ-酰胺键和巯基的三肽,谷胱甘肽通过电子转移途径与自由基反应,自由基与电子结合失去活性,降低体内活性氧自由基(ROS)含量,减少氧化应激反应。
橄榄粉中含有多种生物活性成分,包括甾醇类、萜烯酸与二元醇类、生育酚、酚类化合物等,其中酚类化合物中的羟基酪醇是最强效的天然抗氧化剂,羟基酪醇通过激活二相解毒酶系统触发细胞抗氧化机制,间接保护细胞结构和功能,促进二相酶系统和抗氧化酶系的整体提升,清除体内活性氧自由基(ROS)。
含SOD酵母粉中的超氧化物歧化酶(SOD),是一种源于生命体的活性物质,它可对抗与阻断因氧自由基对细胞造成的损害,并及时修复受损细胞,复原因自由基造成的对细胞伤害,达到延缓衰老,保持健康的目的。SOD可以有效抑制酪氨酸酶的活性达到50%以上,减少黑色素生成,可以抵御黑色素沉积,美白肌肤。
米糠油粉中含有大米神经酰胺,不仅能够有助于保持肌肤水分,减少皱纹产生,同时其具有抑制酪氨酸酶的活性,减少黑色素生成,使肌肤焕亮透白的功能。
烟酰胺是一种水溶性维生素,其参与能量代谢、DNA修复以及氧化应激等生命活动,可以促进角质细胞代谢,抑制黑色素向表皮层角质细胞的转移,从而起到美白肌肤的作用。
另外,本申请还在组合物中添加了膳食纤维和益生元成分,即在组合物中添加了抗性糊精、低聚木糖和壳寡糖,三者的加入可以改善肠道健康,促进体内垃圾毒素排出,从而改善皮肤色素沉积,本发明在实施过程中意外地发现控制抗性糊精、低聚木糖和壳寡糖三者的加入量以及控制其质量比为2:1-1.5:1,不仅可以改善肠道健康,而且使得到的组合物具有更高的稳定性,长期放置组合物中的活性成分几乎无降低。
本发明还提供了上述组合物的制备方法,即将配方用量的上述组分混合后灭菌,灌装,二次灭菌,即得。
本发明还提供了上述组合物在制备食品或保健品中的应用;所述的食品或保健品可以但不限于功能饮料。
所述的功能饮料的制备方法为:将配方用量的上述组分按顺序逐一溶于一定量的水中,溶解完全,并混合均匀后,灭菌,灌装,二次灭菌,即得。
与现有技术相比,本发明的有益效果在于:
(1)本发明公开的组合物中添加了酵母抽提物、SOD酵母粉、N-乙酰神经氨酸、橄榄粉、米糠油粉、烟酰胺,在实施过程中通过控制酵母抽提物、SOD 酵母粉和橄榄粉的质量比为1:1-1.5:1-3明显提高了组合物的美白效果;
(2)本发明还在组合物中添加了抗性糊精、低聚木糖和壳寡糖作为膳食纤维和益生元成分,能够改善肠道健康,促进体内垃圾毒素排出,从而改善皮肤色素沉积;本申请采用抗性糊精作为低分子水溶性膳食纤维,膳食纤维吸收水分后膨胀,增加粪便的体积,增强对肠壁刺激,促进肠蠕动,有利于排便。当肠道中的细菌在膳食纤维中繁殖时,能产生胶状闭块,具有缓泻作用,从而保证粪便在肠道中的滑润性,有利于顺利通过肠道;低聚木糖、壳寡糖作为益生元成分可以在体内促进肠道有益菌的生长和繁殖,形成微生态竞争优势,优化肠道微生态平衡,人体摄入功能性低聚糖导致双歧杆菌的量增多,肠内双歧杆菌发酵低聚糖产生大量醋酸和乳酸等短链脂肪酸,能促进肠道的蠕动、增加粪便湿润度,并保持一定的渗透压,故可以改善和防止便秘;两者从不同方向不同维度产生促排作用,在促进肠道健康方面均具有良好作用。在实施过程意外地发现通过控制抗性糊精、低聚木糖和壳寡糖的质量比为2:1-1.5:1时,得到的组合物在改善肠道健康的基础上具有更好的稳定性。

制备方法:将配方用量的上述组分按顺序逐一溶于50mL的水中,溶解完全,并混合均匀后,灭菌,灌装,二次灭菌,即得。

实施例3一种具有美白作用的功能饮料及其制备方法
按重量份数计包括以下组分:酵母抽提物3份、SOD酵母粉4份、N-乙酰神经氨酸2份、橄榄粉9份、米糠油粉2份、烟酰胺0.02份、抗性糊精4份、低聚木糖2份和壳寡糖3份。
制备方法:将配方用量的上述组分按顺序逐一溶于100mL的水中,溶解完全,并混合均匀后,灭菌,灌装,二次灭菌,即得。
实施例4一种具有美白作用的功能饮料及其制备方法
按重量份数计包括以下组分:酵母抽提物6份、SOD酵母粉6份、N-乙酰神经氨酸3份、橄榄粉8份、米糠油粉1.2份、烟酰胺0.02份、抗性糊精3份、低聚木糖2份和壳寡糖1.5份。
制备方法:将配方用量的上述组分按顺序逐一溶于120mL的水中,溶解完全,并混合均匀后,灭菌,灌装,二次灭菌,即得。
对比例1
与实施例4的区别在于:酵母抽提物、SOD酵母粉和橄榄粉三者比例为1:3:1;即酵母抽提物5份、SOD酵母粉15份、橄榄粉5份,其他组分含量以及制备方法与实施例4相同。
对比例2
与实施例4的区别在于:酵母抽提物、SOD酵母粉和橄榄粉三者比例为 1:2:0.2;即酵母抽提物6.25份、SOD酵母粉12.5份、橄榄粉1.25份,其他组分含量以及制备方法与实施例4相同。
对比例3
与实施例4的区别在于:将SOD酵母粉替换为橄榄粉,即酵母抽提物6份、橄榄粉14份,其他组分含量以及制备方法与实施例4相同。
对比例4
与实施例4的区别在于:将橄榄粉替换为酵母抽提物,即酵母抽提物14份、SOD酵母粉6份,其他组分含量以及制备方法与实施例4相同。
对比例5
与实施例4的区别在于:抗性糊精、低聚木糖和壳寡糖的质量比为2:0.3:0.2,即抗性糊精5.2份、低聚木糖0.78份和壳寡糖0.52份,其他组分含量以及制备方法与实施例4相同。
对比例6
与实施例4的区别在于:抗性糊精、低聚木糖和壳寡糖的质量比为0.2:4:2.3,即抗性糊精0.2份、低聚木糖4份和壳寡糖2.3份,其他组分含量以及制备方法与实施例4相同。
对比例7
与实施例4的区别在于:将抗性糊精替换为α-环状糊精,其他组分含量以及制备方法与实施例4相同。
对比例8
现有技术CN113367255A中实施例1制备的刺梨保湿美白口服液。
效果实验
1、美白效果实验-抑制酪氨酸酶活性实验
实验方法:
1.1试剂:酪氨酸酶、酪氨酸、磷酸二氢钠、磷酸氢二钠、阳性对照(熊果苷粉末)、样品(功能饮料)、去离子水。
1.2试剂配制:
1.2.1配制磷酸盐缓冲溶液(pH=6.8):
先分别配制0.2M的磷酸二氢钠和0.2M的磷酸氢二钠,取51mL磷酸二氢钠+49mL磷酸氢二钠即得0.2M、pH=6.8的磷酸缓冲液。
0.2M磷酸二氢钠的配制方法:称取71.6gNa2HPO4·12H2O,溶于1000mL去离子水,即得;0.2M磷酸氢二钠的配制方法:称取31.2gNaH2PO4·2H2O,溶于 1000mL去离子水,即得。
1.2.2配制L-酪氨酸溶液:称取L-酪氨酸25.6g,用磷酸缓冲液定容于50mL 容量瓶中,即得L-酪氨酸溶液。
1.2.3配制酪氨酸酶溶液:将马铃薯洗净,于4℃预冷4h左右。去皮,切成约1.0cm3丁状,于-20℃冷冻过夜;称重,按1:1(W:V)的比例加入4℃预冷的磷酸钠缓冲液,用组织捣碎机制成匀浆,3层纱布过滤,滤液于4000r/min离心 10min,上清液即为所得的酪氨酸酶粗酶液,4℃保存,2h内用完。
1.2.4配制受试液:将各个实施例和对比例制备的功能饮料用甲醇稀释到 1mg/ml,即得。
1.2.5配制阳性对照:取熊果苷粉末0.01g,溶于10ml的甲醇溶液,即得1mg/ml 的对照品溶液。
1.3实验方法:
按照下表1所示向试管中依次加入磷酸盐缓冲溶液、受试液、酪氨酸溶液,于35℃水浴10分钟。然后加入酪氨酸酶液,混匀,再在35℃下孵育30min,迅速转移至比色皿中,在475nm处测定吸光值。

受试组用空白对照组1调零,阴性对照组用空白对照组2调零,阳性对照组用空白对照组3调零。
抑制率=1-[(A1-A2)/(A3-A2)]*100%=(A3-A1)/(A3-A2)*100%
其中,受试组吸光值为A1,阴性对照组吸光值为A2,阳性对照组吸光值为 A3。
抑制酪氨酸酶活性实验结果,见下表2所示。

根据上表2的检测数据可以看出,本申请实施例1-4制备的功能饮料对酪氨酸酶活性具有较高的抑制性,尤其是实施例4控制酵母抽提物、SOD酵母粉和橄榄粉三者比例为3:3:4,其得到的功能饮料对酪氨酸酶活性的抑制率最高,可以达到 15.87%,实施例1-3中改变各组分的质量比以及配比,得到的功能饮料对酪氨酸酶活性抑制率均在12.4%以上,对比例1和对比例2改变酵母抽提物、SOD酵母粉和橄榄粉三者比例不在本申请保护范围内,得到的功能饮料对酪氨酸酶活性抑制率较实施例4降低;对比例3和对比例4省略组分SOD酵母粉或橄榄粉,得到的功能饮料对酪氨酸酶活性的抑制率也有所降低,说明合适配比的酵母抽提物、SOD 酵母粉和橄榄粉对酪氨酸酶活性抑制率有明显影响。
2、稳定性试验
按照本发明方法实施例1-4和对比例5-8分别制做美白饮料产品,按照 GB/T5009.171-2003的检测方法检测SOD活性,结果如下表3和表4:

根据上表3和表4的记载可以看出,本申请实施例1-4制备的功能饮料的 SOD活性稳定性较高,实施例1-4制备的功能饮料30℃的室内放置4周后SOD 活性下降率分别为4.82%、4.48%、4.31%和3.25%,说明本发明实施例制备的功能饮料具有较好的稳定性,尤其是实施例4通过控制抗性糊精、低聚木糖和壳寡糖的质量比为3:2:1.5,得到的功能饮料在改善肠道健康的基础上具有更好的稳定性;对比例5-7改变抗性糊精、低聚木糖和壳寡糖的质量比,或将抗性糊精替换为α-环状糊精,得到的功能饮料30℃的室内放置4周后SOD活性下降率分别为 20.5%、33.7%、34.9%,说明功能饮料中抗性糊精、低聚木糖和壳寡糖的质量比以及组分的种类均会影响功能饮料的稳定性;对比例8为现有技术中公开的口服液,其稳定性8.18%,也低于本申请实施例1-4的稳定性。
对于本领域技术人员而言,显然本发明不限于上述示范性实施例的细节,而且在不背离本发明的精神或基本特征的情况下,能够以其他的具体形式实现本发明。
因此,无论从哪一点来看,均应将实施例看作是示范性的,而且是非限制性的,本发明的范围由所附权利要求而不是上述说明限定,因此旨在将落在权利要求的等同要件的含义和范围内的所有变化囊括在本发明内。
此外,应当理解,虽然本说明书按照实施方式加以描述,但并非每个实施方式仅包含一个独立的技术方案,说明书的这种叙述方式仅仅是为清楚起见,本领域技术人员应当将说明书作为一个整体,各实施例中的技术方案也可以经适当组合,形成本领域技术人员可以理解的其他实施方式。
Claims (12)
1.一种具有美白作用的组合物,其特征在于:包括以下组分:酵母抽提物、SOD酵母粉、N-乙酰神经氨酸、橄榄粉、米糠油粉和烟酰胺。
2.根据权利要求1所述的组合物,其特征在于:按重量份数计包括以下组分:酵母抽提物2-9份、SOD酵母粉2-9份、N-乙酰神经氨酸1-5份、橄榄粉4-10份、米糠油粉0.5-2份、烟酰胺0.01-0.05份。
3.根据权利要求2所述的组合物,其特征在于:按重量份数计包括以下组分:酵母抽提物3-7份、SOD酵母粉4-7份、N-乙酰神经氨酸2-4份、橄榄粉6-9份、米糠油粉1-2份、烟酰胺0.02-0.04份。
4.根据权利要求2所述的组合物,其特征在于:所述的酵母抽提物、SOD酵母粉和橄榄粉三者比例为1:1-1.5:1-3。
5.根据权利要求4所述的组合物,其特征在于:所述的酵母抽提物、SOD酵母粉和橄榄粉三者比例为3:3:4。
6.根据权利要求2所述的组合物,其特征在于:所述的组合物按重量份数计包括以下组分:酵母抽提物6份、SOD酵母粉6份、N-乙酰神经氨酸3份、橄榄粉8份、米糠油粉1.2份、烟酰胺0.02份。
7.根据权利要求2所述的组合物,其特征在于:所述的组合物按重量份数计还包括:抗性糊精1-5份、低聚木糖1-5份和壳寡糖1-4份。
8.根据权利要求7所述的组合物,其特征在于:所述组合物,按重量份数计还包括如下组分:抗性糊精2-4份、低聚木糖2-4份和壳寡糖1-3份。
9.根据权利要求7所述的组合物,其特征在于:所述的抗性糊精、低聚木糖和壳寡糖的质量比为2:1-1.5:1。
10.根据权利要求9所述的组合物,其特征在于:所述的抗性糊精、低聚木糖和壳寡糖的质量比为3:2:1.5。
11.根据权利要求10所述的组合物,其特征在于:所述的组合物按重量份数计包括如下组分:酵母抽提物6份、SOD酵母粉6份、N-乙酰神经氨酸3份、橄榄粉8份、米糠油粉1.2份、烟酰胺0.02份、抗性糊精3份、低聚木糖2份和壳寡糖1.5份。
12.权利要求1-11任一项所述的组合物在制备食品或保健品中的应用;所述的食品或保健品可以但不限于功能饮料。
