Description

Anti-dandruff composition, shampoo and preparation method thereof
Technical Field
The invention relates to the technical field of household bath products, and in particular relates to an anti-dandruff composition, shampoo and a preparation method thereof.
Background
Shampoo is one of common household bath products, and has the main functions of cleaning scalp and hair and keeping the scalp and the hair clean and sanitary. With the improvement of living standard, people have higher and higher requirements on the quality and the function of the shampoo, and the requirements are not satisfied with the cleaning efficacy of the shampoo alone.
The problem of dandruff has long plagued many people. Dandruff is the exfoliative corneocytes of the scalp. Normally, keratinocytes shed very little, slowly and are not seen significantly. However, under certain stimulation, a large amount of immature keratinocytes are produced, agglomerated and shed, and a phenomenon of 'snowflakes', namely dandruff, which is raised in a dispute is formed. During dandruff formation, two substances play an important role: oil and fat secreted from scalp, and malassezia that feeds on oil and fat. Under certain conditions, such as when the grease secretion in adolescence is vigorous, malassezia can propagate on the surface of the scalp in a large quantity to stimulate scalp cells to secrete more grease; a large amount of grease provides nutrients for further amplification of malassezia. Finally, the normal scalp stratum corneum cell metabolism becomes unstable, and immature keratinocytes aggregate and fall off in a row, and dandruff is generated.

In the specific embodiment of the invention, the principle of the malassezia bacteriostasis ring test is that the bacteriostat is continuously dissolved and is diffused by agar to form different concentration gradients so as to display the bacteriostasis effect. The experiment judges whether the bacteriostatic ability is possessed or not according to the size of the bacteriostatic ring. The malassezia bacteriostasis ring test comprises the following steps:
preparing the antibacterial tablets: for liquid bacteriostat, a sterile and dry filter paper sheet is taken. Dripping 20 μ L of bacteriostatic agent solution with practical use concentration into each piece, placing the filter paper piece in a clean sterile plate, opening the cover, and oven drying in a warm box (36 deg.C), or naturally drying at room temperature for use.
Preparing a negative control sample: and (4) taking a sterile dry filter paper sheet, dripping 20 mu L of sterile distilled water into each sheet, and drying for later use.
Inoculation of test bacteria, namely dipping test bacteria suspension with the concentration of 5 multiplied by 105 cfu/mL-5 multiplied by 106cfu/mL by using a sterile cotton swab, and evenly smearing the test bacteria suspension on the surface of a nutrient agar medium flat plate for 3 times. For each application 1 time, the plate should be rotated 60 and the cotton swab finally applied around the edge of the plate for one revolution. The plate was covered and dried at room temperature for 5 min.
Fourthly, sticking bacteriostatic agent sample pieces: each test is stuck with 1 infectious bacterium plate, and each plate is stuck with 4 test sample plates and 1 negative control sample plate, and the number of the negative control sample plates is 5. A sample was taken with sterile forceps and placed on the surface of the plate. The distance between the centers of the various pieces is more than 25mm, and the distance between the centers of the various pieces and the periphery of the flat plate is more than 15 mm. After the sample is placed, the sample is lightly pressed by using sterile tweezers to be tightly attached to the surface of the flat plate. The plate is covered, the plate is placed in a 36 ℃ incubator, and the result is observed after 24-48 h of culture. The diameter of the antibacterial ring (including the patch) was measured with a vernier caliper and recorded. The experiment was repeated 3 times. When the inhibition zone is measured, the inhibition zone which is uniform and completely aseptically grows is selected for measurement. The diameter of the catheter is measured by taking the outer edge of the bacteriostatic ring as a boundary.
The evaluation of the malassezia bacteriostasis ring test is judged according to the following contents:
the negative control group should have no bacteriostatic ring, otherwise the test is invalid. If the diameter of the bacteriostatic ring in the experimental group is more than 7mm, the bacteriostatic ring is judged to have bacteriostatic action; if the diameter of the bacteriostatic ring is less than or equal to 7mm, judging that the bacteriostatic action is not exerted; and judging the test to be qualified when the three repeated tests have the antibacterial effect results.
The specific procedure of the evaluation of the anti-dandruff effect of the embodiment of the present invention is to collect volunteers and evaluate the anti-dandruff effect of shampoo, and the evaluation criteria are shown in table 3.

Comparative examples 1 to 3
Comparative examples 1 to 3 each provide a shampoo different from the shampoos of examples 1 to 4 in that: on the basis of the basic formula, the anti-dandruff composition provided by the embodiment of the invention is not added. The shampoo of comparative example 1 to comparative example 3 had the respective components in mass percentages shown in table 5.
TABLE 5
Figure BDA0003688938830000111
Figure BDA0003688938830000121
After the shampoo fragrances of examples 1 to 4 and comparative examples 1 to 3 are prepared, the patches, the malassezia bacteriostatic ring tests and the dandruff removing effect evaluations of examples 1 to 4 are carried out; the shampoos of comparative examples 1 to 3 were subjected to the patch test.
The specific method for testing the patch comprises the following steps:
the method comprises the following steps of firstly, collecting subjects, wherein 32 subjects are collected in the collection of the subjects, 2 subjects are removed, 4 male subjects, 26 female subjects, the minimum age is 24 years old, the maximum age is 58 years old, and the average age is about 36 years old.
② selecting the area not more than 50mm 2 And a qualified spot test device with the depth of about 1mm, placing 1% diluted test object on a filter paper sheet, placing the filter paper sheet into a small chamber of a spot test device, wherein the dosage is about 0.02 mL-0.025 mL, a control hole is a negative control (distilled water), applying the spot test device with the test object on the back of a test subject by using a hypoallergenic adhesive tape, and lightly pressing the test object by hands to uniformly apply the test object on the skin for 24 hours.
Thirdly, observing the skin reaction according to the standard of the table 2 after 30 minutes (after the indentation disappears), 24 hours and 48 hours of removing the patch device of the tested object, and recording the observation result.
The test results of the patch test of examples 1 to 4 and comparative examples 1 to 3 are shown in table 6.
TABLE 6
Example 1 3 out of 30 people suspicious reactions
Example 2 1 out of 30 people with suspicious reactions
Example 3 8 out of 30 suspicious reactions
Example 4 4 out of 30 people suspicious reactions
Comparative example 1 2 out of 30 suspicious reactions
Comparative example 2 7 out of 30 suspicious reactions
Comparative example 3 12 out of 30 people with suspicious reactions
Based on the test data of table 6: comparing example 1 and example 2 with comparative example 2, and comparing example 3 and example 4 with comparative example 3, it can be seen that the number of suspicious reactions in the patch test after adding the herbal extracts (cacumen biotae extract, angelica sinensis extract and sophora flavescens extract) is smaller in the case of the same content of the malassezia inhibitor pyrrolidone ethanolamine salt. Comparing example 1 with example 2, and comparing example 3 with example 4, it can be seen that the higher the content of the herbal extracts (cacumen biotae extract, angelica sinensis extract and sophora flavescens extract) is, the less the number of suspicious reactions in the spot-on test of the corresponding shampoo is, in the case that the content of the malassezia inhibitor pyrrolidone ethanolamine salt is the same, thereby demonstrating that the addition of cacumen biotae extract, angelica sinensis extract and sophora flavescens extract is effective in reducing the irritation caused by the addition of pyrrolidone ethanolamine salt. Comparative example 1 above, which does not contain the anti-dandruff composition provided by the present invention, contains only the base formula, and the patch results of comparative example 1 are 2 suspicious responses, indicating that in the shampoo provided by the present invention, the base formula is relatively mild and can provide a vehicle for the anti-dandruff composition provided by the present invention.
The invention also performs a malassezia bacteriostasis ring test on the shampoos obtained in examples 1-4, and specifically comprises the following steps:
preparing the antibacterial tablets: for liquid bacteriostat, a sterile and dry filter paper sheet is taken. Dripping 20 μ L of bacteriostatic agent solution with practical use concentration into each piece, placing the filter paper piece in a clean sterile plate, opening the cover, and oven drying in a warm box (36 deg.C), or naturally drying at room temperature for use.
Preparing a negative control sample: taking a sterile dry filter paper sheet, dripping 20 mu l of sterile distilled water into each sheet, and drying for later use.
Inoculation of test bacteria: a sterile cotton swab is dipped in test bacterial suspension with the concentration of 5 multiplied by 105 cfu/mL-5 multiplied by 106cfu/mL, and the test bacterial suspension is evenly smeared on the surface of a nutrient agar culture medium plate for 3 times. For each application 1 time, the plate should be rotated 60 and the cotton swab finally applied around the edge of the plate for one revolution. The plate was covered and dried at room temperature for 5 min.
And fourthly, sticking bacteriostatic agent sample sheets, namely sticking 1 bacterial staining flat plate on each test, sticking 4 test sample sheets and 1 negative control sample sheet on each flat plate, and sticking 5 test sample sheets.
The results of the malassezia bacteriostasis ring test of examples 1-4 are shown in table 7.
TABLE 7
Malassezia californica antibacterial ring (mm)
Example 1 30.31
Example 2 42.43
Example 3 36.23
Example 4 48.56
As is clear from table 7, examples 1 to 4 had good inhibitory effects on malassezia. Among them, the suppression effects of examples 2 and 4 are relatively better.
In the present invention, 120 persons were also recruited as volunteers and divided into four groups, and the anti-dandruff effect of the shampoos prepared in examples 1 to 4 was evaluated in 30 persons per group, and the evaluation results are shown in Table 8.
TABLE 8
Figure BDA0003688938830000141
As is clear from table 8, examples 1 to 4 all had good chip-removing effects. Among them, the antidandruff effect of example 2 and example 4 is relatively better.
It can be seen from the comprehensive spot-pasting test, the malassezia bacteriostatic ring test and the anti-dandruff effect evaluation that, as a preferable scheme, the shampoos of examples 2 and 4 all have relatively good spot-pasting test results, malassezia inhibition effects and anti-dandruff effects. Further, the malassezia inhibition effect and the anti-dandruff effect of example 4 are relatively superior to those of example 2, but the number of persons who are suspected to be positive on the spot sticker of example 4 is 4, the number of persons who are suspected to be positive on the spot sticker of example 2 is 1, and the formulation price is relatively high since the amount of the pyrrolone ethanolamine salt of example 4 added is twice that of example 2, and therefore, the anti-dandruff regimen of shampoo is more preferable to the anti-dandruff regimen of shampoo.
In conclusion, the anti-dandruff composition and the corresponding shampoo provided by the embodiment of the invention can inhibit the proliferation of malassezia and repair scalp, and have good anti-dandruff effect.
The foregoing is directed to embodiments of the present application and it is noted that numerous modifications and adaptations may be made by those skilled in the art without departing from the principles of the present application and are intended to be within the scope of the present application.
