技术领域
本发明涉及生物技术领域,具体涉及一种强化型复合骨肽及其制备方法与应用。
背景技术
骨骼中有机物的70%~80%是胶原蛋白,骨骼生成时,首先必须合成充足的胶原蛋白纤维来组成骨骼的框架。因此,有人称胶原蛋白为“骨中之骨”。胶原蛋白的流失或缺乏会引起骨质疏松等疾病。筋腱—构成骨骼与肌肉相联的筋腱的成分80%是胶原蛋白,缺乏胶原蛋白会导致筋腱与肌肉的衰弱。
随着年龄的增长,胶原蛋白流失及身体各方面营养供给不足,软骨的营养供给不足会导致骨骼中的无机物增多,进而导致骨骼弹性和韧性降低,关节软骨不可避免的发生退化和磨损,而关节一直处于运动状态,导致软骨的磨损越来越严重,两块骨直接接触,引发炎症,最后导致骨关节疾病。而骨关节炎多发于中老年人,致残率非常高,被称为“不死的癌症”。且据资料显示,随着人老龄化程度的逐渐加大,骨质疏松逐渐成为人类健康生活的一大关键因素。另外,骨折也是骨损伤的重要因素之一,骨折是指骨头或骨头的结构完全或部分断裂,多见于儿童及老年人,中青年也时有发生。
传统的骨关节临床用药主要以止痛+非甾体类抗炎药结合使用为主,但过多的使用药物治疗存在一系列的副作用,不仅会加重肝肾代谢负担,还会产生药物依赖效应,存在较大的健康安全隐患;还有关节置换、修复性治疗、基础治疗、运动支持等治疗方法,但有费用昂贵、治标不治本或疗效不明确等缺陷。
而骨折时通常伴随着重要血管损伤、周围组织损伤以及周围神经损伤,骨折的愈合一般分为肉芽组织修复期、原始骨痂形成期、成熟骨板期和塑形期,肉芽组织修复期。肉芽组织修复期是骨折后骨的正常结构和周围的软组织破坏,形成血肿,血肿的清除、机化,形成肉芽组织,使骨折断端初步连接在一起,这个过程在骨折后两到三个星期内完成。原始骨痂形成期也就是骨折后新骨形成。开始于骨折后七到十天延续到骨折完全愈合。成熟骨板期是将原始的骨痂被改造成坚强有力的板状骨,这个过程需要8到12个星期。塑形期为骨折愈合过程中塑形,使骨折断端愈合的骨骼结实有力,骨折线消失恢复以前的正常结构,通常要几个月甚至几年。骨折修复大多需卧床,不仅神经和肌肉的活动力大大降低,且血液运行不畅,不利于骨折愈合,修复时间过长,还可能会出现痔疮、下肢深静脉血栓、关节僵硬等并发症。
而随着人们生活水平的提高及营养保健意识日益增强,人们更追求有利于自身健康的食品,更接受“食补、食疗”的养生方式,食疗养生逐渐成为人们关注的热点。通过食补养生达到保健目的以及通过食补促进康健效果、缩减康复时间的方法逐渐被大众所接受和尝试。
骨肽即骨胶原蛋白肽,是以骨明胶为原料,应用酶解工艺,通过控制反应条件所制备得到的骨胶原蛋白产品。胶原蛋白能有效抑制骨骼、软骨肉胶原蛋白的分解,并加速骨密度增加,可预防及治疗退化性关节炎、快速消炎、止痛、消肿、逐渐修复关节软骨及促进新骨形成,可有效预防退化性关节炎,并对关节炎患者具保护功能,能用于帮助关节炎病人的治疗,并有效改善关节疼痛,加速断骨修复、骨折的复原。现有实际生产中也有了骨肽片、骨肽注射液和骨肽饮料等产品,但现有的骨肽饮料产品在吸收速率、人体利用率以及效果等方面都还有很大的优化空间。
发明内容
本发明目的在于提供一种强化型复合骨肽,可应用于促进骨骼生长、增强骨密度、改善骨质疏松和骨关节炎,其具有优良的吸收速率和人体利用率。
为达到上述目的,本发明提供一种强化型复合骨肽,包括以下质量份组分:
牛骨胶原蛋白肽10~15份、鸡软骨蛋白肽1.5~2.5份、磷酸氢钙0.001~0.005份、L-乳酸钙5~8份、维生素D 0.01~0.02份、维生素B12 0.005~0.01份、维生素C 0.1~0.2份、姜黄0.1~0.2份、壳寡糖0.2~0.5份、甜菊糖苷0~0.01份、DL-苹果酸0.01~0.04份、果胶0.01~0.03份、调味辅助剂0.01~1份,所述鸡软骨蛋白肽中含硫酸软骨素35wt%以上,含活性Ⅱ型胶原蛋白肽55wt%以上。
进一步优选的,所述调味辅助剂包括三氯蔗糖0.01~0.05份、焦糖色素0.005~0.01份和/或黄原胶0.1~0.3份。
进一步优选的,所述调味辅助剂还包括益生菌0.1~0.2份和/或透明质酸钠0.05~0.2份。
本发明还提供所述强化型复合骨肽的制备方法,包括以下步骤:
步骤一、牛骨脱脂、酶解、灭酶、浓缩干燥得到牛骨胶原蛋白肽粉;
步骤二、鸡胸软骨粉碎、高压蒸煮、脱脂、低温酶解、灭酶后浓缩干燥得到鸡软骨蛋白肽粉,其中低温酶解条件为:复合酶结合超声波辅助酶解,超声波频率25~30Hz,酶解温度为46~48℃,酶解时间为100~120min;所述鸡软骨蛋白肽粉中含硫酸软骨素含量35wt%以上,含活性Ⅱ型胶原蛋白肽55wt%以上;
步骤三、按照质量比,在搅拌机中边搅拌边加入全部量的牛骨胶原蛋白肽粉、鸡软骨蛋白肽粉、磷酸氢钙、L-乳酸钙、维生素D、维生素B12、维生素C、姜黄、壳寡糖、甜菊糖苷、DL-苹果酸、果胶、调味辅助剂,搅拌均匀后采用高温瞬时灭菌技术进行灭菌处理,得到所述强化型复合骨肽。
进一步优选的,步骤二中步骤二中复合酶为胰酶、木瓜蛋白酶和胃蛋白酶,胰酶、木瓜蛋白酶及胃蛋白酶添加量分别为1.2g/1000ml、1g/1000ml、1.1g/1000ml。
本发明还提供所述的强化型复合骨肽在制备药物中的应用,所述的药物用于促进骨骼生长、增强骨密度、缓解关节疼痛、舒缓骨质疏松和改善骨关节炎。
本发明还提供所述的强化型复合骨肽在制备保健品或食品中的应用,所述的保健品或食品用于骨胶原蛋白和钙、磷元素补充,促进钙、磷元素吸收和强健骨骼。
本发明还提供一种用于促进骨骼生长的药物,所述药物中包括质量分数为70~100%的所述强化型复合骨肽和质量分数为0~30%药品领域可接受的辅料,所述药物为片剂、粉剂、颗粒剂、口服液、滴剂、丸剂或胶囊剂。
本发明还提供一种食品或保健品,所述食品或保健品中包括质量分数为30~50%的所述强化型复合骨肽和质量分数为50~70%的食品或保健品领域可接受的辅料,所述保健品或食品为饮品、糖果或粉剂。。

本实施例提供一种强化型复合骨肽,包括以下质量份组分:牛骨胶原蛋白肽15份、鸡软骨蛋白肽1.5份、磷酸氢钙0.001份、L-乳酸钙5.2份、维生素D0.01份、维生素B12 0.005份、维生素C 0.1份、姜黄0.1份、壳寡糖0.2份、甜菊糖苷0.004份、DL-苹果酸0.02份、果胶0.03份、三氯蔗糖0.02份、焦糖色素0.01份、黄原胶0.3份、益生菌0.14份、透明质酸钠0.18份,所述鸡软骨蛋白肽中硫酸软骨素不低于35wt%,含活性Ⅱ型胶原蛋白肽不低于55wt%。
所述强化型复合骨肽的制备方法,包括以下步骤:
步骤一、牛骨脱脂、酶解、灭酶、浓缩干燥得到牛骨胶原蛋白肽粉。
步骤二、鸡胸软骨粉碎、高压蒸煮、脱脂、低温酶解、灭酶后浓缩干燥得到鸡软骨蛋白肽粉,其中低温酶解条件为:使用胰酶、木瓜蛋白酶和胃蛋白酶结合超声波辅助酶解,胰酶、木瓜蛋白酶及胃蛋白酶添加量分别为1.2g/1000ml、1g/1000ml、1.1g/1000ml;超声波频率30Hz,酶解温度为46~48℃,酶解时间为120min。
步骤三、按照质量比,向搅拌机中边搅拌边加入全部量的牛骨胶原蛋白肽粉、鸡软骨蛋白肽粉、磷酸氢钙、L-乳酸钙、维生素D、维生素B12、维生素C、姜黄、壳寡糖、甜菊糖苷、DL-苹果酸、果胶、三氯蔗糖、焦糖色素、黄原胶、益生菌、透明质酸钠,搅拌均匀后采用高温瞬时灭菌技术进行灭菌处理,得到强化型复合骨肽。
1、鸡软骨酶解条件试验
选用木瓜蛋白酶和胰酶配合为复合酶1、胰酶和胃蛋白酶配合为复合酶2、木瓜蛋白酶和胃蛋白酶配合为复合酶3、木瓜蛋白酶和胰酶及胃蛋白酶配合为复合酶4以及木瓜蛋白酶、胰酶、胃蛋白酶分别对鸡软骨进行酶解,酶总添加量相同,酶解温度为酶最适温度,酶解相同时间,以蛋白回收率、Ⅱ型胶原蛋白得率和硫酸软骨素得率为评价标准。结果如下表1。

由表1可得,以木瓜蛋白酶和胰酶及胃蛋白酶配合得复合酶3酶解产物蛋白回收率、Ⅱ型胶原蛋白得率和硫酸软骨素得率最高。
另以步骤二时不以超声辅助酶解,其余工艺条件与实施例3相同为对照制备鸡软骨蛋白肽粉,对照制得的鸡软骨蛋白肽粉的蛋白回收率、Ⅱ型胶原蛋白得率和硫酸软骨素得率均低于实施例3。
2、实施例1~4的强化型复合骨肽治疗骨质疏松功效的研究
1)30只雄性昆明小鼠在SPF级饲养室正常喂食稳定三天后准备建立骨质疏松模型,30只小鼠随机分为三组,分别为正常组、模型组和试验组1~4。除正常组外,其余5组每天灌胃给予150mg/kg的维甲酸建立骨质疏松模型,建模时间持续两周。
维甲酸诱导的骨质疏松模型建立完成后,正常组和模型组小鼠除正常饮食外每天给予自由饮水,试验组1~4除正常饮食饮水外分别每天给予7.5m1/20g体积的实施例1~4的强化型复合骨肽喂养,喂养时间持续三周。
2)小鼠股骨体积及骨钙含量测定
(1)小鼠股骨体积及长、短径的测量
喂食鳄鱼骨肤21天后,处死小鼠,剥离小鼠股骨,置于烘箱中干燥,称量干燥后的小鼠股骨重量,记录骨重数据,股骨干重记为B,随后将小鼠股骨置于lml注射器内,推动针芯,将股骨抵至注射器底部,然后将注射器置于4 0C的水中,抽动针芯,直至股骨被水浸泡,同时翻动注射器以将空气排出,推动针芯使其抵住股骨。在注射器针芯边界处做好标记,用游标卡尺测量针芯至注射器底部的距离记为L,然后称量注射器总重(注射器内仅含水和骨),记录为C。抽出针芯,将股骨及水移出注射器,然后抽水至标记的刻度处,并用滤纸将注射器周围的水吸干,称重,记录注射器及水重为D。股骨在注射器中所占体积V=D+B-C,根据水在4℃水中的体积重量换算方法:1g水的体积为lcm3,即可将股骨重量换算为体积。取出的股骨用游标卡尺测量其横长径与横短径,记录结果,详细结果见表1。

如表1、表2所示,骨质疏松模型组小鼠的股骨体积及横长、短径明显小于正常组,股骨骨钙含量也明显低于正常组,而试验组1~4小鼠无论是股骨体积及横长、短径,还是股骨骨钙含量均与正常组相差无几,说明本申请提供的强化型复合骨肽能为小鼠进行钙元素补充且能促进其对骨钙的吸收,对于骨质疏松疗效良好。
以上所揭露的仅为本发明几种较佳实施例而已,当然不能以此来限定本发明之权利范围,因此依本发明申请专利范围所作的等同变化,仍属本发明所涵盖的范围。
Claims (10)
1.一种强化型复合骨肽,其特征在于,包括以下质量份组分:
牛骨胶原蛋白肽10~15份、鸡软骨蛋白肽1.5~2.5份、磷酸氢钙0.001~0.005份、L-乳酸钙5~8份、维生素D 0.01~0.02份、维生素B12 0.005~0.01份、维生素C 0.1~0.2份、姜黄0.1~0.2份、壳寡糖0.2~0.5份、甜菊糖苷0~0.01份、DL-苹果酸0.01~0.04份、果胶0.01~0.03g、调味辅助剂0.01~1份;
所述鸡软骨蛋白肽中含硫酸软骨素35wt%以上,含活性Ⅱ型胶原蛋白肽55wt%以上。
2.如权利要求1所述的一种强化型复合骨肽,其特征在于,所述调味辅助剂包括三氯蔗糖0.01~0.05份、焦糖色素0.005~0.01份和/或黄原胶0.1~0.3份。
3.如权利要求2所述的一种强化型复合骨肽,其特征在于,所述调味辅助剂还包括益生菌0.1~0.2份和/或透明质酸钠0.05~0.2份。
4.一种强化型复合骨肽的制备方法,用于制备如权利要求1~3所述的强化型复合骨肽,其特征在于,包括以下步骤:
步骤一、牛骨脱脂、酶解、灭酶、浓缩干燥得到牛骨胶原蛋白肽粉;
步骤二、鸡胸软骨粉碎、高压蒸煮、脱脂、低温酶解、灭酶后浓缩干燥得到鸡软骨蛋白肽粉,其中低温酶解条件为:复合酶结合超声波辅助酶解,超声波频率25~30Hz,酶解温度为46~48℃,酶解时间为100~120min;所述鸡软骨蛋白肽粉中硫酸软骨素含量不低于35wt%,含活性Ⅱ型胶原蛋白肽不低于55wt%;
步骤三、按照质量比,在搅拌机中边搅拌边加入全部量的牛骨胶原蛋白肽粉、鸡软骨蛋白肽粉、磷酸氢钙、L-乳酸钙、维生素D、维生素B12、维生素C、姜黄、壳寡糖、甜菊糖苷、DL-苹果酸、果胶、调味辅助剂,搅拌均匀后采用高温瞬时灭菌技术进行灭菌处理,得到所述强化型复合骨肽。
5.如权利要求4所述的强化型复合骨肽的制备方法,其特征在于,步骤二中复合酶为胰酶、木瓜蛋白酶和胃蛋白酶。
6.如权利要求5所述的强化型复合骨肽的制备方法,其特征在于,胰酶、木瓜蛋白酶及胃蛋白酶添加量分别为1.2g/1000ml、1g/1000ml、1.1g/1000ml。
7.权利要求1~3任一项所述的强化型复合骨肽在制备药物中的应用,其特征在于,所述的药物用于促进骨骼生长、增强骨密度、缓解关节疼痛、舒缓骨质疏松和改善骨关节炎。
8.权利要求1~3任一项所述的强化型复合骨肽在制备保健品或食品中的应用,其特征在于,所述的保健品或食品用于骨胶原蛋白和钙、磷元素补充,促进钙、磷元素吸收和强健骨骼。
9.一种用于促进骨骼生长的药物,其特征在于,所述药物中包括质量分数为70~100%的权利要求1~3任一项所述的强化型复合骨肽和质量分数为0~30%药品领域可接受的辅料,所述药物为片剂、粉剂、颗粒剂、口服液、滴剂、丸剂或胶囊剂。
10.一种食品或保健品,其特征在于,所述食品或保健品中包括质量分数为30~50%的权利要求1~3任一项所述的强化型复合骨肽和质量分数为50~70%的食品或保健品领域可接受的辅料,所述保健品或食品为饮品、糖果或粉剂。
