Abstract

本申请涉及噬菌体技术领域,尤其是涉及一种宽裂解谱耐紫外奇异变形杆菌噬菌体及其组合物。奇异变形杆菌噬菌体为奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)。奇异变形杆菌噬菌体PMP1在浓度为102~103PFU/mL的奇异变形杆菌培养基中105~106 PFU/mL的噬菌体对该浓度的奇异变形杆菌的灭菌率达到97%以上,对奇异变形杆菌具有广谱的杀菌能力;奇异变形杆菌噬菌体PMP1的组合物可裂解更多奇异变形杆菌,裂解率达97%以上,具有更强的裂解性。

技术领域
本申请涉及噬菌体技术领域,尤其是涉及一种宽裂解谱耐紫外的奇异变形杆菌噬菌体及其组合物、试剂盒和应用。
背景技术
变形杆菌是广泛分布于自然界的革兰阴性腐生性细菌,属于肠杆菌科(Enterobacteriaceae)。变形杆菌属包括奇异变形杆菌(Proteous mirabilis)、普通变形杆菌(Proteous vulgaris)、潘氏变形杆菌(Proteous penneri)和产黏变形杆菌(Proteousmyxofaciens)。国内流行病学资料显示,食源性微生物病原菌中,主要以沙门氏菌、副溶血性弧菌和变形杆菌为代表。近年来,变形杆菌在细菌性食物中毒中占有较大比重,尤其是奇异变形杆菌引起的食物中毒占变形杆菌食物中毒的70%。
奇异变形杆菌与感染相关的毒力因子主要表现在菌毛与荚膜多糖带来的附力与侵袭力上,另外该菌产生的各种蛋白水解酶、各种细胞毒性溶血素使感染变得更加复杂。奇异变形杆菌属肠道正常菌群,为条件性致病菌。在动物正常情况下,该菌无致病作用,动物不会发病;当肠道功能受到扰乱,或机体抵抗力下降时方可致病。故奇异变形杆病多在应激情况下发生。奇异变形杆菌广泛存在于水、土壤腐败的有机物以及人和动物的肠道中,多体现为继发感染,如慢性中耳炎、创伤感染等,也可引起膀胱炎、婴儿腹泻、食物中毒等。
奇异变形杆菌主要污染动物性食品,特别是肉类,因此应加大对动物性食品中奇异变形杆菌的监测力度,保障消费者的食用安全。
家禽奇异变形杆菌病一般没有明显的临床症状,病重的成年鸡产软壳蛋、沙皮蛋、畸形蛋,有时可引起脱肛,啄肛等。
噬菌体是一类专性寄生或感染细菌的病毒,它们广泛存在于自然界中。烈性噬菌体可以在敏感宿主菌胞内增殖,最后裂解宿主菌释放出子代噬菌体,从而达到破坏宿主菌的作用。在杀菌效果上,烈性噬菌体有广泛应用,尤其是利用其治疗泛耐药的“超级细菌”引起的感染,取得了良好的效果。
现有公告号为CN110684742B的国内发明专利,公开了一株奇异变形杆菌噬菌体RDP-SA-16033,其保藏编号为CGMCC No.18197。该奇异变形杆菌噬菌体RDP-SA-16033对养殖环境中奇异变形杆菌S5具有强裂解效用;RDP-SA-16033具有较高的温度耐受性和较宽的酸碱耐受范围,效价较高,安全有效。
上述中的现有技术方案存在以下缺陷:上述奇异变形杆菌噬菌体的裂解范围均有限且不耐紫外,亟需提供一种宽裂解谱耐紫外的奇异变形杆菌噬菌体。
发明内容
针对现有技术存在的不足,本申请的目的之一是提供一种宽裂解谱耐紫外奇异变形杆菌噬菌体。
本申请的上述发明目的之一是通过以下技术方案得以实现的:一种宽裂解谱耐紫外的奇异变形杆菌噬菌体,所述奇异变形杆菌噬菌体为奇异变形杆菌噬菌体PMP1(Proteusmirabilis phage PMP1),保藏编号为CCTCC NO:M2020902。
通过采用上述技术方案,提供了一种新型的具有宽裂解谱的奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1),该噬菌体同时对紫外具有良好的耐受能力,可以高效杀灭奇异变形杆菌。该噬菌体具有SEQ ID No.1所示的核苷酸序列。
本申请在一较佳示例中可以进一步配置为:所述奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1),MOI=0.0001条件下培养8~10h,其效价为1.8×1011PFU/mL。
本申请在一较佳示例中可以进一步配置为:所述奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1),对紫外具有良好的耐受性,于紫外下照射20min后仍有90%以上的存活率。
本申请的目的之二是提供一种含有上述的宽裂解谱耐紫外奇异变形杆菌噬菌体的组合物及其应用。奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)可以与其他噬菌体组合联用,也可以与其它物质混合,满足特异性需求。
本申请的上述发明目的之二是通过以下技术方案得以实现的:一种含有上述的宽裂解谱奇异变形杆菌噬菌体的组合物,组合物至少含有所述奇异变形杆菌噬菌体(Proteusmirabilis phage PMP1)。
本申请在一较佳示例中可以进一步配置为:所述组合物还包括有大肠杆菌噬菌体EC35P1(Escherichia coli phage EC35P1),保藏编号为CCTCC NO:M2020438;沙门氏菌噬菌体SG8P3(Salmonella pullorum phage SG8P3),保藏编号为CCTCC NO:M 2020205;金黄色葡萄球菌噬菌体J1P2(Staphylococcus aureus phage J1P2),保藏编号为CCTCC NO:M2016285。

综上所述,本申请包括以下至少一种有益技术效果:

奇异变形杆菌噬菌体PMP1(Proteusmirabilis phage PMP1)、大肠杆菌噬菌体EC35P1(Escherichia coli phage EC35P1)、沙门氏菌噬菌体SG8P3(Salmonella pullorumphage SG8P3)和金黄色葡萄球菌噬菌体J1P2(Staphylococcus aureus phage J1P2)的组合物1~组合物7均依实施例6所述方法制备。
奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)、终浓度0.05%的双癸基二甲基氯化铵和终浓度度为0.01%的新洁尔灭的组合物8依实施例6所述方法制备。
实施例1噬菌体的筛选和纯化
(一)样品采集及处理
本申请中的样品采集自山东德州的养殖场污水与附近农田土壤。
采集的样品经过5000r/min离心10min,过0.22μm滤膜。
(二)样品中针对目标奇异变形杆菌的噬菌体富集将上述滤液以1:1的比例与2倍LB液体培养基混合,接种目标奇异变形杆菌菌株100μL,过夜富集。
(三)噬菌体筛选及纯化
将上述富集液离心取上清过0.22μm膜后,取1mL与5mL含有目标奇异变形杆菌的LB半固体培养基混匀,倾倒于盛有LB固体培养基的培养皿上,待半固体培养基凝固后,37℃过夜培养,第二天观察是否有噬斑,记录实验结果。
挑取上述单噬菌斑于1mL SM缓冲液中,150rpm震荡15min,梯度稀释,将稀释液与5mL含有目标奇异变形杆菌的LB半固体培养基混匀,倾倒于盛有LB固体培养基的培养皿上,待半固体培养基凝固后,37℃过夜培养。重复该步骤3~5次,得到噬菌体单克隆样品,命名为奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)。奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)形态如图1所示,形态为一呈多面体对称的头部和可伸缩的尾部,头部直径55~65nm,尾长113~123nm,尾部直径7~12nm。本图中奇异变形杆菌噬菌体PMP1头部直径57nm,尾长117nm,尾部直径10nm[x1]该噬菌体具有SEQ ID No.1所示的核苷酸序列。将奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)的序列在NCBI网站上比对并结合透射电镜观察结果可得该噬菌体属于长尾科(Siphoviridae)噬菌体。
(四)噬菌体颗粒制备
应用宿主菌将奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)扩增,待培养基澄清后,8000g离心10min,去除杂质,加入固体聚乙二醇(PEG8000)至终浓度为10%(w/v),搅拌溶解,4℃过夜,4℃8000g离心20min,将沉淀重悬于SM缓冲液中。将得到的噬菌体悬液4℃保存待用。
实施例2奇异变形杆菌噬菌体PMP1(Proteus mirabilisphage PMP1)对奇异变形杆菌最佳感染复数(MOI)的测定
(1)奇异变形杆菌噬菌体效价的测定
用SM液做稀释液,将奇异变形杆菌噬菌体PMP1(由实施例1制得)的原液10倍梯度逐级稀释至l08倍。分别取l05、l06、l07及l08稀释度的噬菌体培养液l000μL与其宿主菌菌液300μL均匀混合,静置15min使其与细菌表面的受体充分结合。将上述混合液加入5mL冷却至50℃的半固体琼脂培养基中,混匀后立即铺于已凝固的固体琼脂平板上,待琼脂凝固后于37℃倒置培养6~8h。每个稀释度需做三个平行样,计数时取此稀释度的三个平行样的平均数。其中,噬菌体效价(PFU/mL)=平均噬菌斑数×稀释倍数
(二)奇异变形杆菌噬菌体最佳感染复数(MOI)的测定
挑取奇异变形杆菌单个菌落,接种到盛有3mL LB培养液的试管中,37℃150rpm振荡培养8h,得到宿主菌悬液。将菌悬液以1:100比例转接到l0mL LB培养液,37℃150rpm振荡培养至对数前期。按照感染复数比例加入奇异变形杆菌噬菌体PMP1的纯培养液(由实施例1制得)和宿主菌(MOI=噬菌体数量/细菌数量),加入LB液体培养基使各管总体积相同。在37℃摇床中150rpm摇培8h。培养完毕后5000g离心l0min并收集上清液,测定噬菌体效价。各点均作双份复管培养取平均值,以产生最高噬菌体效价的MOI为最佳感染复数。实验重复3次。

结果如表1所示,奇异变形杆菌噬菌体PMP1效价达到最高(1.8x1011PFU/mL)时,其MOI为1:10000。其初始投入量少,繁殖速度快,可应用于工业生产中。
实施例3奇异变形杆菌噬菌体PMP1的毒力基因或不良基因缺失检测试验本实施例选取103种经鉴定源自病原细菌体内溶源性噬菌体的毒力基因如表2所示,通过测定奇异变形杆菌噬菌体PMP1的全基因组并对其进行生物信息学分析,以确定其是否含有下列毒力基因。结果显示,奇异变形杆菌噬菌体PMP1不含有下列毒力基因或不良基因,所以无法编码可能引起潜在健康风险的蛋白,因此奇异变形杆菌噬菌体PMP1不会影响人或动物体的健康。

实施例4毒理实验
实验小鼠20只,雌雄各半,适应性饲养三天后,随机分为两组(噬菌体组、对照组),每组10只(雌雄各5只),给予噬菌体组计量为1010PFU/kg的奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)(实施例2制得),对照组给予等量生理盐水,连续给药15d,将实验鼠断颈处死,检查内脏情况。
实验结果显示,此剂量的奇异变形杆菌噬菌体PMP1(Proteus mirabilis phagePMP1)对小鼠日常行为没有影响。解剖检查内脏未见异常。本申请的奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)具有生物安全性,可应用于饲料添加剂。当然,奇异变形杆菌噬菌体PMP1也可以应用成保健品去运用或药剂去应用,在本毒理验中也证明是安全的。
实施例5奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)的热稳定性试验
各取l00μL噬菌体纯培养液(实施例2制得)装于无菌EP管中,分别于55℃、65℃、75℃水浴中作用2h、24h、48h。作用时间结束后取出样品管并立即置于冰浴中冷却,经适当稀释后采用双层平板法测定噬菌体效价。实验重复3次。

结果如表3所示,实验组中奇异变形杆菌噬菌体PMP1(Proteus mirabilis phagePMP1)在55℃下较易存活。奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)具有良好的耐高温特性,与对照相比,65℃水浴2h仍具有80.8%存活率,75℃下水浴2h仍具106PFU/mL效价。
实施例6:奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)及奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)组合物的制备和裂解范围试验采用点滴法来测定噬菌体的裂解谱。选择大量的奇异变形杆菌株,将5mL含有奇异变形杆菌的LB半固体培养基混匀,分别倾倒于盛有LB固体培养基的培养皿上,待半固体培养基凝固后用所述奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)进行点滴。
分别取效价为2x109PFU/mL奇异变形杆菌噬菌体PMP1(Proteus mirabilis phagePMP1)和大肠杆菌噬菌体EC35P1(Escherichia coli phage EC35P1)的原液,将2株噬菌体等体积1:1均匀混合于SM液中,制成奇异变形杆菌噬菌体PMP1(Proteus mirabilis phagePMP1)和大肠杆菌噬菌体EC35P1(Escherichia coli phage EC35P1)的1:1的组合物(组合物1)。
分别取效价为2x109PFU/mL奇异变形杆菌噬菌体PMP1(Proteus mirabilis phagePMP1)和沙门氏菌噬菌体SG8P3(Salmonella pullorum phage SG8P3)的原液,将2株噬菌体等体积均匀混合制成奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)和沙门氏菌噬菌体SG8P3(Salmonella pullorum phage SG8P3)的1:1的组合物(组合物2)。
分别取效价为2x109PFU/mL奇异变形杆菌噬菌体PMP1(Proteus mirabilis phagePMP1)和金黄色葡萄球菌噬菌体J1P2(Staphylococcus aureus phage J1P2)的原液,将2株噬菌体等体积均匀混合制成奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)和金黄色葡萄球菌噬菌体J1P2(Staphylococcus aureus phage J1P2)的1:1的组合物(组合物3)。

分别取效价为2x109PFU/mL奇异变形杆菌噬菌体PMP1(Proteus mirabilis phagePMP1)、终浓度0.05%的双癸基二甲基氯化铵、终浓度度为0.01%的新洁尔灭,将3者等体积1:1:1均匀混合制成组合物(组合物8)。
挑取140株奇异变形杆菌的单菌落分别接种于盛有3mL LB的试管中,160rpm培养8h,制得各株细菌菌液。取300μL菌悬液分别与半固体培养基混合铺于普通琼脂平板上,分别取10μL奇异变形杆菌噬菌体PMP1培养液及其组合物1~8培养液滴于平板的不同位置上,加样时各噬菌体培养液间不可接触,以免影响试验结果。待自然风干后37℃培养6~8h,观察结果。试验重复三次。
结果如表4所示,奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)具有较宽的宿主范围。奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)可裂解133株奇异变形杆菌,裂解率为95%。而噬菌体PMP1的组合物培养液普遍可识别至少136株溶藻弧菌以上,裂解率至少为97.14%以上。说明奇异变形杆菌噬菌体PMP1(Proteusmirabilis phage PMP1)具有较宽的宿主范围,同时噬菌体PMP1的组合物可弥补噬菌体单一应用时宿主谱的局限性,在噬菌体治疗方面具有极大的应用潜力。

实施例7奇异变形杆菌噬菌体PMP1及其组合物在液体中的杀菌效果1.奇异变形杆菌计数方法(效价)
采用BS奇异变形杆菌选择性培养基,应用稀释涂布的方法计数,37℃培养24h,黑色有金属光泽的菌落为阳性菌落。
2.奇异变形杆菌噬菌体PMP1在液体中杀菌效果实验培养奇异变形杆菌至对数生长期,分装进不同试管中,用等体积的液体LB培养基稀释菌液至奇异变形杆菌的终浓度为3x103cfu/mL,接种进不同浓度的奇异变形杆菌噬菌体PMP1(Proteus mirabilis phagePMP1),使样品中奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)的浓度分别是5x102PFU/mL、5x103PFU/mL、5x104PFU/mL、5x105PFU/mL、5x106PFU/mL。同时设置对照组和空白组(CK),对照组给予终浓度3x103cfu/mL的奇异变形杆菌;空白组给予等量生理盐水。4h后检测奇异变形杆菌的残留量。结果参见表5。

表5的测试结果显示:奇异变形杆菌噬菌体PMP1浓度为105~106PFU/mL时,奇异变形杆菌噬菌体PMP1对液体LB培养基中奇异变形杆菌的杀菌效果最好,杀灭率在97%以上。表明奇异变形杆菌噬菌体PMP1具有应用为生物杀菌剂的潜力。
1.含高浓度奇异变形杆菌噬菌体PMP1的组合物在液体中杀菌效果实验
奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)组合物1~组合物8,其配制方法见实施例6。
培养奇异变形杆菌至对数生长期,分装进不同试管中,用液体LB培养基稀释菌液至使奇异变形杆菌的终浓度为3x103cfu/mL,接种带有终浓度为3x106PFU/mL浓度的奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)的组合物1~组合物7。设置对照组和空白组(CK),对照组给予终浓度1x103cfu/mL的奇异变形杆菌;空白组给予等量生理盐水。4h后检测奇异变形杆菌的残留量,检测方法见本实施例步骤1。结果参见表6。

表6测试结果显示:奇异变形杆菌噬菌体PMP1浓度为高浓度3x106PFU/mL时,带有其的组合物不仅杀菌效果好,而且对其它组合成分无拮抗作用。奇异变形杆菌噬菌体PMP1的组合物具有应用为生物杀菌剂的潜力。
实施例8奇异变形杆菌噬菌体PMP1及其组合物对雏鸡养殖过程中奇异变形杆菌感染的防治
组合物1~组合物8的制备参见实施例6。
实验雏鸡550只,适应性饲养3天后,随机分为11组(噬菌体PMP1组、组合物1~组合物8、对照组和空白组),每组50只,各处理实验组给予剂量为3x108PFU/kg的供试噬菌体和3x108PFU/kg的奇异变形杆菌;对照组给予3x108PFU/kg的奇异变形杆菌;空白组给予等量生理盐水,连续给药15d。统计雏鸡日常生长情况及发病率,结果参见表7。

表7结果显示:对照组在第7天雏鸡全部发病;空白组发病率4%,雏鸡生活有大量的腹泻,脱肛现象;单奇异变形杆菌噬菌体PMP1组雏鸡发病率26%;奇异变形杆菌噬菌体PMP1组合物7雏鸡发病率10%,其它组合物的雏鸡发病率也均为20%以下,表明奇异变形杆菌噬菌体PMP1可与其它组分协同使用,并不影响其性能。该试验也表明奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)及其组合物可以用作为生物杀菌剂或饲料添加剂,有效防治鸡养殖过程中奇异变形杆菌的感染。
实施例9奇异变形杆菌噬菌体PMP1(Proteus mirabilisphagePMP1)及其组合物对鸡肉样品保存过程中奇异变形杆菌污染的防治
1.噬菌体计数方法:同实施例2所述的方法。
2.奇异变形杆菌计数方法:同实施例7所描述的方法。
3.组合物1~组合物8的配制方法:同实施例6所描述的方法。
4.将高压蒸汽灭菌的鸡胸肉切成1cm见方的小方块50块,分为11组(单噬菌体PMP1组、组合物1~组合物8、对照组和空白组),每组5块,置于无菌平皿中。实验组每块接种3x105PFU奇异变形杆菌和剂量为3x108PFU/kg的供试噬菌体;对照组每块接种3x105PFU奇异变形杆菌和等量无菌水;空白组给与等量无菌生理盐水。将各处理置于37℃培养箱中培养。每2h取一块鸡肉置于10mL无菌水中,充分震荡,测定该液体的奇异变形杆菌含量,结果参见表8。

表8结果显示:8h后对照组的鸡肉表面生长了大量的奇异变形杆菌;而各实验组由于奇异变形杆菌噬菌体PMP1及其组合物的添加,鸡肉表面的奇异变形杆菌始终控制在极低的水平。奇异变形杆菌噬菌体PMP1及其组合物可以用作为生物杀菌剂,有效杀灭鸡肉表面的奇异变形杆菌并防治鸡肉在保存过程中奇异变形杆菌的污染。
实施例10奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)的紫外耐受实验
取10mL效价为3×109PFU/mL的噬菌体PMP1(由实施例1制得)平铺于90mm的无菌培养皿中,于灭过菌的超净工作台内将其置于紫外灯(20w,30cm,50μW/cm2)下进行照射。分别在0min,20min,40min,60min,2h,3h,4h,5h,6h,7h,8h时取样并于室温下将其置于暗处30min后采用双层平板法测定噬菌体效价。实验重复3次。效价测定方法见实施例7,结果见表9。

表9结果显示:奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)对紫外具有良好的耐受性,于紫外下照射20min后仍有90%以上的存活率;照射1h后其效价降低一个数量级;照射8h后PMP1仍具有104PFU/mL效价。
实施例11奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)及其组合物的试剂盒的制备及使用
试剂盒中含有5~10mL的效价为3×109的奇异变形杆菌噬菌体PMP1(Proteusmirabilis phage PMP1)或奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)的组合物,1L LB半固体培养基,1L LB固体培养基。
试剂盒的使用方法为:取效价为3×109PFU/mL奇异变形杆菌噬菌体PMP1(Proteusmirabilis phage PMP1)噬菌体液体或奇异变形杆菌噬菌体PMP1(Proteus mirabilisphage PMP1)的组合物的液体,采用双层平板点滴法来测定供试噬菌体的裂解谱。挑取待检测单菌落,将其接种于目标液体培养基中,于目标温度下结合待检测菌株生长特性进行振荡培养,制得待检测菌株菌液。取300μL待检测菌株菌悬液分别与5mL LB半固体培养基混合铺于LB固体平板上,取10μL奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)液体或奇异变形杆菌噬菌体PMP1(Proteus mirabilis phage PMP1)的组合物点滴于平板上。待自然风干后根据待检测菌株生长特性于目标温度下进行培养,观察结果即可。

2.一种含有权利要求1所述的宽裂解谱耐紫外的奇异变形杆菌噬菌体的组合物,其特征在于:组合物至少含有所述的奇异变形杆菌噬菌体(Proteus mirabilis phage) PMP1。
3.根据权利要求2所述宽裂解谱耐紫外的奇异变形杆菌噬菌体的组合物,其特征在于:所述组合物还包括有大肠杆菌噬菌体(Escherichia coli phage)EC35P1,保藏编号为CCTCC NO: M 2020438;沙门氏菌噬菌体(Salmonella pullorum phage)SG8P3,保藏编号为CCTCC NO: M 2020205;金黄色葡萄球菌噬菌体(Staphylococcus aureus phage)J1P2,保藏编号为CCTCC NO: M 2016285。
4.据权利要求2所述宽裂解谱耐紫外的奇异变形杆菌噬菌体的组合物,其特征在于:所述组合物还包括化学性消毒剂。
5.根据权利要求4所述宽裂解谱耐紫外的奇异变形杆菌噬菌体的组合物,其特征在于:采用的化学性消毒剂为终浓度为 0.05%的双癸基二甲基氯化铵或终浓度为0.01%的新洁尔灭中的一种。
6.一种含有奇异变形杆菌噬菌体的试剂盒,其特征在于:所述试剂盒中含有权利要求1所述的奇异变形杆菌噬菌体(Proteus mirabilis phage)PMP1或权利要求2~5任意一项所述的含有奇异变形杆菌噬菌体(Proteus mirabilis phage)PMP1的组合物。
7.一种权利要求1所述宽裂解谱耐紫外的奇异变形杆菌噬菌体的应用,其特征在于:所述奇异变形杆菌噬菌体(Proteus mirabilis phage ) PMP1在制备生物杀菌剂、消毒剂中的应用,所述生物杀菌剂、消毒剂是用来杀灭或抑制奇异变形杆菌(Proteus mirabilis)或溶藻弧菌(Vibrio alginolyticus)。
