Abstract

本发明公开了一株裂解性、高效价副溶血弧菌噬菌体RDP‑VP‑21010及其应用,其宿主为副溶血弧菌,透过电镜观察,该噬菌体头部呈正六边形,头部直径约为56nm,尾部长度为103nm,为长尾科噬菌体,基因组全长为12558bp,保藏编号为CGMCC No.22481。副溶血弧菌噬菌体RDP‑VP‑21010对水体中副溶血弧菌具有强裂解作用,具有较高的温度耐受性和较宽的酸碱耐受范围,可以高效治疗该病原菌在水产养殖中所引起的副溶血弧菌疾病,以及可以作为消毒剂,为工业化、规模化、程序化生产噬菌体及治疗副溶血弧菌引起的水产养殖细菌病提供了噬菌体来源。

技术领域
本发明涉及生物技术领域,特别是涉及一株裂解性、高效价副溶血弧菌菌噬菌体及其在水产养殖中副溶血弧菌感染中的应用。
背景技术
副溶血弧菌又名致病性嗜盐菌,是一种海洋致病菌,为人畜共患病原菌。在1950年在日本的爆发性食物中毒中分离发现。副溶血弧菌分布极广,主要分布在海水和水产品中,据统计,我国华东地区沿岸海水中副溶血性弧菌的检出率是47.5%~66.5%,海产鱼虾的平均带菌率是45.6%~48.7%,在夏季时可高达90%以上。除了海产品以外,在畜禽肉、咸菜、咸蛋、淡水鱼等都发现有副溶血性弧菌的存在。
近年来,我国的水产养殖业获得迅猛发展。随着海水养殖规模的日益扩大和集约化养殖方式的不断推广,鱼虾疾病的频繁发生对水产品的规模化养殖的健康发展造成了严重的威胁。水产动物的发病要素主要有细菌、病毒,寄生虫等,其中细菌病是对水产动物度危害程度较为严重的疾病。目前,对于副溶血性弧菌的预防和控制的主要方法是使用抗生素,虽然抗生素使用方便,广谱抑菌,见效快,但是,抗生素的滥用也导致了细菌的严重耐药性,在副溶血性弧菌所造成的疾病防治中的效果愈发下降,因此,急需开发新型抗副溶血性弧菌的药物。
噬菌体是一种细菌病毒,在自然界中具有较高的丰度,具有宿主专一性强,不会造成微生态环境的破坏。噬菌体侵染宿主菌后,裂解时会释放出新的噬菌体,而少量的噬菌体就能起到防治的效果。副溶血性弧菌噬菌体的作用靶点在原核微生物的细胞壁,而对于动植物细胞上没有相应的作用位点,增加了噬菌体使用的安全性。并且噬菌体和病原菌可以共同进化,防止细菌耐药性的出现。因此,利用噬菌体来解决细菌具有广阔前景的生物防治途径。
本发明在于响应国家全面禁抗的号令,解决细菌耐药性,抗生素滥用的问题,提供一株治疗副溶血弧菌的且安全可靠的裂解性副溶血弧菌噬菌体,扩充了噬菌体储备库为开发治疗副溶血弧菌的抗生素替代品奠定了基础。
发明内容
本发明的目的在于提供一种专一性裂解性副溶血性弧菌噬菌体RDP-VP-21010,以替代抗生素在防治水产养殖中的应用,并解决水产动物发病急而治疗不及时,且因致病菌耐药性而导致用药无效等问题。
为实现上述目的,本发明采用如下技术方案:一株裂解性副溶血性弧菌噬菌体RDP-VP-21010,透过电镜观察该噬菌体头部呈正六边形,头部直径约为56nm,尾部长度为103nm,保藏编号为CGMCC No.22481。
本发明利用双层琼脂平板法从养殖淤泥中分离到一株副溶血弧菌噬菌体,其基因组全长为50642bp,该噬菌体在pH为5-10中作用6小时,其效价较高,可在2.2×1010pfu/mL之上,具有明显的嗜碱性。在pH为12时,效价可达到106pfu/mL,在pH为4时,效价仍可达到107pfu/mL,可见该噬菌体的酸碱耐受范围比较宽。该噬菌体的最适温度在37℃,效价能达到1.7×1012pfu/mL,该噬菌体在﹣80℃的条件下可长期保存,效价维持稳定。该噬菌体可特异性地裂解本实验室从青岛即墨养殖户取得的患病虾病灶处分离的致病性副溶血性弧菌BV-21002,其最佳感染复数为1:1。将该噬菌体命名为副溶血性弧菌噬菌体RDP-VP-21010,该噬菌体于2021年06月03日保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏地址为:北京市朝阳区北辰西路1号院3号,保藏编号CGMCC No.22481。
本发明还提供了所述的副溶血性弧菌噬菌体RDP-VP-21010在制备抗副溶血弧菌制剂中的应用。
本发明还提供了所述的副溶血性弧菌噬菌体RDP-VP-21010在制备抗副溶血弧菌BV-21002药物中的应用。
本发明的另一目的在于提供该副溶血性弧菌噬菌体RDP-VP-21010在制备预防和或治疗副溶血弧菌引起的疾病的药物中的用途。
本发明的另一目的在于提供该副溶血性弧菌噬菌体RDP-VP-21010在制备预防和或治疗副溶血性弧菌引起的疾病的饲料中的用途。
本发明的另一个目的是提供一种用于防治副溶血弧菌感染的杀菌组合物,包括有效量的副溶血性弧菌噬菌RDP-VP-21010。
本发明的另一目的在于提供一种副溶血弧菌噬菌体的应用,该目的是通过将副溶血性弧菌噬菌RDP-VP-21010用作为由副溶血弧菌引起的细菌感染提供潜在的治疗药物或保健品来实现的。
本发明的另一目的在于提供一种预防和或治疗副溶血弧菌引起的疾病的添加剂,该目的是通过其有效成分包括副溶血性弧菌噬菌RDP-VP-21010来实现的。
本发明还提供了一种清洁剂或消毒剂,包括副溶血性弧菌噬菌RDP-VP-21010。
本发明的有益效果体现在:
(1)本发明发现并分离了一株致病性副溶血性弧菌BV-21002,并以该致病性副溶血性弧菌BV-21002为宿主分离得到了副溶血弧菌噬菌体RDP-VP-21010,该噬菌体RDP-VP-21010对水产养殖环境中致病性副溶血性弧菌BV-21002具有强裂解效用,为工业化生产噬菌体用于水产养殖环境中致病性副溶血性弧菌BV-21002的防治提供了噬菌体来源;
(2)本发明所提供的副溶血弧菌噬菌体RDP-VP-21010是从自然界中分离的烈性噬菌体供试噬菌体不含毒力基因或不良基因;本发明未对供试噬菌体进行任何遗传修饰;
(3)本发明提供的副溶血性弧菌噬菌体RDP-VP-21010,当水体中加入副溶血弧菌噬菌体RDP-VP-21010后,8小时内有明显的裂解效果,具有较强的裂解活性,表现出了良好的应用前景。
(4)本发明提供的副溶血性弧菌噬菌体RDP-VP-21010,在pH为5-10中作用6小时,其效价较高,可在2.2×1010pfu/mL之上,具有明显的嗜碱性。在pH为12时,效价可达到106pfu/mL,在pH为4时,效价仍可达到107pfu/mL,可见该噬菌体的酸碱耐受范围比较宽,表现出了较高的温度耐受性和较宽的酸碱耐受范围,效价较高,安全有效;
(5)本发明提供的副溶血性弧菌噬菌体RDP-VP-21010在防控水产动物致病性副溶血性弧菌中具有良好的应用前景。
(6)本发明所提供的副溶血性弧菌噬菌体RDP-VP-21010可作为应用于环境消毒的各种产品的有效成分。
(7)本发明副溶血性弧菌噬菌体RDP-VP-21010可作为应用于环境消毒的各种产品的有效成分,例如包括但不限于以液体浸泡、喷洒、与含水性载体联合使用等形式对配水系统、医疗设施、养殖业设施、公共及私人设施或其他环境表面进行消毒去污,可有效控制目标细菌的生长及活性。所述液体浸泡、喷洒形式包括但不限于洗涤剂、消毒剂、去污剂等;所述含水性载体包括但不限于磷酸盐缓冲液、LB培养基、氯游离水等。
(6)本发明的副溶血性弧菌噬菌体RDP-VP-21010,可以有效杀灭海产表面的副溶血性弧菌并防治海产在保存过程中副溶血性弧菌的污染,可作为海产食品防护的各种产品的有效成分。本发明包括但不限于以液体浸泡、喷洒、与合成组分联合使用等形式对由副溶血弧菌侵染所导致的食品腐坏进行预防,尤其适用于熟食或不宜灭菌的食品。所述液体浸泡、喷洒形式包括但不限于食品除菌剂、食品消毒剂、食品防腐剂等;本发明中合成组分包括但不限于苯甲酸、苯甲酸钠、山梨酸、山梨酸钾、丙酸钙等。
(7)本发明的副溶血性弧菌噬菌体RDP-VP-21010,可用于制备组合物,试剂,或试剂盒;可应用于副溶血性弧菌的快速检测,其包括但不限于以试纸、试剂盒等形式对目标样本中的副溶血性弧菌进行检测,或对临床样本中的目标致病菌进行筛选,可有效确保检测的灵敏度;
(8)本发明的副溶血性弧菌噬菌体RDP-VP-21010可以单独或混合使用,用作为生物杀菌剂,饲料添加剂,或因副溶血性弧菌引起的细菌感染的治疗药物、保健品及医疗器械;副溶血性弧菌噬菌体RDP-VP-21010可应用于治疗或预防由副溶血弧菌引起的感染性疾病;

具体实施方式
下面结合附图和实施例对本发明做进一步说明。以下实施例将有助于本领域的技术人员进一步理解本发明,但不以任何形式限制本发明。应当指出的是,对本领域的普通技术人员来说,在不脱离本发明构思的前提下,还可以做出若干变形和改进。这些都属于本发明的保护范围。
本发明使用从青岛即墨一养殖场的患病虾病灶处分离副溶血弧菌BV-21002,并通过实验确定了BV-21002的致病性和耐药性,以BV-21002为宿主,从养殖淤泥中分离得到一株裂解性副溶血弧菌噬菌体,并通过全基因组序列确定了RDP-VP-21010的基因型。
实施例1病原菌及噬菌体的分离及鉴定
(1)致病性副溶血弧菌BV-21002的分离及其鉴定
从发病对虾的鳃取样,采用无菌操作的方法,在TCBS培养基上涂布,37℃培养18-24h后,在TCBS琼脂平板上呈现边缘整齐的圆形绿色菌落,直径约2~3mm;然后挑取典型菌落在科玛嘉弧菌显色培养基上划线,37℃培养18-24h后,在科玛嘉弧菌显色培养基上呈现紫色菌落,挑取典型菌落继续划线纯化3次,然后挑取单菌落接种于5mL的2216E液体培养基中,37℃,200rpm振荡培养6h,得到均匀浑浊的细菌悬浮液。再通过16sRNA分子鉴定,确定为副溶血弧菌,将其中一株命名为BV-21002,保存于﹣80℃冰箱。
(2)噬菌体分离及鉴定
本发明试验用淤泥样品为2021年从青岛即墨养殖户水底淤泥中采集,作为噬菌体分离的样品。
样品处理:取适量淤泥浸泡于20mL 2216E液体中,10000rpm离心10分钟,除去较大杂质及大部分细菌后,再用0.22μm微孔滤膜过滤除菌。
制备混合菌悬液:取2mL滤液和1mL宿主菌菌悬液,一起加入27mL高压灭菌后的2216E液体培养基中,然后置37℃培养箱中,过夜培养。培养后取3mL,10000rpm离心5分钟,再用0.22μm微孔滤器过滤除菌,收集滤液备用,滤液即为拟含有噬菌体的原液。
噬菌体分离:采用双平板法进行噬菌体的分离,取混合菌悬液滤出液0.1mL和副溶血弧菌菌悬液0.1mL混合均匀后,加入加热至45-54℃3mL4%的2216E琼脂中,然后铺双平板,放于37℃培养箱培养6-8h后,观察结果,如果有噬菌体,那么平板上会有透明的噬菌斑。挑取噬菌斑于1mL3%盐水中浸泡10min后,取0.1mL浸出液和0.1mL菌悬液,加入加热至45-54℃3mL的2216E琼脂中,铺双平板,37℃培养箱培养4-6h纯化,照此步骤纯化2-3次,至噬菌斑大小均匀为止,由此得到一株噬菌体,并将其命名为RDP-VP-21010。RDP-VP-21010在双平板形成的噬菌斑直径在4mm-5mm,圆形透明,有晕圈。
(3)菌体效价的测定
将纯化好的噬菌体铺双层平板,然后取单个噬菌斑,加入2216E液体培养基中,同时加入宿主菌悬液,将噬菌体增殖后,再将增殖液10000rpm离心5分钟后再用0.22μm微孔过滤器过滤,将滤液做10倍梯度稀释至10-8倍,取最后3个稀释度的噬菌体稀释液0.1mL和宿主菌悬液0.1mL,铺双层平板,测定噬菌体效价。噬菌体效价(pfu/mL)=噬菌斑数*稀释倍数÷0.1。结果表明,其噬菌斑形态、大小和清晰度一致,有典型的裂解性噬菌体特征;该噬菌体的效价可达10^12pfu/mL以上。
(4)噬菌体的保藏
将噬菌体增殖液与60%的甘油按照1:1的比例混合后,于液氮中保存。
实施例2噬菌体的电镜观察
取20μL含噬菌体粗制颗粒的液体滴于铜网上,自然沉淀15min,并用滤纸从侧面吸去多余液体,加一滴2%的磷钨酸(PTA)于铜网上对噬菌体染色10min,然后用滤纸从侧面吸去染色液,待样品干燥后用电子显微镜观察噬菌体形态。
由图1可知,该噬菌体RDP-VP-21010为长尾科噬菌体,头部呈正六边形,头部直径约为56nm,尾部长度为103nm。
实施例3噬菌体基因组测序
通过对单株噬菌体的富集培养后,在4℃条件下8000g离心15分钟,加入10%PEG8000和0.5M NaCl静置过夜,再加入等量氯仿混匀,静置分层后5000g下离心10min,去除氯仿层和PEG层后,加入限制性核酸内切酶消化处理,并采用梯度密度氯化铯条件下悬浮噬菌体,后期采用TM缓冲液透析3次,每次30min,送华大基因生物测序公司进行全基因组测序。结果如序列表所示。
实施例4:噬菌体pH稳定性的测定
用稀盐酸和稀NaOH溶液调节生理盐水的酸碱度,配成pH值为2、3、4、5、6、7、8、9、10、11、12的缓冲液,再用配置好的缓冲溶液将噬菌体稀释成1×1010pfu/mL,将稀释液在37℃下,水浴1h,用生理盐水10倍比稀释后铺双平板;37℃倒置培养4-6h后计数。
由图2可知,在不同pH条件下培养1h后,噬菌体RDP-VP-21010在pH4.0-11.0范围能维持较好的活性,噬菌体滴度没有明显变化,而且在pH为8.0时,噬菌体效价最高,达5×109pfu/ml。随着pH值的升高或降低,噬菌体的滴度下降明显。上述结果表明,噬菌体RDP-VP-21010能耐受弱酸和强碱条件,具有良好的酸碱耐受性。
实施例5:最佳感染复数的测定
按照感染复数为10:1、1:1、1:10、1:100、1:1000、1:10000的比例将噬菌体增殖液和宿主加入2216E中,并且确保培养体系的总体积相同。于37℃200rpm振荡培养8h后,常温下12000r/min离心5min,取上清液铺双平板测定其效价。结果如表1所示。

由表1数据可知,当感染复数为1:1时,培养8h后,增殖液较为清澈,效价最高,表明本发明副溶血弧菌噬菌体RDP-VP-21010的最佳感染复数为1:1。
实施例6:噬菌体对宿主裂解曲线
将培养6h的宿主菌悬液以1:1的比例接种到100mL 2216E培养基中,37℃200rpm振荡培养2h(OD约0.45,约6.0×108cfu/mL)将菌悬液分装在4个灭菌的锥形瓶中,以最佳感染复数,向其中3个加入噬菌体增殖液0.2mL(约4.0×1011pfu/mL,经0.22μm膜过滤),同时以不加噬菌体的作为对照,取其平均值,结果如图3所示。
由图3可知,副溶血弧菌噬菌体RDP-VP-21010加入到宿主菌BV-21002菌悬液中,作用1h后出现一定的裂解效果,约1h后,OD600值开始下降,持续作用至8.5h,9h后混合菌液开始变得浑浊,OD值逐渐升高,至12h达到平衡,但仍较宿主菌的OD值低。由于前期,噬菌体处于对宿主菌的吸附、侵染阶段,只有少部分噬菌体开始裂解宿主,因此,其裂解宿主的量远远小于宿主的增长量,因此,OD600在前1h,依旧保持一定的上升趋势,伴随着噬菌体数量的指数增加,在1h后,其裂解宿主数量大于宿主的增殖速度,所以OD600值开始逐渐降低,混合菌液达最澄清。持续作用9小时后,混合菌液出现返浊,原敏感菌被噬菌体裂解,被突变的耐受菌所取代的一种菌群交替。
实施例7:噬菌体热稳定性的测定
将噬菌体原液分装到EP管中,分别在40℃、50℃、60℃和70℃的条件下孵育30min,然后用生理盐水10倍比稀释后,铺双平板测定其效价。结果如图4所示。
由图4可知,噬菌体在40℃~60℃以内活性保持稳定,效价在1012pfu/mL以上;在70℃条件下作用30min后,效价仍可达到7.4*10^4pfu/mL,表明噬菌体具有一定的温度耐受性。
实施例8:稳定性遗传实验
将RDP-VP-21010连续传代15次,每代培养基中加入等量的RDP-VP-21010和BV-21002,每代培养时间6h,然后铺双平板,测定其效价。结果如图5所示。
由图5可知,将RDP-VP-21010连续传15代后,测定其效价,其效价稳定在1011pfu/mL以上,说明该噬菌体具有较好的遗传稳定性,适合工业化生产。
实施例9:裂解率实验
无菌条件下,分别取1mL样品和1mL宿主菌菌液(1×106CFU/mL),37℃孵育15分钟,混匀后用生理盐水稀释至10-1-10-3个梯度,每个梯度取100μL涂布于2216E琼脂平板上,置于37℃,培养24小时,每个梯度重复两次。同时取1mL3%盐水和1mL宿主菌菌液(1×106CFU/mL)作为空白对照,重复以上步骤。选取30-300个菌落数的平板进行计数。该试验重复3次,取其平均值。噬菌体裂解率=(1-处理组菌落数/对照组菌落数)×100%。
经计算,RDP-VP-21010的裂解率达93.1%,对宿主有较好的裂解效果,适合在养殖过程中使用。
实施例10:RTD实验
在平板上划分区域,在每一区域中心处滴加一滴宿主菌(约10μL),晾干,将不同稀释度的一滴噬菌体液滴加到宿主菌的斑点上,晾干,37℃培养过夜。根据宿主菌生长状况,凡是不能连片生长的,称为CL浓度(CL浓度是可以完全性裂解环境中宿主菌的浓度,可以用于指导生产实践的稀释方案),结果如图6所示。
由图6可知,RDP-VP-21010在稀释105倍后对宿主菌仍有较好的裂解效果。
实施例11:噬菌体大规模生产的方法
该工艺流程简便,可靠有效,有利于工业化生产。
具体步骤
1、种子制备
(1)宿主种子制备:无菌条件下,挑取单菌落划线于2216E琼脂培养基,37℃培养过夜,
然后挑取培养好的单菌落于5mL 2216E液体培养基中,37℃,180rpm震荡培养6h后,以1%的接种量接种于2216E液体培养基中,37℃,180rpm震荡培养6h。做好的种子液置于4℃冰箱保存备用。
(2)噬菌体种子制备:将保存的噬菌体种子适当稀释后,取100μL与100μL宿主菌置于上层培养基中,倒双层平板,37℃,培养4-6h,然后用无菌涂布棒将其刮取,将其在10000rpm,5min离心,然后过0.22μm的滤膜除菌,将收集到的过滤液置于4℃冰箱中保存备用。
噬菌体和宿主分别以1%和5%的比例接种于2216E液体培养基中,37℃,180rpm,培养6-8h,至增殖液澄清出现大量絮状物为止。
2、噬菌体增殖:在种子罐中接种2%的宿主菌,培养2-3h后,OD600在0.6-0.8,再以1%的接种量接种噬菌体种子,培养6-8h,至溶氧反升,pH趋于稳定为止。
在种子罐中接种2%的宿主菌,培养4-5h,待溶氧开始缓慢下降为止。
将准备好的宿主菌以5%的接种量,接种到大发酵罐中,培养2-3h,待溶氧开始缓慢下降,然后再以5%的接种量接入噬菌体种子,将通气量提高至1vvm,转速提高至150rpm,培养8-10h,待溶氧反升,pH升高,结束增殖。
注:发酵罐参数:种子罐200-300rpm,37℃,通气量0.6-1vvm。发酵罐:120-150pm,37℃,通气量0.6-1vvm。
培养基成分:酵母浸粉0.5%,蛋白胨1%,牛肉膏0.1%,甘油0.5%,氯化钠0.5%,磷酸二氢钾0.1%,磷酸氢二钾0.1%,氯化钙20ppm,氯化镁20ppm。
3、噬菌体后处理工艺
(1)过滤:分别采用采用800nm和500nm的陶瓷膜进行过滤,以去除细胞碎片和未裂解的宿主菌,过滤压力小于0.4mpa,减小剪切力对噬菌体的损伤。
(2)浓缩:采用中空纤维膜进行浓缩,以提高噬菌体效价。
(3)稳定剂:加入5%的甘油,海藻糖3%,聚山梨醇酯3%,磷酸二氢钠3%,磷酸钠2%,搅拌至完全溶解。
(4)除菌:采用三级过滤(玻璃纤维0.45μm,玻璃纤维0.2μm,聚醚砜0.22μm)除菌。
(5)将过滤除菌的噬菌体液体置于4℃条件下储存。
应当指出的是,具体实施方式只是本发明比较有代表性的例子,显然本发明的技术方案不限于上述实施例,还可以有很多变形。本领域的普通技术人员,以本发明所明确公开的或根据文件的书面描述毫无异议的得到的,均应认为是本专利所要保护的范围。

2.根据权利要求1所述的副溶血弧菌噬菌体,其特征在于,透过电镜观察,该副溶血弧菌噬菌体(Vibrio Parahemolyticus phage)RDP-VP-21010头部呈正六边形,头部直径约为56nm,尾部长度为103nm,为长尾科噬菌体。
3.根据权利要求1所述的副溶血弧菌噬菌体,其特征在于,该副溶血弧菌噬菌体(Vibrio Parahemolyticus phage)RDP-VP-21010的最适温度为37℃,效价最高达到1.7*10^12pfu/mL。
4.根据权利要求1所述的副溶血弧菌噬菌体,其特征在于,该副溶血弧菌噬菌体(Vibrio Parahemolyticus phage)RDP-VP-21010对水体中副溶血弧菌具有强裂解作用。
