背景技术
口蹄疫(Foot-and-Mouth Disease,FMD)是一种影响偶蹄动物(偶蹄目Artiodactyla)的病毒性疾病,包括牛、猪、绵羊、山羊、鹿等家畜和各种野生动物,其特征是在嘴巴、鼻子、乳头、蹄子等处形成水泡。口蹄疫具有高度传染性,死亡率高达40%或以上,这使其成为一种具有社会经济影响的疾病。为此,世界动物卫生组织(OIE)将FMD归类为A类疾病并进行管理,在大多数国家,FMD已被定为一类传染病。此外,在过去十年中,几乎每个大陆都发生了FMD疫情,造成了严重的经济损失。

根据VP1编码区的序列,FMDV可分为O、A、C、SAT1、SAT2、SAT3和亚洲1(Asia 1)7个血清型,VP1在结构蛋白基因区中具有最多的突变,并且对于确定免疫原性(如中和抗体的形成)很重要。此外,根据遗传特征,每个血清型可分为多达约80个亚型。
在亚洲(包括韩国、东南亚)和欧洲,O血清型的FMD最为常见,因此针对O血清型的FMD疫苗已被积极研究和接种。然而,迄今为止开发的针对O血清型的FMD疫苗并不能有效地防御最近在包括韩国在内的亚洲出现的口蹄疫病毒。因此,迫切需要开发一种针对O血清型FMDV的新型疫苗。
另一方面,由于主要发生在韩国的口蹄疫是O血清型,因此对A血清型病毒的准备不足。然而,2017年2月在韩国延川市发生的口蹄疫为A血清型,这使得情况变得严重。
此外,已知Asia 1血清型在口蹄疫血清型中相对较弱,但自2005年中国出现Asia1血清型口蹄疫以来,Asia 1型在东亚地区开始不断出现(Hammond J.2009.OIE/FAO世界参考实验室报告,1月-3月2009,口蹄疫.世界参考实验室,皮尔布赖特:1-33)。这些属于基因型V的病毒,被证实与在印度分离的病毒有遗传关系(Jong hyun Park等,2008a,"世界口蹄疫分子流行病学分析及近期分布(Molecular epidemiological analysis and recentdistribution of foot-and-mouth disease in the world)",预防兽医学杂志)。考虑到韩国周边地区口蹄疫的现状,除了A血清型和O血清型外,还需要为Asia1血清型进入韩国的情况做好准备。
针对特定血清型病毒的疫苗不具有针对其他血清型病毒的免疫力,并且存在属于同一血清型的两个毒株的DNA序列差异高达总基因的30%的情况。因此,由于FMDV有多种血清型和亚型,疫苗应针对每种血清型单独应用。此外,由于一种血清型存在许多突变,因此很难开发出有效的疫苗。而且,即使接种口蹄疫疫苗,也存在免疫力只能维持几个月的缺点。

技术方案
为了实现上述目的,本发明一方面提供了一种多肽,其包含SEQ ID NO:1的氨基酸序列或与其具有90%以上同一性的氨基酸序列。SEQ ID NO:1的氨基酸序列或与其具有90%以上序列同一性的氨基酸序列可以是对应于O血清型口蹄疫病毒的VP1区的序列。

作为一个实例,在二价或三价疫苗中,A血清型FMDV的VP1区包含SEQ ID NO:88、89或90的核苷酸序列。SEQ ID NOs:88、89和90所示的核苷酸序列分别是对应于A血清型FMDV的VP1区的重组序列。特别地,SEQ ID NO:89的核苷酸序列是SEQ ID NO:88的核苷酸序列用猪密码子优化后的序列。

术语“疫苗”是指含有抗原的生物制剂,可使受试者免疫。即,疫苗是指免疫原性或抗原性物质,例如通过注射或口服给予人或动物以预防感染,从而为受试者提供免疫。
该疫苗可以是DNA疫苗。术语“DNA疫苗”是指通过人为地复制病原体、病毒等的某些基因并对其进行施用而引起免疫应答的疫苗。这些DNA疫苗与现有蛋白质疫苗相比具有许多优势,包括如下优势:(i)DNA疫苗可以仅利用纯目标病原体抗原的遗传信息即可合成,因此无需处理有害病原体本身,(ii)由于仅使用了一些诱导毒性所需的基因,因此DNA疫苗在施用于受试者时不会显示任何意外毒性,(iii)由于质粒DNA的结构简单,可以通过对突然发生的各种传染病的快速反应来开发疫苗。
用于FMD的疫苗组合物可以对FMDV,特别是O血清型FMDV具有保护性免疫。
本发明的疫苗组合物可以包含兽医学上可接受的载体。术语“兽医学上可接受的载体”包括任何和所有溶剂、分散介质、涂层、免疫佐剂、稳定剂、稀释剂、防腐剂、抗菌剂、抗真菌剂、等渗剂、吸附延迟剂等。可包含在疫苗组合物中的载体、赋形剂和稀释剂的实例包括乳糖、葡萄糖、蔗糖、山梨糖醇、甘露糖醇、木糖醇、麦芽糖醇、淀粉、甘油、阿拉伯胶、海藻酸盐、明胶、磷酸钙、硅酸钙、纤维素、甲基纤维素、微晶纤维素、聚乙烯吡咯烷酮、水、羟基苯甲酸甲酯、羟基苯甲酸丙酯、滑石粉、硬脂酸镁和矿物油。另外,本发明的疫苗组合物可以按照常规方法制成口服剂型,例如散剂、颗粒剂、片剂、胶囊剂、混悬剂、乳剂、糖浆剂、气雾剂等或无菌注射液。当配制疫苗组合物时,可以使用任何稀释剂或赋形剂制备组合物,例如常用的填充剂、增量剂、粘合剂、润湿剂、崩解剂、表面活性剂等。特别优选地,可以使用聚-L-谷氨酸帮助稳定。
疫苗组合物可以通过多种形式注射到受试者体内。“注射”可以通过选自皮下注射、肌肉注射、皮内注射、腹腔注射、鼻内给药、静脉内注射、经皮给药和口服给药所组成的组中的任一种来进行。
疫苗组合物可以包含一种或多种佐剂以改善或增强免疫应答。合适的佐剂包括肽、氢氧化铝、磷酸铝、氧化铝、矿物油如Marcol 52、植物油、乳化剂或表面活性物质如溶血卵磷脂、聚阳离子、聚阴离子等。
本发明的一个方面提供了一种针对两种或更多种血清型的FMDV的多价疫苗组合物。
疫苗组合物对其具有保护性免疫的FMDV可以是两种或更多种,例如,对选自O、A、C、SAT1、SAT2、SAT3和Asia1血清型FMDV中的2、3、4、5、6或7种。作为一个具体的实例,FMDV可以是O和A血清型,在这种情况下,疫苗组合物作为二价疫苗发挥作用。作为另一个实例,FMDV可能是O、A和Asia1血清型,在这种情况下,疫苗组合物作为三价疫苗发挥作用。

本发明的效果
本发明所述的疫苗组合物可以提供对FMDV显示出高免疫原性的DNA疫苗组合物。如果施用本发明的疫苗组合物,则在接种疫苗的个体中产生的针对FMDV的抗体水平非常高,因此有效地表现出免疫应答。特别是对O血清型FMDV具有极好的保护性免疫应答,特别是最近在亚洲出现的O血清型FMDV。
因此,当对猪或牛等个体给药时,可以实现极好的预防FMD的效果。
此外,本发明所述的疫苗组合物可以与其他血清型FMD的疫苗、抑制剂或用于治疗或预防其他血清型FMD药剂组合使用。或者,它可以用作多价疫苗以预防两种或多种血清型FMD。

实施例1.FMDV血清型O的序列分析
WRLFMD(世界口蹄疫参考实验室)网站,ME报告中(分子流行病学报告)对最近在亚洲出现的O血清型FMDV的VP1区进行了研究。
FMDV的VP1区在FMDV的P1结构蛋白基因区中突变最多,是决定免疫原性的重要位点,例如中和抗体的形成。
该研究在21个国家和地区进行,包括孟加拉国、不丹、柬埔寨、中国、印度尼西亚、印度、日本、老挝、马来西亚、蒙古、缅甸、尼泊尔、菲律宾、俄罗斯、新加坡、韩国、斯里兰卡、泰国和越南。

实施例2.新型DNA疫苗的制备
通过Clustal/muscle对SEQ ID NOs:4至84的81个氨基酸序列进行比对。根据比对结果,推导出VP1的共有(consensus)序列。将衍生的共有序列与常规的O血清型FMD DNA疫苗pGX9014的序列进行比较,结果证实存在5个氨基酸错配。
在现有O血清型FMD DNA疫苗pGX9014的VP1区域中,替换了与共有序列不匹配的五个氨基酸。图1显示通过替换得到的VP1的氨基酸序列(SEQ ID NO:1),图2显示了通过替换获得的VP1的核苷酸序列(SEQ ID NO:2)。
使用由SEQ ID NO:2的核苷酸序列所示的多核苷酸制备了一种新型FMD血清型O疫苗,整体示意图如图3所示。如图3所示,制备的重组核苷酸中,Msc I位点、VP3 C-末端位点、弗林蛋白酶切割位点、由SEQ ID NO:2的核苷酸序列表示的重组VP1位点、弗林蛋白酶切割位点、2A和Xho I位点顺序连接。所制备的重组核苷酸的序列和位点如图4所示,由SEQ IDNO:3表示。包含SEQ ID NO:3的核苷酸的DNA质粒被命名为“PLS-FMD-O”。
将具有PLS-FMD-O的大肠杆菌接种于2.5L的LB培养基中,然后在37℃下过夜培养。然后使用EndoFree Plasmid Giga试剂盒(QIAGEN,Cat#12391)从培养的大肠杆菌中提取质粒。将提取的质粒溶解在无内毒素水中并储存在-70℃下。然后将储存的质粒用于抗体形成实验,以确认动物的疫苗接种效果。
实施例3.新型DNA疫苗的抗体形成效应
3.1 DNA疫苗接种
将15只C57BL/6小鼠(8周龄,雌性)分成三组,每组5只,如下所示,每组每隔两周接种本发明疫苗、常规疫苗和模拟质粒3次。使用电穿孔装置(cellectra 2000)进行接种,DNA疫苗以30μg/小鼠的浓度给药。
·第1组(对照组)-模拟质粒DNA
·第2组(对照组)-O血清型FMD DNA(pGX1414,常规疫苗)
·第3组(实验组)-O血清型FMD DNA(PLS-FMD-O,本发明疫苗)
在给药前(第0周)、第二次给药后(第4周)和第三次给药后(第6周)使用眼眶后采血法采集血液。从收集的血液中分离血清并在-80℃下储存。将储存的血清溶解并进行SPC(固相竞争)ELISA分析。
3.2.测量抗体形成-SPC ELISA分析
根据试剂盒制造商提供的实验方案,使用PrioCHECK A血清型FMDV ELISA试剂盒(Prionics lelystad B.V/荷兰)进行SPC ELISA分析。
结果如图5所示。图5中可见,本发明的疫苗PLS-FMD-O具有比现有疫苗pGX1414更高的PI(抑制百分比)值。此处,PI值越高,产生的针对O血清型FMDV的特异性抗体就越多。与现有疫苗pGX1414相比,本发明的疫苗PLS-FMD-O在同一时间产生的抗体明显更高。
如上所述,FMD具有高度传染性,并且由于FMD病毒在出现任何临床症状之前就已经从体内排出,因此很容易发生动物间的传播。而且FMD的潜伏期非常短。因此,为了发挥作为FMD疫苗的有效功能,不仅要抑制FMD病毒的效果优异,而且要快速产生针对FMD病毒的抗体。在施用根据本发明的疫苗的情况下,该疫苗不仅在抑制FMD病毒方面非常出色,而且产生抗体的速度也明显快于传统疫苗,因此有可能在早期阶段抑制FMD的传染。因此,根据本发明的疫苗是一种非常有效的针对FMD的疫苗。
实施例4.新型DNA二价疫苗的制备
使用先前制备的重组序列制备针对A和O血清型FMD的二价DNA疫苗。
图6显示了制备二价疫苗的示例。在二价疫苗中,A血清型FMD的VP1区可能包含SEQID NO:88、89或90的核苷酸序列,O血清型FMD的VP1区可能包含SEQ ID NO:2的核苷酸序列。
使用SEQ ID NO:89的核苷酸序列作为A血清型FMD的VP1区,和SEQ ID NO:2的核苷酸序列作为O血清型FMD的VP1区,制备一种抗A和O血清型FMD的二价DNA疫苗,将其命名为“PLS-FMD-A,O”。
将具有PLS-FMD-A,O的大肠杆菌接种至2.5L的LB培养基中,在37℃下过夜培养。然后使用EndoFree Plasmid Giga试剂盒(QIAGEN,Cat#12391)从培养的大肠杆菌中提取质粒。将提取的质粒溶解在无内毒素水中并储存在-70℃下。然后将储存的质粒用于抗体形成实验,以确认动物的疫苗接种效果。
实施例5.新型DNA二价疫苗的抗体形成效应
5.1 DNA疫苗接种

图8显示了制备三价疫苗的示例。在三价疫苗中,A血清型FMD的VP1区可以包含SEQID NO:88、89或90的核苷酸序列,O血清型FMD的VP1区可以包含SEQ ID NO:2的核苷酸序列,Asia1血清型FMD的VP1区可以包含常规已知的核苷酸序列。
使用SEQ ID NO:89的核苷酸序列作为A血清型FMD的VP1区,SEQ ID NO:2的核苷酸序列作为O血清型FMD的VP1区,以及,SEQ ID NO:93的核苷酸序列作为Asia1血清型FMD的VP1区,制备针对FMD的A、O和Asia1血清型的三价DNA疫苗,将其命名为“PLS-FMD-A,O,Asia1”。作为Asia1血清型FMD的VP1区的SEQ ID NO:93的核苷酸序列是美国INOVIO提供的pGX9010中包含的Asia1血清型FMD的VP1区。
以与实施例4相同的方式制备PLS-FMD-A,O,Asia1质粒。
实施例7.新型DNA三价疫苗的抗体形成效果
7.1.DNA疫苗接种
实施例7的制备方法与实施例5相同,但第1组至第3组设置如下:

因此,根据本发明的重组O血清型疫苗已被证明既可作为单价疫苗又可作为多价疫苗。

2.一种多肽,所述多肽包含SEQ ID NO:1的氨基酸序列或包含与SEQ ID NO:1的氨基酸序列具有90%以上同一性的氨基酸序列,以及选自口蹄疫病毒的VP2、VP3和VP4蛋白的氨基酸序列中的至少一种。
3.根据权利要求2所述的多肽,其中,所述口蹄疫病毒的VP2、VP3和VP4蛋白来源于O血清型的口蹄疫病毒。
4.一种多核苷酸,所述多核苷酸包含编码SEQ ID NO:1的氨基酸序列或与SEQ ID NO:1的氨基酸序列具有90%以上同一性的氨基酸序列的核苷酸序列,或者SEQ ID NO:2的核苷酸序列或与SEQ ID NO:2的核苷酸序列具有90%以上同一性的核苷酸序列。
5.一种包含权利要求4所述的多核苷酸的质粒。
6.根据权利要求5所述的质粒,所述质粒包含SEQ ID NO:3的核苷酸序列。
7.根据权利要求5所述的质粒,所述质粒还包含编码口蹄疫病毒的VP2、VP3和VP4蛋白中的至少一种的核苷酸序列。
8.根据权利要求7所述的质粒,其中,所述口蹄疫病毒的VP2、VP3和VP4蛋白来自于O血清型口蹄疫病毒。
9.一种用于口蹄疫的疫苗组合物,所述疫苗组合物包含权利要求1-3中任一项所述的多肽、权利要求4所述的多核苷酸或权利要求5-8中任一项所述的质粒。
10.根据权利要求9所述的疫苗组合物,其中,所述疫苗是单价疫苗或多价疫苗。
11.根据权利要求9所述的疫苗组合物,其中,所述多核苷酸还包含口蹄疫病毒的VP1蛋白的氨基酸序列中的至少一种,所述口蹄疫病毒选自A、C、SAT1、SAT2、SAT3和Asia1血清型口蹄疫病毒。
12.根据权利要求9所述的疫苗组合物,其中,所述多核苷酸还包含口蹄疫病毒的VP2、VP3和VP4蛋白的氨基酸序列中的至少一种,所述口蹄疫病毒选自A、C、SAT1、SAT2、SAT3和Asia1血清型口蹄疫病毒。
13.一种在受试者中引发对FMDV的免疫应答的方法,包括:向受试者施用权利要求9所述的疫苗组合物。
14.根据权利要求13所述的方法,其中,所述施用通过电穿孔进行。
15.一种在受试者中预防口蹄疫病毒的方法,包括:向受试者施用权利要求9所述的疫苗组合物。
16.根据权利要求15所述的方法,其中,所述施用通过电穿孔进行。
17.一种用于预防口蹄疫病毒的药物组合物,所述药物组合物包含权利要求1-3中任一项所述的多肽、权利要求4所述的多核苷酸或权利要求5-8中任一项所述的质粒。
