Abstract
本发明公开了一种多糖及其美白用途。本发明提供的多糖为一种蓝花参种子多糖,多糖含量为(94.2±1.9)%。酪氨酸酶抑制试验显示该蓝花参种子多糖为一种有效的酪氨酸酶抑制剂,可以抑制酪氨酸酶活性,进而抑制黑色素合成,具有美白活性。

技术领域
本发明属于化妆品领域,涉及化妆品活性成分的开发,具体涉及一种多糖及其美白用途。
背景技术
以植物提取物作为活性成分配制的化妆品与传统化妆品相比,具有很多优点,诸如:克服了传统化妆品依赖化学合成品的缺点,使产品的安全性能更高;天然组分更容易被皮肤吸收,使产品的作用效果更显著;功能性更突出等。因此,植物提取物应用于化妆品中是市场发展的必然趋势。另外,植物提取物具有许多明显的优势:来源广、作用机理针对性强。特别是随着现代提取技术和分析技术的进步,其功效成分及相关作用机理已逐渐被人们发现并证实,在防晒、美白、抗衰老、杀菌等方面有着广泛应用。
多糖是生物体中广泛存在的物质,是一类由醛糖或酮糖通过糖苷键连接而成的天然高分子多聚物,它是生物体内重要的生物大分子,是维持生命活动正常运转的基本物质之一。植物多糖,又称植物多聚糖,是植物细胞代谢产生的聚合度超过10个的聚糖。
以多糖等植物提取物为活性成分的化妆品已经成为化妆品市场的重要组成部分。
发明内容
本发明是为了提供一种多糖及其美白用途。
本发明技术方案如下:
一种多糖,通过如下方法制备:
取蓝花参种子,用沸程60~90℃的石油醚热回流提取脱脂,收集脱脂后的种子,挥干溶剂,再用去离子水加热提取,提取液过滤,滤液减压浓缩,冷却后离心,取上清液;向该上清液中加入4倍体积无水乙醇,混匀静置,离心,收集醇沉物,Sevag法脱蛋白,得总多糖;
用去离子水将总多糖溶解配制成多糖溶液,上样于DEAE-Sepharose Fast Flow柱(内径2.5cm,柱床高度30cm),依次用去离子水、0.1mol/LNaCl溶液洗脱和0.3mol/LNaCl溶液以2mL/min的流速分级洗脱,每种洗脱剂洗脱500mL,收集0.3mol/LNaCl溶液洗脱液,透析、浓缩、冻干,即得。
优选地,所述蓝花参种子干燥粉碎过60目筛。
优选地,加热提取温度为80~90℃。
优选地,离心条件为3000r/min离心15min。
优选地,上样的多糖溶液浓度为20mg/mL。
上述多糖的美白用途。
上述多糖在制备美白化妆品中的应用。
有益技术效果:
1、本发明提供了一种蓝花参种子多糖,多糖含量为(94.2±1.9)%。
2、本发明提供的蓝花参种子多糖为一种有效的酪氨酸酶抑制剂,可以抑制酪氨酸酶活性,进而抑制黑色素合成,具有美白活性。

具体实施方式
下面通过具体的实施例详细介绍本发明实质性内容,但不能以此限定本发明的保护范围。
一、材料
将桔梗科蓝花参属植物蓝花参(Wahlenbergia marginata)的种子干燥粉碎,过60目筛。
β-熊果苷购自上海源叶生物科技有限公司,纯度99%。
二、方法
1、蓝花参多糖的制备
取1kg蓝花参种子,加入8L沸程为60~90℃的石油醚,热回流提取3h,收集脱脂后的种子,挥干溶剂,再用8L去离子水85℃加热提取3h,提取液过滤,滤液减压浓缩至2L,冷却后以3000r/min的转速离心15min,取上清液;向该上清液中加入4倍体积无水乙醇,混匀静置12h,3000r/min的转速离心15min,收集醇沉物,Sevag法脱蛋白,即得总多糖。
用去离子水将总多糖溶解配制成浓度为20mg/mL的多糖溶液,取20mL多糖溶液上样于DEAE-Sepharose Fast Flow柱(内径2.5cm,柱床高度30cm),依次用去离子水、0.1mol/LNaCl溶液洗脱、0.3mol/LNaCl溶液洗脱和0.5mol/LNaCl溶液以2mL/min的流速分级洗脱,每种洗脱剂洗脱500mL,每10mL洗脱液收集1管,根据多糖显色反应结果绘制洗脱曲线(图1)。
合并0.3mol/LNaCl溶液洗脱液,透析、浓缩、冻干,得到蓝花参多糖Ⅰ。合并0.5mol/LNaCl溶液洗脱液,透析、浓缩、冻干,得到蓝花参多糖Ⅱ。
2、酪氨酸酶抑制活性检测
参考文献方法(肉桂酸-香豆素酯类似物的合成及抑制酪氨酸酶活性研究,有机化学,Chin.J.Org.Chem.2020,40)测定蓝花参多糖Ⅰ、蓝花参多糖Ⅱ和阳性对照对酪氨酸酶的抑制率:
将蓝花参多糖Ⅰ、蓝花参多糖Ⅱ和阳性对照分别用50%DMSO水溶液溶解,并稀释成不同浓度的溶液。将130μL磷酸缓冲溶液(50mM,pH=6.8)、10μL酪氨酸酶溶液(终浓度33.3U/mL)和10μL待测多糖或阳性对照混匀后,加入50μL L-Dopa(终浓度0.5mM)混匀,10min后于490nm波长下测定OD值。同时设置对照组,对照组加入10μL溶媒代替多糖或阳性对照溶液。各设5个复孔,计算均值,根据如下公式计算不同浓度多糖或阳性对照对酪氨酸酶的抑制率:
抑制率(%)=(OD对照-OD药物)/OD对照×100%。
3、统计学分析
数据以平均值±SD表示,组间比较采用t检验,P<0.05代表具有显著性差异。
三、结果
1、蓝花参多糖的制备
苯酚-硫酸法测定蓝花参多糖Ⅰ中多糖含量为(94.2±1.9)%,蓝花参多糖Ⅱ中多糖含量为(95.5±1.6)%。SEM图(放大倍数10000×)如图2,其中A为蓝花参多糖ⅠSEM图,B为蓝花参多糖ⅡSEM图,蓝花参多糖Ⅰ、蓝花参多糖Ⅱ呈不规则颗粒状,但颗粒大小分布较为均一。
2、酪氨酸酶抑制活性
蓝花参多糖Ⅰ、蓝花参多糖Ⅱ和阳性对照对酪氨酸酶的抑制率如表1所示,蓝花参多糖Ⅰ具有略强于阳性对照β-熊果苷的酪氨酸酶抑制活性,蓝花参多糖Ⅱ对酪氨酸酶无明显抑制作用。

酪氨酸酶是调控黑色素生成的限速酶,直接影响黑色素的合成。抑制酪氨酸酶的活性可以有效抑制黑色素的合成。目前市面上大多数美白化妆品中均含有酪氨酸酶抑制剂。上述试验结果说明,本发明提供的蓝花参多糖Ⅰ为一种有效的酪氨酸酶抑制剂,可以抑制酪氨酸酶活性,进而抑制黑色素合成,具有美白活性。
Claims (2)
1.一种多糖,其特征在于,通过如下方法制备:
取1kg蓝花参种子,加入8L沸程为60~90℃的石油醚,热回流提取3h,收集脱脂后的种子,挥干溶剂,再用8L去离子水85℃加热提取3h,提取液过滤,滤液减压浓缩至2L,冷却后以3000r/min的转速离心15min,取上清液;向该上清液中加入4倍体积无水乙醇,混匀静置12h,3000r/min的转速离心15min,收集醇沉物,Sevag法脱蛋白,即得总多糖;
用去离子水将总多糖溶解配制成浓度为20mg/mL的多糖溶液,取20mL多糖溶液上样于内径2.5cm、柱床高度30cm的DEAE-Sepharose Fast Flow柱,依次用去离子水、0.1mol/LNaCl溶液、0.3mol/L NaCl溶液和0.5mol/L NaCl溶液以2mL/min的流速分级洗脱,每种洗脱剂洗脱500mL,每10mL洗脱液收集1管,合并0.3mol/L NaCl溶液洗脱液,透析、浓缩、冻干,即得。
2.权利要求1所述的多糖在制备美白化妆品中的应用。
