Abstract

本发明公开了一种姜味草多糖及用于制备美白淡斑化妆品的用途,姜味草多糖通过下述制备方法方法制备:取洗净干燥的姜味草全草,粉碎过筛,加入去离子水常温浸泡提取,收集提取液,过滤,减压浓缩,离心收集上清液,用4倍体积的无水乙醇醇析,沉淀用去离子水复溶至原上清液体积,重复用4倍体积的无水乙醇醇析一次,沉淀依次用乙醇、丙酮交替洗涤,挥干溶剂后复溶于去离子水中,用Sevag法脱蛋白,冷冻干燥即得。本发明提供的姜味草多糖对酪氨酸酶具有较高的抑制作用,具有用于制备美白淡斑化妆品的前景。

一种姜味草多糖及用于制备美白淡斑化妆品的用途
技术领域
本发明属于化学领域,具体涉及一种姜味草多糖及用于制备美白淡斑化妆品的用途。
背景技术
酪氨酸酶抑制剂是目前公认的用于美白类化妆品的美白添加剂,如美白淡斑成分曲酸。因此,加强酪氨酸酶抑制剂的开发可以丰富化妆品中美白淡斑添加剂的种类。
植物多糖是大自然对人类的馈赠,许多植物多糖具有生物活性,如免疫调节、抗肿瘤、降血糖、降血脂、抗辐射、抗菌抗病毒、保护肝脏等保健作用。所以植物多糖早已被广泛运用到医学界、餐饮界等大众生活领域中。将植物多糖应用于化妆品具有安全无刺激的优点。
姜味草(Micromeria biflora benth.)为唇形科姜味草属植物,主要分布于贵州、云南、广西等地,是西南地区布依族、苗族常用草药,含有多糖、多肽和众多小分子化合物。
姜味草全草中多糖的研究极少,没有其对酪氨酸酶抑制作用的活性报道。
发明内容
本发明的目的是为了提供一种姜味草多糖及用于制备美白淡斑化妆品的用途。
本发明目的通过以下技术方案实现:
一种姜味草多糖,通过下述制备方法方法制备:
取洗净干燥的姜味草全草,粉碎过筛,加入去离子水常温浸泡提取,收集提取液,过滤,减压浓缩,离心收集上清液,用4倍体积的无水乙醇醇析,沉淀用去离子水复溶至原上清液体积,重复用4倍体积的无水乙醇醇析一次,沉淀依次用乙醇、丙酮交替洗涤,挥干溶剂后复溶于去离子水中,用Sevag法脱蛋白,冷冻干燥即得。
优选地,粉碎过40目筛。
优选地,按照料液比1kg:15~25L加入去离子水进行常温浸泡提取36~60h。
优选地,按照1kg全草对应1L浓缩液进行减压浓缩。
优选地,减压浓缩后的离心参数为4000r/min条件下离心10min。
上述的姜味草多糖用于制备美白淡斑化妆品的用途。
有益效果:
综合本发明实施例可知,本发明从姜味草全草中制备得到了一种多糖含量在90%以上的姜味草多糖,测试结果表明,姜味草多糖对酪氨酸酶具有较高的抑制作用,具有用作化妆品添加剂提高化妆品美白淡斑效果的前景。

具体实施方式
下面结合附图和实施例具体介绍本发明实质性内容,但并不以此限定本发明的保护范围。
实施例1:姜味草多糖的提取
取洗净干燥的姜味草全草,粉碎,过40目筛,按照料液比1kg:20L加入去离子水,常温浸泡提取48h,收集提取液,过滤,按照1kg全草对应1L浓缩液进行减压浓缩,4000r/min条件下离心10min,上清液用4倍体积的无水乙醇醇析12h,沉淀用去离子水复溶至原上清液体积,重复用4倍体积的无水乙醇醇析一次,沉淀依次用乙醇、丙酮按照1g沉淀对应50mL溶剂交替洗涤3次,挥干溶剂后复溶于去离子水中,用Sevag法脱蛋白,冷冻干燥即得。
经苯酚-硫酸法测定,多糖含量为93.88%,多糖含量较高,扫描电镜图如图1。
实施例2:姜味草多糖的提取
取洗净干燥的姜味草全草,粉碎,过40目筛,按照料液比1kg:15L加入去离子水,常温浸泡提取60h,收集提取液,过滤,按照1kg全草对应1L浓缩液进行减压浓缩,4000r/min条件下离心10min,上清液用4倍体积的无水乙醇醇析12h,沉淀用去离子水复溶至原上清液体积,重复用4倍体积的无水乙醇醇析一次,沉淀依次用乙醇、丙酮按照1g沉淀对应50mL溶剂交替洗涤3次,挥干溶剂后复溶于去离子水中,用Sevag法脱蛋白,冷冻干燥即得。

实施例3:姜味草多糖的提取
取洗净干燥的姜味草全草,粉碎,过40目筛,按照料液比1kg:25L加入去离子水,常温浸泡提取36h,收集提取液,过滤,按照1kg全草对应1L浓缩液进行减压浓缩,4000r/min条件下离心10min,上清液用4倍体积的无水乙醇醇析12h,沉淀用去离子水复溶至原上清液体积,重复用4倍体积的无水乙醇醇析一次,沉淀依次用乙醇、丙酮按照1g沉淀对应50mL溶剂交替洗涤3次,挥干溶剂后复溶于去离子水中,用Sevag法脱蛋白,冷冻干燥即得。
经苯酚-硫酸法测定,多糖含量为91.82%,多糖含量较高。
实施例4:姜味草多糖对酪氨酸酶的抑制活性
将实施例1~3制备的姜味草多糖和阳性药物曲酸分别配制成不同浓度的样品溶液,酪氨酸酶抑制活性的测定按照王加利等提供的方法进行(驱虫斑鸠菊化学成分及酪氨酸酶抑制活性研究,中药材,第40卷第8期2017年8月)。该反应在96孔培养板中进行,总反应体系为200μL:不同浓度的样品溶液40μL;67mmol/L磷酸盐缓冲液80μL;100U/mL酪氨酸酶溶液40μL;2.5mmol/L左旋多巴40μL。调零孔仅加67mmol/L磷酸盐缓冲液200μL。加样完毕后,混匀,将其置于37℃水浴孵箱中孵育20min,用酶联免疫检测仪检测反应产物在490nm波长处的特异性吸光值(A490),重复测试3次。按照下式计算姜味草多糖和阳性药物曲酸对酪氨酸酶的抑制率:酪氨酸酶抑制率(%)=[(A-B)-(C-D)]/(A-B)×100%。其中,A=A490-药+酶;B=A490-药-酶;C=A490+药+酶;D=A490+药-酶。同时以抑制率对抑制剂浓度作图,根据抑制曲线求得姜味草多糖和阳性药物曲酸的IC50值。结果如表1和图2所示。

综合上述实施例可知,本发明从姜味草全草中制备得到了一种多糖含量在90%以上的姜味草多糖,测试结果表明,姜味草多糖对酪氨酸酶具有较高的抑制作用,具有用作化妆品添加剂提高化妆品美白淡斑效果的前景。
上述实施例的作用在于具体介绍本发明的实质性内容,但本领域技术人员应当知道,不应将本发明的保护范围局限于该具体实施例。

1.一种姜味草多糖用于制备美白淡斑化妆品的用途,该姜味草多糖通过如下方法制备:取洗净干燥的姜味草全草,粉碎,过40目筛,按照料液比1kg:(15~25)L加入去离子水,常温浸泡提取36~60h,收集提取液,过滤,按照1kg全草对应1L浓缩液进行减压浓缩,4000r/min条件下离心10min,上清液用4倍体积的无水乙醇醇析12h,沉淀用去离子水复溶至原上清液体积,重复用4倍体积的无水乙醇醇析一次,沉淀依次用乙醇、丙酮按照1g沉淀对应50mL溶剂交替洗涤3次,挥干溶剂后复溶于去离子水中,用Sevag法脱蛋白,冷冻干燥。
