Abstract
本发明涉及一种近红外纳米光敏剂及其制备方法和应用,涉及光敏剂技术领域。本发明所述的近红外纳米光敏剂通过对氟硼二吡咯母核进行共轭延长修饰,使其吸收和发射光谱接近于近红外光区;将多氟烷烃基团和聚乙二醇基团引入上述氟硼二吡咯结构中从而获得两亲性光敏剂;利用多氟烷烃基团间强的氟‑氟作用和聚乙二醇基团的亲水作用,最终构建具有超低CMC值的纳米光敏胶束;利用氟…氟作用诱导氟硼二吡咯发生J‑聚集从而使其最大吸收峰红移至近红外区域,有利于肿瘤深部光治疗。
Description
技术领域
本发明涉及光敏剂技术领域,尤其涉及一种近红外纳米光敏剂及其制备方法和应用。
背景技术
肿瘤是机体在各种致瘤因子作用下,局部组织细胞增生所形成的螯合物,发展新型肿瘤治疗方式是目前医药领域的重要方向。近年来,随着激光技术的发展和应用,光动力治疗逐渐成为一种新型肿瘤治疗手段。光敏剂作为光动力治疗的核心要素,能将激发光的能量传递给组织中的氧,使其转变为活性氧物质(如单线态氧),继而氧化脂类、蛋白质和核酸等内源性分子,导致组织不可逆损伤。然而,大多数光敏剂普遍存在光稳定性差、水溶性差、靶向性低、治疗深度浅等缺陷,严重限制了光动力治疗的临床使用。
临界胶束浓度(CMC)作为一项关键的性能参数,对于描述纳米胶束或囊泡的稳定性起着重要的作用。在光敏药物递送中,常规纳米胶束往往被体液稀释到临界胶束浓度以下,纳米胶束提前发生解聚并释放光敏药物,导致光敏药物在肿瘤部位的富集程度有限。此外,绝大多数光敏剂,如卟啉、二氢卟吩和稠环醌类化合物等,其最大吸收峰位于紫外和可见波段。这些波段的光能被血红蛋白、黑色素以及体内其它含色素的蛋白吸收,致使无法到达肿瘤组织内部。因此,设计合成具有超低临界胶束浓度的近红外纳米光敏胶束是光动力治疗领域亟待解决的重要问题。
中国专利CN109078185A公开了一种高效纳米光敏剂的组装方法,即通过引入长链烷基基团和适当长度的聚乙二醇片段构建两亲性光敏分子。但该专利仍未解决两个问题:一是未能构建具有较低CMC值的光敏胶束,导致稳定性较差;二是制备的光敏胶束的最大吸收峰无法实现红移至近红外区域,致使其无法到达肿瘤组织内部。
由于氟原子具有较小的原子半径和最强的电负性,氟原子的引入往往会带来意想不到的物理和化学性质,已被广泛用于材料、原子能、航空航天等多个领域。近年来,含氟化合物在生物医药领域的应用备受关注,如含氟药物、含氟麻醉剂、含氟示踪剂、药物载体等。利用多氟烷烃基团之间的“亲氟”作用,可用于生物检测、固相分离、均相催化、标记合成等领域。除上述领域外,氟…氟相互作用亦可用于纳米结构的高效自组装,如氟化金纳米粒自组装体、氟化DNA胶束、氟化纳米滴等。
现有技术不足之处主要分为两方面:(1)现有光敏剂虽然在临床上已经取得了一定的疗效,但是仍存在组织靶向性差、光稳定性差、治疗深度不够、代谢缓慢造成的长期光毒性等问题;(2)氟硼二吡咯是一类新型的有机染料分子,具有强摩尔消光系数、较高的荧光量子产率等优点。可以通过不同策略调控氟硼二吡咯的辐射跃迁率和系间窜越几率,获得长寿命的三线态激发态。但是其最大吸收峰往往处于可见光区,因而限制该类化合物在光治疗方面的运用;(3)虽然通过利用高分子聚合物、人血清白蛋白、无机纳米材料等作为递送载体,从而改善光敏剂水溶性、光稳定性和靶向性。但注射体内后,此类纳米光敏剂被常因不稳定,因此开发新型光敏剂药物仍然面临极大的挑战。
发明内容
为此,本发明所要解决的技术问题在于克服现有技术中光稳定性差、治疗深度浅的问题。
为解决上述技术问题,本发明提供了一种近红外纳米光敏剂及其制备方法和应用。
本发明的第一个目的是提供一种近红外纳米光敏剂,具有如式(I)所示的结构式:

本发明的第二个目的是提供一种所述的近红外纳米光敏剂的制备方法,包括以下步骤,
(2)将步骤(1)所述的式(III)所示的化合物与叠氮化聚乙二醇在五水硫酸铜和抗坏血酸钠存在的条件下,在溶剂中发生反应,得到所述式(I)所示的化合物;

在本发明的一个实施例中,所述溶剂为乙腈、甲苯、苯、N,N-二甲基甲酰胺、二甲基亚砜中的一种或多种。
在本发明的一个实施例中,在步骤(1)中,所述反应的温度为80-120℃;反应的时间为12-48h。
在本发明的一个实施例中,在步骤(2)中,所述式(III)所示的化合物、叠氮化聚乙二醇、五水硫酸铜和抗坏血酸钠的摩尔比为1:1-3:0.1-0.2:0.1-0.2。
在本发明的一个实施例中,在步骤(2)中,所述反应的温度为40-60℃;反应的时间为12-24h。
本发明的第三个目的是提供一种纳米胶束溶液,所述纳米胶束溶液是所述的近红外纳米光敏剂于水中经分子自组装形成的。
本发明的第四个目的是提供一种所述的近红外纳米光敏剂或所述的纳米胶束在制备用于治疗肿瘤的药物中的应用。
本发明的技术方案相比现有技术具有以下优点:
(1)本发明所述的近红外纳米光敏剂利用氟原子的特殊性,氟原子具有原子半径小、极化性小、电负性高等性质,导致多氟烷基或烷烃具有既不亲水又不亲油的特性,因此多氟烷基或烷烃倾向于通过氟-氟作用聚集在一起,从而产生了有别于水相、油相的第三相,即氟相。通过对氟硼二吡咯母核进行共轭延长修饰,使其吸收和发射光谱接近于近红外光区;将多氟烷烃基团和聚乙二醇基团引入上述氟硼二吡咯结构中从而获得两亲性光敏剂;利用多氟烷烃基团间强的氟-氟作用和聚乙二醇基团的亲水作用,最终构建具有超低CMC值的纳米光敏胶束;利用氟…氟作用诱导氟硼二吡咯发生J-聚集从而使其最大吸收峰红移至近红外区域,有利于肿瘤深部光治疗。
(2)本发明所述的近红外纳米光敏剂的制备过程中,首先通过缩合反应延长了氟硼二吡咯的共轭体系,并在末端引入亲水的乙二醇链,解决了当下氟硼二吡咯类化合物在应用方面存在的问题,改善了其水溶性。其次通过反应引入氟原子,提高分子自组装的驱动力,又同时引入了光热和光动力治疗相结合的多模式治疗概念。其对提高单线态氧产率,抗肿瘤效果,和肿瘤靶向性的有益效果在细胞实验结果中也到了证明。
(3)本发明所述的近红外纳米光敏剂利用氟原子来提高光敏剂的高效自组装体系,提高肿瘤靶向性。同时,通过氟原子来调控BODIPY光敏剂光物理活性,使得新型BODIPY光敏剂吸收峰红移至近红外区,提高治疗深度。

具体实施方式
下面结合附图和具体实施例对本发明作进一步说明,以使本领域的技术人员可以更好地理解本发明并能予以实施,但所举实施例不作为对本发明的限定。
在本发明中,除非另有说明,目标分子化合物中R1基团为(CH2CH2O)nCH2CH2OCH3,且n为113,即R1基团为(CH2CH2O)113CH2CH2OCH3。
实施例1
如图1所示,一种近红外纳米光敏剂及其制备方法,具体包括以下步骤:
(1)合成化合物1:将含炔基的对羟基苯甲醛和2,4-二甲基吡咯,按摩尔比1:1加入反应容器中,然后加入2,4-二甲基吡咯重量的100倍的四氢呋喃作为溶剂和6-10滴三氟乙酸作为催化剂。在25℃下搅拌24h。随后在上述反应液中加入与2,4-二甲基吡咯摩尔比1:1.5的2,3-二氯-5,6-二氰对苯醌,室温搅拌反应24h。最后加入2,4-二甲基吡咯重量的50倍的三乙胺,并在冰水浴条件下逐滴加入2,4-二甲基吡咯重量50倍的三氟化硼·乙醚反应过夜。反应结束后,用乙酸乙酯和水萃取,经旋转蒸发仪浓缩后,经快速柱层析法得到粗产物,再次经柱层析(SiO2;洗脱剂为石油醚/二氯甲烷)得到化合物1,产率70%。
(2)合成化合物2:将化合物1和N-碘代丁二酰亚胺按摩尔比1:2于反应容器中,然后加入50mL的二氯甲烷作为溶剂反应15min。反应结束后经旋转蒸发仪浓缩,再用进行萃取并收集有机层,经干燥浓缩后得到粗产物。粗产物经硅胶柱层析分离(洗脱剂为石油醚-二氯甲烷体系),得到化合物2,产率80%。
(3)合成化合物3a:化合物2和CHO-1按摩尔比1:4于反应容器中,用10mL乙腈作为溶剂。最后与化合物2摩尔比1:15:15的哌啶和醋酸在氮气保护下加热至100℃回流反应12h。反应结束后经浓缩、萃取、柱层析(石油醚:二氯甲烷)后得到化合物3a,产率为70%。
(4)合成化合物F9-BDP:化合物3a和叠氮聚乙二醇(R1N3)按摩尔比(1:1.5)于反应容器中。加入与化合物3a摩尔比1:0.1:0.1的五水硫酸铜和抗坏血酸钠,并加入5mL的DMSO作为溶剂在氮气保护下于55℃反应24h。反应结束后经浓缩、萃取、柱层析(甲醇:二氯甲烷)后得到化合物F9-BDP,产率50%。
实施例2
一种近红外纳米光敏剂及其制备方法,具体包括以下步骤:
(1)合成化合物1:将含炔基的对羟基苯甲醛和2,4-二甲基吡咯,按摩尔比1:3加入反应容器中,然后加入2,4-二甲基吡咯重量的50倍的四氢呋喃作为溶剂和3-5滴三氟乙酸作为催化剂。在40℃下搅拌24h。随后在上述反应液中加入与2,4-二甲基吡咯摩尔比1:3的2,3-二氯-5,6-二氰对苯醌,室温搅拌反应24h。最后加入2,4-二甲基吡咯重量的100倍的三乙胺,并在冰水浴条件下逐滴加入2,4-二甲基吡咯重量100倍的三氟化硼·乙醚反应过夜。反应结束后,用乙酸乙酯和水萃取,经旋转蒸发仪浓缩后,经快速柱层析法得到粗产物,再次经柱层析(SiO2;洗脱剂为石油醚/二氯甲烷)得到化合物1,产率60%。
(2)合成化合物2:将化合物1和N-碘代丁二酰亚胺按摩尔比1:3于反应容器中,然后加入化合物1重量的100倍的二氯甲烷作为溶剂反应1h。反应结束后经旋转蒸发仪浓缩,再用进行萃取并收集有机层,经干燥浓缩后得到粗产物。粗产物经硅胶柱层析分离(洗脱剂为石油醚-二氯甲烷体系),得到化合物2,产率70%。
(3)合成化合物3a:化合物2和CHO-1按摩尔比1:4于反应容器中,用化合物2重量的50倍的甲苯作为溶剂。最后与化合物2摩尔比1:20:20的哌啶和醋酸在氮气保护下加热至80℃回流反应24h。反应结束后经浓缩、萃取、柱层析(石油醚:二氯甲烷)后得到化合物3a,产率为50%。

实施例3
一种近红外纳米光敏剂及其制备方法,具体包括以下步骤:
(1)合成化合物1:将含炔基的对羟基苯甲醛和2,4-二甲基吡咯,按摩尔比1:5加入反应容器中,然后加入2,4-二甲基吡咯重量的30倍的二氯甲烷作为溶剂和6-10滴三氟乙酸作为催化剂。在60℃下搅拌48h。随后在上述反应液中加入与2,4-二甲基吡咯摩尔比1:2的2,3-二氯-5,6-二氰对苯醌,室温搅拌反应24h。最后加入2,4-二甲基吡咯重量的50倍的三乙胺,并在冰水浴条件下逐滴加入2,4-二甲基吡咯重量50倍的三氟化硼·乙醚反应过夜。反应结束后,用乙酸乙酯和水萃取,经旋转蒸发仪浓缩后,经快速柱层析法得到粗产物,再次经柱层析(SiO2;洗脱剂为石油醚/二氯甲烷)得到化合物1,产率45%。
(2)合成化合物2:将化合物1和N-碘代丁二酰亚胺按摩尔比1:2于反应容器中,然后加入化合物1重量的50倍的四氢呋喃作为溶剂反应6h。反应结束后经旋转蒸发仪浓缩,再用进行萃取并收集有机层,经干燥浓缩后得到粗产物。粗产物经硅胶柱层析分离(洗脱剂为石油醚-二氯甲烷体系),得到化合物2,产率80%。
(3)合成化合物3a:化合物2和CHO-1按摩尔比1:6于反应容器中,用化合物2重量的100倍的甲苯作为溶剂。最后与化合物2摩尔比1:10:10的哌啶和醋酸在氮气保护下加热至80℃回流反应48h。反应结束后经浓缩、萃取、柱层析(石油醚:二氯甲烷)后得到化合物3a,产率为75%。
(4)合成化合物F9-BDP:化合物3a和叠氮聚乙二醇(R1N3)按摩尔比(1:2)于反应容器中。加入与化合物3a摩尔比1:0.1:0.1的五水硫酸铜和抗坏血酸钠,并加入化合物3a重量的50倍的DMSO作为溶剂在氮气保护下于60℃反应12h。反应结束后经浓缩、萃取、柱层析(甲醇:二氯甲烷)后得到化合物F9-BDP,产率45%。
实施例4
一种近红外纳米光敏剂及其制备方法,具体包括以下步骤:
(1)合成化合物1:将含炔基的对羟基苯甲醛和2,4-二甲基吡咯,按摩尔比1:2加入反应容器中,然后加入2,4-二甲基吡咯重量的40倍的二氯甲烷作为溶剂和5-8滴三氟乙酸作为催化剂。在20℃下搅拌24h。随后在上述反应液中加入与2,4-二甲基吡咯摩尔比1:3的2,3-二氯-5,6-二氰对苯醌,室温搅拌反应12h。最后加入2,4-二甲基吡咯重量的30倍的三乙胺,并在冰水浴条件下逐滴加入2,4-二甲基吡咯重量30倍的三氟化硼·乙醚反应过夜。反应结束后,用乙酸乙酯和水萃取,经旋转蒸发仪浓缩后,经快速柱层析法得到粗产物,再次经柱层析(SiO2;洗脱剂为石油醚/二氯甲烷)得到化合物1,产率25%。
(2)合成化合物2:将化合物1和N-碘代丁二酰亚胺按摩尔比1:3于反应容器中,然后加入化合物1重量的30倍的四氢呋喃作为溶剂反应12h。反应结束后经旋转蒸发仪浓缩,再用进行萃取并收集有机层,经干燥浓缩后得到粗产物。粗产物经硅胶柱层析分离(洗脱剂为石油醚-二氯甲烷体系),得到化合物2,产率45%。
(3)合成化合物3a:化合物2和CHO-1按摩尔比1:3于反应容器中,用化合物2重量的50倍的苯作为溶剂。最后与化合物2摩尔比1:10:10的哌啶和醋酸在氮气保护下加热至120℃回流反应24h。反应结束后经浓缩、萃取、柱层析(石油醚:二氯甲烷)后得到化合物3a,产率为60%。
(4)合成化合物F9-BDP:化合物3a和叠氮聚乙二醇(R1N3)按摩尔比(1:1)于反应容器中。加入与化合物3a摩尔比1:0.1:0.1的五水硫酸铜和抗坏血酸钠,并加入化合物3a重量的50倍的DMF作为溶剂在氮气保护下于40℃反应24h。反应结束后经浓缩、萃取、柱层析(甲醇:二氯甲烷)后得到化合物F9-BDP,产率48%。
实施例5
一种近红外纳米光敏剂及其制备方法,具体包括以下步骤:
(1)合成化合物1:将含炔基的对羟基苯甲醛和2,4-二甲基吡咯,按摩尔比1:3加入反应容器中,然后加入2,4-二甲基吡咯重量的80倍的四氢呋喃作为溶剂和10滴三氟乙酸作为催化剂。在50℃下搅拌12h。随后在上述反应液中加入与2,4-二甲基吡咯摩尔比1:5的2,3-二氯-5,6-二氰对苯醌,室温搅拌反应24h。最后加入2,4-二甲基吡咯重量的50倍的三乙胺,并在冰水浴条件下逐滴加入2,4-二甲基吡咯重量50倍的三氟化硼·乙醚反应过夜。反应结束后,用乙酸乙酯和水萃取,经旋转蒸发仪浓缩后,经快速柱层析法得到粗产物,再次经柱层析(SiO2;洗脱剂为石油醚/二氯甲烷)得到化合物1,产率33%。
(2)合成化合物2:将化合物1和N-碘代丁二酰亚胺按摩尔比1:5于反应容器中,然后加入化合物1重量的50倍的四氢呋喃作为溶剂反应24h。反应结束后经旋转蒸发仪浓缩,再用进行萃取并收集有机层,经干燥浓缩后得到粗产物。粗产物经硅胶柱层析分离(洗脱剂为石油醚-二氯甲烷体系),得到化合物2,产率90%。
(3)合成化合物3a:化合物2和CHO-1按摩尔比1:6于反应容器中,用化合物2重量的100倍的甲苯作为溶剂。最后与化合物2摩尔比1:20:20的哌啶和醋酸在氮气保护下加热至120℃回流反应24h。反应结束后经浓缩、萃取、柱层析(石油醚:二氯甲烷)后得到化合物3a,产率为56%。
(4)合成化合物F9-BDP:化合物3a和叠氮聚乙二醇(R1N3)按摩尔比(1:1.5)于反应容器中。加入与化合物3a摩尔比1:0.1:0.1的五水硫酸铜和抗坏血酸钠,并加入化合物3a重量的50倍的DMF作为溶剂在氮气保护下于60℃反应24h。反应结束后经浓缩、萃取、柱层析(甲醇:二氯甲烷)后得到化合物F9-BDP,产率62%。
对比例1
如图1所示,一种纳米光敏剂及其制备方法,具体包括以下步骤:
(1)合成化合物1:将含炔基的对羟基苯甲醛和2,4-二甲基吡咯,按摩尔比1:1加入反应容器中,然后加入2,4-二甲基吡咯重量的100倍的四氢呋喃作为溶剂和6-10滴三氟乙酸作为催化剂。在25℃下搅拌24h。随后在上述反应液中加入与2,4-二甲基吡咯摩尔比1:1.5的2,3-二氯-5,6-二氰对苯醌,室温搅拌反应24h。最后加入2,4-二甲基吡咯重量的50倍的三乙胺,并在冰水浴条件下逐滴加入2,4-二甲基吡咯重量50倍的三氟化硼·乙醚反应过夜。反应结束后,用乙酸乙酯和水萃取,经旋转蒸发仪浓缩后,经快速柱层析法得到粗产物,再次经柱层析(SiO2;洗脱剂为石油醚/二氯甲烷)得到化合物1,产率70%。
(2)合成化合物2:将化合物1和N-碘代丁二酰亚胺按摩尔比1:2于反应容器中,然后加入50mL的二氯甲烷作为溶剂反应15min。反应结束后经旋转蒸发仪浓缩,再用进行萃取并收集有机层,经干燥浓缩后得到粗产物。粗产物经硅胶柱层析分离(洗脱剂为石油醚-二氯甲烷体系),得到化合物2,产率80%。
(3)合成化合物3b:化合物2和CHO-1按摩尔比1:4于反应容器中,用10mL乙腈作为溶剂。最后与化合物2摩尔比1:15:15的哌啶和醋酸在氮气保护下加热至100℃回流反应12h。反应结束后经浓缩、萃取、柱层析(石油醚:二氯甲烷)后得到化合物3b,产率为65%。
(4)合成化合物C16-BDP:化合物3b和叠氮聚乙二醇(R1N3)按摩尔比(1:1.5)于反应容器中。加入与化合物3b摩尔比1:0.1:0.1的五水硫酸铜和抗坏血酸钠,并加入5mL的DMSO作为溶剂在氮气保护下于55℃反应24h。反应结束后经浓缩、萃取、柱层析(甲醇:二氯甲烷)后得到化合物C16-BDP,产率50%。
测试例1
对实施例1的目标产物F9-BDP和对比例1制备的C16-BDP进行核磁鉴定并进行归属。
如图2-3所示,表明目标产物已顺利合成得到。
测试例2
对于纳米光敏剂C16-BDP-NPs或F9-BDP-NPs纳米胶束利用透射电镜进行表征和考察。
采用薄膜分散法取少量的实施例1的化合物C16-BDP或对比例1的化合物F9-BDP,将C16-BDP或F9-BDP溶于有机试剂(如二氯甲烷或丙酮),使用旋转蒸发仪旋干。在超声条件下加入适量去离子水,继续超声10-60min,即可制备出纳米光敏剂C16-BDP-NPs或F9-BDP-NPs纳米胶束。
分别制备等浓度的纳米胶束溶液,滴于铜网中,挥干后使用透射电镜进行拍摄,如图4-5所示,表明纳米光敏剂C16-BDP-NPs或F9-BDP-NPs具有均一的纳米尺寸。
测试例3
对于制备出来的C16-BDP-NPs或F9-BDP-NPs纳米胶束进行光谱性质测定。
如图6-7所示,C16-BDP-NPs纳米胶束的最大吸收波长约600nm,C16-BDP-NPs的吸收波长出现蓝移现象。F9-BDP-NPs纳米胶束的最大吸收波长约720nm,F9-BDP-NPs纳米胶束的吸收波长红移到近红外区域。同时通过荧光强度初步证明本设计方案成功的实现了高效近红外BODIPY光敏剂,且F9-BDP-NPs纳米胶束的荧光强度明显高于C16-BDP-NPs纳米胶束。
测试例4
对于制备出来的不同浓度的C16-BDP-NPs或F9-BDP-NPs纳米胶束溶液进行临界胶束浓度(CMC)测定。
由于传统的芘测试法难以测得低于10-3mM的临界胶束浓度(CMC),因此采用纳米金临界胶束浓度法测试了F9-BDP-NPs和C16-BDP-NPs纳米胶束的CMC值,观察化合物在不同浓度下的紫外-可见吸收光谱的变化情况,结果如图8所示,F9-BDP-NPs和C16-BDP-NPs的临界胶束值分别为7.4×10-5mM和1.5×10-4mM,F9-BDP-NPs的临界胶束值远低于专利CN109078185A中的2-IBMs(10-3mM)和传统的高分子胶束(>10-3mM)。
可见F9-BDP-NPs纳米胶束溶液具有极低的CMC,可在低浓度时保持纳米胶束状态。
测试例5
对于制备出来的C16-BDP-NPs或F9-BDP-NPs纳米胶束进行单线态氧产率的考察。
分别制备等浓度的纳米胶束溶液,其中包含等摩尔的1,3-二苯基异苯并呋喃,利用卤素灯(>660nm,0.1W/cm2)作为光照实验的激发光源,照射180s,每隔30s测量溶液的紫外-可见吸收光谱,观察415nm处,溶液的紫外-可见吸收的变化,由变化情况绘图得到单线态氧产率折线图,如图9所示,C16-BDP-NPs或F9-BDP-NPs纳米胶束均具有较高的单线态氧产率,以F9-BDP-NPs纳米胶束为最佳,因此F9-BDP-NPs纳米胶束是更好的实现光动力治疗的基础。
测试例6
对于制备出来的C16-BDP-NPs或F9-BDP-NPs纳米胶束进行细胞毒性实验。
取对数生长期的4T1细胞铺96孔板,接种密度为6×103/mL,每孔100μL,放入细胞培养箱恒温培养12h,确定细胞贴壁后,倒掉培养液,用磷酸盐缓冲液洗1-2次,加入用培养基配好的C16-BDP-NPs或F9-BDP-NPs纳米胶束溶液,每孔100微升,光照组配置浓度梯度为0.625、1.25、2.5、5.0、10、20μM,每个浓度4个复孔:非光照组配置同样浓度梯度0.625、1.25、2.5、5.0、10、20μM,每个浓度4个复孔放入培养箱培养24h后,更换培养液,光照组分别在0.1W/cm2条件下,光照30min,放回培养箱中继续培养24h,加MTT的PBS溶液(5mg/mL,20μL),4h后弃去培养液,加入DMSO(150μL),振荡10min,酶标仪490nm处测定吸光度值,如图10-11所示,光照下F9-BDP-NPs纳米胶束具有较低的IC50值,相比之下,暗毒性很低,证明只有特定激光才能引起药物发挥毒性作用。
测试例7
对于制备出来的C16-BDP-NPs或F9-BDP-NPs纳米胶束进行小鼠体内组织分布实验,具体步骤如下:
(1)荷瘤小鼠模型的建立:提前选取体重为20g的BALB/c雌性小鼠,然后收集生长状态良好的4T1细胞,每只小鼠接种107个细胞,体积为50μL,皮下注射到小鼠背侧部。
(2)纳米胶束的放射性标记:将C16-BDP-NPs或F9-BDP-NPs纳米胶束溶液(400μM),分别取500μL放到2mL的EP管中。另外准确称取5mg的Iodogen,放到2mL的EP管里,加入1mL三氯甲烷溶解。溶解后用氮气吹干,使其均匀铺到管底部。随后,从铅罐里取出1mCi的125I放到上述EP管中,并加入C16-BDP-NPs或F9-BDP-NPs纳米胶束,放入振荡器于室温震荡20min。20min后终止反应,将EP管里溶液转移到超滤管里进行超滤离心(3500rpm,15min)。离心结束后,使用放射性活度计测试超滤管上管和下管的放射性活度,离心三次即可。
(3)以建立好的雌性BALB/c荷瘤小鼠,每组3只,尾静脉注射上述放射性标记后的纳米胶束(400μM,核素含量20μCi)。给药设置时间点。待到时间,处死小鼠,取出组织。将各个组织称重后放到流式管内,使用伽马免疫计数仪测试组织中的放射性含量,并通过计算进行定量,如图12所示,含F9-BDP-NPs纳米胶束药物在小鼠体内具有较好的肿瘤靶向性。
由此可见,F9-BDP-NPs纳米胶束溶液具有更优异的性能和更好的应用前景。
显然,上述实施例仅仅是为清楚地说明所作的举例,并非对实施方式的限定。对于所属领域的普通技术人员来说,在上述说明的基础上还可以做出其它不同形式变化或变动。这里无需也无法对所有的实施方式予以穷举。而由此所引申出的显而易见的变化或变动仍处于本发明创造的保护范围之中。
Claims (10)
2.据权利要求1所述的近红外纳米光敏剂,其特征在于,所述y为3、5或7。
4.根据权利要求3所述的近红外纳米光敏剂的制备方法,其特征在于,所述溶剂为乙腈、甲苯、苯、N,N-二甲基甲酰胺、二甲基亚砜中的一种或多种。
6.根据权利要求3所述的近红外纳米光敏剂的制备方法,其特征在于,在步骤(1)中,所述反应的温度为80-120℃;反应的时间为12-48h。
7.根据权利要求3所述的近红外纳米光敏剂的制备方法,其特征在于,在步骤(2)中,所述式(III)所示的化合物、叠氮化聚乙二醇、五水硫酸铜和抗坏血酸钠的摩尔比为1:1-3:0.1-0.2:0.1-0.2。
8.根据权利要求3所述的近红外纳米光敏剂的制备方法,其特征在于,在步骤(2)中,所述反应的温度为40-60℃;反应的时间为12-24h。
9.一种纳米胶束溶液,其特征在于,所述纳米胶束溶液是由权利要求1或2所述的近红外纳米光敏剂于水中经分子自组装形成的。
10.权利要求1或2所述的近红外纳米光敏剂或权利要求9所述的纳米胶束在制备用于治疗肿瘤的药物中的应用。
