本发明属于功能性食品的加工技术领域,涉及一种含有花胶和轮叶党参低聚肽的抗皮肤衰老组合物及制备方法。
背景技术
由于年龄的增长,人体皮肤的形态结构与生理功能会出现退行性改变。依据影响因素的不同分为外源性衰老与内源性衰老。皮肤的衰老主要表现为暴露部位粗糙、弹性下降、皱纹加深加粗、不规则性色素沉着、血管扩张、表皮角化不良和异常增殖等,未暴露部分皮肤萎缩、细小皱纹和干燥。皮肤衰老不仅有损于美容,且与许多皮肤病如脂溢性角化、日光性角化、基底细胞癌、鳞状细胞癌的发生也有着病因学的关系。
遗传基因是皮肤衰老的根本,然而很多因素加速了皮肤的衰老,如紫外线/蓝光照射、不良饮食结构、不规律的作息习惯等。这些外界因素只是表象,根本上的原因是过量的自由基。自由基具有极强的氧化能力,自由基主要通过对细胞生物膜、细胞核和线粒体的攻击引起细胞的衰老,从而导致身体及皮肤表现出衰老症状。
自由基对细胞生物膜的攻击主要有两个途径:一是可使生物膜中不饱和脂类发生过氧化,形成过氧化脂质,其终产物丙二醛是强的交联剂,与蛋白质、核酸或脂类结成难溶性物质,使生物膜硬化导致通透性降低,影响细胞物质交换,继而使细胞破裂、死亡;二是对生物膜上受体的攻击造成蛋白质交联变性,空间立体结构发生改变,丧失了与配体的结合能力,不能发生自体磷酸化及后继信号传导功能。
自由基使组成DNA的胸腺嘧啶5,6位双键氧化、交联,导致DNA基因突变,导致转录异常,受体合成减少并产生异种蛋白质,甚者导致细胞癌变。
线粒体是细胞能量代谢中心、自由基代谢中心,也是细胞凋亡调控中心,并在信号转导和基因表达调控中发挥重要作用。线粒体一旦受损,一方面能量代谢发生紊乱,另一方面产生大量自由基,影响细胞的正常生长,并导致细胞甚至机体的衰老自由基对线粒体的攻击会导致线粒体的功能异常,进而影响这个细胞的代谢功能。
发明内容
本发明的目的在于提供一种含有花胶和轮叶党参低聚肽的抗皮肤衰老组合物及制备方法,通过给真皮层补充优质胶原蛋白、促进成纤维细胞增殖、提高皮肤持水能力、去除自由基、提高抗氧化酶活性、增强皮肤代谢能力,达到预防、延缓或改善皮肤衰老的目的。
皮肤衰老的根本原因在于自由基攻击导致的细胞代谢及修复能力的下降,因此自由基是否能够有效去除,决定了皮肤的抗衰老能力。
本发明选择从去除自由基、提高皮肤抗氧化能力、给皮肤补充必需营养(胶原蛋白和透明质酸)、促进成纤维细胞增殖等方面入手,研究评价对皮肤更新及修复的引导和促进作用。
本发明的组合物可以为口服溶液、冲剂、口服乳液、片剂、悬浮液、丸剂、软胶囊等形式,优选以口服溶液或冲剂的形式产品。
本发明的方案如下:一种含有花胶和轮叶党参低聚肽的抗皮肤衰老组合物,由以下组分组成:花胶低聚肽100-150份、雨生红球藻粉10-20份、海参多糖15-35份、葡萄籽提取物10-20份、人参低聚肽25-50份、透明质酸钠10-20份、酵母抽提物5-10份、轮叶党参低聚肽80-120份、维生素E粉5-10份、维生素C1-5份、茶多酚1-10份。
进一步的,花胶低聚肽是由花胶酶解而形成的复合物。
进一步的,花胶低聚肽含有抗氧化肽段序列NH2-Gly-Pro-Ala-Gly-Pro-Cys。
进一步的,花胶低聚肽基于如下方式制备而得:将干花胶粗粉碎后,加入15~20倍质量体积的水制成匀浆液置于酶解罐中,然后加入花胶蛋白质质量的3~5%的复合蛋白酶,在50~55℃下酶解3~5小时,酶反应pH值控制在7.0~8.0,酶解结束后升温至90℃灭酶10分钟,得到花胶酶解液,离心去除杂质,然后采用截留分子量为3000Da的陶瓷膜进行膜分离,精制脱盐脱重金属,浓缩至可溶性固形物含量占浓缩液总质量的30~40%;所述复合蛋白酶的质量配比为:中性蛋白酶:胰蛋白酶:菠萝蛋白酶:风味蛋白酶=(4~6):(3~5):(1~3):(2~4)。

进一步的,轮叶党参低聚肽基于如下方式制备而得:轮叶党参样品粗粉碎处理后,加入10~15倍质量体积的水制成匀浆液置于酶解罐中,然后加入轮叶党参质量0.5~1.0%的纤维素酶,在45~50℃下酶解1~2小时,酶反应pH值控制在5.0~6.0;再加入轮叶党参蛋白质质量3~5%的复合蛋白酶,在50~60℃下酶解3~4小时,酶反应pH值控制在8.5~9.5,酶解结束后升温至90℃灭酶10分钟,得到轮叶党参酶解液,离心去除杂质,然后采用截留分子量为3000Da的陶瓷膜进行膜分离,精制脱盐脱重金属,浓缩至可溶性固形物含量占浓缩液总质量的30~40%,所述复合蛋白酶的质量配比为:木瓜蛋白酶:中性蛋白酶:碱性蛋白酶=(2~4):(1~2):(4~6)。
进一步的,海参多糖基于如下方式制备而得:取海参原料制浆,得到海参浆料;加水,调节pH至6.5~8.0,加复合蛋白酶,酶解2~6小时;离心分离,收集上清液;采用超滤膜反复浓缩过滤,收集最终浓缩液即为海参多糖浓液;将海参多糖浓液冷冻干燥后,填充到树脂柱中,先采用65%酒精过柱,再采用95%酒精过柱;取出树脂柱中的海参多糖,置于硅胶干燥箱中干燥,即得到精制海参多糖。

有益效果:为了实现抗皮肤衰老的目的,发明人从调节皮肤氧化平衡、激活成纤维细胞、促进皮肤代谢、提高皮肤保水能力等方面入手,找到具有有益的抗皮肤衰老的最佳组合,本发明所述的组合物具有优越的抗皮肤衰老性能且不产生副作用。
本发明选择花胶低聚肽和透明质酸,直接给皮肤补充必需营养,提高皮肤的代谢与更新能力。花胶低聚肽、雨生红球藻粉、葡萄籽提取物、轮叶党参低聚肽等能够进一步地去除过量的自由基,同时提高皮肤自有抗氧化酶的活性,抑制皮肤自然老化与光老化。人参低聚肽促进皮肤营养输送,酵母抽提物提供谷胱甘肽促进成纤维细胞地增殖,两者协同进一步地促进受损皮肤的自我修复。海参多糖分子链中单糖分子的羟基还能与金属离子螯合,抑制脂质过氧化。维生素C、维生素E和茶多酚协同保护其他成分的活性。

含有花胶低聚肽的抗皮肤衰老组合物,组分为:花胶低聚肽120份、雨生红球藻粉20份、海参多糖25份、葡萄籽提取物20份、人参低聚肽40份、透明质酸钠15份、酵母抽提物10份、维生素E粉10份、维生素C 2份、茶多酚6份。
制备方法:取120g花胶低聚肽,加入雨生红球藻粉20g、海参多糖25g、葡萄籽提取物20g、人参低聚肽40g、透明质酸钠15g、酵母抽提物10g、维生素E粉10g、维生素C 2g、茶多酚6g,在反应釜中搅拌20分钟,然后进行喷雾干燥,得到粉剂,喷雾干燥条件为:进风温度130℃,温差为2℃;出风温度70℃,温差为1℃,粉剂采用粉体灌装机进行分装。
其中花胶低聚肽的制备方法为:将干花胶粗粉碎后,加入15倍质量体积的水制成匀浆液置于酶解罐中,然后加入花胶蛋白质质量的5%的复合蛋白酶,在52℃下酶解4小时,酶反应pH值控制在7.5,酶解结束后升温至90℃灭酶10分钟,得到花胶酶解液,离心去除杂质,然后采用截留分子量为3000Da的陶瓷膜进行膜分离,精制脱盐脱重金属,浓缩至可溶性固形物含量占浓缩液总质量的35%。所述复合蛋白酶的质量配比为:中性蛋白酶:胰蛋白酶:菠萝蛋白酶:风味蛋白酶=5:4:2:2。
其中海参多糖的制备方法为:取海参原料制浆,得到海参浆料;加水,调节pH至6.5~8.0,加复合蛋白酶,酶解2~6小时;离心分离,收集上清液;采用超滤膜反复浓缩过滤,收集最终浓缩液,即为海参多糖浓液;将海参多糖浓液冷冻干燥后,填充到树脂柱中,先采用65%酒精过柱,再采用95%酒精过柱;取出树脂柱中的海参多糖,置于硅胶干燥箱中干燥,即得到精制海参多糖。
实施例2
含有轮叶党参低聚肽的抗皮肤衰老组合物,组分为:轮叶党参低聚肽100份、雨生红球藻粉10份、海参多糖15份、葡萄籽提取物15份、人参低聚肽30份、透明质酸钠20份、酵母抽提物7份、维生素E粉8份、维生素C 3份、茶多酚5份。
制备方法:取100g轮叶党参低聚肽,加入雨生红球藻粉10g、海参多糖15g、葡萄籽提取物15g、人参低聚肽30g、透明质酸钠20g、酵母抽提物7g、维生素E粉8g、维生素C3g、茶多酚5g,加入反应釜中搅拌25分钟,然后进行喷雾干燥,得到粉剂,喷雾干燥条件为:进风温度130℃,温差为2℃;出风温度70℃,温差为1℃,粉剂采用粉体灌装机进行分装。
其中轮叶党参低聚肽的制备方法为:轮叶党参样品粗粉碎处理后,加入13倍质量体积的水制成匀浆液置于酶解罐中,然后加入轮叶党参质量(以干重计)0.7%的纤维素酶,在50℃下酶解2小时,酶反应pH值控制在5.0;再加入轮叶党参蛋白质质量4%的复合蛋白酶,在55℃下酶解4小时,酶反应pH值控制在8.5,酶解结束后升温至90℃灭酶10分钟,得到轮叶党参酶解液,离心去除杂质,然后采用截留分子量为3000Da的陶瓷膜进行膜分离,精制脱盐脱重金属,浓缩至可溶性固形物含量占浓缩液总质量的30%。所述复合蛋白酶的质量配比为:木瓜蛋白酶:中性蛋白酶:碱性蛋白酶=3:2:4。
海参多糖的制备方法同实施例1中的制备方法。
实施例3
含有花胶及轮叶党参低聚肽的抗皮肤衰老组合物,组分为:花胶低聚肽120份、轮叶党参低聚肽100份、雨生红球藻粉20份、海参多糖30份、葡萄籽提取物20份、人参低聚肽40份、透明质酸钠15份、酵母抽提物10份、维生素E粉10份、维生素C 2份、茶多酚6份。
制备方法:取120g花胶低聚肽和100g轮叶党参低聚肽,加入雨生红球藻粉20g、海参多糖30g、葡萄籽提取物20g、人参低聚肽40g、透明质酸钠15g、酵母抽提物10g、维生素E粉10g、维生素C 2g、茶多酚6g,在反应釜中搅拌30分钟,然后进行喷雾干燥,得到粉剂,喷雾干燥条件为:进风温度130℃,温差为2℃;出风温度70℃,温差为1℃,粉剂采用粉体灌装机进行分装。

实施例4
一种含有花胶和轮叶党参低聚肽的抗皮肤衰老组合物,包含花胶低聚肽、雨生红球藻粉、海参多糖、葡萄籽提取物、人参低聚肽、透明质酸钠、酵母抽提物、轮叶党参低聚肽、维生素E粉、维生素C、茶多酚。
具体的,花胶低聚肽100-150份、雨生红球藻粉10-20份、海参多糖15-35份、葡萄籽提取物10-20份、人参低聚肽25-50份、透明质酸钠10-20份、酵母抽提物5-10份、轮叶党参低聚肽80-120份、维生素E粉5-10份、维生素C1-5份、茶多酚1-10份。
花胶低聚肽的原料来源为黄花鱼(Larimichthys)、鳕鱼、赤嘴鳘、石斑鱼、黄姑鱼、鲟鱼、鲈鱼等其中的一种或几种,轮叶党参(Codonopsis lanceolata)低聚肽的原料来源为长白山地区的轮叶党参。
组合物具有调节皮肤氧化平衡的作用,其特征在于组分包括花胶低聚肽、雨生红球藻粉、海参多糖、葡萄籽提取物、人参低聚肽、轮叶党参低聚肽。
进一步地,该组合物具有消除、抑制DPPH自由基和羟自由基的作用。
组合物的制备方法包括:
(1)花胶低聚肽的制备:将干花胶粗粉碎后,加入15~20倍质量体积的水制成匀浆液置于酶解罐中,然后加入花胶蛋白质质量的3~5%的复合蛋白酶,在50~55℃下酶解3~5小时,酶反应pH值控制在7.0~8.0,酶解结束后升温至90℃灭酶10分钟,得到花胶酶解液,离心去除杂质,然后采用截留分子量为3000Da的陶瓷膜进行膜分离,精制脱盐脱重金属,浓缩至可溶性固形物含量占浓缩液总质量的30~40%。所述复合蛋白酶的质量配比为:中性蛋白酶:胰蛋白酶:菠萝蛋白酶:风味蛋白酶=(4~6):(3~5):(1~3):(2~4)。花胶低聚肽指花胶酶解而来的复合物,进一步是指90%以上肽分子量<1000Da的花胶低聚肽,进一步含有重复的Gly-X-Y,进一步指含有抗氧化肽段序列NH2-Gly-Pro-Ala-Gly-Pro-Cys。
(2)海参多糖的制备:取海参原料制浆,得到海参浆料;加水,调节pH至6.5~8.0,加复合蛋白酶,酶解2~6小时;离心分离,收集上清液;采用超滤膜反复浓缩过滤,收集最终浓缩液,即为海参多糖浓液;将海参多糖浓液冷冻干燥后,填充到树脂柱中,先采用65%酒精过柱,再采用95%酒精过柱;取出树脂柱中的海参多糖,置于硅胶干燥箱中干燥,即得到精制海参多糖。
(3)轮叶党参低聚肽的制备:轮叶党参(Codonopsis lanceolata)样品粗粉碎处理后,加入10~15倍质量体积的水制成匀浆液置于酶解罐中,然后加入轮叶党参质量(以干重计)0.5~1.0%的纤维素酶,在45~50℃下酶解1~2小时,酶反应pH值控制在5.0~6.0;再加入轮叶党参蛋白质质量3~5%的复合蛋白酶,在50~60℃下酶解3~4小时,酶反应pH值控制在8.5~9.5,酶解结束后升温至90℃灭酶10分钟,得到轮叶党参酶解液,离心去除杂质,然后采用截留分子量为3000Da的陶瓷膜进行膜分离,精制脱盐脱重金属,浓缩至可溶性固形物含量占浓缩液总质量的30~40%。所述复合蛋白酶的质量配比为:木瓜蛋白酶:中性蛋白酶:碱性蛋白酶=(2~4):(1~2):(4~6)。轮叶党参低聚肽指轮叶党参酶解而来的复合物,进一步是指90%以上肽分子量<1000Da的轮叶党参低聚肽,进一步指含有抗氧化肽段序列NH2-Lys-Gly-Ser-Phe-Phe-Arg-Pro。
(4)取花胶低聚肽100-150份、雨生红球藻粉10-20份、海参多糖15-35份、葡萄籽提取物10-20份、人参低聚肽25-50份、透明质酸钠10-20份、酵母抽提物5-10份、轮叶党参低聚肽80-120份、维生素E粉5-10份、维生素C1-5份、茶多酚1-10份,加入反应釜中搅拌15-30分钟,然后进行喷雾干燥,得到粉剂,喷雾干燥条件为:进风温度130℃,温差为2℃;出风温度70℃,温差为1℃。
本发明还可以加工为片剂、丸剂、口服溶液,等,如片剂可进一步包含粘合剂,如麦芽糊精、羧甲基纤维素钠、明胶,且如有调味剂、甜味剂等,片剂可通过常用的混合、制粒或包衣的方法进行制备。
功效实验
抗氧化活性实验
通过测定花胶低聚肽(含有肽段序列NH2-Gly-Pro-Ala-Gly-Pro-Cys)和轮叶党参低聚肽(含有肽段序列NH2-Lys-Gly-Ser-Phe-Phe-Arg-Pro)对DPPH自由基和羟自由基的清除能力,验证其抗氧化活性。
将实施例1、实施例2、实施例3、实施例4中的花胶低聚肽和轮叶党参低聚肽分别喷雾干燥,记为花胶低聚肽1、2、3和轮叶党参低聚肽1、2、3,然后配置成10mg/mL浓度的花胶低聚肽样品溶液和轮叶党参低聚肽样品溶液,备用。
DPPH自由基清除能力测定:
取样品溶液2mL,加入2mL 1×10-4mol/L的DPPH溶液,混匀,37℃下避光反应20min,5000r/min离心8min,用95%的乙醇调零,然后在517nm波长处测其吸光度A1;空白样品为去离子水,吸光度记为A0。平行测定3次。计算DPPH自由基的清除率,公式为:DPPH自由基的清除率(%)=[1-(A1-A2)/A0]×100%
羟自由基清除能力测定:
取样品溶液1mL加入离心管中,然后依次分别加入2mL 1.8mmol/L的硫酸亚铁、1.5mL 1.8mmol/L水杨酸-乙醇、0.1mL 8.8mmol/L的H2O2,在37℃恒温培养箱中孵化30min。随后取200μL反应液于酶标仪的96孔板上,在510nm波长处测定其吸光度A1;空白样品为去离子水,吸光度记为A0。平行测定3次。计算羟自由基清除率,公式为:羟自由基清除率(%)=(A0-A1)/A0×100%
DPPH自由基的清除结果如图1所示,羟自由基的清除结果如图2所示,实验显示花胶低聚肽和轮叶党参低聚肽均对DPPH自由基和羟自由基有很好的清除效果,其中DPPH自由基清除率最高达92.18%,羟自由基清除率最高达94.15%,表明该工艺制备的花胶低聚肽和轮叶党参低聚肽有较好的抗氧化活性。
动物实验
通过动物实验探讨验证本发明组合物抗皮肤衰老的效果。
造模:每日给小鼠(3月龄,18-22g)颈背部皮下注射5%D-半乳糖1 000mg/kg,连续给D-半乳糖42d。同时给小鼠背部正中皮肤脱毛,暴露约3cm×5cm的皮肤,用短波紫外线(UVC)灯(峰波长值为265nm)和金属钨灯(长波紫外线UVA占70%)照射,功率150V/A。
每次照射量分别为UVC 56mJ/cm2(约2个最小红斑量)、UVA 2.86mJ/cm2,隔天1次,连续42d,制备小鼠皮肤衰老模型。
造模成功表观指标:干燥、粗糙、下垂,出现小细纹,松弛、变薄、面色晦暗无光泽。
造模成功病理指标:弹力纤维染色采用Weigert.s间苯二酚品红法,胶原纤维染色采用Mallory-Heidenhain.s改良法。真皮层厚度明显减小、真皮层胶原纤维及弹力纤维缺失、排列紊乱、结构松散,出现断裂、卷曲。
造模成功病理指标:血液中HYP、GSH-Px、SOD的水平降低,MDA的水平升高。
实验阶段,在5周实验模型建立后,开始给予动物发明的组合物,分为空白组、实施例1组、实施例2组、实施例3组。分别按照1.5g/kg/d灌胃小鼠。空白组正常喂食。
测定指标:
皮肤指标:水分含量、羟脯氨酸含量、透明质酸含量、弹性纤维含量。(见表1)
血清指标:MMP、GSH-Px、SOD、MDA。(见表2)
对比空白组,低、高剂量组小鼠皮肤水分、羟脯氨酸、透明质酸、弹性纤维的含量均明显升高,MDA明显降低,具有统计学意义(数据见表1和表2)。

根据抗皮肤衰老的实验数据可以看出,本发明的组合物在预防、延缓或改善皮肤衰老方面效果突出。
尽管本发明的描述是通过特定实施例进行的,但本领域技术人员应当理解的是,本发明也可以涵盖如文中所说的本发明的界限之内的其他实施方式。

1.一种含有花胶和轮叶党参低聚肽的抗皮肤衰老组合物,其特征在于,由以下组分组成:
其中:
花胶低聚肽含有抗氧化肽段序列NH2-Gly-Pro-Ala-Gly-Pro-Cys;
轮叶党参低聚肽含有抗氧化肽段序列NH2-Lys-Gly-Ser-Phe-Phe-Arg-Pro;
其中:花胶低聚肽基于如下方式制备而得:将干花胶粗粉碎后,加入15~20倍质量体积的水制成匀浆液置于酶解罐中,然后加入花胶蛋白质质量的3~5%的复合蛋白酶,在50~55℃下酶解3~5小时,酶反应pH值控制在7.0~8.0,酶解结束后升温至90℃灭酶10分钟,得到花胶酶解液,离心去除杂质,然后采用截留分子量为3000Da的陶瓷膜进行膜分离,精制脱盐脱重金属,浓缩至可溶性固形物含量占浓缩液总质量的30~40%;所述复合蛋白酶的质量配比为:中性蛋白酶:胰蛋白酶:菠萝蛋白酶:风味蛋白酶=(4~6):(3~5):(1~3):(2~4);
其中,轮叶党参低聚肽基于如下方式制备而得:轮叶党参样品粗粉碎处理后,加入10~15倍质量体积的水制成匀浆液置于酶解罐中,然后加入轮叶党参质量0.5~1.0%的纤维素酶,在45~50℃下酶解1~2小时,酶反应pH值控制在5.0~6.0;再加入轮叶党参蛋白质质量3~5%的复合蛋白酶,在50~60℃下酶解3~4小时,酶反应pH值控制在8.5~9.5,酶解结束后升温至90℃灭酶10分钟,得到轮叶党参酶解液,离心去除杂质,然后采用截留分子量为3000Da的陶瓷膜进行膜分离,精制脱盐脱重金属,浓缩至可溶性固形物含量占浓缩液总质量的30~40%,所述复合蛋白酶的质量配比为:木瓜蛋白酶:中性蛋白酶:碱性蛋白酶=(2~4):(1~2):(4~6)。
2.根据权利要求1所述的组合物,其特征在于,海参多糖基于如下方式制备而得:取海参原料制浆,得到海参浆料;加水,调节pH至6 .5~8 .0,加复合蛋白酶,酶解2~6小时;离心分离,收集上清液;采用超滤膜反复浓缩过滤,收集最终浓缩液即为海参多糖浓液;将海参多糖浓液冷冻干燥后,填充到树脂柱中,先采用65%酒精过柱,再采用95%酒精过柱;取出树脂柱中的海参多糖,置于硅胶干燥箱中干燥,即得到精制海参多糖。
3.根据权利要求2所述的组合物,其特征在于,海参多糖的纯度达到80%以上。
