本发明涉及中药技术领域,具体而言,涉及一种降糖药及其制备方法。
背景技术
糖尿病是一种以高血糖为特征的代谢性疾病,是威胁人类健康的常见慢性病,糖尿患者普遍存在于广大社会人群中。近年来,物质资源越来越丰富,而体力劳动量越来越少,肥胖和糖尿病患者人数逐年增加。对于糖尿病患者来说,严格控制血糖非常重要。糖尿病是威胁人类健康的常见慢性病,糖尿患者普遍存在于广大社会人群中,中国有将近1.3亿糖尿病患者,是糖尿病高发国家之一。近年来,物质资源越来越丰富,而体力劳动量越来越少,肥胖、糖尿病患者人数逐年增加。尿病初期患者主要出现饥饿、多食、体重下降、手脚麻痹、血压高、视力下降等临床症状。后期主要有失明、心衰、心肌梗塞、糖尿病足、脑梗塞脑出血等症状,极大降低了患者的生活质量。糖尿病是世界上主要的疾病和死亡的原因。
正常情况下,人体能够通过激素调节和神经调节这两大调节系统确保血糖的来源与去路保持平衡,使血糖维持在一定水平,但是在遗传因素如糖尿病家族史与环境因素如不合理的膳食、肥胖等的共同作用下,两大调节功能发生紊乱,就会出现血糖水平的升高。当血糖值高于正常范围即为高血糖。高血糖也是通常大家所说“三高”中的一高。另外“两高”分别是高血压和高脂血症。空腹血糖正常值在6.1mmol/L以下,餐后两小时血糖的正常值在7.8mmol/L以下,如果高于这一范围,称为高血糖。
一般情况下,身体通过激素调节和神经调节系统,使得血糖维持在正常水平。但会因为受到遗传因素、不合理的饮食和肥胖等使得调节功能发生紊乱,从而导致血糖水平升高。短时间和一次性的高血糖对身体没有严重损伤,但长期高血糖会使得全身各个组织器官发生病变,从而引起急慢性并发症。
长期血糖高会导致:1、伤害心脏,心脏冠状动脉主要为身体各个组织和器官供给新鲜血液,当冠状动脉狭窄时,会引起心肌梗死和冠心病。2、伤害大脑,大脑血管狭窄时可引起脑缺血或脑梗死,糖尿病患者患上脑血管疾病的风险比正常人高达好几倍,若患者出现不明原因头痛头晕以及头部跳痛等需立马就医,看看是否跟高血糖有关。3、伤害眼睛,持续性高血糖会引起微血管病变,导致视网膜发生病变,易引起增殖性视网膜疾病,从而导致视力下降或失明。4、伤害肾脏,高血糖可影响肾脏血管,增加尿液中白蛋白排泄量,导致大量白蛋白尿和血肌酐水平升高,最终引起肾功能衰竭,这种情况下需通过血液透析和肾移植来维持生命。
对于糖尿病的治疗,现有已经有许多化学药物被广泛应用。这些化学药物非机体主动吸收的物质,在给药以后会被机体迅速地排除,因此仅仅能够发挥有限的治疗价值。为了获得想要的治疗效果,就必须加大给药剂量,大剂量的质量药物又会引起机体的负反馈,降低药物的效果。比如,二甲双胍为大多数临床指南推荐的基础初始用药,是临床上广泛的降糖药物之一。但该药也伴随着恶心、呕吐、腹泻、低血糖和乳酸中毒等不良反应。中药具有多靶点,多途径,多环节等特点,可以从整体角度出发,调节机体机能,达到治本目的。
中药天然药材为天然存在的植物、动物药材,特别是植物药材和一般食物具有很好的相似性,容易为人体吸收利用,具有很好的亲和性。研究中药天然药材在降低血糖中的应用,具有很好的发掘潜力,有希望提供一种具有良好的友好性的天然药物组合物,特别是天然植物药材组合物。
天然植物药材在中药应用领域中要想发挥出真正的药效功能,还需要深入的研究分析,一方面要研究药物的治疗机理,另一方面要对药效成分和非药效成分加以区别,确定有效的成分,排出不良或无效的成分的干扰。在提升药物组合物的药效的同时,降低药物的不良反应影响等。
发明内容
本发明的目的在于提供一种降糖药,此降糖药具有降血糖效果好和使用安全的优点。
本发明的另一目的在于提供一种降糖药的制备方法,以获得此降糖药。
本发明解决其技术问题是采用以下技术方案来实现的。
一方面,本申请实施例提供一种降糖药,按重量份计,包括如下原料:桑叶10-50份、桑枝5-35份、地骨皮5-35份、水皂角5-35份、川芎5-35份和葛根3-43份。
另一方面,本申请实施例提供一种降糖药的制备方法,包括如下步骤:

本发明的降糖药具体是将其泡在水中,或者加入水中加热,用来泡脚,通过透皮给药的方式进行使用,能够极大提高药效,缩短起效时间,且泡脚能够促进血液循环,也能促进药物吸收,进而能够促进血糖降低。

需要说明的是,在不冲突的情况下,本申请中的实施例及实施例中的特征可以相互组合。下面将参考具体实施例来详细说明本发明。
桑叶:味甘、苦,性寒。归肺、肝经。有疏散风热,清肺润燥,清肝明目之效。用于风热感冒,肺热燥咳,头晕头痛,目赤昏花。《本草经疏》:“桑叶,甘所以益血,寒所以凉血,甘寒相合,故下气而益阴,是以能主阴虚寒热及因内热出汗。其性兼燥,故又能除脚气水肿,利大小肠,除风。经霜则兼清肃,故又能明目而止渴。发者血之余也,益血故又能长发,凉血故又止吐血。合痈口,罨穿掌,疗汤火,皆清凉补血之功也。”
桑枝:味微苦,性平。归肝经。有壮肺气,燥湿,滋肾水,通经,止咳除烦,消肿止痛之效。
地骨皮:味甘,性寒。归肺、肝、肾经。有凉血除蒸,清肺降火之效。用于阴虚潮热,骨蒸盗汗,肺热咳嗽,咯血,衄血,内热消渴。《本草正》:“地骨皮,枸杞根也,南者苦味轻,微有甘辛,北者大苦性劣,入药惟南者为佳。其性辛寒,善入血分,凡不因风寒而热在精髓阴分者最宜。此物凉而不峻,可理虚劳,气轻而辛,故亦清肺。”
水皂角:味甘、微苦,性平。归肝、肾、脾经。健脾利湿,止咳化痰。用于夜盲,慢性肾炎,咳嗽痰多,慢性便秘。《东北常用中草药手册》:“清热利尿,健胃通便。治水肿,肾炎,口渴,咳嗽,慢性便秘。”
川芎:味辛,性温。归肝、胆、心包经。有燥湿,活血行气,祛风止痛之效。用于月经不调,经闭痛经,症瘕腹痛,胸胁刺痛,跌扑肿痛,头痛,风湿痹痛。《日华子本草》:“治一切风,一切气,一切劳损,一切血,补五劳,壮筋骨,调众脉,破癥结宿血,养新血,长肉,鼻洪,吐血及溺血,痔瘘,脑痈发背,瘰疬瘿赘,疮疥,及排脓消瘀血。”
葛根:味甘、辛,性凉。归脾、胃经。有解肌退热,生津,透疹,升阳止泻之效。用于外感发热头痛、项背强痛,口渴,消渴,麻疹不透,热痢,泄泻;高血压颈项强痛。《本经疏证》:“葛根之用,妙在非徒如栝蒌但浥阴津,亦非徒如升麻但升阳气,而能兼擅二者之长,故太阳阳明合病,自下利者(葛根汤证),太阳被下,利遂不止,脉促喘汗者(葛根芩连汤证)咸用之。盖两者之利,为阳盛于外,不与阴交,阴遂不固而下溜,起其阴气,使与阳泱,得曳以上行,则非但使利止,并能使阳之遏于外者,随胃阳鼓荡面散矣。”
荔枝核:味甘、微苦,性温。归肝、肾经。有行气散结,祛寒止痛之效。用于寒疝腹痛,睾丸肿痛。
紫杉:味淡,性平。归肾经。有利尿消肿之效。用于肾炎浮肿,小便不利,糖尿病。
一年蓬:味苦,性凉。有清热解毒,抗疟之效。内服急性胃肠炎,疟疾;外用治齿龈炎,蛇咬伤。《浙江民间常用草药》:“消食,止泻,解毒,止血。”
河套大黄:味苦,性寒。有消食化滞,通腑泄热之效。用于食积不化,脘腹胀满,腹痛泄泻不爽,热结便秘。
都咸子:味甘,性平,无毒。归肺经。有润肺止咳,止渴,除烦之效。治咳逆,心烦,口渴。
竹节参:味甘、微苦,性温。归肝、脾、肺经。有滋补强壮,散瘀止痛,止血祛痰之效。用于病后虚弱,劳嗽咯血,咳嗽痰多,跌扑损伤。

实验例
(一)、刺激性实验
选择120只成年、健康、应使用成年、健康且皮肤无损伤的白色家兔,雌性和雄性各一半,雌性家兔是未孕且未曾产仔白色家兔单笼饲养,试验前白色家兔在实验动物房环境中适应性喂养7d,均分为12组,每组10只家兔。
分别取50g实施例1-12制备的降糖药,将其加入温度为100℃的水中,浸泡20min,获得试验液体。
试验前24h,将白色家兔背部脊柱两侧毛剪掉,去毛范围左、右各约3cm×3cm。取试验液体约0.5mL直接涂在皮肤上,然后用二层纱布(2.5cm×2.5cm)和一层玻璃纸覆盖,再用无刺激性胶布和绷带加以固定。另一侧皮肤作为对照。采用封闭试验,敷用时间为4h。试验结束后用温水清除残留受试物,观察对比皮肤情况。
无刺激:无红斑,无水肿。
轻微刺激:勉强可见轻微红斑或轻微水肿。
轻度刺激:明显红斑或皮肤水肿轮廓清晰。
中度刺激:重度红斑或皮肤隆起约1mm。
重度刺激:严重红斑或轻微焦痂,皮肤隆起超过1mm。

根据表1,可以看出本申请实施例制备的降糖药对皮肤刺激性弱,使用安全。
(二)、疗效观察
收集患有血糖高的患者,60例,其中,男性37例,女性23例,年龄在45-73岁。
将患者随机分为6组,每组10人,分别用实施例1、实施例4、实施例6、实施例7、实施例10和实施例12制备的降糖药进行治疗。
治疗方式:用50g降糖药浸泡在100℃的水中,待水温降温至人体可以接受的温度后,将脚方进行,保温浸泡30min,持续3个月,观察患者血糖情况。
在治疗前,患者的空腹血糖均大于≥7.0mmol/L,治疗后,统计患者血糖情况,结果如表2所示。

根据表2,可以看出,患者经过降糖药的治疗后,空腹血糖降低明显,证明本发明具有较好的降糖作用;对比来看,可以看出实施例4的降糖效果最佳,这说明实施例4制备的降糖药对于血糖的降低效果最佳。

本发明的降糖药具体是将其泡在水中,或者加入水中加热,用来泡脚,通过透皮给药的方式进行使用,能够极大提高药效,缩短起效时间,且泡脚能够促进血液循环,也能促进药物吸收,进而能够促进血糖降低。

1.一种降糖药,其特征在于,按重量份计,包括如下原料:桑叶10-50份、桑枝5-35份、地骨皮5-35份、水皂角5-35份、川芎5-35份和葛根3-43份。
2.根据权利要求1所述的一种降糖药,其特征在于,所述降糖药按重量份计,包括如下原料:桑叶20-40份、桑枝10-30份、地骨皮10-30份、水皂角10-30份、川芎10-30份和葛根8-38份。
3.根据权利要求1所述的一种降糖药,其特征在于,所述降糖药按重量份计,包括如下原料:桑叶30份、桑枝20份、地骨皮20份、水皂角20份、川芎20份和葛根18份。
4.根据权利要求1所述的一种降糖药,其特征在于,所述降糖药还包括如下重量份的原料:荔枝核5-25份、紫杉10-40份、一年蓬10-40份、河套大黄5-35份、都咸子10-35份和竹节参5-35份。
5.根据权利要求4所述的一种降糖药,其特征在于,所述降糖药按重量份计,包括如下原料:桑叶20-40份、桑枝10-30份、地骨皮10-30份、水皂角10-30份、川芎10-30份、葛根8-38份、荔枝核10-20份、紫杉15-35份、一年蓬15-35份、河套大黄10-30份、都咸子15-30份和竹节参10-30份。
6.根据权利要求4所述的一种降糖药,其特征在于,所述降糖药按重量份计,包括如下原料:桑叶30份、桑枝20份、地骨皮20份、水皂角20份、川芎20份和葛根18份、荔枝核15份、紫杉25份、一年蓬25份、河套大黄20份、都咸子23份和竹节参20份。
7.如权利要求1-6任意一项所述的一种降糖药的制备方法,其特征在于,包括如下步骤:
将原料破碎后混合,得到降糖药成品。
8.根据权利要求7所述的一种降糖药的制备方法,其特征在于,所述原料破碎后的尺寸为50-200目。
