Description

Preparation for promoting digestion of animals and preparation method thereof
Technical Field
The invention relates to the technical field of feed additives, in particular to a preparation for promoting digestion of animals and a preparation method and application thereof.
Background
At present, the most widely used method for treating animal clinical diseases is that the problems of antibiotics, bacterial drug resistance and the like are more serious, and the situation that the clinical treatment effect is not obvious and even no drug is available appears. The long-term treatment with large dose of antibiotics not only causes serious burden to the liver and kidney of animals, but also induces a series of problems of aplastic anemia, flora disorder and the like. Antibiotics cause drug resistance problem, so that the generation of super bacteria threatens not only animal medical industry but also human public health safety.
The hormone medicine is also a common medicine for animal clinic, and has the characteristics of quick response, low dosage and the like, but the problems of endocrine disorder, immunosuppression, metabolic disorder and the like of animals caused by the hormone medicine become more serious day by day, and even serious iatrogenic diseases are caused, such as skin fragility syndrome caused by long-term use of glucocorticoid medicine.
With the continuous occurrence of reports of toxic and side effects of chemical drugs and the like, when animal owners treat diseases of animals, the animal owners are not limited to the treatment effect of the drugs, and the animal owners also have toxic and side effects caused by the loss of the drugs to organisms, particularly when the animals are sick and need long-term treatment, and the chemical drugs only can play a treatment role temporarily and cannot achieve the effects of long-term regulation and health care. In view of the above situations, the advantages of traditional Chinese medicine in animal clinical treatment are slowly highlighted, and the unique theoretical system and the drug treatment characteristics of traditional Chinese medicine are in disagreement with the requirements of animal medical industry at present. In view of the above problems, the present invention aims to disclose an animal digestion promoting preparation and a preparation process thereof, wherein the preparation has more excellent stability and thickening property and obviously improved palatability compared with the traditional animal digestion promoting preparation. In the traditional animal digestion-promoting preparation, for example, sodium carboxymethylcellulose (NaCMC) is used as a stabilizer and a thickening agent, the NaCMC is greatly influenced by temperature and pH value, the preparation cannot be stabilized when the addition amount of the NaCMC is small, and researches report that the absorption of trace elements is influenced by the NaCMC.

To the extent that the terms "includes," including, "" comprises, "and" having "or variants thereof are used in either the detailed description and/or the claims, these terms are intended to be inclusive in a manner similar to the term" comprising.
As used herein, "about" refers to a value within a range of + -10% of one particular value. For example, "about 20" includes ± 10% of 20, or from 18 to 22, including 18 and 22.
As described in the background section, the existing common drugs for preventing and/or treating animal dyspepsia and inappetence have obvious toxic and side effects, are easy to generate drug resistance, and easily cause the problems of animal endocrine disorder, immunosuppression and metabolic disorder. Experiment of drug effect
Experiment one-Individual test
The separate test means that the test dogs and cats received the food of one of the oral suspension liquids prepared in preparation example 4 without any other preparation.
Number of experimental dogs: 20 dogs and cats of different ages
Wherein the amounts of the suspension oral liquid samples prepared in preparation example 4 provided for the test were: and (4) the cat: 5g, dog: 15 g.
Days of the experiment: 5 days
The experimental method comprises the following steps: and providing a food basin for the experimental dogs and cats at the same time, and placing a known amount of samples to be tested in the food basin. The amount of feed taken on the sample during one feeding session (9: 00-15: 00) was weighed and recorded.
And the feed intake (%) is the sample feed amount (g)/total sample supply amount (g). times.100% as shown in the formula
And calculating the feed intake rate.
Wherein: the food bowls are placed and exchanged every other day.
The results of the independent tests show that (see figure 2), the feed intake rate of the suspension oral liquid prepared by the preparation example 4 is more than 94%, and the recommended dosage in the specification can be basically eaten; but differences between the races cannot be determined from the feed intake results, palatability between the product and the races can be further tested by the first bite preference test.
Experiment two-first Port preference test
Meanwhile, two kinds of oral liquid are provided for experimental animals, and the animals are observed to eat which kind of oral liquid firstly in the first bite. This test is primarily directed to the first impression of the smell or appearance of food by the animal.
Number of experimental dogs: 20 dogs and cats of different ages
Wherein the amount of the suspension oral liquid sample prepared in preparation example 4 and the amount of the Haka khauso water P-O nutrient solution sample provided for the test are both: and (4) the cat: 5g, dog: 15 g.
Days of the experiment: five days
The experimental method comprises the following steps: two basins were provided for the test dogs and cats at the same time, each basin containing the same amount of different amounts of the suspended oral liquid, and the first choice of each test dog and cat was observed and recorded.
Wherein: the food bowls are placed and exchanged every other day.
The results are shown in Table 1.
Table 1 results of two suspension oral liquid animal palatability tests:
Figure BDA0003374869860000151
wherein: A. a suspension oral liquid prepared from preparation example 4 (without adding sodium carboxymethylcellulose); B. hai Jia local tyrant water P-O nutrient solution (with sodium carboxymethyl cellulose added).
As can be seen from table 1, in the feeding experiment lasting five days, the suspension oral liquid a prepared by preparation example 4 was selected for the first bite of most of the test dogs and cats; the feed intake of the oral suspension liquid prepared in preparation example 4 by dogs and cats is continuously higher than that of Hai coffee local tyrant water P-O nutrient solution B, and the consumption rate of the oral suspension liquid A prepared in preparation example 4 is stable for more than 93 percent, and the continuous feed intake is good. The first taste preference test result shows that the flavor presented by the suspension oral liquid A prepared in the preparation example 4 is the flavor which is liked by dogs and cats, which is probably caused by the unique flavor brought by the preparation containing the choerospondias axillaris fruit in a specific weight part ratio.
Experiment III-viscosity stability test
A sample of the suspension oral liquid was prepared according to preparation example 4, and its viscosity (25 ℃ C.) was measured.
Samples of the suspension formulation of the raw formulation of Geofur (self-developed by the Applicant company, Lilin Biotechnology Ltd.) were prepared and the viscosity (25 ℃) was measured.
After the two samples were sealed and allowed to stand for three months, their viscosities were measured again (15 ℃).
The results are shown in Table 2.
Table 2 viscosity test results for two kinds of oral suspensions
Figure BDA0003374869860000161
As can be seen from Table 2, the Choerospondias axillaris suspension formulation prepared by preparation example 4 was stable in viscosity for 3 months, and no matter whether the temperature was changed, it was revealed that the system was less affected by the ambient temperature. However, the viscosity of the galenical suspension oral preparation is obviously changed by temperature, and the stability is relatively low.
In the process of pouring the two preparation samples on white paper respectively, the invention discovers that: the choerospondias axillaris suspension preparation prepared in preparation example 4 is stable and uniform in liquid state after 3 months, good in fluidity and free from caking and lumping. However, the suspension formulation of the raw galenical formulation uses sodium carboxymethylcellulose as a thickening agent, the viscosity of the system is greatly changed by the influence of the ambient temperature, and the system also has the phenomenon of little caking unevenness under the condition of low ambient temperature, so that the viscosity of the suspension formulation is increased by reducing the indoor temperature by 10 ℃ within 3 months, and little caking particles exist, and the liquid state is uneven (as shown by arrows in fig. 3).
