Abstract
The invention discloses a cosmetic composition with whitening, moisturizing and antioxidant effects and a preparation method thereof, wherein cell walls of raw materials of radix rehmanniae, burdock and black bean are damaged by biological enzyme, so that active substances in raw material cells are easier to extract by a solvent, and then the biological enzyme is inactivated by using ethanol with higher concentration, thereby avoiding the problems of degradation of the active substances in the raw materials and energy waste caused by enzyme inactivation due to high-temperature heating in the extraction process of the traditional enzyme method, and better preserving the active substances in the raw materials; the cosmetic composition can be widely applied to cream emulsion and water as a cosmetic raw material, has a wide application range and is convenient to use, and the traditional ancient formula adopts a medicinal liquor oral form, so that the use limitation is great; the invention has practical efficacy experiment as an adjuvant and stronger persuasion compared with the ancient formula. Description
Technical Field

step 1: adding 5-10 times of deionized water into 1-2 parts of radix rehmanniae recen, 2-4 parts of burdock and 6-8 parts of black bean according to a proportion, adding 1-3% of cellulase and 1-3% of pectinase, carrying out enzymolysis for 1-2 hours at 30-40 ℃, then adding 5-10 times of 95% ethanol, carrying out water bath for 5-7 hours at 50-70 ℃, filtering with a 1-3 mu m filter paper plate to obtain a filtrate, and detecting the solid content of the filtrate; the burdock and the black bean contain abundant pectin, so that cellulase and pectinase are required to be used together;
step 2: concentrating the filtrate obtained in the step 1 until the filtrate is 1/3-1/5 of the original weight, adding an organic solvent to make the total amount of the organic solvent reach 30-50% of the stock solution, and preparing the stock solution;
and step 3: using 20-30% of organic solvent aqueous solution to prepare the stock solution according to the ratio of 1: diluting at ratio of 5-1:10, and filtering with 0.22-0.45 μm filter membrane to obtain the cosmetic composition.

step 2: concentrating the filtrate obtained in the step 1 until the filtrate reaches 1/5 of the original weight, adding dipropylene glycol to enable the total amount of the dipropylene glycol to reach 50% of the original liquid, and preparing the original liquid;
and step 3: the stock solution was diluted with 30% dipropylene glycol aqueous solution at a ratio of 1:10, and filtered through a 0.45 μm filter to obtain the cosmetic composition.

Assay for inhibition of tyrosinase Activity by SNH Complex
1. The purpose is as follows: the half inhibitory concentration of the SNH complex on tyrosinase activity was determined.

Sample preparation: buffer solution: k2HPO4(8.71g/500mL)+KH2PO4(6.805g/500mL)→pH=6.80 (±0.02)
Substrate: about 0.3mg/mL of L-tyrosine
A spectrophotometer: uv, 480nm
3. The method comprises the following steps:
(1) enzyme concentration selection:
taking a small amount of enzyme, dissolving the enzyme by using a buffer solution, wherein the color is brown; adding 500. mu.L of (L-tyrosine (0.3 mg/mL)), 500. mu.L of buffer solution, 450. mu.L of pure water and 50. mu.L of enzyme into a 1.5mL centrifuge tube; before adding enzyme, at 37 deg.C for 20 min; after the addition of the enzyme, the absorbance was measured at a wavelength of 480nm in a spectrophotometer at 37 ℃ for 10min (enzyme addition-test). The enzyme concentration was adjusted to give an Absorbance (ABS) of between 0.45 and 0.55. The enzyme solution at this concentration was used as an experimental enzyme solution.
