and step 3: stirring the mixture for enzymolysis for 1-3 hours at 35-40 ℃, destroying cell walls of the raw materials, then adding 1000-1500 parts of 95% ethanol, and mixing uniformly; the cell walls of the raw materials are damaged, so that the subsequent extraction by using an ethanol water solution is convenient, the extraction efficiency is improved, and the extraction time is shortened; the ethanol is added to inactivate and denature the cellulase and the pectinase which are added before, so that the defects of reduction of active substances of an extracting solution and deepening of color caused by long-time high-temperature inactivation after enzyme extraction in the traditional enzyme extraction technology are overcome;
and 4, step 4: the mixture is bathed in water for 3 to 5 hours at the temperature of 60 to 70 ℃;
and 5: filtering the mixture to 1 μm, detecting solid content, concentrating at 70 deg.C until no alcohol exists, diluting with butanediol water solution, filtering, and bottling to obtain the final product.
Further, the enzymes added in the step 2 are cellulase and pectinase, the dosage of the cellulase is 0.2-0.5% of the mass of the substrate, the dosage of the pectinase is 0.2-0.5% of the mass, the cellulase activity index is more than or equal to 3500u/g, and the pectinase activity index is 60000u/g.
Further, in the step 5, when the solution is concentrated to be free of alcohol at 70 ℃, the solution is subjected to rotary evaporation at 70 ℃ by a rotary evaporator to obtain a solution, and the solution is diluted to 10 times by using a butanediol aqueous solution.
Application of a cosmetic composition containing peach blossom and safflower four-ingredient decoction with skin whitening, antioxidant and anti-wrinkle effects in cosmetics is provided.
The composition can be made into any selected dosage form of cream, lotion, and smoothing toner.
Compared with the prior art, the invention has the following beneficial effects:
1. according to the invention, the cellulase and the pectinase are adopted to destroy the cell wall of the material, the extraction efficiency is improved, the cellulase and the pectinase are inactivated by using the ethanol with higher concentration, and the reduction of active ingredients and energy loss caused by high-temperature inactivation are ingeniously avoided.
2. Compared with the traditional water extraction and decoction, the method utilizes the ethanol with higher concentration, and the ethanol aqueous solution has stronger penetrability, higher extraction efficiency and higher content of active ingredients of the extracting solution.
3. According to the invention, the prepared rehmannia root, the Chinese angelica, the white peony root, the peach seed, the ligusticum wallichii and the safflower are organically combined, the whitening and antioxidation effects are achieved through the blood circulation promoting and blood stasis removing effects of the prepared rehmannia root, the Chinese angelica, the white peony root, the peach seed, the ligusticum wallichii and the safflower, the raw materials are all traditional Chinese medicines, compared with products on the market, the components are safer and more effective, the application range is wider, no toxic or side effect is caused to a human body, the requirements of people on the safety of cosmetics are met, and the application prospect is wide.

Detailed Description
Example 1
A peach red four-ingredient soup cosmetic composition with whitening, antioxidant and anti-wrinkle effects comprises the following components: according to the weight portion, the traditional Chinese medicine composition comprises 15 portions of prepared rehmannia root, 15 portions of angelica, 10 portions of white peony root, 9 portions of peach seed, 8 portions of ligusticum wallichii and 6 portions of safflower.
The cosmetic composition is prepared by the steps of:
step 1: mixing radix rehmanniae Preparata, radix Angelicae sinensis, radix Paeoniae alba, semen Persicae, rhizoma Ligustici Chuanxiong and Carthami flos at a certain proportion;
and 2, step: adding the raw materials into 1000 parts of pure water, sequentially adding 0.2% of cellulase and 0.2% of pectinase, wherein the cellulase activity index is 3500u/g, and the pectinase activity index is 60000u/g, and uniformly mixing;

and 4, step 4: the mixture is bathed in water for 3 hours at the temperature of 60 ℃;
and 5: filtering the mixture to 1 μm, detecting solid content, rotary concentrating at 70 deg.C with rotary evaporator until no alcohol exists, diluting with butanediol water solution at a ratio of 10 times, filtering, and bottling to obtain the final product.
Example 2
A peach blossom and safflower four-ingredient soup cosmetic composition with whitening, antioxidant and anti-wrinkle effects comprises the following components: according to the weight portion, 20 portions of prepared rehmannia root, 15 portions of Chinese angelica, 15 portions of white peony root, 10 portions of peach kernel, 10 portions of ligusticum wallichii and 8 portions of safflower.
The cosmetic composition is prepared by the steps of:
step 1: mixing radix rehmanniae Preparata, radix Angelicae sinensis, radix Paeoniae alba, semen Persicae, rhizoma Ligustici Chuanxiong and Carthami flos at a certain proportion;
and 2, step: adding the raw materials into 1500 parts of pure water, sequentially adding 0.5% of cellulase and 0.5% of pectinase, wherein the activity index of the cellulase is 3550u/g, the activity index of the pectinase is 60000u/g, and uniformly mixing;
and step 3: stirring and carrying out enzymolysis on the mixture for 3 hours at the temperature of 40 ℃, destroying cell walls of the raw materials, facilitating subsequent extraction by using an ethanol water solution, improving the extraction efficiency and shortening the extraction time, then adding 1500 parts of 95% ethanol, and uniformly mixing to inactivate and denature the previously added cellulase and pectinase, thereby avoiding the defects of reduction of active substances of an extracting solution and deepening of color caused by long-time high-temperature inactivation after enzyme extraction in the traditional enzyme extraction technology;
and 4, step 4: the mixture was water-bathed for 5 hours at 70 ℃;
and 5: filtering the mixture to 1 μm, detecting solid content, rotary concentrating at 70 deg.C with rotary evaporator until no alcohol exists, diluting with butanediol water solution at a ratio of 10 times, filtering, and bottling to obtain the final product.
Example 3
A peach red four-ingredient soup cosmetic composition with whitening, antioxidant and anti-wrinkle effects comprises the following components: 15 parts of prepared rehmannia root, 10 parts of Chinese angelica, 10 parts of white paeony root, 8 parts of peach seed, 8 parts of szechuan lovage rhizome and 6 parts of safflower.
A cosmetic composition with skin whitening, antioxidant and anti-wrinkle effects, TAOHONGSIWUTANG, is prepared by the following steps:
step 1: mixing radix rehmanniae Preparata, radix Angelicae sinensis, radix Paeoniae alba, semen Persicae, rhizoma Ligustici Chuanxiong and Carthami flos at a certain proportion;
step 2: adding the raw materials into 1200 parts of pure water, sequentially adding 0.4% of cellulase and 0.4% of pectinase, wherein the activity index of the cellulase is 3540u/g, the activity index of the pectinase is 60000u/g, and uniformly mixing;
and step 3: stirring and carrying out enzymolysis on the mixture for 2 hours at the temperature of 37 ℃, destroying cell walls of the raw materials, facilitating subsequent extraction by using an ethanol water solution, improving the extraction efficiency and shortening the extraction time, then adding 1200 parts of 95% ethanol, and uniformly mixing to inactivate and denature the previously added cellulase and pectinase, thereby avoiding the defects of reduction of active substances of an extracting solution and deepening of color caused by long-time high-temperature inactivation after enzyme extraction in the traditional enzyme extraction technology;
and 4, step 4: the mixture was water-bathed for 4 hours at 65 ℃;
and 5: filtering the mixture to 1 μm, detecting solid content, rotary concentrating at 70 deg.C with rotary evaporator until no alcohol exists, diluting with butanediol water solution at a ratio of 10 times, filtering, and bottling to obtain the final product.
According to the invention, the cell walls of the materials are destroyed by adopting the cellulase and the pectinase, the extraction efficiency is improved, the cellulase and the pectinase are inactivated by using the ethanol with higher concentration, and the reduction of active ingredients and energy loss caused by high-temperature inactivation are ingeniously avoided; compared with the traditional water extraction and decoction, the ethanol with higher concentration has stronger penetrability of an ethanol aqueous solution, higher extraction efficiency and higher content of active ingredients in the extracting solution; the prepared rehmannia root, the Chinese angelica, the white paeony root, the peach seed, the ligusticum wallichii and the safflower are organically combined, the whitening and antioxidation effects are achieved through the blood circulation promoting and blood stasis removing effects of the prepared rehmannia root, the Chinese angelica, the white paeony root, the peach seed, the ligusticum wallichii and the safflower, the raw materials are all traditional Chinese medicines, compared with products on the market, the components are safer and more effective, the application range is wider, no toxic or side effect is caused to a human body, the requirements of people on the safety of cosmetics are met, and the application prospect is wide.
To better illustrate the beneficial effects of the present invention, efficacy evaluation experiments were performed:
experiment for removing DPPH free radical by using Taohong Siwu decoction
1. Reagent preparation
(1) 3.943mgDPPH is made up to 50mL with 95% ethanol
(2) The vitamin E of about 10mg is added with 95 percent ethanol to reach the constant volume of 100mL
(3) Taohong Siwu Tang-2
2. Experimental procedure
(1) UV zero calibration, 95% ethanol as blank
(2) Preliminary experiments (DPPH concentration selection)
Shaking 500 μ L DPPH +1000 μ L95% ethanol for 10s, standing for 10min, and measuring absorbance (absorbance at 520nm of 0.45-0.55, can be used as DPPH solution for the following experiment)
(3) Determination of vitamin E
Adding DPPH or 95% ethanol, shaking for 10s, standing for 10min, and measuring absorbance
(4) Experiment of
Adding DPPH or 95% ethanol, shaking for 10s, and standing for 10min
(5) The absorbance at 520nm was measured, and a table was prepared and plotted to obtain IC50 values.
Calculating the formula: inhibition rate (%) = [1- (S-SB)/(C-CB) ] = 100
3. Results
The DPPH radical scavenging rate of Taohong Siwu decoction is shown in FIG. 1.
As a result: not only active oxygen in the living body but also a system capable of suppressing excess active oxygen in the skin and removing the generated active oxygen is required for sedation and inhibition of skin aging. The concentration-dependent inhibition of free radicals was confirmed in DPPH experiments with an IC50 value of about 5.5uL/mL.
Therefore, it was judged that the peach blossom decoction of four ingredients-2 has high antioxidant activity and is recommended as a cosmetic material having anti-aging effect.
Evaluation of whitening efficacy of Taohong Siwu decoction (cell experiment)
1. Purpose of the experiment
In order to evaluate the whitening effect of the Taohong Siwu decoction, the inhibition effect of the melanin generation of cells was evaluated after quantifying the amount of melanin in the melanin generation cells or the total melanin amount inside and outside the cells.
2. Sample information
Peach red Siwu decoction (TSS-01070722)
3. Experimental method
1) Evaluation of cytotoxicity (MTT assay)
The 6-well plate was inoculated with B16 melanoma cells and cultured, the culture medium of the cultured cells was replaced with a new one, and the samples were treated at different concentrations and cultured. After the completion of the culture, the cells were reacted with an MTT solution, the reaction was completed, the supernatant was removed, and MTT formazan crystals formed by dissolving them in dimethyl sulfoxide (DMSO) were added to measure the absorbance using a microplate reader.
2) Measurement of amount of melanin inside and outside cell
The 6-well plate was inoculated with B16 melanoma cells and cultured, the culture medium of the cultured cells was replaced with a new one, and the samples were treated at different concentrations and cultured. After completion of the culture, the medium of the cells was recovered, transferred to a 96-well plate, and the amount of extracellular melanin was measured by measuring absorbance. The plate from the recovered medium was washed with PBS, treated with NaOH, and the cells were transferred to a tube. After the completely lysed cells were transferred to a 96-well plate, the absorbance was measured to determine the amount of melanin in the cells.
4. Results of the experiment
1) Cytotoxicity
The cytotoxicity results of the peach blossom decoction for whitening test are shown in figure 2.
Cytotoxicity appeared from 3%.
2) Rate of melanin production per cell
The results of the melanin production rate per cell of TAOHONGSIWU decoction are shown in FIG. 3.
The concentration dependence inhibits the production of melanin per cell.
3) Unit cell melanin secretion rate
The results of melanin secretion rate per cell of Taohong Siwu decoction are shown in FIG. 4.
5. Conclusion
The TAOHONGSIWU decoction can inhibit skin pigmentation by inhibiting melanin generation at full concentration, and can be used as cosmetic material for whitening skin.
Evaluation of anti-wrinkle efficacy of Taohong Siwu decoction (cell experiment)
1. Purpose of experiment
Experiments were performed to evaluate the anti-wrinkle efficacy of the pink decoction of four ingredients.
2. Experimental methods
1) Evaluation of cytotoxicity (MTT assay)
After 24 hours of cell culture, the cells were inoculated on a 24-well plate, and then replaced with a serum-free medium, treated at different concentrations, and cultured. After completion of the culture, the cells were replaced with a medium containing MTT solution to carry out a reaction, and after completion of the reaction, the supernatant was removed, and MTT formazan crystals formed by dissolution in dimethyl sulfoxide (DMSO) were added, and then absorbance was measured at a wavelength of 570nm using a microplate reader.
2) Method for evaluating expression of genetic gene related to wrinkle resistance
Human fibroblasts (dermal fibroplast) were seeded in a 60 mm culture dish and cultured, after washing with buffer, UVA irradiation, sample treatment after replacement with serum-free medium and culture in a 37 ℃ 5% CO2 microbial incubator. After the RNA isolation was quantified, cDNA was synthesized and subjected to Real-time PCR (Applied Biosystems kit, rapid Real-time quantitative PCR instrument).
3. Results of the experiment
1) Evaluation of cytotoxicity (MTT assay)
The anti-wrinkle test cytotoxicity results of Taohong Siwu Tang are shown in FIG. 5.
Cell deformation was observed at 5% concentration in the pink decoction of four drugs, the maximum concentration of this experiment was performed to 4%.
2) Evaluation of expression of genetic genes involved in wrinkle resistance
The result of inhibition of MMP-1 gene expression by Taohong Siwu decoction is shown in FIG. 6.
The effect of inhibiting MMP-1 expression was confirmed in Taohong Siwu decoction at a concentration of 2%.
4. Conclusion
In order to use the pink-red four-ingredient soup as a cosmetic raw material, the anti-wrinkle effect evaluation was performed in cells. Research shows that ultraviolet rays (UVA) decompose matrix components such as cells of the dermis of the skin and elastic fibers by regulating the synthesis of various MMPs (MMPs) through specific decomposition of the main matrix components Collagen I, III and V of the dermis and the main matrix components Collagen IV, VII, proteolycans, laminin and the like of the basement membrane, and induce the generation of skin wrinkles and photoaging.
Therefore, the Taohong Siwu Tang has anti-aging effect judged by the activity of MMP-1 which induces skin wrinkles and photoaging in human skin fibroblasts which block ultraviolet irradiation at a concentration of 2%.
By combining the three efficacy test experiments, the invention has remarkable whitening, anti-oxidation and anti-wrinkle effects.
The above-mentioned embodiments are merely illustrative of the preferred embodiments of the present invention, and do not limit the scope of the present invention, and various modifications and improvements made to the technical solution of the present invention by those skilled in the art without departing from the spirit of the present invention should be included in the protection scope defined by the claims of the present invention.
Claims (7)
1. A peach blossom and safflower four-ingredient soup cosmetic composition with whitening, antioxidant and anti-wrinkle effects is characterized in that: the composition comprises the following components: prepared rehmannia root, chinese angelica, white peony root, peach kernel, szechuan lovage rhizome and safflower, wherein the prepared rehmannia root is 15 to 20 parts, the Chinese angelica is 10 to 15 parts, the white peony root is 10 to 15 parts, the peach kernel is 8 to 10 parts, the Szechuan lovage rhizome is 8 to 10 parts and the safflower is 6 to 8 parts by weight.

The peach blossom four-ingredient decoction cosmetic composition with whitening, antioxidant and anti-wrinkle effects according to claim 1, which is characterized in that: the composition comprises the following components: according to the weight portion, the traditional Chinese medicine composition comprises 15 portions of prepared rehmannia root, 15 portions of angelica, 10 portions of white peony root, 9 portions of peach seed, 8 portions of ligusticum wallichii and 6 portions of safflower.
