背景技术
近十几年来,我国对新型肥料的研究不断深入,控释肥、缓释肥、生物有机肥等多种新型肥料不断研制成功,对促进粮食增产发挥了积极的促进作用,由于农民的习惯性施肥方法,忽视了抗病毒肥料的施用,农业和肥料研究单位对生物质抗病毒肥料的研究也受到了较大的影响,特别是近年来重茬种植导致的病原菌积累越来越多,致使近几年农作物的病虫害日趋严重,研究一种抑制或杀死多种作物病原菌的新型生物质防病肥料是十分必要的。
发明内容
针对现有技术中存在的市场空白等问题,本发明提供了一种含枯草芽孢杆菌生防尿素的生产方法,本发明针对目前农民施肥习惯及土壤中病原菌积累的情况,根据枯草芽孢杆菌的生理特征,制备了一种新型的生防尿素。
本发明为了实现上述目的所采用的技术方案为:
本发明提供了一种含枯草芽孢杆菌生防尿素的生产方法,包括以下步骤:
(1)将纯种枯草芽孢杆菌液涂布于琼脂平板上,网状划线,置于恒温培养箱内,恒温培养,培养基表面形成菌落,从平板中挑选出长势优良的单菌落,用接种环挑起移种到装有25ml液体培养基的摇甁中,置于摇床内,恒温振荡得到优良菌种液,再涂布琼脂平板上,恒温培养,从中挑选长势良好的单菌落,移种到液体培养基中,恒温培养,得到二代优良菌种,三次涂布于琼脂平板上,恒温培养,从平板中挑选出最为饱满的单菌落,移种到液体培养基中,培养,即得到三代优良菌种;
(2)将得到的三代优良菌种移植到种子培养基中,进行种子培养,即得到枯草芽孢杆菌发酵种子液;
(3)将枯草芽孢杆菌发酵种子液接种到含有发酵培养基的发酵罐,火焰保护接种后,启动搅拌机,发酵,即得到枯草芽孢杆菌发酵液;
(4)将枯草芽孢杆菌发酵液再经三效蒸发浓缩至75%,得浓缩发酵也,蒸发压力控制在-0.096Mpa,温度86℃,用高压尿素液泵将尿素液打入造粒塔顶部旋流喷射头,尿素液喷洒为细雾状,同时用高压计量泵,将浓缩发酵液打入造粒塔顶另一旋流喷雾器,喷洒出的细雾状枯草芽孢杆菌浓缩发酵液和细雾状尿液混合,在下落100米的过程中,水分蒸发,混合的细微颗粒聚集成大颗粒,枯草芽孢杆菌被包裹在尿素颗粒中,处于休眠状态,即得到含枯草芽孢杆菌生防尿素。
进一步的,步骤(1)中,所述琼脂平板培养基配方:牛肉膏30g/L;蛋白胨10g/L,NaCL5g/L,琼脂15g/L;所述液体培养基配方:葡萄糖20g/L,蛋白胨15g/L, NaCL5g/L,牛肉膏5g/L。
进一步的,步骤(1)中,所述恒温培养或恒温振荡的条件为56℃下培养或振荡24h。

进一步的,步骤(4)中,所述浓缩发酵液占尿素液重量的0.2%-0.5%。
本发明将枯草芽孢杆菌菌体包含于尿素颗粒中,施入土壤后,菌体在养分丰富的土壤中迅速生长繁殖,抑制和杀死病原菌。将市售的枯草芽孢杆菌纯种,经平板-液体培养-平板-液体培养三代驯化筛选,得到枯草芽孢杆菌优良菌种,利用优良菌种进行发酵,得到的枯草芽孢杆菌发酵液,负压浓缩,通过高塔喷雾造粒将芽孢杆菌菌体包含于尿素颗粒中,制得含枯草芽孢杆菌生防尿素。
本发明的有益效果为:
(1)本发明制备的含枯草芽孢杆菌的尿素施入土壤中,其中休眠状态的菌体,得到充足的水分,氧气和养分,特别在夏季高温时期,会迅速生长繁殖,在生长过程中会产生枯草素、多粘素、短杆素等活性物质,这些活性物质对植物致病菌有显著的抑制作用,在生长过程中还会产生溶菌物质,从而溶解病原菌的菌丝体,致使菌丝体断裂解体或细胞质消解致死。
(2)本发明制备的生防尿素,经大田实验,枯草芽孢杆菌菌株对不同作物的众多真菌和细菌病毒具有非常有效的抑制和灭除作用,如粮食作物中的水稻纹枯病,稻瘟病、小麦纹枯病、豆类根腐病,在蔬菜和水果中,如草莓叶斑病,番茄叶病,黄瓜枯萎病,苹果霉心病,柑橘青霉病,油桃褐腐病等多种多样的病毒。在不改变农民施肥习惯的情况下,将抗菌微生物包含于尿素颗粒中,达到生物防治农作物病害的作用。
具体实施方式
下面通过具体的实施例对本发明的试验方案做进一步的解释和说明。
本发明所使用的枯草芽孢杆菌购自环凯微生物科技有限公司。
实施例1
(1)枯草杆菌在液体培养基中生长繁殖尤其迅速,将纯种枯草芽孢杆菌液分别涂布于6块琼脂平板上,网状划线,置于恒温培养箱内,56℃下恒温培养24h,培养基表面形成菌落,从6块平板中挑选出长势优良的单菌落,用接种环挑起移种到装有25ml液体培养基的摇甁中,置于摇床内,56℃恒温振荡24h得到优良菌种液,再涂布6块琼脂平板上,56℃恒温培养24h,从6块中挑选长势良好的单菌落,移种到液体培养基中,56℃恒温培养24h,得到二代优良菌种,三次涂布于6块琼脂平板上,恒温培养24h,从6块平板中挑选出最为饱满的单菌落,移种到液体培养基中,培养24h,即得到三代优良菌种。分别封存于干燥无菌的沙土保藏管中;
琼脂平板培养基配方:牛肉膏30g/L;蛋白胨10g/L,NaCL5g/L,琼脂15g/L ;
液体培养基配方:葡萄糖20g/L,蛋白胨15g/L, NaCL5g/L,牛肉膏5g/L;
(2)将三代优良菌种,按常规手段复活后,移植到种子培养基中,一般种子培养用4-8只2000ml的摇甁,装有800ml培养基,接种量为1%,温度56℃,摇床转速150-200r/min,培养24-28h,即得到枯草芽孢杆菌发酵种子液;
种子培养基配方:葡萄糖30g/L、大豆粉30g/L、酵母粉10g/L、(NH4)2SO4 5g/L,NaCl5g/L,K2HPO4 3g/L、KH2PO4 2g/L;
(3)得到的种子液按接种量的5%接种到发酵罐,发酵罐提前空罐灭菌115℃,25min,加入培养基后进行实罐灭菌115℃、25min,空气过滤器等相关管道阀门都须灭菌,发酵液培养基配方:葡萄糖30g/L,大豆粉30g/L,酵母粉10g/L, (NH4)2SO4 5g/L, NaCL5g/L,K2HPO4 3g/L、KH2PO4 2g/L。火焰保护接种后,启动搅拌机转速200r/min,温度控制65℃,PH=7,通气量2.5vvm,发酵48h,即得到枯草芽孢杆菌发酵液;
(4)将枯草芽孢杆菌发酵液再经三效蒸发浓缩至75%,蒸发压力控制在-0.096Mpa,温度86℃。用高压尿素液泵将浓度为75%的尿素液(含有占其重量5%的魔芋甘露聚糖、1%的阿拉伯胶及1%的柠檬酸钙)打入造粒塔顶部旋流喷射头,尿素液喷洒为细雾状,同时用高压计量泵,将浓缩发酵液按尿液的0.2%-0.5%打入造粒塔顶另一旋流喷雾器,喷洒出的细雾状枯草芽孢杆菌浓缩发酵液和细雾状尿液混合,在下落100米的过程中,水分蒸发,混合的细微颗粒聚集成大颗粒,芽孢杆菌被包裹在尿素颗粒中,处于休眠状态,即得到含枯草芽孢杆菌生防尿素。
对比例1
(1)枯草杆菌在液体培养基中生长繁殖尤其迅速,将纯种枯草芽孢杆菌液分别涂布于6块琼脂平板上,网状划线,置于恒温培养箱内,56℃下恒温培养24h,培养基表面形成菌落,从6块平板中挑选出长势优良的单菌落,用接种环挑起移种到装有25ml液体培养基的摇甁中,置于摇床内,56℃恒温振荡24h得到优良菌种液,再涂布6块琼脂平板上,56℃恒温培养24h,从6块中挑选长势良好的单菌落,移种到液体培养基中,56℃恒温培养24h,得到二代优良菌种,三次涂布于6块琼脂平板上,恒温培养24h,从6块平板中挑选出最为饱满的单菌落,移种到液体培养基中,培养24h,即得到三代优良菌种。分别封存于干燥无菌的沙土保藏管中;
(2)将三代优良菌种,按常规手段复活后,移植到种子培养基中,一般种子培养用4-8只2000ml的摇甁,装有800ml培养基,接种量为1%,温度56℃,摇床转速150-200r/min,培养24-28h,即得到枯草芽孢杆菌发酵种子液;
(3)得到的种子液按接种量的5%接种到发酵罐,发酵罐提前空罐灭菌115℃,25min,加入培养基后进行实罐灭菌115℃、25min,空气过滤器等相关管道阀门都须灭菌,发酵液培养基配方:葡萄糖30g/L,大豆粉30g/L,酵母粉10g/L, (NH4)2SO4 5g/L, NaCL5g/L,K2HPO4 3g/L、KH2PO4 2g/L。火焰保护接种后,启动搅拌机转速200r/min,温度控制65℃,PH=7,通气量2.5vvm,发酵48h,即得到枯草芽孢杆菌发酵液;
(4)将枯草芽孢杆菌发酵液再经三效蒸发浓缩至75%,蒸发压力控制在-0.096Mpa,温度86℃。用高压尿素液泵将浓度为75%的尿素液(含有占其重量1%的柠檬酸钙)打入造粒塔顶部旋流喷射头,尿素液喷洒为细雾状,同时用高压计量泵,将浓缩发酵液按尿液的0.2%-0.5%打入造粒塔顶另一旋流喷雾器,喷洒出的细雾状枯草芽孢杆菌浓缩发酵液和细雾状尿液混合,在下落100米的过程中,水分蒸发,混合的细微颗粒聚集成大颗粒,芽孢杆菌被包裹在尿素颗粒中,处于休眠状态,即得到含枯草芽孢杆菌生防尿素。
效果实施例
(一)将实施例1及对比例1制备的含枯草芽孢杆菌生防尿素进行有效活菌数检测,按照GB20287-2006及NY/T2321-2013对样品进行检测,统计最终的活菌率,具体结果将表1。

(二)将实施例1及对比例1制备的生防尿素进行实际使用效果检测, 肥效试验使用盆栽,每个盆中种植草莓苗,使用的底肥相同,分为3组,每组50盆,设置为实施例1组、对比例1组及空白对照组,实施例1及对比例1分别追加制备的尿素,空白对照组仅采用底肥,施加量按照3kg底土中加入10g尿素,观察各组草莓生长情况。
通过观察,实施例1组及对比例1组草莓叶色浓绿,植株健壮,生长势好,且无白粉病、叶斑病发生;空白对照组草莓生长势明显弱于实施例及对比例组,同时,发生草莓叶斑病及白粉病。
待草莓开花期,随机选取10盆草莓,进行地下部分根长、根粗统计,具体结果见表2。

Claims (9)
1.一种含枯草芽孢杆菌生防尿素的生产方法,其特征在于,包括以下步骤:
(1)将纯种枯草芽孢杆菌液涂布于琼脂平板上,网状划线,置于恒温培养箱内,恒温培养,培养基表面形成菌落,从平板中挑选出长势优良的单菌落,用接种环挑起移种到装有25ml液体培养基的摇甁中,置于摇床内,恒温振荡得到优良菌种液,再涂布琼脂平板上,恒温培养,从中挑选长势良好的单菌落,移种到液体培养基中,恒温培养,得到二代优良菌种,三次涂布于琼脂平板上,恒温培养,从平板中挑选出最为饱满的单菌落,移种到液体培养基中,培养,即得到三代优良菌种;
(2)将得到的三代优良菌种移植到种子培养基中,进行种子培养,即得到枯草芽孢杆菌发酵种子液;
(3)将枯草芽孢杆菌发酵种子液接种到含有发酵培养基的发酵罐,火焰保护接种后,启动搅拌机,发酵,即得到枯草芽孢杆菌发酵液;
(4)将枯草芽孢杆菌发酵液再经三效蒸发浓缩至75%,得浓缩发酵也,蒸发压力控制在-0.096Mpa,温度86℃,用高压尿素液泵将尿素液打入造粒塔顶部旋流喷射头,尿素液喷洒为细雾状,同时用高压计量泵,将浓缩发酵液打入造粒塔顶另一旋流喷雾器,喷洒出的细雾状枯草芽孢杆菌浓缩发酵液和细雾状尿素液混合,在下落100米的过程中,水分蒸发,混合的细微颗粒聚集成大颗粒,枯草芽孢杆菌被包裹在尿素颗粒中,处于休眠状态,即得到含枯草芽孢杆菌生防尿素。
2.根据权利要求1所述的生产方法,其特征在于,步骤(1)中,所述琼脂平板培养基配方:牛肉膏30g/L;蛋白胨10g/L,NaCL5g/L,琼脂15g/L;所述液体培养基配方:葡萄糖20g/L,蛋白胨15g/L, NaCL5g/L,牛肉膏5g/L。
3.根据权利要求1或2所述的生产方法,其特征在于,步骤(1)中,所述恒温培养或恒温振荡的条件为56℃下培养或振荡24h。
4.根据权利要求1所述的生产方法,其特征在于,步骤(2)中,所述种子培养基配方:葡萄糖30g/L、大豆粉30g/L、酵母粉10g/L、(NH4)2SO4 5g/L,NaCl 5g/L,K2HPO4 3g/L、KH2PO42g/L;所述接种量为1%。
5.根据权利要求1或4所述的生产方法,其特征在于,步骤(2)中,所述种子培养的条件为在温度56℃,摇床转速150-200r/min条件下,培养24-28h。
6.根据权利要求1所述的生产方法,其特征在于,步骤(3)中,所述发酵液培养基配方:葡萄糖30g/L,大豆粉30g/L,酵母粉10g/L, (NH4)2SO4 5g/L, NaCL5g/L,K2HPO4 3g/L、KH2PO4 2g/L。
7.根据权利要求1或6所述的生产方法,其特征在于,步骤(3)中,所述发酵培养基中的接种量为3-5%;所述搅拌机的转速为200r/min;所述发酵的温度控制65℃,PH=7,通气量2.5vvm,发酵48h。
8.根据权利要求1所述的生产方法,其特征在于,步骤(4)中,所述尿素液的浓度为70-75%;所述尿素液中含有占其重量3-5%的魔芋甘露聚糖、1%的阿拉伯胶及1%的柠檬酸钙。
9.根据权利要求1或8所述的生产方法,其特征在于,步骤(4)中,所述浓缩发酵液占尿素液重量的0.2%-0.5%。
