技术领域
本发明涉及基因工程技术领域,具体涉及一种筛选LO2细胞的方法。
背景技术
目前瞬时转染是细胞中最常用的转染技术之一,瞬时转染虽然能够快速生产得到微量至中量的重组蛋白;实验成本低;一个宿主可以带有多个拷贝,表达效率高等特点。但是其外源基因并没有转染到细胞的染色体上而是存在于游离的载体上,这样可以在短时间内获得基因的表达产物,但是随着细胞的不断分裂增殖外源基因最终会丢失,无法继续进行重组蛋白的生产。
慢病毒感染细胞可以得到稳定表达的细胞系,而且稳定转染是外源基因转染至细胞染色体上,目的基因不会随着细胞传代而消失,能够长期稳定的生产目的蛋白。通过稳定转染(稳定细胞系构建)能够实现长期、稳定的生产重组蛋白,得到稳转株,后续生产蛋白的成本降低能够对基因进行基因插入、基因敲除等编辑操作。使用慢病毒感染细胞后需要对细胞进行筛选,嘌呤霉素常用于转染细胞进行筛选。嘌呤霉素是一种由白黑链霉菌产生的氨基苷类抗生素。在原核和真核细胞中,嘌呤霉素都是强效翻译抑制剂,它具有与tRNA分子末端类似的结构,能够共价连接到核糖体延长的多肽链上,从而产生不成熟的多肽链,使蛋白翻译过早终止。此外有研究表明,嘌呤霉素诱导的细胞凋亡并不归因于抑制蛋白合成,而是通过抑制调控细胞分裂周期的氨肽酶,导致细胞在G/M期积累,最终抑制了细胞分裂进程诱导其凋亡。如果嘌呤霉素抗性基因被引入动物细胞并表达相关的酶活性,则细胞可以在嘌呤霉素环境下存活。嘌呤霉素抗性基因被用作在抗生素选择下能够生长的转染哺乳动物细胞的主要标记物。不同于遗传霉素(G418)和湿霉素,嘌呤霉素作用迅速,低浓度嘌呤霉素即可快速导致细胞死亡,因此常用嘌呤霉素进行瞬态选择。嘌呤霉素筛选浓度过高容易杀死阳性转化株,浓度过低又不易杀死阴性转染细胞,筛选时间过长或过短都不易于阳性转化株的筛选。嘌呤霉素处理观察时间通常为3~5天,为保证杀死所有阴性细胞株,通常将药物的筛选浓度提高为原来的2倍作为最佳筛选浓度。由于细胞种类不同,嘌呤霉素筛选的最佳浓度有所不同,不同厂家及批次的嘌呤霉素浓度也有所差异,因此需要进行浓度梯度实验摸索转染的最佳嘌呤霉素筛选浓度。
LO2细胞是人正常肝细胞,具有典型的肝细胞形态学特征,可用于多种实验研究,近年来在生物医药领域中逐渐引起关注。解决LO2细胞池的构建问题,对于使用LO2细胞作为宿主细胞生产药物蛋白具有重要的意义。
发明内容
本发明的目的在于克服现有技术的不足,提供一种筛选LO2细胞的方法,以及利用该方法构建LO2细胞池,稳定地表达目的蛋白,缩短实验研发周期。
为实现上述目的,本发明采取的技术方案为:一种筛选LO2细胞的方法,包括以下步骤:在LO2细胞中先后加入具有第一浓度的抗生素和第二浓度的抗生素进行培养;所述第一浓度的抗生素能够富集细胞,所述第二浓度的抗生素能够筛选细胞。
发明人经大量的实验发现,先后加入不同浓度的抗生素,且第一次加入的抗生素浓度低于第二次加入的抗生素浓度,使得第一次加入的抗生素对细胞产生富集作用,第二次加入的抗生素对细胞进行筛选,避免一次加入的抗生素浓度过高时,由于细胞无法直接耐受高浓度的抗生素而或率下降乃至死亡,或由于一次加入的抗生素浓度过低,使抗生素无法对细胞进行筛选,导致大量的假阳性克隆。
作为本发明所述筛选LO2细胞的方法的优选实施方式,所述LO2细胞为基因工程改造的LO2细胞。
作为本发明所述筛选LO2细胞的方法的优选实施方式,所述抗生素为嘌呤霉素。
作为本发明所述筛选LO2细胞的方法的优选实施方式,所述第一浓度的抗生素和第二浓度的抗生素均采用LO2细胞培养基作为溶剂配制而成。
作为本发明所述筛选LO2细胞的方法的优选实施方式,所述嘌呤霉素的第一浓度为0.5~3μg/mL,第二浓度为3.5~5μg/mL。

(1)目的基因质粒的获取;
(2)制备慢病毒上清液;
(3)慢病毒上清液去感染LO2细胞,获得基因工程改造的LO2细胞;
(4)采用上述筛选LO2细胞的方法对步骤(3)的LO2细胞进行筛选。
本发明的有益效果:本发明提供了一种筛选LO2细胞的方法,通过先后两次加入不同浓度的嘌呤霉素对经过基因工程改造的LO2细胞进行筛选,快速的筛选出阳性转化的LO2细胞,避免高浓度抗生素杀死阳性转化株和低浓度抗生素产生大量假阳性,节约了实验时间;本发明还提供该筛选LO2细胞的方法在LO2细胞池的构建方法中的应用,本发明的筛选LO2细胞的方法使细胞提前进入细胞活率的持续增长期。

具体实施方式
以下通过实施例形式的具体实施方式,对本发明的上述内容再作进一步的详细说明。但不应将此理解为本发明上述主题的范围仅限于以下的实例。凡基于本发明上述内容所实现的技术均属于本发明的范围。
实施例1
本实施例采用不同抗生素浓度对经过转染的LO2细胞进行筛选。
试验组1:在经过转染的LO2细胞中先加入第一浓度为0.5μg/mL,再加入第二浓度为4.2μg/mL的嘌呤霉素进行培养,观察细胞形态变化。
试验组2:在经过转染的LO2细胞中先加入第一浓度为0.5μg/mL,再加入第二浓度为4.5μg/mL的嘌呤霉素进行培养,观察细胞形态变化。
试验组3:在经过转染的LO2细胞中先加入第一浓度为0.5μg/mL,再加入第二浓度为5.3μg/mL的嘌呤霉素进行培养,观察细胞形态变化。
试验组4:在经过转染的LO2细胞中先加入第一浓度为0.5μg/mL,再加入第二浓度为6.4μg/mL的嘌呤霉素进行培养,观察细胞形态变化。
试验组5:在经过转染的LO2细胞中先加入浓度为0.5μg/mL的嘌呤霉素进行培养,观察细胞形态变化。
实验结果发现,试验组5的LO2细胞生长状态良好,未见明显生长抑制和细胞死亡;试验组1的LO2细胞在培养48h后细胞明显减少;试验组3的LO2细胞在培养48h后开始出现显著凋亡增多,细胞数量可见显著减少;试验组4的LO2细胞在最初24h对细胞抑制效果并不显著,随着处理时间的延长,存活的细胞数越来越少,在处理48h后,绝大多数细胞正常的梭形形状消失,转变为圆形,呈漂浮状态。试验组2的LO2细胞在最初24h时可见大量漂浮细胞,处理48h后细胞全部变圆死亡,呈成簇堆积形态。细胞形态发生显著变化,显微镜下可见细胞核增大,细胞质变浑浊,细胞膜粗糙有褶皱,最后细胞膜破裂,细胞裂解死亡,正常的梭形形状消失,转变为圆形,呈漂浮状态。
实施例2
本实施例采用实施例1试验组2的筛选方法构建LO2细胞池,包括以下步骤:
(1)293T/LO2细胞的复苏
温度为37℃的水浴,从液氮罐中取出冻存的细胞(需戴上棉手套,防止被冻伤),迅速丢入水浴锅中并快速晃动,尽量在1~2min内使细胞溶液完全溶解;将细胞溶液转移到15mL离心管中,并在其中加上2mL新鲜的完全培养基,混匀后1000rpm离心3min;去掉上清,加入5mL新鲜的完全培养基,混匀沉淀后,转入6cm培养皿;将培养皿平稳放入37℃、5%CO2和95%相对湿度的培养箱中培养;第二天观察细胞存活率,给细胞换培养基,以后每天观察细胞生长情况,为后边实验提供状态良好的细胞。
(2)载体构建、重组
在Genebank中找到人GSTZ1的基因序列,通过Crispr设计工具及sgRNA的设计原则,评估基因序列上得分较高的靶位点设计sgRNA,其评估原则包括:1)序列下游是否有PAM(NGG),2)脱靶效率低,不易脱靶。根据设计的sgRNA,在其5末端加上CACC得到正向寡核苷酸序列,在其互补链的5末端加上AAAC得到反向寡核苷酸序列,分别合成正向和反向寡核苷酸序列,然后将合成的序列通过变性、退火,得到具有Esp3I粘性末端的双链DNA片段。将双链目的DNA片段和用Esp3I酶切过的lentilCRISPR v2载体进行连接;将连接产物转化到大肠杆菌DH5a感受态细胞中,涂布于带有氨苄青霉素抗性的LB平板上,筛选阳性菌落,提取阳性菌落质粒进行分析及测序,确定sgRNA表达载体构建成功。
(3)慢病毒包装
在转染前将已复苏状态良好的293T细胞铺在中皿中,等到密度70%-80%时进行转染。以lentiCRISPR v2-gRNA:psPAX2:pMD2.G=4:3:1的比例进行慢病毒包装。取两个1.5ml的离心管,各加入500uloptiMEM,向其中一个管中加入12ul LipoFiter,轻柔混匀,另一个管中加入目的质粒、psPAX2、pMD2.G轻柔混匀,然后将后管的全部轻轻加入前管中,轻柔混匀。室温静置20-25min,转染复合物制备完成;将转染复合物轻轻滴入细胞无抗生素培养基中,轻柔混匀;继续培养24小时后,换成新鲜无抗生素培养基,48小时后收集上清;3000转离心5分钟,用0.45μm的滤膜过滤上清后分装冻存于-80℃备用。
(4)靶细胞感染
在感染前一天将LO2细胞铺在6孔板中,等到密度为70%-80%时进行感染;移除细胞培养基,加入备好的慢病毒,感染6-8小时后移除慢病毒,换成新鲜无抗生素培养基。
(5)嘌呤霉素筛选LO2细胞
质粒转染LO2细胞后,先加入嘌呤霉素0.5μg/mL培养24小时,再加入嘌呤霉素4.5μg/mL。每隔24小时换一次液,连续筛选至无细胞死亡。
(6)Western Blot检测细胞池蛋白表达
取筛选48小时的细胞上清液进行凝胶电泳实验。
由图1~图4可知,LO2细胞在第一浓度为0.5μg/mL,第二浓度为4.5μg/mL的嘌呤霉素处理0h时细胞生长状态良好,当处理时间为24h时,出现大量漂浮细胞,当处理48h后细胞全部变圆死亡,呈成簇堆积形态。细胞形态发生显著变化,显微镜下可见细胞核增大,细胞质变浑浊,细胞膜粗糙有褶皱,最后细胞膜破裂,细胞裂解死亡,正常的梭形形状消失,转变为圆形,呈漂浮状态。由图5可知,该LO2细胞GSTZ1蛋白表达缺失,即成功获得GSTZ1表达缺失LO2细胞。
最后应当说明的是,以上实施例仅用以说明本发明的技术方案而非对本发明保护范围的限制,尽管参照较佳实施例对本发明作了详细说明,本领域的普通技术人员应当理解,可以对本发明的技术方案进行修改或者等同替换,而不脱离本发明技术方案的实质和范围。
Claims (10)
1.一种筛选LO2细胞的方法,其特征在于,包括以下步骤:在LO2细胞中先后加入具有第一浓度的抗生素和第二浓度的抗生素进行培养;所述第一浓度的抗生素能够富集细胞,所述第二浓度的抗生素能够筛选细胞。
2.如权利要求1所述筛选LO2细胞的方法,其特征在于,所述LO2细胞为基因工程改造的LO2细胞。
3.如权利要求1所述筛选LO2细胞的方法,其特征在于,所述抗生素为嘌呤霉素。
4.如权利要求1所述筛选LO2细胞的方法,其特征在于,所述第一浓度的抗生素和第二浓度的抗生素均采用LO2细胞培养基作为溶剂配制而成。
5.如权利要求3所述筛选LO2细胞的方法,其特征在于,所述嘌呤霉素的第一浓度为0.5~3μg/mL,第二浓度为3.5~5μg/mL。
6.如权利要求4所述筛选LO2细胞的方法,其特征在于,所述嘌呤霉素的第一浓度为0.5μg/mL,第二浓度为4.5μg/mL。
7.如权利要求1所述筛选LO2细胞的方法,其特征在于,所述加入所述第一浓度的抗生素与加入所述第二浓度的抗生素所间隔的时间为20~30小时。
8.如权利要求7所述筛选LO2细胞的方法,其特征在于,所述加入所述第一浓度的抗生素与加入所述第二浓度的抗生素所间隔的时间为24小时。
9.如权利要求7~8任一项所述筛选LO2细胞的方法,其特征在于,所述加入所述第一浓度的抗生素与加入所述第二浓度的抗生素所间隔的时间包括细胞培养时间。
10.一种LO2细胞池的构建方法,其特征在于,包括以下步骤:
(1)目的基因质粒的获取;
(2)制备慢病毒上清液;
(3)慢病毒上清液去感染LO2细胞,获得基因工程改造的LO2细胞;
(4)采用权利要求1~9任一项所述的筛选LO2细胞的方法对步骤(3)的LO2细胞进行筛选。
