Abstract
本发明提供了唐古特大黄叶柄在制备保健食品或药物中的用途。具体提供了唐古特大黄叶柄在制备抗氧化的保健食品中的用途,以及唐古特大黄叶柄在制备抗氧化、降尿酸、清热解毒的药物中的用途。本发明研究发现唐古特大黄叶柄具有良好的抗氧化、降尿酸、清热解毒功效,可以预防和/或治疗痛风或消炎。因此可以将唐古特大黄叶柄制备成具有相关功效的保健食品或药物。本发明还提供了一种唐古特大黄叶柄汁的制备方法,采用该方法可以克服唐古特大黄叶柄涩的口感,得到口感优良的唐古特大黄叶柄汁,该方法生产工艺简便、易于实现工业化生产。本发明开拓了唐古特大黄叶柄的经济价值,具有良好的市场前景。
Description
技术领域
本发明涉及唐古特大黄叶柄在制备保健食品或药物中的用途。
背景技术
大黄是应用广泛的重要中药材之一,也是中医常用药物,《神农本草经》、《伤寒杂病论》及《本草纲目》中已有记载。大黄又名将军、火参等,是我国传统常用中药,主要来源于廖科植物掌叶大黄、唐古特大黄和药用大黄。大黄为多年生草本,药用部分为干燥根和根茎。有解毒,清热清火,通经逐瘀,凉血止血等多种功效,一直以来具有巨大的市场需求。唐古特大黄,蓼科,大黄属,主要分布于青海、西藏、四川、甘肃海拔1600-4500m的高海拔地区,是大黄的主要生药来源之一。
由于大黄的药用部分为干燥根和根茎,因此对其叶柄部位研究较少。但是,青海西藏当地农民有食用唐古特大黄叶柄的习俗,当地农牧民剥去唐古特大黄叶柄的皮后用于生吃或是水浸泡后饮用,其味酸,加入糖后酸甜可口,拥有很好的风味。同时,民间认为叶柄具有耐缺氧的能力,农牧民将其作为登山缓解缺氧的食品使用。
但是,目前对唐古特大黄叶柄的药用价值缺乏研究,造成大量唐古特大黄叶柄浪费。探究唐古特大黄叶柄的药用价值,对于扩展唐古特大黄市场以及其经济价值具有重要意义。
发明内容
本发明的目的是提供唐古特大黄叶柄在制备保健食品或药物中的用途。为唐古特大黄生物资源的可持续利用提供科学依据。
本发明提供了唐古特大黄叶柄在制备抗氧化的保健食品中的用途。
进一步地,所述保健食品为唐古特大黄叶柄粉末、或叶柄汁、或叶柄汁冻干粉、或者采用水或有机溶剂提取后得到的提取物为活性成分,加上保健食品上可接受的辅料或辅助性成分制备而成的保健食品。
进一步地,所述保健食品为饮料、糖果。
本发明还提供了唐古特大黄叶柄在制备抗氧化、降尿酸、清热解毒的药物中的用途。
进一步地,所述药物为预防和/或治疗痛风的药物;
和/或,所述药物为消炎的药物。
进一步地,所述药物为唐古特大黄叶柄粉末、或叶柄汁、或叶柄汁冻干粉、或者采用水或有机溶剂提取后得到的提取物为活性成分,加上药学上可接受的辅料或辅助性成分制备而成的制剂;
优选地,所述制剂为口服制剂;
更优选地,所述口服制剂为散剂、片剂、丸剂、颗粒剂、胶囊剂、口服液。
本发明还提供了一种唐古特大黄叶柄汁的制备方法,它包括如下步骤:
取唐古特大黄叶柄,压榨叶柄后收集叶柄汁,对叶柄汁进行均质、过滤、灭菌,即得;
优选地,所述唐古特大黄叶柄汁的制备方法包括如下步骤:
取唐古特大黄叶柄,压榨叶柄后收集叶柄汁,采用胶体磨对叶柄汁进行混合分散均质,研磨后采用膜分离设备进行除杂,收集膜分离设备透过侧液体,收集到的液体经过灌装灭菌后,即得。
进一步地,
所述唐古特大黄叶柄采用8-9月唐古特大黄叶柄;
和/或,所述胶体磨转速为1500-3500r/min,流量为0.01-20m3/h;优选转速为2500r/min,流量为10m3/h;
和/或,所述膜分离设备中陶瓷膜的孔径为0.1~0.5μm;优选为0.2μm;
和/或,所述灭菌采用压力超高压技术杀菌,所述超高压为100~1000MPa。
本发明还提供了一种唐古特大黄叶柄汁,它是由前述的制备方法制备得到的唐古特大黄叶柄汁。
本发明还提供了一种唐古特大黄叶柄保健食品,它包括前述的唐古特大黄叶柄汁;
优选地,所述保健食品为饮料、糖果。
与现有技术相比,本发明的有益效果是:
本发明研究发现唐古特大黄叶柄具有良好的抗氧化、降尿酸、清热解毒功效,可以预防和/或治疗痛风或消炎。因此可以将唐古特大黄叶柄制备成具有相关功效的保健食品或药物。本发明还提供了一种唐古特大黄叶柄汁的制备方法,采用该方法可以克服唐古特大黄叶柄涩的口感,得到口感优良的唐古特大黄叶柄汁,该方法生产工艺简便、易于实现工业化生产。本发明开拓了唐古特大黄叶柄的经济价值,具有良好的市场前景。

具体实施方式
本发明具体实施方式中使用的原料、设备均为已知产品,通过购买市售产品获得。
实施例1、唐古特大黄叶柄汁的制备
取8月唐古特大黄叶柄后剥皮,采用压榨法压榨叶柄后收集叶柄汁,采用胶体磨对叶柄汁进行混合分散均质,胶体磨功率为转速为1500r/min,流量为0.01m3/h。研磨后采用陶瓷膜的孔径为0.1μm膜分离设备进行除杂,收集膜分离设备透过侧液体,收集到的液体经过灌装后100MPa超高压技术杀菌后即得唐古特大黄叶柄汁。
实施例2、唐古特大黄叶柄汁的制备
取9月唐古特大黄叶柄后剥皮,采用压榨法压榨叶柄后收集叶柄汁,采用胶体磨对叶柄汁进行混合分散均质,胶体磨功率为转速为3500r/min,流量为20m3/h。研磨后采用陶瓷膜的孔径为0.5μm膜分离设备进行除杂,收集膜分离设备透过侧液体,收集到的液体经过灌装后在压力为1000MPa超高压技术杀菌后即得唐古特大黄叶柄汁。
实施例3、唐古特大黄叶柄汁的制备
取9月唐古特大黄叶柄后剥皮,采用压榨法压榨叶柄后收集叶柄汁,采用胶体磨对叶柄汁进行混合分散均质,胶体磨功率为转速为2500r/min,流量为10m3/h。研磨后采用陶瓷膜的孔径为0.2μm膜分离设备进行除杂,收集膜分离设备透过侧液体,收集到的液体经过灌装后在压力为500MPa超高压技术杀菌后即得唐古特大黄叶柄汁。
以下通过具体的试验例证明本发明的有益效果。
试验例1、唐古特大黄叶柄的抗氧化性研究
1、待测样品
取实施例1唐古特大黄叶柄汁经过冷冻干燥后的样品。
2、检测方法
过氧化物自由基(ROO-)是过氧化氢自由基(HOO-)的类似物,是由HOO-中的氢原子被有机基团替代而得到的,ROO-通常是在蛋白质以及脂类的氧化过程中产生的,此外,在与氧化应激相关的炎症反应中,嗜中性粒细胞被激活后也可产生ROO-。ROO-能够破坏细胞膜和蛋白质的结构,因此是许多与脂类自氧化和酶失活相关的各种疾病的致病因子。目前,有研究表明,ROO-与肿瘤生长过程中的血管形成有很大的关系。
本发明抗氧化性检测参照文献(Huang D.J.,Ou B.,Hampsch-Woodill.M.,Flanagan.J.A.,and Prior.R.high-throughput assay of oxygen radical absorbancecapacity(ORAC)using a multichannel liquid handing system coupled withamicroplate fluorescence reader in 96-well format.Journal of agricultural andfood chemistry.2002,50:4437-4444)的检测方法,该方法是根据ROO-破坏荧光探针,使荧光强度产生变化的原理,对待测样品的抗ROO-能力进行检测。以AAPH作为产生ROO-的来源,荧光素为荧光探针,维生素E水溶性类似物Trolox为标准对照品,进行体外抗ROO-能力测定。“时间-荧光信号图”中的曲线下面积(AUC)越大,表示待测样品抗ROO-的能力越强。该方法适用于植物提取物、食品、化妆品、药品等中的水溶性物质的抗过氧化物自由基的活性评价。
3、检测结果
经检测,唐古特大黄叶柄的抗氧化值为3500μmole TE/g。该检测结果说明唐古特大黄叶柄具有较强的抗氧化性。
试验例2、唐古特大黄叶柄的降尿酸作用研究
1、待测样品
取实施例1唐古特大黄叶柄汁经过冷冻干燥后的样品。
2、检测方法
高尿酸血症是由体内嘌呤代谢紊乱从而导致尿酸水平过高的一类代谢性疾病。尿酸生成增加或者排清不足导致其浓度超过6.8mg/mL即可被诊断为高尿酸血症。尿酸在体内一般以尿酸单钠盐的形式存在,由于溶解度较低,尿酸单钠极易以晶体形式析出并沉积在关节腔内,造成炎症,形成痛风。
黄嘌呤氧化酶是嘌呤代谢中的一种关键酶,是人体中嘌呤分解代谢的末端酶,将次黄嘌呤与黄嘌呤催化反应生成尿酸。尿酸水平升高导致各种疾病,包括痛风和肾结石的形成。黄嘌呤氧化物酶抑制剂通过抑制黄嘌呤氧化物酶的生物活性,减少尿酸的形成,起到治疗痛风的效果。
精密称取唐古特大黄叶柄汁冻干粉适量,用磷酸盐缓冲液溶解备用,活性测试使用酶标仪,溶液总体积为100μL,反应时先依次向96孔板加入样品20μL,黄嘌呤氧化酶液40μL,在酶标仪内37摄氏度孵育三分钟,之后加入底物黄嘌呤40μL,启动反应,立即在295nm处记录吸收度值(每隔30s一次),共计5min。本实验设置样品组(加样品、底物和酶)、酶组(加底物和酶但不加样品)、空白组(只加底物和样品不加酶)和阳性对照组(别嘌呤醇)。黄嘌呤氧化酶抑制率(%)用下列公式计算:抑制率(%)=(ΔA酶-ΔA样品)/(ΔA酶-ΔA空白)×100。其中ΔA指一定时间内吸光度的差值。
3、检测结果
经检测,唐古特大黄叶柄的在浓度为15.0μg/ml时,对黄嘌呤氧化物酶的抑制率为75%。别嘌呤醇在浓度为2μg/ml时,对黄嘌呤氧化物酶的抑制率为57%。说明唐古特大黄叶柄能够显著降低尿酸含量,可以预防和/或治疗痛风。
试验例3、唐古特大黄叶柄的清热解毒作用研究
1、待测样品
取实施例1唐古特大黄叶柄汁经过冷冻干燥后的样品。样品使用DMSO配制成不同浓度的溶液。
2、检测方法
细胞毒性测定
采用四甲基偶氮唑蓝比色法对的体外肿瘤增殖抑制作用进行评价。
RAW264.7细胞株均培养于即完全培养基中,加新生牛血清、链霉素和青霉素,选择对数生长期细胞,将细胞接种于孔板中,置37℃,5%二氧化碳培养箱中,培养24小时后加入不同浓度样品溶液,在加药培养24小时后进行MTT检测。在37℃下培养后取出培养板,吸去孔内培养上清液,每孔加入MTT溶液100微升,振荡10min,使结晶物充分溶解。在自动酶标仪上测定各孔吸收值(OD),各孔光吸收值。实验重复次,记录各组的值,算细胞相对存活率。
相对抑制细胞生长的表述如下:百分比(%)=(1-[OD]测试/[OD]对照)×100%。
NO含量测定
RAW264.7细胞贴壁培养24小时,轻轻吸取培养液,加入新的培养液及样品溶液,继续培养24小时,培养结束时,吸取100μl培养液,并加入100ulGriess reagent试剂,室温放置10min,用酶标仪测定570nm处的吸光值,以亚硝酸钠作为对照品测定NO的浓度。
PEG2含量测定
RAW264.7细胞贴壁培养24小时,轻轻吸取培养液,加入新的培养液及样品溶液和LPS,继续培养24小时,培养结束时,吸取培养液,按照小鼠前列腺素E2酶联免疫检测盒说明书测定PEG2含量。
3、检测结果
经检测,唐古特大黄叶柄的IC50值大于2000μM,说明其对RAW264.7细胞无明显的抑制生长作用,细胞相容性好。从表1看出,唐古特大黄叶柄具有消炎作用,进一步说明唐古特大黄叶柄具有清热解毒功效。

试验例4、唐古特大黄叶柄汁制备方法的筛选
鲜榨的唐古特大黄叶柄汁口感较涩,经过胶体磨混合使影响口感的鞣制类成分分散均质,研磨后采用陶瓷膜的分离设备进行去除影响鞣制类化合物,收集膜分离设备透过侧液体,即得到口感优良的唐古特大黄叶柄汁。
综上,本发明研究发现唐古特大黄叶柄具有良好的抗氧化、降尿酸、清热解毒功效,可以预防和/或治疗痛风或消炎。因此可以将唐古特大黄叶柄制备成具有相关功效的保健食品或药物。本发明还提供了一种唐古特大黄叶柄汁的制备方法,采用该方法可以克服唐古特大黄叶柄涩的口感,得到口感优良的唐古特大黄叶柄汁,该方法生产工艺简便、易于实现工业化生产。本发明开拓了唐古特大黄叶柄的经济价值,具有良好的市场前景。

1.唐古特大黄叶柄在制备抗氧化的保健食品中的用途。
2.根据权利要求1所述的用途,其特征在于:所述保健食品为唐古特大黄叶柄粉末、或叶柄汁、或叶柄汁冻干粉、或者采用水或有机溶剂提取后得到的提取物为活性成分,加上保健食品上可接受的辅料或辅助性成分制备而成的保健食品。
3.根据权利要求2所述的用途,其特征在于:所述保健食品为饮料、糖果。
4.唐古特大黄叶柄在制备抗氧化、降尿酸、清热解毒的药物中的用途。
5.根据权利要求4所述的用途,其特征在于:所述药物为预防和/或治疗痛风的药物;
和/或,所述药物为消炎的药物。
6.根据权利要求4所述的用途,其特征在于:所述药物为唐古特大黄叶柄粉末、或叶柄汁、或叶柄汁冻干粉、或者采用水或有机溶剂提取后得到的提取物为活性成分,加上药学上可接受的辅料或辅助性成分制备而成的制剂;
优选地,所述制剂为口服制剂;
更优选地,所述口服制剂为散剂、片剂、丸剂、颗粒剂、胶囊剂、口服液。
7.一种唐古特大黄叶柄汁的制备方法,其特征在于:它包括如下步骤:
