Abstract
本发明公开了一株耐低营养乳酸菌及其应用。该耐低营养乳酸菌的名称为乳酸菌(Lactobacillus plantarum)SCLN1,保藏于广东省微生物菌种保藏中心,保藏地点为广州市先烈中路100号大院59号楼5楼广东省微生物研究所,保藏号为GDMCC No:60944,保藏日期为2020年1月3日。该乳酸菌为格兰氏染色阳性杆菌,为葡萄糖同型发酵,耐酸,生长速度快,生长温域宽,尤为耐低营养,可显著提高水溶性碳水化合物含量低的牧草青贮发酵品质,因此可在我国各地使用,特别对于含糖量低的饲料效果独特,克服了因水溶性碳水化合物含量较低造成的青贮饲料发酵不佳的问题。

技术领域
本发明涉及微生物应用和饲料调制加工技术领域,具体涉及一株耐低营养乳酸菌及其应用。
背景技术
青贮饲料由于气味香甜、柔软多汁、营养保存好、适口性佳等特点,已成为畜牧业不可或缺的基础性饲料。青贮饲料不仅能提高肉、奶的品质,而且能充分利用不可直接饲用的植物材料,以及减少农产品加工副产物的浪费。因此,青贮饲料已被世界各国所重视,特别是畜牧业发达国家。青贮饲料是依靠原料上附着的乳酸菌在厌氧条件下将原材料中的水溶性碳水化合物转化成以乳酸为主的有机酸,使pH下降,从而抑制有害菌的生长和繁殖,使饲料得以长期保存。因此,水溶性碳水化合物和乳酸菌对青贮发酵品质起关键作用。在生产实践中,对于青贮原材料水溶性碳水化合物不足时,无论是否添加乳酸菌,均难以得到优质的青贮饲料。针对这一问题,大多采用添加糖源或产糖物质,如糖类、含糖物质多的副产物或纤维素酶等,但需要人力物力和成本相对较高。另外,现有添加剂的乳酸菌,都是在正常营养含量的培养基中筛选得来,未针对耐低营养条件。若有耐低营养的乳酸菌菌株,不仅能解决原材料含糖量低或乳酸菌数量少导致的发酵品质差,且能有效降低生产成本。
发明内容
本发明的首要目的在于克服现有技术的缺点与不足,提供一株耐低营养乳酸菌。
本发明的另一目的在于提供上述耐低营养乳酸菌在青贮调制中的应用。
为实现上述目的,本发明通过以下述技术方案实现:
一株耐低营养乳酸菌,名称为乳酸菌(Lactobacillus plantarum)SCLN1,保藏于广东省微生物菌种保藏中心,保藏地点为广州市先烈中路100号大院59号楼5楼广东省微生物研究所,保藏号为GDMCC No:60944,保藏日期为2020年1月3日。
上述乳酸菌(Lactobacillus plantarum)SCLN1的生物学特性为革兰氏染色阳性杆菌,葡萄糖同型发酵,耐酸性强(在pH3.5微弱生长),生长速度快(MRS培养基培养24小时pH可降到4.0以下),生长温域宽(15~45℃可正常生长),在纯培养的葡萄糖浓度为0.002g/L时正常生长。
上述乳酸菌(Lactobacillus plantarum)SCLN1的获得方法是富集培养该菌,稀释平板法筛选获得:将装有500mL蒸馏水的锥形瓶、装有9mL蒸馏水的试管、MRS固体培养基及平板灭菌,灭菌后再将MRS培养基倒入平板内。无菌条件下,取10g样品(如青贮苏丹草)于塑料袋内,再加入90mL灭菌蒸馏水,使其终浓度为100g/L,放置于200rpm的摇床振荡30min,然后再取稀释后样品液1mL加入含有9mL灭菌蒸馏水的试管振荡均匀,使其稀释10倍,此时取该液体0.02mL涂于平板中,37℃厌氧培养24~48h,再用接种针挑取单菌落,接种到MRS液体培养基中,37℃厌氧培养24~48h,反复进行平板划线分离,直至得到单菌落,将单菌落用接种针插入含有MRS固体培养基的试管中心,于4℃冰箱内保存。其中MRS培养基为:蛋白胨(Proteose peptone NO.3)10.0g、牛肉膏(Beef extract)10.0g、酵母提取物(Yeastextract)5.0g、葡萄糖(Dextrose)20.0g、吐温(Polysorbate 80)1mL、柠檬酸铵(Ammoniumcitrate)2.0g、醋酸钠(NaAc)5.0g、硫酸镁(MgSO4·7H2O)0.1g、硫酸锰(MnSO4·4H2O)0.05g、磷酸氢二钾(K2HPO4)2.0g,固体培养基再加15g/L琼脂(Agar),蒸馏水(H2O)定容至1000mL,121℃灭菌20min。
本发明通过调低培养基中葡萄糖浓度至0.002g/L筛选出耐低糖、耐酸及生长速度快的乳酸菌Lactobacillus plantarum SCLN1。提取乳酸菌Lactobacillus plantarum全长基因,然后运用PCR扩增引物25F(SEQ ID NO:1)和1492R(SEQ ID NO:2),扩增出16S rDNA基因,在NCBI上对比相关序列,确定其相似菌种。最后,将筛选出的菌株及对照菌(在国内使用最多的商品菌剂)分别添加到不同饲草中进行青贮,开封后分析青贮发酵品质并与对照菌进行对比,确认Lactobacillus plantarum SCLN1可以显著提高含糖量低的牧草青贮发酵品质。
上述耐低营养乳酸菌在青贮调制中的应用,优选包括如下步骤:
(1)将待发酵饲料切短;
(2)将切短处理后的待发酵饲料和上述耐低营养乳酸菌混合均匀;
(3)将步骤(2)最终得到的饲料脱气密封后贮藏。
作为一种优选方案,步骤(1)中所述的切短的规格优选为切短至3cm以下;更优选为切至切短至2.5cm以下;最优选为切短至1~2cm。
步骤(2)中所述的耐低营养乳酸菌的添加量优选按每千克待发酵饲料配比1.0×108个以上所述的耐低营养乳酸菌计算。
与现有技术相比,本发明具有如下有益效果:
(1)本发明提供了一株耐低营养乳酸菌,菌株可在我国各地使用,特别对于含糖量低的饲料效果独特,克服了因水溶性碳水化合物含量较低造成的青贮饲料发酵不佳的问题。
(2)本发明利用微生物乳酸菌Lactobacillus plantarum SCLN1改善青贮饲料发酵品质,成本低,安全,可靠,易于利用。
(3)本发明使用的乳酸菌Lactobacillus plantarum SCLN1青贮效果比市售乳酸菌添加剂效果更佳。

具体实施方式
下面结合实施例及附图对本发明作进一步详细的描述,但本发明的实施方式不限于此。
实例1:乳酸菌的分离、筛选以及生理生化测定
将燕麦、小麦、象草、黄燕麦、苏丹草等牧草分别称取10g,加入90mL无菌水,放置200rpm摇床中震荡30min,吸取1mL到9mL无菌水中,依次稀释至10-1~10-3浓度,分别取0.02mL涂至MRS固体培养基,37℃厌氧培养24~48h,挑取菌落不一的单菌落,再分别将单菌落接种到5mL MRS液体培养基中,37℃厌氧培养24~48h,在MRS固体培养基上反复划线分离,通过液-固-液3代以上的步骤分离纯化出92株菌株,进行革兰氏染色和细胞形状观察,并通过将菌株分别接种到0.002g/L葡萄糖的MRS液体培养基(pH 6.0)中,37℃厌氧培养24h,进行OD600测定,筛选出10株生长好的菌株,得到的结果如图1所示。从图1中可以看出,SCLN1的OD600值显著高于其它菌株,表明SCLN1菌株在培养基葡萄糖含量为0.002g/L可生长,具有较强的耐低营养能力。再将SCLN1分别接种到pH为3.5、4.0、4.5、7.5、8.0、8.5的MRS液体培养基(葡萄糖为20g/L),从表1可以看出SCLN1在低至pH4的MRS培养基中生长良好,具有强的耐酸性。
乳酸菌SCLN1的分离、生理生化试验、培养基及其配制的方法如下:
1)革兰氏染色、发酵类型及形状观察参照东秀珠主编的《常见细菌系统鉴定手册》。
2)乳酸菌生长pH调节使用4mol/L NaOH和4mol/L HCl。
3)糖发酵采用API 50CHL(bioMérieux,l'Etoile,France)分析。
4)培养基:
MRS培养基(用于乳酸菌的培养):
蛋白胨(Proteose peptone)10.0g、牛肉膏粉(Beef extract)10.0g、酵母膏粉(Yeast extract)4.0g、葡萄糖(Dextrose)20.0g、吐温(Polysorbate 80)1mL、柠檬酸三铵(Ammonium citrate)2.0g、乙酸钠(NaAc)5.0g、硫酸镁(MgSO4·7H2O)0.1g、硫酸锰(MnSO4·4H2O)0.05g、磷酸氢二钾(K2HPO4)2.0g。用蒸馏水将前述成分溶解,接着再用蒸馏水定容至1000mL,得到MRS液体培养基;MRS固体培养基则是在MRS液体培养基的基础上加15g/L琼脂(Agar),121℃灭菌20min。
API 50CHL培养基:
蛋白胨(Proteose peptone)10.0g、酵母膏粉(Yeast extract)5.0g、吐温(Polysorbate 80)1mL、乙酸钠(NaAc)5.0g、柠檬酸三铵(Ammonium citrate)2.0g、硫酸镁(MgSO4·7H2O)0.2g、磷酸氢二钾(K2HPO4)2.0g、硫酸锰(MnSO4·4H2O)0.05g、溴甲酚紫(Bromcresol purple)0.17g。用蒸馏水将前述成分溶解,接着再用蒸馏水定容至1000mL,分装在15mL的试管中,每试管10mL,121℃灭菌20min。
检测结果如表1所示:

实例2:乳酸菌的鉴定
实施例1所得菌株在5mL的MRS培养基中37℃培养过夜,菌液移入1.5mL的离心管中,10000rpm离心3min~5min收集菌,用TE(10mmol/L Tris–HCl、0.1mmol/L EDTA,pH 8.0)清洗两次,提取菌株全长基因,然后运用PCR扩增引物25F(5`-AACTGAAGAGTTTGATCCTGGCTC-3`)和1492R(5`-TACGGCTACCTTGTTACG ACT-3`),扩增出16S rDNA基因,送至华大基因(中国)测序,在NCBI基因库上对比相关序列,序列如下所示,确定SCLN1为植物乳杆菌(Lactobacillus plantarum)。

实例3:添加到青贮饲料中的效果
以柱花草184和苏丹草为材料进行了青贮添加试验。将材料切短至<2.5cm,混合均匀,设对照、SCLN1、商品菌剂(为植物乳杆菌菌剂),每kg新鲜材料分别加入菌株约1.0×108cfu,对照添加等量无菌水。装入30cm×20cm的聚乙烯青贮袋,每个处理4个重复,每袋约200g,用真空密封机(SINBO Vacuum Sealer,Hong Tai Home Electrical ApplianceCo.Ltd.)抽气、密封,置于黑暗处贮藏60d,开封后分析青贮发酵品质并进行对比分析。
结果如下:柱花草水溶性碳水化合物含量不到5%(表2),添加SCLN1其青贮料的pH降至4.67,氨态氮降至11.64%TN,均显著低于对照和商品菌剂(表3),乳酸含量显著高于对照和商品菌剂(表3)。虽然苏丹草水溶性碳水化合物含量较高,但添加SCLN1处理的发酵品质也显著优于对照和商品菌剂。因此,添加SCLN1显著提高了两种牧草的发酵品质。

营养成分及青贮发酵指标测定方法:
1)牧草营养成分测定及微生物分析:干物质(DM)含量采用张丽英的饲料分析中70℃烘干法,粗蛋白含量采用凯氏定氮法测定(定氮仪KN680,ALVA仪器有限公司),粗纤维、中性洗涤纤维和酸性洗涤纤维含量采用滤袋法测定,粗脂肪含量采用乙醚提取法测定(SLF-06,杭州托普仪器有限公司),粗灰分含量采用灼烧法测定。水溶性碳水化合物(Watersoluble-carbohydrates,WSC)含量采用蒽酮-硫酸法测定,氨态氮含量用凯氏定氮法测定,缓冲能采用盐酸、氢氧化钠滴定法测定,乳酸菌采用MRS(de-Man Rogosa Sharpe)琼脂培养基培养计数,细菌采用营养琼脂培养基(Nutrient agar,广东环凯微生物有限公司)培养计数,酵母菌和霉菌数量采用马铃薯葡萄糖琼脂培养基(Potato-dextrose agar,广东环凯微生物有限公司)培养计数。乳酸菌用厌氧箱37℃培养1d~2d;细菌、酵母菌、霉菌用生化培养箱30℃培养2d~4d。
发酵品质分析:青贮袋开封后,取20g混匀的青贮料放入自封袋中,加入80mL蒸馏水,置于4℃冰箱浸泡18h后过滤,用pH计(Mettler Toledo FE28 pH计)测定浸提液pH。乳酸菌、细菌、酵母菌和霉菌数量测定同上,有机酸含量采用岛津LC-20AT型高效液相色谱仪测定:色谱条件:色谱柱(Eleven Organic Acids on Transgenomic COREGel 87H3),检测器:RID-10A,流动相为0.1mmol/L的磷酸溶液,流速1mL/min,柱温40℃,检测波长210nm,进样量20μL。
上述实施例为本发明较佳的实施方式,但本发明的实施方式并不受上述实施例的限制,其他的任何未背离本发明的精神实质与原理下所作的改变、修饰、替代、组合、简化,均应为等效的置换方式,都包含在本发明的保护范围之内。

Claims (5)
1.一株耐低营养乳酸菌,其特征在于:所述的耐低营养乳酸菌的名称为植物乳杆菌(Lactobacillus plantarum)SCLN1,保藏于广东省微生物菌种保藏中心,保藏地点为广州市先烈中路100号大院59号楼5楼广东省微生物研究所,保藏号为GDMCC No: 60944,保藏日期为2020年1月3日。
2.权利要求1所述的耐低营养乳酸菌在青贮调制中的应用。
3.根据权利要求2所述的耐低营养乳酸菌在青贮调制中的应用,其特征在于包括如下步骤:
(1)将待发酵饲料切短;
(2)将切短处理后的待发酵饲料和权利要求1所述的耐低营养乳酸菌混合均匀;
(3)将步骤(2)最终得到的饲料脱气密封后贮藏。
4.根据权利要求3所述的耐低营养乳酸菌在青贮调制中的应用,其特征在于:步骤(1)中所述的切短的规格为切短至3 cm以下。
5.根据权利要求3所述的耐低营养乳酸菌在青贮调制中的应用,其特征在于:步骤(2)中所述的耐低营养乳酸菌的添加量按每千克待发酵饲料配比1.0×108个以上所述的耐低营养乳酸菌计算。
