Abstract
The invention provides an anti-hair loss and hair-growing composition and a preparation method and application thereof. The anti-hair loss and hair-nourishing composition comprises: liquiritin, Bordeaux leaf extract and oleanolic acid. The three components are mutually matched and synergized, so that the activity of the 5 alpha-reductase is effectively inhibited, the 5 alpha-reductase is greatly prevented from catalyzing testosterone to be converted into DHT, and the problems that the high-activity DHT is combined with an androgen receptor and then acts on hair follicles to generate the effect of rapidly entering a catagen, being miniaturized, falling off hairs and the like are solved; meanwhile, the anti-hair loss and hair-growing composition can also inhibit the expression of IL-1 beta and TNF-alpha inflammatory factors in skin hair follicles, so that the aims of preventing hair loss and promoting hair growth are finally fulfilled.

In response to sebaceous gland dyssecretosis, generally medically in response to hyperandrogenism, the use of increasing estrogen regulation or the use of some drugs such as cimetidine tincture against androgens that cause sebaceous gland dyssecretosis, and the use of finasteride, dutasteride and epristeride to inhibit 5 α -reductase. CN105232410A discloses an oil-control and hair-loss-prevention shampoo product and a preparation method thereof, wherein the product comprises the following substances: lauroyl wheat amino acid potassium, cocamidopropyl betaine, soapberry peel extract, camellia seed extract, gleditsia sinensis extract, ginseng root extract, sophora flavescens root extract, cowherb seed extract, mulberry leaf extract, garden balsam stem extract, licorice root extract, safflower flower extract, rheum palmatum root extract betaine, panthenol, allantoin, hydrolyzed collagen, hydroxypropyl guar hydroxypropyl trimethyl ammonium chloride, magnolia flower oil, a preservative and water. The oil-controlling and hair loss-preventing shampoo product can reduce the secretion of scalp grease and reduce hair loss, but has poor effects on preventing hair loss and controlling skin diseases such as acne, seborrheic dermatitis and the like.
CN113116960A discloses a composition for treating alopecia and a preparation method thereof, the traditional Chinese medicine composition is prepared from the following raw materials: water extract, alcohol extract, VC1, VE1, baicalin, oleanolic acid, glycyrrhizic acid, tea polyphenol, menthol and borneol. The hair blackening agent can supplement nutrition required by hair roots to a certain extent, stimulate hair follicle activity, promote black hair growth, and is suitable for juvenile alopecia and sparse hair of young and middle-aged women; for alopecia in middle and old aged people and thin hair, it is also not effective in preventing alopecia and controlling skin diseases such as acne and seborrheic dermatitis.
Therefore, the development of a head care product that fundamentally solves and fundamentally prevents the problem of hair loss is a major research focus in the art.

In the invention, the liquiritin can inhibit peptic ulcer and prevent premature graying and alopecia, and can remarkably improve the anti-hair loss effect while diminishing inflammation, inhibiting bacteria, dissipating stagnation and relieving exterior syndrome when being added into the hair-care, anti-hair loss and hair-nourishing composition; the Bordeaux leaves extract activates the synthesis of skin beta-defensin (an endogenous antibacterial peptide), enhances the natural antibacterial system of skin, reduces the breeding of bacteria, inhibits inflammatory factors, refines pores and improves oily scalp; the main component of oleanolic acid comprises oleanolic acid, can inhibit the activity of 5 alpha-reductase so as to achieve the effect of controlling oil, and can show the advantages of quick effect taking of oil control, long oil control time and difficult repetition.
Preferably, the hair loss preventing and hair growing composition comprises the following components in parts by weight: 0.01-1 part of liquiritin, 1-3 parts of Bordeaux leaf extract and 0.01-2 parts of oleanolic acid.

In the invention, the mixed solution of specific alcohol and water with a proper proportion is selected for dilution, the exertion of the self-ability of each functional component in the hair loss preventing and growing composition can be further improved, the oil control, anti-inflammation and bacteriostasis abilities of the composition are further improved, and the hair loss problem can be better solved and prevented.

(3) the hair loss preventing and nourishing composition can prevent and improve the condition of hair loss, keep the cortex sanitary, breathable and non-greasy, and can be used for converging excessively fat sebaceous glands, reducing sebum secretion and promoting blood circulation, and has the effects of cleaning and stimulating fallen hair pores.

The specific test results are shown in Table 1
(wherein, the code C2 represents hair loss prevention and hair growth promotion composition, the DPO represents diaminopyrimidine oxide, and the two are diluted with water to 100%):

The test results in table 1 show that the inhibition rate of 100 wt% of the hair loss prevention and hair growth promotion composition on the activity of 5 alpha reductase is as high as more than 188%, and the synergistic compounding of the liquiritin, the boldo leaf extract and the oleanolic acid active ingredients fully indicates that the composition can effectively penetrate into the scalp and hair follicles, so that the activity of the 5 alpha reductase is effectively inhibited, the 5 alpha reductase is prevented from catalyzing testosterone to convert into DHT to the maximum extent, and the problems that the high-activity DHT acts on the hair follicles after being combined with an androgen receptor and quickly enters a catagen period, is miniaturized, falls off hairs and the like are avoided.
In addition, in dilution experiments, the anti-hair loss and hair growth composition disclosed by the invention is found to show a change relation of different concentrations of the inhibition rate of the 5 alpha reductase activity of cosmetics in different diluents; wherein in pure water, the composition shows low-concentration high inhibition, the concentration of C2 is less than 5 wt%, and the inhibition rate of the hair loss prevention and growth promotion composition on the activity of 5 alpha reductase is the highest and can reach more than 75%; in a mixed solution of water and alcohol, the concentration of C2 is about 10 wt%, and the inhibition rate of the hair loss prevention and growth promotion composition on the activity of 5 alpha reductase is the highest and can reach more than 105%; after DPO is additionally added into a mixed solution of water and alcohol, the concentration of C2 is about 15 wt% at a high concentration, and the anti-hair-regrowth composition has the highest inhibition rate on 5 alpha reductase activity and can reach more than 185%.
In conclusion, 100 wt% of the anti-hair loss and hair growth composition has extremely obvious synergistic effect of the components; however, the hair loss preventing and hair growing composition of the present invention exhibits not only concentration dependence but also partial or irregular antagonism between components in different fold of diluted solvent, which in turn results in a decrease in the inhibition rate of 5 α reductase activity of partial examples. Therefore, the most suitable scheme is to adopt a mixed solvent of alcohol and water, and additionally add diaminopyrimidine oxide, so that the inhibition rate of 5 alpha reductase activity which is equivalent to that of 100 wt% can be achieved, and the defects and waste caused by excessive addition amount can be avoided.
Test example 2
Inflammatory factor inhibition test
Test samples: the hair loss preventing and growing compositions prepared in examples 1 to 7 and comparative examples 1 to 7;
the test method comprises the following steps: lipopolysaccharide (LPS) is utilized to stimulate human immortalized keratinocytes (HaCaT cells) of an administration group and a blank group respectively, and protein expression of relevant inflammatory factors interleukin-1 beta (IL-1 beta) and tumor necrosis factor-alpha (TNF-alpha) in supernatant of the HaCaT cells of a pretreatment group and a control group of the administration group is detected by ELISA.
HaCaT cells were routinely cultured in DMEM complete medium containing 10% (volume fraction) fetal bovine serum (supplemented with 1% diabody: penicillin 100U/mL and streptomycin 100. mu.g/mL) at 37 ℃ in 5% (volume fraction) CO2 saturated humidity incubator, with liquid change once every 2-3 days and passage once every 3-7 days. Taking HaCaT cells in logarithmic phase, digesting the cells with 0.25% trypsin-EDTA for 10min, cleaning the cells, adding a complete culture medium into the cell sediment, and gently blowing to obtain cell suspension. Fresh culture medium was used to adjust the cell concentration to 4X 104And (4) inoculating the cells to a 24-well plate, incubating for 24 hours in a cell incubator, and adding LPS after the adherence of the cells reaches about 60-70%. Setting a blank group, a Lipopolysaccharide (LPS) 20 mu g/mL stimulation 24h group, a pretreatment 24h + LPS 20 mu g/mL stimulation 24h group of different administration groups, setting 5 multiple holes in each group, sucking cell supernatant of each hole after stimulation is finished, subpackaging the cell supernatant into 1.5mL EP tubes, and immediately using or freezing at-20 ℃ for standby. The expression level of TNF-alpha and IL-1 beta in the supernatant was measured by ELISA kit method (Jiangsu enzyme-labeled Biotechnology Co., Ltd.).
The specific test results are shown in table 2:

As shown in the test data in Table 2, the hair loss prevention and hair growth promotion composition has a good inhibition effect on the generation of inflammatory factors, the content of the inflammatory factors IL-beta is reduced to be below 112 after the hair loss prevention and hair growth promotion composition is used, and the content of the inflammatory factors TNF-alpha is reduced to be below 110 after the hair loss prevention and hair growth promotion composition is used. Fully shows that the synergistic compounding of the liquiritin, the boldo leaf extract and the oleanolic acid can inhibit over-expressed IL-1 beta and TNF-alpha 2 inflammatory factors in skin hair follicles and has an anti-inflammatory effect.
Test example 3
Mouse experiment
Test samples: the hair loss preventing and growing compositions prepared in examples 1 to 7 and comparative examples 1 to 7;
the test method comprises the following steps: c57 mice (male, 5 weeks old) were kept in an environment of temperature (23 + -2) ° C, humidity (54 + -10)% and free diet and drinking water. Except for a blank group, selecting the back of a mouse for subcutaneous injection of the drug, injecting testosterone propionate of 5 mg/kg. d, successfully modeling for 4 weeks, then administering, smearing normal saline on the model group and the blank group, smearing 5% minoxidil tincture of 0.2mL/d on the minoxidil group, smearing 5 wt% of aqueous solution of composition C2 on the sample group, 1 time per day, and a period of 4 weeks.
(1) Ratio of terminal hair/vellus hair value: after the observation is finished in the 8 th week of the experiment, the mice are killed, the skin of the hair-shedding area at the back of each group of mice is taken and is flatly pasted on a fixed plate after being about 1cm multiplied by 1cm, paraffin section dewaxing, hematoxylin-eosin staining and dehydration mounting experiment operation are carried out, the morphological change of the hair follicles of the mice is observed under the 100-time microscope visual field and 3 non-overlapping visual fields on average, the length and the size of the hair follicles are measured by using Image-Pro Plus, and the ratio (A/T) of the number of the hair follicles in the growth phase to the number of the hair follicles in the resting phase is counted;
(2) ELISA assay of Dihydrotestosterone (DHT) and 5 α reductase content in skin tissue: dissecting the collected tissues of each group of mice to prepare tissue homogenate, measuring the content of DHT and 5 alpha-reductase in each group of samples by using an Elisa kit according to a method in an ELISA kit specification, recording data, and calculating the average value by using Excel;
(3) immunohistochemical determination of TGF-. beta.1 and TNF-. alpha.content in skin: taking 2.0cm multiplied by 0.8cm long-strip-shaped skin of each group of mice, placing the mice in physiological saline to fully rinse residual blood, spreading the residual blood on filter paper, drying the blood at room temperature, wrapping the mice with tin foil paper, carrying out an experiment for measuring the contents of TGF-beta 1 and TNF-alpha in the skin of the mice by immunohistochemical detection according to the using method of Anti-TGFB1 and TNF-alpha, recording data, and calculating the average value of the data by using Excel;
the specific test results are shown in table 3:

As can be seen from the test data in Table 3, the ratio of the terminal hair/vellus hair value of the HE staining result of the test area tissues on the back of the mouse is obviously increased to more than 4 after the anti-hair loss composition of the invention is used; the ELISA assay shows that the content of Dihydrotestosterone (DHT) and 5 alpha reductase in skin tissues is obviously reduced, and the immunohistochemical assay shows that the content of TGF-beta 1 and TNF-alpha in skin is obviously reduced. Therefore, the synergistic combination of the liquiritin, the boldo leaf extracting solution and the oleanolic acid can effectively penetrate into the scalp and hair follicles, effectively inhibit 5 alpha-reductase and improve hair follicle atrophy caused by DHT (DHT); meanwhile, IL-1 beta and TNF-alpha inflammatory factors which are over-expressed due to the breeding of anaerobic bacteria acne bacillus in skin hair follicles are inhibited, the effects of controlling oil and resisting inflammation can be achieved by treating both symptoms and root causes, the microcirculation of the hair follicles is fundamentally improved, the cell metabolism is stimulated, and the hair loss problem is fundamentally solved.
The applicant states that the present invention is illustrated by the above examples to show the hair loss prevention and hair growth promotion composition of the present invention, and the preparation method and application thereof, but the present invention is not limited to the above examples, i.e. it is not meant that the present invention is implemented only by relying on the above examples. It should be understood by those skilled in the art that any modification of the present invention, equivalent substitutions of the raw materials of the product of the present invention, addition of auxiliary components, selection of specific modes, etc., are within the scope and disclosure of the present invention.
Claims (10)
1. An anti-alopecia hair-growing composition, comprising: liquiritin, Bordeaux leaf extract and oleanolic acid.
2. The hair loss prevention and hair growth composition of claim 1, wherein the hair loss prevention and hair growth composition comprises in parts by weight: 0.01-1 part of liquiritin, 1-3 parts of Bordeaux leaf extract and 0.01-2 parts of oleanolic acid.
3. The hair loss prevention and hair growth promoting composition according to claim 1 or 2, further comprising any one or a combination of at least two of an angelica sinensis extract, a zanthoxylum bungeanum fruit extract, a sophora flavescens extract or a forsythia suspensa extract, preferably a combination of an angelica sinensis extract, a zanthoxylum bungeanum fruit extract, a sophora flavescens extract and a forsythia suspensa extract.
4. The anti-alopecia hair-growing composition according to any one of claims 1-3, wherein the anti-alopecia hair-growing composition comprises, in parts by weight: 0.01-1 part of liquiritin, 1-3 parts of boldo leaf extract, 0.01-2 parts of oleanolic acid, 5-10 parts of angelica extract, 1-5 parts of zanthoxylum fruit extract, 3-8 parts of sophora flavescens extract and 3-8 parts of forsythia extract.
5. The anti-alopecia hair care composition according to any one of claims 1-4, wherein the anti-alopecia hair care composition further comprises a solvent;
preferably, the solvent is a mixed solution of alcohol and water;
preferably, the mass ratio of the alcohol to the water is 1 (1-2);
preferably, the alcohol comprises any one or a combination of at least two of ethanol, isoprene glycol, caprylyl glycol, propylene glycol, butylene glycol, hexylene glycol, or glycerol, preferably butylene glycol and/or hexylene glycol.
6. The hair loss prevention and hair growth promoting composition according to any one of claims 1 to 5, wherein the hair loss prevention and hair growth promoting composition comprises the following components in percentage by mass: 0.01-1% of liquiritin, 1-3% of Bordeaux leaves extract, 0.01-2% of oleanolic acid, 5-10% of Chinese angelica extract, 1-5% of Chinese prickly ash fruit extract, 3-8% of radix sophorae flavescentis extract, 3-8% of fructus forsythiae extract, 10-30% of butanediol, 10-30% of hexanediol and the balance of water.
7. The method of preparing the anti-alopecia hair-growing composition according to any one of claims 1-6, wherein the method of preparing the anti-alopecia hair-growing composition comprises: dissolving liquiritin, Bordeaux leaves extracting solution and oleanolic acid, and filtering to obtain the hair loss preventing and hair growing composition.
8. The method of preparing the hair loss prevention and hair growth composition of claim 7, wherein the method of preparing the hair loss prevention and hair growth composition comprises: dissolving liquiritin, Bordeaux leaves extracting solution, oleanolic acid, Chinese angelica extracting solution, Chinese prickly ash fruit extracting solution, radix sophorae flavescentis extracting solution and fructus forsythiae extracting solution in mixed solution of alcohol and water, mixing and stirring, and filtering to obtain the hair loss preventing and hair nourishing composition.
9. Use of the anti-alopecia hair-care composition according to any one of claims 1-6 for preparing a cosmetic.
10. A cosmetic comprising the hair loss preventing and hair growth promoting composition according to any one of claims 1 to 6;
preferably, the cosmetic comprises any one or combination of at least two of shampoo, cream shampoo, essence or hair conditioner;
preferably, the addition amount of the anti-hair loss and hair growth promoting composition accounts for 1-20% of the total mass of the cosmetic.
