背景

现在将详细参考当前公开的主题的各种实施方案,其中的一个或更多个实例如下所述。每个实施方案都是以解释而非限制主题的方式提供。事实上,对于本领域的技术人员来说明显的是,在不脱离本公开内容的范围或精神的情况下,可以对本公开内容进行多种修改和变化。例如,作为一个实施方案的一部分示出或描述的特征可以用在另一个实施方案中,以产生又一个实施方案。因此,本公开内容意图是包括在所附权利要求及其等同物的范围内的修改和变化。
本公开内容整体上涉及能够特异性结合弹性蛋白的表位的抗弹性蛋白抗体及其抗原结合片段。更具体地,公开了一种分离的抗体或抗原结合片段,该分离的抗体或抗原结合片段对大鼠、小鼠、猪、马、狗和人以及其它形式的无定形交联弹性蛋白具有特异性。具体地,所公开的抗体及抗原结合片段特异性识别和结合以下中的一种或更多种的表位序列:GALGPGGKPPKPGAGLL (SEQ ID NO:1)、LGYPIKAPKLPGGYGLPYTTGKLPYGYPGGVAGAAGKAGYPTTGTGV(SEQ ID NO:2)或PGGYGLPYTTGKLPYGYP(SEQ ID NO:3)。还公开了递送剂,该递送剂可以掺入抗弹性蛋白抗体及其抗原结合片段作为靶向剂,用于将生物活性剂递送至包含弹性蛋白的区域。
以SEQ ID NO:1-3为例的表位序列是弹性纤维的无定形的、交联的弹性蛋白组分的多肽组分,其在弹性纤维降解,并且特别是在弹性纤维的微原纤维支架结构降解时,可以变得暴露和可接近。因此,在一种实施方案中,所公开的靶向剂可以用于结合受损的弹性纤维,并且可以表现出很少或不结合可能在血液中循环的健康的弹性纤维或可溶性弹性蛋白前体或分解组分。例如,包括特异性识别和结合SEQ ID NO:1-3中的一种或更多种的抗体或其抗原结合片段的靶向剂可以表现出很少或不结合α-弹性蛋白降解产物。在一种实施方案中,靶向剂可以结合不再可溶但未完全交联并形成弹性纤维的未成熟弹性蛋白,例如动脉粥样硬化纤维帽中的未成熟弹性蛋白。

修饰可以包括N-末端修饰和/或C-末端修饰。例如,修饰可以包括N-末端生物素化和/或C-末端生物素化。在一种实施方案中,抗体或抗原结合片段的可分泌形式包含允许结合到免疫球蛋白(Ig)铰链区的N-末端修饰。在另一种实施方案中,Ig铰链区来自但不限于IgA铰链区。在另一种实施方案中,抗体或抗原结合片段的可分泌形式包括允许结合到酶促可生物素化位点的N-末端修饰和/或C-末端修饰。在另一种实施方案中,所述位点的生物素化可以使该位点功能化,以结合至任何用链霉亲和素、亲和素、亲和素衍生部分或第二试剂包覆的表面。
修饰可以包括,例如,添加N-连接或O-连接的碳水化合物链,将化学部分附接到氨基酸主链上,对N-连接或O-连接的碳水化合物链进行化学修饰,以及添加或缺失N-末端甲硫氨酸残基。
抗体或抗原结合片段可以通过诸如本领域周知的任何合成或重组方法产生。抗体或抗原结合片段可以通过本领域已知的技术手段进一步修饰以改变生物物理或生物特性。例如,抗体可以被修饰以增加其抗蛋白酶的稳定性,或改变其亲脂性、溶解性或对SEQ IDNO:1-3中的一种或更多种的结合亲和力。
例如,抗体可以通过用靶抗原免疫接种各种动物,包括小鼠、大鼠、兔、山羊、灵长类动物、鸡和人来产生,该靶抗原诸如所描述的完整肽序列或包括至少一个抗弹性蛋白表位的、包含所描述的序列中的一种或更多种的弹性蛋白的肽片段。在一种实施方案中,可以在免疫接种动物之前纯化抗原或包含抗原的肽片段。免疫接种后获得的抗体或抗原结合片段可以通过本领域已知的方法例如凝胶过滤、离子交换、亲和层析等纯化。亲和层析或本领域已知的许多其它技术中的任何一种可用于从血清、腹水或杂交瘤上清液中分离多克隆抗体或单克隆抗体。
“纯化的”是指抗体与通常与抗体结合的蛋白质中的至少一些分离,并且优选与除蛋白质以外的所有细胞物质分离。
抗体或其抗原结合片段可以通过使用基因重组技术产生。例如,在形成嵌合抗体、人源化抗体、抗体的功能片段等,诸如Fv、SCA、scFv-Fc等中,可以使用遗传重组技术。
在一种实施方案中,用于产生掺入重链的可变区(SEQ ID NO:7)和轻链的可变区(SEQ ID NO:25)的全部或一部分的靶向剂的方法可以通过使用遗传重组技术来进行,所述靶向剂例如包括一个或更多个CDR区(SEQ ID NO:9、11、13、27、29、31),例如在嵌合抗体的形成中。

由于密码子冗余,存在许多理论上可能的可以编码本文所描述的抗体或抗原结合片段的cDNA序列变体。此外,修饰天然蛋白序列同时保留结合活性的变体还增加了这一数目。对于这些实施方案,遗传修饰可以导致肽(例如,SEQ ID NO:7)或保留天然肽的结合功能的肽变体的表达。

在一种实施方案中,重组技术可用于产生包含特异性CDR的抗体,该特异性CDR包括SEQ ID NO:9、11、13、27、29或31中的一种或更多种。例如,可以使用一种方法来形成人源化抗体,如本文所用,人源化抗体是指具有来源于除人以外的动物的CDR和来源于人的其它区域(框架区、恒定区等)的抗体。
例如,可以制备编码抗体的重链CDR(SEQ ID NO:9、11、13)和轻链CDR(SEQ ID NO:27、29、31)的核苷酸序列。作为DNA,示例了对应于由SEQ ID NO:8、10、12、26、28、30表示的每个CDR核苷酸序列的序列;然而,如上所讨论的,可以使用具有其他核苷酸序列的那些。DNA可以通过已知的方法制备,诸如PCR。DNA可以通过化学合成来制备。

包含少于完整抗体的全部CDR的靶向剂可以在类似的过程中产生。例如,可以以类似的方式产生仅包括抗体的VH区或仅包括抗体的VL区而不存在恒定区的靶向剂。
用于纯化根据本文描述的方法形成靶向剂的方法没有特别限制,并且可以采用已知的技术。例如,可以收集杂交瘤或重组细胞的培养上清液,并且抗体或抗原结合片段可以通过已知技术诸如各种层析、盐沉淀、透析、膜分离等的组合来纯化。当抗体的同型是IgG时,可以通过使用蛋白A的亲和层析方便地纯化抗体。
在使用所公开的物质时,抗体或抗原结合片段可以可操作地连接到第二物质,用于靶向和将剂递送到降解的弹性纤维或递送到降解的弹性纤维附近的区域。如本文所用,术语“可操作地连接”是指直接或间接连接,其可以是永久或暂时的(例如可降解的)连接,其中两个或更多个分子、序列、颗粒或其组合以确保组分的正常功能的方式附接,并且特别是以抗体或其抗原结合片段可结合其表位的方式附接。这样,抗体或其抗原结合片段可将任何种类的有用的剂递送至结缔组织中或结缔组织附近的区域,诸如动脉、肺、皮肤等。此外,在一些实施方案中,抗体或抗原结合片段可以直接连接至可以携带和递送一种或更多种活性剂的载体(例如,如本文进一步描述的颗粒)。因此,可以使用组合物在延长的时间内经由剂从载体中的控制释放而递送活性剂。
抗体或其抗原结合片段可用于在治疗或诊断以弹性蛋白降解为标志的疾病中递送生物活性剂,所述疾病包括心血管疾病(如动脉粥样硬化和动脉硬化),以及肺部疾病(诸如慢性支气管炎、COPD和肺气肿)。可以包括弹性纤维降解并且抗体或其抗原结合片段可用于剂递送的其他状况可包括与动脉瘤、动脉硬化、动脉粥样硬化、遗传紊乱、钝器伤、马方氏综合征、弹性假黄色瘤、皮肤老化等相关的状况。在一种实施方案中,所述物质可用于治疗在衰老以及许多遗传和代谢紊乱中常见的血管钙化。血管钙化现在被认为是在患有其他紊乱,诸如糖尿病和慢性肾病(CKD)以及在普通人群中的心血管事件的强预测因子。所述物质可用于治疗中层动脉钙化(medial arterial calcification,MAC),中层动脉钙化可独立于动脉粥样硬化而存在,并且通常与弹性纤维降解有关。弹性蛋白特异性中层钙化导致收缩血压(SBP)和脉压(PP)升高,并促成单纯收缩期高血压(ISH)。在一种实施方案中,所公开的物质可用于同时靶向未成熟的弹性蛋白纤维和/或内膜钙化和中层钙化中受损的弹性蛋白纤维。例如,当受试者中存在动脉粥样硬化钙化和中层钙化二者时,所公开的物质可以同时靶向两种钙化。

所述物质可应用于皮肤护理,诸如用于包括疤痕、皮肤下垂和皱纹的状况,这些情况通常由于弹性纤维的损失/降解而随着年龄增长出现,包括由于日晒(sun exposure)或其他疾病状态引起的弹性纤维的损失/降解。可能受益于递送剂的使用的患者还可以包括患有皮肤动脉状况诸如皮肤血管炎的那些患者。皮肤血管炎可引起在皮肤的小动脉中的弹力膜(elastic lamina)损伤,并且使用所述物质递送治疗组合物可减轻这种损伤。
可以通过使用抗体或其抗原结合片段递送的剂可以包括生物活性剂,诸如但不限于抗凝血剂、抗血小板剂、抗炎剂、SMC增殖抑制剂、MMP和组织蛋白酶抑制剂、细胞抑制剂(cytostatic agent)、抗氧化剂、螯合剂、弹性蛋白稳定和再生剂、细胞因子、酶、趋化因子、放射性同位素、酶活性毒素或化疗剂。
在一种实施方案中,所述物质可用于递送可包括DNA和/或RNA核酸构建体的遗传物质。可以通过使用所描述的靶向物质递送的遗传物质可以包括但不限于微RNA、转运RNA、核糖体RNA、沉默RNA、调节RNA、反义RNA、RNA干扰、非编码RNA和编码RNA、DNA片段、包括基因连同调控序列的质粒、功能构建体的前体(例如mRNA前体)、DNA/RNA探针等,以及类似遗传物质。

在一种实施方案中,用于靶向递送的生物活性化合物可以包括可用于直接处理降解的弹性蛋白的化合物。这样的化合物可以包括可以促进弹性蛋白交联从而为结缔组织提供额外的结构支持的那些化合物,以及能够上调弹性蛋白形成,特别是通过增加原弹性蛋白的形成和/或交联来上调弹性蛋白形成的化合物。例如,弹性蛋白交联剂,诸如五没食子酰葡萄糖(PGG)可以通过使用抗体或其抗原结合片段来递送。能够促进原弹性蛋白的形成和/或交联从而促进新弹性纤维形成的生物活性化合物包括赖氨酰氧化酶和/或增加赖氨酰氧化酶活性的剂,诸如铜离子或为环AMP(cAMP)诱导剂的福司可林(forskolin)。另一种可用于促进原弹性蛋白交联的化合物是TGF-β,其已被证明能增加赖氨酰氧化酶活性。铜离子(Cu2+)通过使电子能够从氧转移以促进弹性蛋白中赖氨酸残基的氧化脱氨和醛形成,可以增强内源性赖氨酰氧化酶的细胞外转运以及内源性和外源性赖氨酰氧化酶的功能活性。因此,抗体或其抗原结合片段可以直接或间接地与铜离子连接,用于递送至降解的弹性纤维。
在一种实施方案中,可递送可以溶解矿物质的剂,诸如例如,乙二胺四乙酸(EDTA),其已经被证明是多用途的螯合剂(versatile chelating agent);钙特异性螯合剂乙二醇-双(β-氨基乙基醚)-N,N,N’,N’-四乙酸(EGTA);乙二醇四乙酸;腈基三乙酸,羟乙基乙二胺三乙酸;8-羟基-7-碘-5-喹啉磺酸;聚(γ-谷氨酸);硫代硫酸钠;α-硫辛酸;双膦酸盐;二乙烯三胺五乙酸(DTPA);和/或本领域已知的其他螯合剂。
抗体或其抗原结合片段可以直接或间接地连接至成像剂。当经由抗体与降解的弹性纤维结合时,成像剂可用于确定弹性纤维降解的位置和程度以及诊断相关或不相关的疾病状况。成像剂可以包括本领域已知的用于CT或MRI扫描或SPECT成像的那些成像剂。可直接或间接地连接到物质的可检测标志物可以包括光活化剂(photoactivatable agents)、荧光团、放射性同位素、生物发光蛋白或肽、荧光标签(例如荧光素、异硫氰酸酯(FITC)、花青染料等)、荧光蛋白或肽、亲和标记物(例如生物素、亲和素、蛋白A等)、酶标记物(例如辣根过氧化物酶或碱性磷酸酶)、或同位素标记物(例如125I)、金颗粒、棒(rod),x-射线不透明物质,和微气泡(例如用于超声成像),或允许检测抗体和任选地对区域进行成像的任何其他这种可检测部分。

松散载体物质的选择可用于提供生物活性剂从负载颗粒中的释放速率的控制。例如,选择可生物降解的物质可以用来控制剂释放的速率,并提供可以在很大程度上通过颗粒降解速率并且在较小程度上通过活性剂扩散透过和离开大颗粒来控制的释放机制。可以使用所述物质,使得活性剂释放速率受到扩散(例如,不可降解颗粒)或纳米颗粒降解速率(例如,由于小的基质网孔尺寸,活性剂基本上不扩散透过颗粒)之一的限制,或者受到其可以被设计成期望的释放速率的某种组合的限制。
颗粒可以是微粒或纳米颗粒。如本文所用,术语纳米颗粒通常是指其尺寸(即平均直径)可为约1000纳米(nm)或更小,通常为约500nm或更小,例如约200nm或更小,或约100nm或更小的颗粒。在一种特定的实施方案中,纳米颗粒的尺寸可以为约50nm或更小,例如平均直径约20nm。在一种实施方案中,纳米颗粒可以具有约50nm至约400nm,或从约100nm至约300nm的平均直径。
可选地可以使用更大的颗粒。例如,在其他实施方案中,具有高达约50微米(μm)的平均尺寸的微粒可以用作载体。
通常,颗粒的优选尺寸可取决于具体应用,例如剂的具体递送方法,诸如经由表面应用(如在霜剂或洗剂中)、经由使用循环系统或消化道的肠胃外注射、经由吸入等,以及剂从颗粒中的期望释放速率。例如,颗粒可以具有阻止细胞摄取的尺寸,从而保留在细胞外基质中,并可用于与受损的弹性纤维相互作用。因此,在一种实施方案中,颗粒可以为约100nm或更大,因为更小的颗粒已被证明具有较高的细胞摄取。颗粒也可以足够小,以穿透内皮和穿透基底膜,从而接触结缔组织的弹性纤维。例如,在一种实施方案中,颗粒的平均直径可以为约400nm 或更小,以便穿透内皮和基底膜。当意图用于静脉施用制剂时,通常不偏向大颗粒(例如,大于约1μm),因为它们会滞留在微血管中。此外,较大的颗粒可在体内积聚或聚集。因此,对于静脉内施用,通常使用低于1μm的颗粒。
通常,颗粒载体的形状可以大体上是球形的,尽管其他形状也适合使用,包括但不限于板、棒、条、不规则形状等。如本领域技术人员将理解的,颗粒的组成、形状、尺寸和/或密度可以广泛变化。
颗粒可以被设计成具有期望的表面电荷,以更好地靶向受损的弹性纤维。例如,与带负电的颗粒相比,带正电的纳米颗粒显示出更好的细胞摄取。因此,在一种实施方案中,颗粒可以用负表面电荷研制,以将颗粒保持在细胞外基质中并避免细胞摄取。
颗粒可以根据任何合适的方法负载一种或更多种剂。例如,沉淀方法可用于在一步形成方法中形成负载颗粒。根据该方法,颗粒松散物质(例如生物相容性聚合物,诸如聚(D,L-丙交酯-共-乙交酯或PGA/PLA共聚物)可以溶解在溶剂中。合适的溶剂可以取决于所涉及的具体物质。例如,可以使用有机溶剂,包括丙酮、四氢呋喃、二甲基亚砜、二甲基甲酰胺或乙腈等。该溶液可以经过标准处理,诸如声处理等,以充分溶解聚合物。然后,可以将溶液添加,通常逐滴添加到第二溶液中。自发地或在乳化方法之后,例如在声处理之后,松散物质(bulk material)的颗粒可以在第二溶液中形成。
当使用一步形成方法时,用于递送的剂(例如治疗剂)也可以包含在第一溶液或第二溶液中。在颗粒形成时,剂可以与松散物质一起掺入颗粒中。
颗粒内或颗粒上的剂的初始浓度将根据剂的性质、递送速率等明显变化。例如,在一种实施方案中,颗粒中/颗粒上的生物活性剂的负载浓度可以从按颗粒的重量计的约4wt.%变化至大于约40wt.%,根据具体的剂、颗粒松散物质等,可以具有更高或更低的浓度。例如,在用于递送的剂在颗粒松散物质中表现出高溶解度的实施方案中,可以获得非常高的负载水平,特别是当两种物质都是高度疏水时。
形成方法可以包括两步方法,其中首先形成颗粒,随后是第二负载步骤,其中一种或更多种活性剂被负载到形成的颗粒中或负载到形成的颗粒的表面上。例如,方法可以包括在包含用于递送的剂的溶液中将预成型的、任选地交联的聚合物颗粒溶胀,以便经由扩散过程负载颗粒。在另一种实施方案中,负载方法可以包括双乳液聚合,双乳液聚合能够将亲水性化合物负载至疏水性颗粒中。用于纳米颗粒的形成方法没有特别限制,并且可以使用本领域已知的其他形成方法,例如声处理法、溶剂沉淀法等。
可以形成负载颗粒,以便控制活性化合物从颗粒中释放的速率。合适的控制机制是本领域技术人员已知的。例如,如已知的,释放速率可以取决于用于递送的剂与松散颗粒物质的相对浓度、松散纳米颗粒物质的分子量和降解特性、聚合物颗粒基质的网孔尺寸、颗粒表面和剂之间的结合机制等。在任何这些情况下,本领域的普通技术人员能够设计一种系统以获得期望的释放速率。例如,在纯扩散限制释放的情况下,这种控制可以通过改变颗粒内的剂的浓度和/或颗粒尺寸、颗粒聚合物网孔尺寸等来实现。在纯降解限制释放的情况下,聚合物单体单元,例如PLGA聚合物的乙醇酸含量,和/或颗粒松散物质的分子量以及颗粒尺寸可以被调节以“微调”活性化合物释放速率。例如,使用具有较高乙醇酸含量和较低分子量的PLGA聚合物可以导致用该聚合物形成的颗粒的降解速率增加。使用上述参数可以调节剂从颗粒中的释放速率,从而产生能够持续从几天到几个月变化的时间段持续释放的载体,最大释放速率通常从几小时到几周变化。
用于递送的剂不一定掺入到松散物质内部。例如,在一种实施方案中,剂可以键合至颗粒的表面。例如,使用类似于以下关于表位结合抗体或片段与颗粒结合的更详细描述的化学,可以将剂键合至颗粒的表面。
抗体或抗原结合片段可以根据任何合适的方法与载体缀合。例如,颗粒可以包括促进颗粒与抗体缀合的表面反应基团。表面活性基团可以包括但不限于醛、羧基、氨基、羟基等。如本领域中通常已知的,表面活性基团可以存在于形成的颗粒表面上,或者可以在形成后添加到形成的颗粒的表面上,例如经由氧化、胺化等。然后抗体或片段可以与颗粒缀合,例如通过与马来酰亚胺反应,其中抗体是巯基化抗体。
抗体或片段可以经由非特异性吸附或共价键附接到载体(例如颗粒)上。优选的附接方法通常可以取决于形成的缀合物的期望的应用。例如,在那些系统被设计成在体内发挥作用的实施方案中,预期载体颗粒会发生与各种生物制剂和组织的多次碰撞。因此,在这样的实施方案中,共价结合可以是优选的,以更好地确保抗体/片段将不会经由颗粒与其他物质的碰撞而移位。
用于将抗体/片段(以及任选地,另一种剂诸如活性治疗剂)结合到载体表面的具体化学没有特别限制。例如,在一种实施方案中,抗体或片段可以根据多肽的胺基团和氯甲基化颗粒的烷基氯之间的亲核取代反应附接到颗粒上。在另一种实施方案中,可溶性碳二亚胺(EDC)和戊二醛化学可分别用于实现多肽的胺基团与羧基化颗粒和胺化颗粒的共价结合。根据又另一种实施方案,肽可以通过链霉亲和素单层与颗粒初始共价附接,随后通过可控附接所需量的生物素化抗体而与颗粒键合。根据又另一种实施方案,抗体/片段可以使用能够交联肽的胺基团和聚合物颗粒的C-H或C-C键的光反应性试剂交联剂共价附接到颗粒,例如叠氮苯交联剂,诸如从Pierce Inc.可得的磺基-HSAB(N-羟基磺基琥珀酰亚胺基-4-叠氮基苯甲酸酯)。
在一种实施方案中,分子间隔物,例如亲水性间隔物,可用于将抗体/片段拴系(tether)到颗粒。使用间隔物可以防止共价结合的肽与颗粒表面的相互作用,并且因此防止抗体/片段的可以导致部分或完全丧失功能的结构变化。间隔物可以包括长的(例如,重均分子量在约2,000和约20,000Da之间)亲水性聚合物,诸如但不限于聚(乙二醇)、聚乙烯醇、多糖等。
间隔物和颗粒可以包含或被加工成包含官能度(functionality),以便于促进彼此结合。例如,PEG间隔物可以包含醛官能度,并且可以通过间隔物的醛基团和颗粒的胺基团之间的共价反应结合至胺化颗粒。然后可以根据简单的方法将巯基化抗体/片段附接至间隔物,所述简单的方法包括在存在马来酰亚胺的情况下将包含巯基化抗体的溶液与颗粒的水性悬浮液混合。
在缀合的最后阶段,载体颗粒可以被例如用表面活性剂,诸如20、或右旋糖酐封闭,表面活性剂可以被吸附在颗粒上以封闭暴露于溶液中的任何疏水表面并置换(displace)任何未附接的剂。低浓度的这样的物质通常不会干扰剂的活性。表面活性剂的存在可以减少不需要的蛋白-颗粒相互作用并防止颗粒聚集。在使用可能会使系统失活的物质的环境中,它还可以防止颗粒表面被其他蛋白非选择性地“污染(fouling)”。
在一种实施方案中,载体可以被设计成对于需要的应用表现出锚定特性。例如,结合能力和载体颗粒可以保持附接于降解的弹性纤维的时间长度可以通过改变颗粒尺寸和/或颗粒表面上靶向抗体/片段的浓度来设计。
缀合化合物可以根据任何合适的方法递送至降解的弹性纤维,通常取决于靶纤维的位置。例如,当考虑全身递送方法时,诸如静脉内递送途径,缀合物可以循环,直到接触到受损的弹性纤维,这仅仅是为了说明的目的,而非意图限制,如在弹性组织变性的情况下。当经由抗体/片段结合至弹性纤维时,剂可以促进检测或者可以经由例如颗粒降解、扩散等从颗粒中释放以提供所需的活性。可以根据多种递送方法应急地(acutely)或长期地递送或施用化合物,包括结合血管周(perivascular)或血管内斑块的持续释放方法、局部应用、静脉内递送、渗透泵、吸入等。

形成方法
单克隆抗体形成
将匙孔戚血蓝蛋白(KLH)与大鼠弹性蛋白的选定氨基酸肽片段(即SEQ ID NO:1-3之一)缀合。使用标准方案,对RBF/dnj或balb/c小鼠用总体积为200μL的在磷酸盐缓冲盐水(PBS)中的100μg总KLH-肽蛋白和佐剂的初始剂量进行皮下(s.c.)致敏。14天后,在弗氏不完全佐剂中给予随后的增强剂。然后以21天间隔给予不含佐剂的增强剂,共进行4次免疫接种。最后一次免疫接种通过腹膜内(i.p.)注射给予。最后一次免疫接种后5天,在CO2室中对小鼠实施安乐死,并且收获脾以获得细胞,然后在聚乙二醇(PEG)存在下将细胞与FOX-NY或Sp2/0-Ag14骨髓瘤融合,以制备将在96孔微量滴定板中使用标准细胞培养程序培养的杂交瘤。融合后14天,通过ELISA步骤筛选来自这些粗杂交瘤混合物的上清液针对未缀合的游离肽片段的免疫反应性。通过限制稀释进一步培养和克隆阳性杂交瘤以获得分泌单克隆抗体的细胞系(杂交瘤)。然后再次检查杂交瘤培养物上清液的特异性,并对同种型和技术应用进行全面表征。
多克隆抗体形成
将匙孔戚血蓝蛋白(KLH)与大鼠弹性蛋白的选定氨基酸肽片段(即SEQ ID NO:1-3之一)缀合。用总体积为200μL的在磷酸盐缓冲盐水(PBS)中的100μg总KLH-肽蛋白和佐剂的初始剂量对新西兰白兔进行皮下(s.c.)致敏,在两个肩部区域的每一个和两个后臀部区域的每一个处给予。14天后,在弗氏不完全佐剂中给予随后的增强剂。然后以21天间隔给予不含佐剂的增强剂,共进行5次免疫接种。10天后,对兔实施安乐死并抽血,让血液凝结,并且离心后收集血清。然后对血清进行抗体滴度表征。
纳米颗粒形成
1)负载DiR染料的纳米颗粒(用于荧光标记和体内成像)
通过凝聚作用制备负载DiR染料的颗粒。简而言之,通过将250mg牛血清白蛋白(BSA)(SeraCare,MA)溶解在4mL去离子水中,获得负载荧光红外染料1,1-二十八烷基-3,3,3,3-四甲基吲哚三碳菁碘(DIR)的纳米颗粒(DIR-NP)。然后,将2.5mg DIR溶解在100μL丙酮中,并加入至BSA溶液。搅拌一小时后,在连续声处理(Omni Ruptor 400UltrasonicHomogenizer,Omni International Inc,Kennesaw,GA)下,经半小时将混合物逐滴加入到24mL乙醇中。为了交联,在搅拌过程中加入戊二醛(EM级70%,EMS,PA)(42μg每mg BSA)。接下来,将10mg DIR-NP与2.5mg异双官能交联剂α-马来酰亚胺-ω-N-羟基琥珀酰亚胺酯聚(乙二醇)(马来酰亚胺-PEG-NHS酯,MW 2000Da,Nanocs Inc.,NY)孵育,以获得巯基反应性颗粒系统。使用特劳特试剂(34μg,G-Biosciences,Saint Louis,MO)将10μg作为对照的兔抗大鼠弹性蛋白抗体(United States Biological,Swampscott,MA)或针对本文所述序列形成的抗体硫醇化,并且将混合物在HEPES缓冲液(20mM,pH=9.0)中室温孵育1小时。将硫醇化抗体用HEPES缓冲液冲洗,并添加至纳米颗粒(4μg抗体每1mg NP),并孵育过夜以缀合。
2)负载EDTA (螯合剂)的纳米颗粒
通过将200mg BSA (SeraCare,MA)和100mg乙二胺四乙酸二钠盐(EDTA)(FisherScientific,NJ)溶解在4mL去离子水中,并将pH调节至8.5,获得负载EDTA的纳米颗粒(EDTA-NP)。在探针声处理持续1小时下,将水溶液逐滴加入至16mL乙醇。为了交联,在声处理过程中加入戊二醛(10μg每mg BSA)。弹性蛋白抗体缀合过程类似于DIR-NP的过程。
3)负载PGG的纳米颗粒(用于稳定弹性蛋白)

4)负载有MMP抑制剂BB-94的纳米颗粒
使用基于溶剂扩散的纳米沉淀法制备聚(D,L-丙交酯)(PLA)纳米颗粒。将PLA(平均MW 75k-120k)(Sigma Aldrich,St.Louis,MO)溶解在丙酮(VWR International,Radnor,PA)中。将1,2-二硬脂酰-sn-甘油-3-磷酸乙醇胺-N-[甲氧基(聚乙二醇)-2000](DSPE-PEG(2000)马来酰亚胺)(Avanti Polar Lipids,Inc.,Alabaster,AL)和BB-94(SigmaAldrich,St.Louis,MO)溶解于二甲基亚砜(DMSO)(Sigma Aldrich,St.Louis,MO)中,然后将上述溶液加入至PLA溶液。保持在4℃声处理(Omni Ruptor 4000)20分钟下,将聚合物溶液逐滴加入(16μl/sec)至水中。声处理后,将颗粒用蒸馏水洗涤两次,在4℃、14000x g离心30分钟,并且然后在蒸馏水中重悬。所有实验的非溶剂(水)与溶剂(丙酮)的比为1:15。制备包含5:1、10:1和15:1的聚合物与BB-94的比的三个不同批次,其中两种聚合物(PLA:DSPE-PEG(2000)马来酰亚胺)的比为4:1。弹性蛋白抗体缀合过程类似于DIR-NP的缀合过程。
5)负载盐酸多西环素的纳米颗粒
负载盐酸多西环素(Sigma Aldrich,St.Louis,MO)的BSA纳米颗粒用类似的过程制备。简而言之,将25mg盐酸多西环素(<DOXTot)与100mg BSA(BSATot)一起溶解于2mL水中,并允许以500rpm搅拌30分钟。随后,使用自动分液器以1ml/min的速度逐滴加入4mL乙醇,这使得溶液浑浊。向其中加入8%戊二醛(40μg/mg BSA)以交联白蛋白,并且将混合物在室温搅拌2小时。所得溶液以14,000rpm离心10分钟,以分离形成的纳米颗粒。将纳米颗粒用DI水洗涤三次,然后继续抗弹性蛋白抗体缀合。通过使用UV分光光度计(BioTek InstrumentsInc.,Winooski,VT)测量273nm处的吸光度,使用从洗脱获得的上清液来估计游离多西环素的量(DOXF)。DOXTot和DOXF之间的差异给出了约17%的封装的多西环素的量(DOXNP)。
实施例1
如所描述的,针对SEQ ID NO:1(GALGPGGKPPKPGAGLL)形成单克隆抗体和多克隆抗体。
大鼠和小鼠主动脉(n=8)购自BioChemed Inc。弹性蛋白酶溶液通过将猪胰腺弹性蛋白酶(10U/mL)溶解在DI水中制备。将主动脉系在缝合线上,并且将主动脉的下半部分在37℃悬浮在弹性蛋白酶溶液中持续1小时。然后在盐水中彻底洗涤主动脉,并将整个主动脉在10mg/mL的用SEQ ID NO:1的单克隆抗体加标签的DiR-NP的溶液中孵育过夜。随后,主动脉在摇床上用盐水洗涤90分钟,并使用IVIS成像系统成像。图1示出了大鼠主动脉的结果,并且图2示出了小鼠主动脉的结果。如所示的,抗弹性蛋白抗体优先地键合到包含降解的弹性纤维的主动脉部分。
8只6周龄的斯普拉格-杜勒雄性大鼠通过外膜周应用0.5M CaCl2 3次,每次5分钟,经受腹主动脉损伤。损伤10天后,向大鼠注射浓度为10mg/kg、用SEQ ID NO:1的抗弹性蛋白单克隆抗体加标签的BSA-DiR NP。注射后24小时,对动物实施安乐死,并使用IVIS成像系统对它们的主动脉进行纳米颗粒靶向成像。如图3所示,抗体成功地靶向主动脉中受损的弹性蛋白(正方形区域),同时放过主动脉其他部分中的健康弹性蛋白。
实施例2
处理钙化的人腿部动脉的一部分并包埋在石蜡中。切取5μm的切片并封固在带正电的载玻片上并进行组织学分析。使用商购可得的IHC试剂盒(Enzo Life Sciences)进行免疫组化(IHC),以SEQ ID NO:1的单克隆抗体作为一抗(10μg/mL)检测弹性蛋白。此外,伟郝夫范吉森(VVG)染色被用来识别断裂(或受损)的弹性蛋白纤维。IHC显示,SEQ ID NO:1的抗体能够成功地标记动脉中存在的受损的弹性蛋白(由图4的较暗区域指示)。伟郝夫范吉森(VVG)染色示出了弹性蛋白断裂和受损(图6,图7)。尽管用VVG可以看到受损的弹性蛋白纤维,但是当与仅对照抗体孵育时,弹性蛋白的IHC没有显示出任何暗染色(图5),有效地证实了SEQ ID NO:1的抗体能够识别和结合受损弹性纤维的人弹性蛋白,而对照抗体不能。
实施例3
在6周龄雄性斯普拉格-杜勒(SD)大鼠(n=6)中建立肺气肿模型,大鼠接受气管内注射溶解于磷酸盐缓冲盐水(PBS)并过滤灭菌的50U猪胰腺弹性蛋白酶(PPE)(ElastinProducts Company Inc.,Owensville,MO)。经过弹性蛋白酶处理的大鼠在四周内出现了弹性蛋白损伤。对动物实施安乐死并用盐水冲洗全身后,小心地分离主动脉。肺的一部分被处理并包埋在石蜡中。制作五微米厚的切片,并使用IHC试剂盒(Enzo Lifesciences),使用本文描述的单克隆抗体作为一抗,对多种组织进行免疫组化。石蜡包埋切片的IHC根据制造商的方案进行。对于所有实验,抗体浓度保持在10μg/mL。
类似的方案被用于开发小鼠的肺气肿模型和小鼠的AngII动脉瘤模型。图8-图11示出了包括大鼠肺的弹性蛋白酶肺气肿模型(图8)、小鼠肺的弹性蛋白酶肺气肿模型(图9)、小鼠主动脉的AngII动脉瘤模型(图10)和弹性蛋白酶处理的小鼠皮肤(图11)的动物模型中,与针对SEQ ID NO:1的单克隆抗体孵育后多种组织的IHC染色。如所示的,针对SEQ IDNO:1产生的抗体能够标记多种不同组织类型中受损的弹性蛋白。

实施例4

实施例6

实施例7
蛋白印迹和银染用于检测所公开的抗体与原弹性蛋白的结合。使用细胞培养基、α弹性蛋白标准品和来自弹性蛋白酶消化的大鼠主动脉的上清液,通过针对SEQ ID NO:1产生的单克隆抗体和作为对照产生的小鼠单克隆弹性蛋白抗体(在杂交瘤之前从小鼠产生的单克隆抗体)来测试弹性蛋白的检测。银染(图25)示出了可溶性弹性蛋白在75kDa和50kDa蛋白标志物之间,但是两种抗体都不能检测可溶性弹性蛋白。当使用弹性蛋白酶消化的主动脉样品时,兔多克隆抗体显示在50kDa蛋白质标志物附近的条带。对于弹性蛋白酶消化的主动脉样品,小鼠单克隆在150kDa标志物以上有一个非常特异的单一条带,但细胞培养基没有条带。
实施例8
针对人弹性蛋白序列SEQ ID NO:3(PGGYGLPYTTGKLPYGYP)形成单克隆IgG1同种型抗体。
使用H&E染色的IHC(图26)示出了对患有轻度动脉瘤的人主动脉中降解的弹性蛋白的特异性。弹性蛋白VVG染色中的圈出部分(图27)显示弹性蛋白降解,并且同一区域显示抗体与切片结合(图28)。仅对照二抗显示无信号(图29)。
使用H&E染色的IHC(图30)示出了对患有轻度动脉瘤的人主动脉中降解的弹性蛋白的特异性。弹性蛋白VVG染色中的圈出部分(图31)显示弹性蛋白降解,并且同一区域显示抗体与切片结合(图32)。仅对照二抗显示无信号(图33)。
实施例9
新鲜的人颈动脉内膜切除术(CEA)样品分成两部分:一部分用于使用负载针对SEQID NO:3产生的抗体的DiR-NP的靶向(ELN组),并且另一部分用于使用不含弹性蛋白抗体的DiR-NP的靶向(对照组)。将样品在4℃孵育12小时。然后用PBS洗涤样品,并且用IVIS测试纳米颗粒附接。IVIS结果(图34)示出,ELN缀合的DiR-NP附接于动脉中降解的和未成熟的弹性蛋白,而没有抗体的对照NP不附接。
然后样品经受脱钙以进行组织学制备,并包埋在中以获得冰冻切片用于染色。如图35和图36所示,通过直接检查(左图)、H&E染色(中图)和VVG染色(右图)可以看到纳米颗粒对动脉粥样硬化中的弹性蛋白的靶向。没有抗体的对照NP(下图)没有被靶向。
实施例10
表征了表达针对人弹性蛋白序列SEQ ID NO:3形成的单克隆IgG1同种型抗体的杂交瘤。按照试剂的技术手册从杂交瘤细胞中分离总RNA。然后按照PrimeScriptTM第一链cDNA合成试剂盒的技术手册,使用同种型特异性反义引物或通用引物将总RNA逆转录成cDNA。根据GenScript的cDNA末端快速扩增(RACE)的标准操作程序,扩增抗体片段VH、VL、CH和CL。将扩增的抗体片段分别克隆至标准克隆载体中。进行菌落PCR以筛选具有正确大小插入物的克隆。对每个片段测序不少于5个具有正确大小的插入物的克隆。对不同克隆的序列进行比对,并且获得共有序列。
图37示意性地示出单克隆抗体。通过对恒定区的序列进行分析,同种型为小鼠IgG/κ。对于共有序列,对五个克隆的VH、CH、VL和CL序列进行测序,所有序列具有大于99%的序列同一性。以下的表1提供了对V(D)J连接的IMGT分析。所有可变序列都是有效的,并且没有检测到D区段。

实施例11
柠檬酸盐加帽的金纳米颗粒(citrate capped gold nanoparticle,GNP)购自Meliorum Technologies,Rochester,NY,平均尺寸为150±25nm。将异双官能硫醇-PEG-酸(SH-PEG-COOH)(2000MW,Nanocs,New York,NY)以4:1的重量比加入至GNP,并且混合物在4℃伴随温和摇动孵育48小时,以实现聚乙二醇化。在室温以10,000rpm离心20分钟后收集聚乙二醇化的GNP,并重悬在0.1M MES(pH:5.5)中。如本文描述的,使用EDC/NHS化学将聚乙二醇化的GNP与抗弹性蛋白抗体缀合。简而言之,将EDC(N-(3-二甲基氨基丙基)-N’-乙基碳二亚胺盐酸盐)(Oakwood Chemical,Estill,SC)和磺基-NHS(N-羟基磺基琥珀酰亚胺)(SigmaAldrich,St.Louis,MO)分别以2:1和4:1的重量比加入至聚乙二醇化的GNP。将该混合物伴随温和涡旋在室温孵育6小时。在室温以10,000rpm离心20分钟后,收集得到的GNP,并重悬在1mL PBS(pH 7.8)中。向每mg GNP添加4μg抗弹性蛋白抗体,并且混合物在4℃缓慢摇动孵育过夜。通过将所得溶液以10,000rpm离心20分钟来除去过量的抗体。将得到的抗体/纳米颗粒缀合物重悬在盐水中至3mg/mL的浓度用于注射。
从杰克逊实验室(Bar Harbor,ME)获得15只雄性低密度脂蛋白受体缺陷型(LDLr)(-/-)小鼠(2月龄,在C57BL/6背景上)。11只小鼠用于动脉瘤研究,而另外4只小鼠用作健康的年龄对照。通过全身输注血管紧张素II(AngII,Bachem Americas,Torrance,CA)结合具有饱和脂肪(21%wt/wt)和胆固醇(0.2%wt/wt;目录号TD88137;Harlan Teklad)的饮食诱导动脉瘤。简而言之,在AngII输注前1周和AngII输注期间的6周向小鼠饲喂高脂饮食。在异氟醚麻醉下,将充满AngII的渗透泵(Model 2004;Alzet,Cupertino,CA)通过小鼠右后肩处的切口皮下植入。在整个手术过程中,小鼠吸入2%至3%的异氟醚作为麻醉剂。AngII的泵送速率被设定为1000ng/kg/min。植入后4周移出(explant)泵,并且允许小鼠恢复2周。通过使用线性阵列探头(MS-550D,宽带频率22MHz-55MHz),使用高频超声机FujifilmVisualSonics Vevo 2100(Fujifilm VisualSonics,Toronto,ON,Canada)监测疾病进展。
在吸入2%-3%异氟醚的情况下,通过眶后注射给予小鼠(n=15)10mg/kg动物体重的剂量的EL-GNP作为对比剂。注射后24小时对小鼠实施安乐死,并分离整个主动脉(从升主动脉到髂总动脉杈)。在显微CT扫描前,清除主动脉周围的结缔组织。将主动脉浸入玉米油中,并用高性能体内显微CT系统(Skyscan 1176,Bruker,Billerica,MA)成像(90kV,250mAs,300ms,0.2mm Al滤器)。使用基于Feldkamp算法的Skyscan Nrecon软件进行重建。主动脉的重建图像被可视化,并使用DataViewer和CT-Vox软件测量动脉瘤的尺寸。获得3D最大强度投影(MIP)图像(图38,左)以确定EL-GNP在主动脉内的分布,同时获得衰减图像(图38,右)以研究由EL-GNP和组织两者给出的信号的强度。使用CT-An软件进一步量化信号强度。
用CytoViva的增强暗场显微镜光学系统(CytoViva,Inc.,Auburn,AL)对冰冻切片组织样品(5μm)进行检查。该系统(Olympus BX51)采用浸油(类型A,nd>1.515,CargilleBrand)超暗场聚光器和具有在1.2至1.4的可调节数值孔径的40×空气平面-FL物镜。照明由Fiber-Lite DC-950调节层压机提供。使用Exponent 7软件以2.8的增益和53ms的曝光时间来获得增强的暗场显微图像(图39),以可视化EL-GNP。使用超光谱成像仪(安装在显微镜上,并由Exelis Visual Information Solutions,Inc.的Environment forVisualization软件控制)以曝光时间为0.25ms,全视场(643行)提取光谱信息,用于绘制样品中的EL-GNP(图41)。对阴性对照样品进行成像和分析,以建立光谱库作为参考。通过从阳性对照的光谱库减去阴性对照的光谱库以应用过滤的光谱库来实现绘制。
动脉瘤和健康主动脉的冰冻切片用于组织学分析(图40)。主动脉在缓冲的福尔马林中固定,在DI水中洗涤后包埋在最佳切割温度(OCT)化合物(Sakura Finetek,Torrance,CA)中,并按照标准程序切片。将五微米的切片封固在带正电荷的玻璃载玻片上。将载玻片放入100%预冷丙酮(Fisher Science Education,Nazareth,PA)中10分钟,以将组织粘附到载玻片上。随后,用自来水冲洗载玻片3分钟,以去除OCT化合物用于进一步染色。载玻片用伟郝夫范吉森(VVG)染色,以确定不同样品中的弹性蛋白损伤。
图39和图40呈现了主动脉的两个切片(图像中的左和右)的暗场图像(图39)和VVG染色图像(图40)。如可以看出的,在图39的左图中可以看到更强的暗场图像信号,显示了相比每一个图右侧的组织切片所示的更多的弹性纤维受损(左,图40),每一个图右侧的组织切片主要包含完整的弹性蛋白纤维(右,图40)。在降解的弹性蛋白暴露的位置发现由金纳米颗粒给出的信号,由图像中较暗的箭头指示,而健康和完整的弹性蛋白纤维没有GNP信号,如由图像中较亮的箭头指示的。

8.根据权利要求7所述的组合物,其中所述第二物质包括生物活性剂,诸如抗凝血剂、抗血小板剂、抗炎剂、SMC增殖抑制剂、基质金属蛋白酶抑制剂、组织蛋白酶抑制剂、细胞抑制剂、抗氧化剂、胶原蛋白稳定剂、弹性蛋白稳定和再生剂、细胞因子、酶、趋化因子、放射性同位素、毒素、免疫调节剂、螯合剂、核酸构建体、化疗剂、弹性蛋白交联剂或原弹性蛋白交联剂。
9.根据权利要求7所述的组合物,其中所述第二物质包括成像剂,诸如光活化剂、荧光团、放射性同位素、生物发光蛋白或肽、荧光标签、荧光蛋白或肽、亲和标记物、酶标记物、MRI剂、金颗粒、x射线不透明物质或同位素标记物。
10.根据权利要求7所述的组合物,其中所述第二物质包括载体,诸如颗粒形式的载体,例如纳米颗粒,该纳米颗粒任选地包括包含聚合物的可降解颗粒。
11.一种杂交瘤或遗传修饰细胞,所述杂交瘤或遗传修饰细胞包含编码权利要求1至6中任一项所述的抗体或抗原结合片段的核酸序列。
12.根据权利要求11所述的杂交瘤或遗传修饰细胞,所述核酸序列包括SEQ ID NO:8、SEQ ID NO:10、SEQ ID NO:12、SEQ ID NO:26、SEQ ID NO:28和SEQ ID NO:30中的一种或更多种,或其中所述核酸序列包括SEQ ID NO:6和/或SEQ ID NO:24。
13.一种方法,所述方法包括将降解的弹性纤维与权利要求1至6中任一项所述的抗体或其抗原结合片段接触,其中所述抗体或其抗原结合片段可操作地连接至第二物质。
14.根据权利要求13所述的方法,其中所述第二物质包括载体和掺入到所述载体中的生物活性剂,在所述抗体或其抗原结合片段结合至所述降解的弹性纤维之后,所述生物活性剂从所述载体中释放。
15.根据权利要求13所述的方法,所述第二物质包括成像剂。
