本发明公开了一种醋红景天及其炮制工艺和应用,涉及中医药领域,炮制工艺具体为:取净红景天片进行炒制,炒制的同时喷淋醋液,炒干后冷却过筛,制得醋红景天,同时还公开了其在固体植物饮品上的应用。该发明是通过在红景天炒制时进行喷淋醋液,首先由于醋液具有引药入肝的功效,更利于红景天中有效成分的溶出,另外,由于红景天苷具有很强的耐氧性能,将其制备成固体植物饮品,有效预防和改善高原地区旅游人群的高原反应。

技术领域
本发明涉及中药领域,具体讲是一种醋红景天炮制工艺及其应用。
背景技术
红景天为景天科植物大花红景天的干燥根和根茎。秋季花茎凋枯后采挖,除去粗皮,洗净,晒干。功能主治:益气活血,通脉平喘。用于气虚血瘀,胸痹心痛,中风偏瘫,倦怠气喘。
红景天的应用已有悠久的历史,《神农本草经》记载“上品、无毒、多服久服不伤人,有轻身益气之功效”。藏《四部医典》记载其“性平、味涩、善润肺,能补肾、理气养血,主治周身乏困、胸闷、恶心、体虚等症。”明李时珍《本草纲目》中记载“景天、上品,甘、涩、寒、无毒,具有祛邪恶气、补诸不足之功效”。红景天的化学成分主要有多糖类、皂甙类、黄酮类等。现代研究表明,红景天具有增强免疫、抗缺氧、抗氧化、抗寒冷等药理作用。其中红景天苷独特的结构和显著的生物活性,受到人们的关注,
但是如何将其活性成分更好地进行应用或是更易在应用中发挥作用,是值得研究的课题。
发明内容
本发明的目的在于至少解决现有技术中存在的技术问题之一,提供一种醋红景天及其炮制工艺和应用。
本发明的技术解决方案如下:
一种醋红景天炮制工艺,取净红景天片进行炒制,炒制的同时喷淋醋液,炒干后冷却过筛,制得醋红景天。
优选地,所述醋液的乙酸浓度为20-30g/L。
优选地,每100kg红景天片,喷淋10-20kg醋液。
优选地,所述炒制的温度为80-120℃。
本发明还公开了一种醋红景天,采用如上所述的炮制工艺制得。
本发明还公开了一种如上所述的醋红景天在制备固体植物饮品上的应用,包含以下步骤:
步骤一:先将醋红景天烘干粉碎,得粗粉,往粗粉中加入提取溶剂进行浸泡,浸泡的同时进行超声处理,处理完成后放入高压容器内,在高压下进行保压,保压完成后恢复常压,进行高速离心,取滤液,减压蒸发浓缩,得醋红景天提取物;
步骤二:将醋红景天提取物、麦芽糊精以及矫味剂,混匀后,喷雾干燥,得浸膏粉;
步骤三:将步骤二中的浸膏粉经干法造粒后,过筛整粒,得到固体植物饮品。
优选地,所述步骤一中,高压的压力为200-500Mpa,保压时间为5-15min。
优选地,所述步骤一中,提取溶剂为含有低共熔溶剂的乙醇溶液。
优选地,所述低共熔溶剂是由氢键受体与氢键给体组合而成的;其中氢键受体为四乙基氯化铵;氢键给体为木糖醇或葡萄糖。
优选地,所述矫味剂为山梨醇和/或甘露醇。
本发明的有益效果是:
(1)本发明的一种醋红景天的炮制工艺,由于醋主要成分是醋酸,还有维生素、灰分、琥珀酸、草酸山梨醇等,其味酸苦、性温,具有引药入肝、理气、止血、行水、消肿、解毒。红景天中的具有止痛和镇静作用的生物碱,以游离的形式存在于药材中,难溶于水,因此,醋炙后,在煎煮时易与醋酸结合生成易溶于水的醋酸盐,即易于溶出使生物碱含量增加,因此,止痛作用明显增加。
(2)本发明的一种醋红景天在制备植物饮品上的应用,通过先对醋红景天进行超声浸泡,然后在高压下进行提纯,一方面,超声浸泡能够尽快使得原料组织软化,高压只需要在常温下进行,避免了高温使得醋红景天中的活性物质失活以及挥发油的挥发,另一方面,高压能够使得提纯溶剂迅速进入醋红景天固体颗粒内部,使其有效成分及时溶解,同时在高压作用下,细胞的细胞壁、细胞膜以及细胞内液等结构发生变化,能够使得细胞内溶物和提取溶剂进行充分接触,从而达到快速高效提取的作用,高压能够对非共价键(氢键、离子键、疏水键)产生影响,而对共价键影响较小,故生物大分子成分可发生变性,而对苷类等小分子物质的结构没有影响,因此,高压可以使蛋白质变性、酶失活、微生物死亡等,而不破坏药物中小分子的有效成分的生理活性,可以达到提取的目的。
(3)本发明的一种醋红景天在制备固体植物饮品上的应用,由于红景天苷具有很强的耐氧性能,将其制备成固体植物饮品,有效预防和改善高原地区旅游人群的高原反应。
(4)本发明的一种醋红景天在制备固体植物饮品上的应用,采用含有低共熔溶剂的提取溶剂,由于低共熔溶剂可以沉淀部分鞣质、脂肪、蛋白质和多糖类胶质等成分,减少大分子物质进入提取液,因此,能够提高红景天苷的提取率。
具体实施方式
以下以具体实施例对本发明的技术方案进行说明。
实施例1
取1kg净红景天片进行炒制,在80℃下进行炒制,炒制的同时喷淋0.1g醋液,醋液的乙酸浓度为20g/L,炒干后冷却过筛,制得醋红景天。
取醋红景天进行烘干,然后粉碎至50目,得粗粉,往粗粉中加入提取溶剂(含有占总比4wt%的低共熔溶剂的乙醇溶液,低共熔溶剂为质量比1:1的四乙基氯化铵和木糖醇)浸泡,浸泡的同时进行超声处理,处理完成后放入高压容器内,在255Mpa的高压下保压8min,泄压至常压,在3000rpm的转速下进行高速离心,取滤液,减压蒸发浓缩,得醋红景天提取物;将醋红景天提取物、麦芽糊精以及甘露醇按照质量比30:20:7混合,混匀后,喷雾干燥,得到浸膏粉,浸膏粉经干法造粒后,过筛整粒,制得固体植物饮品。
实施例2
取1kg净红景天片进行炒制,在90℃下进行炒制,炒制的同时喷淋0.2g醋液,醋液的乙酸浓度为20g/L,炒干后冷却过筛,制得醋红景天。
取醋红景天进行烘干,然后粉碎至50目,得粗粉,往粗粉中加入提取溶剂(含有占总比3wt%的低共熔溶剂的乙醇溶液,低共熔溶剂为质量比1:1的四乙基氯化铵和木糖醇)浸泡,浸泡的同时进行超声处理,处理完成后放入高压容器内,在275Mpa的高压下保压7min,泄压至常压,在3500rpm的转速下进行高速离心,取滤液,减压蒸发浓缩,得醋红景天提取物;将醋红景天提取物、麦芽糊精以及甘露醇按照质量比30:15:4混合,混匀后,喷雾干燥,得到浸膏粉,浸膏粉经干法造粒后,过筛整粒,制得固体植物饮品。
实施例3
取1kg净红景天片进行炒制,在100℃下进行炒制,炒制的同时喷淋0.1g醋液,醋液的乙酸浓度为30g/L,炒干后冷却过筛,制得醋红景天。
取醋红景天进行烘干,然后粉碎至50目,得粗粉,往粗粉中加入提取溶剂(含有占总比4wt%的低共熔溶剂的乙醇溶液,低共熔溶剂为质量比1:1的四乙基氯化铵和木糖醇)浸泡,浸泡的同时进行超声处理,处理完成后放入高压容器内,在285Mpa的高压下保压5min,泄压至常压,在3200rpm的转速下进行高速离心,取滤液,减压蒸发浓缩,得醋红景天提取物;将醋红景天提取物、麦芽糊精以及甘露醇按照质量比30:17:6混合,混匀后,喷雾干燥,得到浸膏粉,浸膏粉经干法造粒后,过筛整粒,制得固体植物饮品。
实施例4
取1kg净红景天片进行炒制,在80℃下进行炒制,炒制的同时喷淋0.15g醋液,醋液的乙酸浓度为30g/L,炒干后冷却过筛,制得醋红景天。
取醋红景天进行烘干,然后粉碎至50目,得粗粉,往粗粉中加入提取溶剂(含有占总比4wt%的低共熔溶剂的乙醇溶液,低共熔溶剂为质量比1:1的四乙基氯化铵和葡萄糖)浸泡,浸泡的同时进行超声处理,处理完成后放入高压容器内,在355Mpa的高压下保压4min,泄压至常压,在3100rpm的转速下进行高速离心,取滤液,减压蒸发浓缩,得醋红景天提取物;将醋红景天提取物、麦芽糊精以及甘露醇按照质量比30:19:6混合,混匀后,喷雾干燥,得到浸膏粉,浸膏粉经干法造粒后,过筛整粒,制得固体植物饮品。
实施例5
取1kg净红景天片进行炒制,在80℃下进行炒制,炒制的同时喷淋0.1g醋液,醋液的乙酸浓度为25g/L,炒干后冷却过筛,制得醋红景天。
取醋红景天进行烘干,然后粉碎至50目,得粗粉,往粗粉中加入提取溶剂(含有占总比3wt%的低共熔溶剂的乙醇溶液,低共熔溶剂为质量比1:2的四乙基氯化铵和葡萄糖)浸泡,浸泡的同时进行超声处理,处理完成后放入高压容器内,在455Mpa的高压下保压2min,泄压至常压,在3600rpm的转速下进行高速离心,取滤液,减压蒸发浓缩,得醋红景天提取物;将醋红景天提取物、麦芽糊精以及甘露醇按照质量比30:19:4混合,混匀后,喷雾干燥,得到浸膏粉,浸膏粉经干法造粒后,过筛整粒,制得固体植物饮品。
对比例1(无喷淋醋液)
取1kg净红景天片进行炒制,在80℃下进行炒制,炒干后冷却过筛,制得红景天。
取红景天进行烘干,然后粉碎至50目,得粗粉,往粗粉中加入提取溶剂(含有占总比4wt%的低共熔溶剂的乙醇溶液,低共熔溶剂为质量比1:1的四乙基氯化铵和葡萄糖)浸泡,浸泡的同时进行超声处理,处理完成后放入高压容器内,在255Mpa的高压下保压5min,泄压至常压,在3000rpm的转速下进行高速离心,取滤液,减压蒸发浓缩,得醋红景天提取物;将醋红景天提取物、麦芽糊精以及甘露醇按照质量比30:20:7混合,混匀后,喷雾干燥,得到浸膏粉,浸膏粉经干法造粒后,过筛整粒,制得固体植物饮品。
对比例2(无低共熔溶剂)
取1kg净红景天片进行炒制,在80℃下进行炒制,炒制的同时喷淋0.1g醋液,醋液的乙酸浓度为3g/L,炒干后冷却过筛,制得醋红景天。
取醋红景天进行烘干,然后粉碎至50目,得粗粉,往粗粉中加入提取溶剂(含有占总比4wt%的低共熔溶剂的乙醇溶液,低共熔溶剂为质量比1:1的四乙基氯化铵和葡萄糖)浸泡,浸泡的同时进行超声处理,在3000rpm的转速下进行高速离心,取滤液,减压蒸发浓缩,得醋红景天提取物;将醋红景天提取物、麦芽糊精以及甘露醇按照质量比30:20:7混合,混匀后,喷雾干燥,得到浸膏粉,浸膏粉经干法造粒后,过筛整粒,制得固体植物饮品。
对比例3(无高压)
取1kg净红景天片进行炒制,在80℃下进行炒制,炒制的同时喷淋0.1g醋液,醋液的乙酸浓度为3g/L,炒干后冷却过筛,制得醋红景天。
取醋红景天进行烘干,然后粉碎至50目,得粗粉,往粗粉中加入提取溶剂(乙醇溶液)浸泡,浸泡的同时进行超声处理,处理完成后放入高压容器内,在255Mpa的高压下保压5min,泄压至常压,在3000rpm的转速下进行高速离心,取滤液,减压蒸发浓缩,得醋红景天提取物;将醋红景天提取物、麦芽糊精以及甘露醇按照质量比30:20:7混合,混匀后,喷雾干燥,得到浸膏粉,浸膏粉经干法造粒后,过筛整粒,制得固体植物饮品。
对上述实施例和对比例的粗红景天提取物中进行红景天苷进行含量测定,具体采用高效液相色谱仪测定,计算其提取率。
测试值见表1。

从表1中可以看出,实施例中的提取物红景天苷高于对比例,主要原因可能如下,通过对比例1的分析可知,醋液中能够将药材中以游离形式的物质结合生成易溶于水的醋酸盐形式,有利于活性物质的溶出;通过对对比例2的分析可知,采用含有低共熔溶剂的提纯溶剂,可以沉淀部分鞣质、脂肪、蛋白质和多糖类胶质等大分子成分,由于红景天苷属于小分子物质,因此,含有低共熔溶剂能够将大分子物质沉淀,减少了大分子物质进入提取物中,减少对红景天苷的成分干扰,从而提高了红景天苷的提取率,同时也能提高固体饮料的适度感;通过对对比例3的分析可知,通过先对醋红景天进行超声浸泡,然后在高压下进行提纯,一方面,超声浸泡能够尽快使得原料组织软化,高压只需要在常温下进行,避免了高温使得醋红景天中的活性物质失活以及挥发油的挥发,另一方面,高压能够使得提纯溶剂迅速进入醋红景天固体颗粒内部,使其有效成分及时溶解,同时在高压作用下,细胞的细胞壁、细胞膜以及细胞内液等结构发生变化,能够使得细胞内溶物和提取溶剂进行充分接触,从而达到快速高效提取的作用,高压能够对非共价键(氢键、离子键、疏水键)产生影响,而对共价键影响较小,故生物大分子成分可发生变性,而对小分子物质(皂苷、黄酮、生物碱、挥发油)的结构没有影响,因此,高压可以使蛋白质变性、酶失活、微生物死亡等,而不破坏药物中小分子的有效成分的生理活性,可以达到提取的目的。
同时对实施例2和对比例的固体植物饮品进行耐氧实验,具体:取体重20.0±2g的小鼠40只,雌性,随机分成4组,经口给予小鼠固体植物饮品(5g固体植物饮品溶于100ml温水中)一个月,进行常压耐缺氧实验,对所测定的常压耐缺氧实验中存活时间进行方差齐性检验,满足方差齐性要求,用单因素方差分析方法中多个实验组与一个对照组间均数的两两比较方法进行统计处理。测试见表2。空白对照组为未饮用固体植物饮品的实验组。

由表2可见,小鼠常压耐缺氧实验中,实施例2的存活时间最长,差异有统计学意义(P<0.05),一方面主要是由于实施例中红景天苷的含量高于对比例,另一方面是由于,醋液具有引药入肝的功效,即能够有利于药效的发挥,结合红景天苷具有优异的耐氧能力,因此能够有效提高红景天苷在体内的药效发挥能力,对预防和改善高原地区旅游人群的高原反应有很好的效果。
同时对实施例2和对比例1固体植物饮品进行大鼠尾尖部压痛实验观察,具体取体重180±2g的大鼠40只,雌雄各20只,随机分成两组,经对大鼠尾尖进行压制,然后口服固体植物饮品(5g固体植物饮品溶于100ml温水中),5min之内观察大鼠尾部扭动状态。见表3。

结果显示实施例2大鼠(20只)均扭动较少,对比例1的大鼠(20只)均扭动频繁,可能的原因是由于实施例2中红景天中具有止痛和镇静作用的生物碱,以游离的形式存在于药材中,难溶于水,因此,醋炙后,在煎煮时易与醋酸结合生成易溶于水的醋酸盐,即易于溶出使生物碱含量增加,因此,止痛作用明显增加。

Claims (10)
1.一种醋红景天炮制工艺,其特征在于,取净红景天片进行炒制,炒制的同时喷淋醋液,炒干后冷却过筛,制得醋红景天。
2.根据权利要求1所述的一种醋红景天炮制工艺,其特征在于,所述醋液的乙酸浓度为20-30g/L。
3.根据权利要求1所述的一种醋红景天炮制工艺,其特征在于,每100kg红景天片,喷淋10-20kg醋液。
4.根据权利要求1所述的一种醋红景天炮制工艺,其特征在于,所述炒制的温度为80-120℃。
5.一种醋红景天,其特征在于,采用如权利要求1-4任一项所述的炮制工艺制得。
6.一种如权利要求5所述的醋红景天在制备固体植物饮品上的应用,其特征在于,包含以下步骤:
步骤一:先将醋红景天烘干粉碎,得粗粉,往粗粉中加入提取溶剂进行浸泡,浸泡的同时进行超声处理,处理完成后放入高压容器内,在高压下进行保压,保压后恢复常压,进行高速离心,取滤液,减压蒸发浓缩,得醋红景天提取物;
步骤二:将醋红景天提取物、麦芽糊精以及矫味剂,混匀后,喷雾干燥,得浸膏粉;
步骤三:将步骤二中的浸膏粉经干法造粒后,过筛整粒,得到固体植物饮品。
7.根据权利要求6所述的一种醋红景天在制备固体植物饮品上的应用,其特征在于,所述步骤一中,高压的压力为200-500Mpa,保压时间为5-15min。
8.根据权利要求6所述的一种醋红景天在制备固体植物饮品上的应用,其特征在于,所述步骤一中,提取溶剂为含有低共熔溶剂的乙醇溶液。
9.根据权利要求8所述的一种醋红景天在制备固体植物饮品上的应用,其特征在于,所述低共熔溶剂是由氢键受体与氢键给体组合而成的。
10.根据权利要求6所述的一种醋红景天在制备固体植物饮品上的应用,其特征在于,所述矫味剂为山梨醇和/或甘露醇。
