Abstract
The invention relates to a fresh dendrobe antioxidant composition, an antioxidant preparation method and application, wherein the fresh dendrobe antioxidant composition comprises 20-40 parts of fresh dendrobe, 10-20 parts of kudzu root, 10-20 parts of coix seed, 2-5 parts of saffron crocus, 5-10 parts of ginseng and 5-10 parts of liquorice; taking fresh dendrobium officinale, smashing, mixing with other components of the fresh dendrobium antioxidant composition, decocting in water, extracting, taking filtrate, and concentrating to obtain antioxidant which can be used for preparing cosmetics. The invention has the beneficial effects that: the antioxidant extracted from the fresh dendrobium composition has high capability of removing OH free radicals.
Description
Technical Field
The invention belongs to the field of biological preparations, and particularly relates to a fresh dendrobe antioxidant composition, an antioxidant preparation method and application.
Background
As people age, various organs and cells of the body gradually age, the skin serving as a first barrier of the human body is the first thing, and particularly, the skin care of beauty-conscious women is important. Research proves that the antioxidant system of a human body is a system which is comparable to the immune system and has perfect and complex functions, the stronger the antioxidant capacity of the body is, the healthier the body is, and when the antioxidant system of the body has problems, a formula capable of resisting oxidation is required to solve the problems.
The Chinese herbal medicine as the treasure of China is from nature, and the above problems can be solved by taking the theory of traditional Chinese medicine as guidance. Dendrobium officinale (Dendrobium huoshanense) is a famous and precious Chinese traditional medicine, and has the effects of nourishing yin, clearing heat, benefiting stomach, promoting fluid production, moistening lung, relieving cough and the like according to records of compendium of materia medica. Can greatly improve the level of SOD (main substance for delaying senility) in human bodies, and is very suitable for people who often stay up all night, excessively use brain, smoke and wine, and have weak and hypodynamia. The dendrobium officinale has the effects of improving eyesight, harmonizing yin and yang, tonifying yang and kidney, and beautifying and maintaining beauty, so that the effects of health care and life prolonging are achieved. A method for preparing antioxidant comprises collecting fresh herba Dendrobii, crushing, mixing with other components of fresh herba Dendrobii antioxidant composition, decocting in water, collecting filtrate, and concentrating to obtain antioxidant.
Preferably, the water extraction is carried out twice, the ratio of the first water extraction to the liquid is 1:20-25, the extraction time is 2-3h, the ratio of the second water extraction to the liquid is 1:10-15, and the extraction time is 0.5-1 h.
Preferably, the filtrate is concentrated to 1kg crude drug/L concentrated solution, and is stored in lyophilized powder form after freeze drying.
Antioxidant preparation method can be used for preparing antioxidant for cosmetic.
The invention has the advantages and positive effects that: the antioxidant extracted from the fresh dendrobium composition has high capability of removing OH free radicals, has good antioxidation effect when being used in cosmetics, does not stimulate the skin, and has warm-keeping function.

Detailed Description
The invention relates to a fresh dendrobium antioxidant composition, and a preparation method and application of an antioxidant. The fresh dendrobium antioxidant composition comprises 20-40 parts of fresh dendrobium, 10-20 parts of kudzu root, 10-20 parts of coix seed, 2-5 parts of saffron, 5-10 parts of ginseng and 5-10 parts of liquorice.
Dendrobe: is a rare Chinese medicinal material in China, and has the effects of nourishing yin, clearing heat, benefiting stomach, promoting fluid production, moistening lung, relieving cough and the like according to records of compendium of materia medica. The fresh dendrobium officinale strips can be directly used, so that the original effective components can be kept to the maximum extent; the dried product needs to be processed to a certain extent, which can affect the integrity of the original effective components of the dendrobium officinale more or less.
Kudzu root: the kudzu root contains starch as main component, flavone compound in 12%, amino acids and trace elements essential for human body. As for the health care value of breast enlargement and beauty, the isoflavone in the kudzuvine root is a phytoestrogen which is most similar to the estrogen secreted by the human body on the molecular structure at present, so the isoflavone can be combined with estrogen receptors on the surfaces of various systems and organ tissue cells of the body to play the effect. Ginseng: contains more than 100 effective components, mainly including ginsenoside, small amount of volatile oil, various amino acids and ginseng polypeptide, glucose, fructose, pectin, vitamins B1, B2, nicotinic acid, pantothenic acid, etc. Wherein ginsenoside is the most active substance, and has antiaging and antiinfectious effects. The ginseng exudate has no any adverse stimulation to the skin, can effectively resist cholesterol, help metabolism, promote skin blood circulation, enhance skin nutrition, and prevent skin dehydration, sclerosis and wrinkling, thereby keeping moisture in skin stratum corneum and enabling the skin to be tender, smooth and elastic. Coix seed, semen coicis, sweet and bland in taste, cool in nature, promote diuresis and eliminate dampness, invigorate spleen and check diarrhea, remove arthralgia, expel pus, detoxify and dissipate stagnation, and is an adjuvant drug in the formula for eliminating dampness and expelling toxin. Saffron, sweet in flavor, neutral in nature, has the effects of promoting blood circulation to remove blood stasis, cooling blood and removing toxicity, and relieving depression and tranquilizing mind. Is an adjuvant drug in the formula and regulates qi and blood of the organism. The liquorice is sweet and neutral, and has the effects of tonifying spleen and qi, clearing away heat and toxic materials and harmonizing the drugs in the recipe. The obtained antioxidant has good free radical scavenging ability, and can be used for preparing cosmetics.
The present invention will be described with reference to specific examples, wherein the experimental methods without specific descriptions of the operation steps are performed according to the corresponding commercial specifications, and the instruments, reagents, and consumables used in the examples can be purchased from commercial companies without specific descriptions.
Example 1: preparation of antioxidants
The formula is as follows: 20-40 parts of fresh dendrobium, 10-20 parts of kudzu root, 10-20 parts of coix seed, 2-5 parts of saffron, 5-10 parts of ginseng and 5-10 parts of liquorice.

Comparative example 1:

Comparative example 2:
selecting dry dendrobium officinale in the same producing area, and preparing the dry dendrobium officinale extract according to the preparation method in the comparative example 1.
Example 2: DPPH free radical scavenging experiment
0.1g of raw material freeze-dried powder is precisely weighed for example 1, comparative example 1 and comparative example 2 respectively, and dissolved by adding methanol in a dark environment to prepare a DPPH methanol solution with the concentration of 0.1 mmol/L. Firstly, precisely measuring 1mL of methanol and 1mL of double distilled water, fully and uniformly mixing, placing a mixed solution in a cuvette, taking the mixed solution as a blank solution at a wavelength of 517nm on an ultraviolet spectrophotometer, and carrying out zero setting; precisely measuring 1.0mL of 0.1mmol/LDPPH methanol solution, precisely measuring 1.0mL of sample solutions with different concentrations, adding the sample solutions, immediately and fully mixing, reacting for 30min in a dark place At room temperature, measuring and recording the absorbance value under the same condition, and marking as At; meanwhile, an Ac tube is arranged, 1.0mL of 0.1mmol/LDPPH methanol solution and 1.0mL of double distilled water are precisely measured and mixed, the mixture is fully and uniformly mixed, the positive control medicament L-ascorbic acid is treated by the same method, and the absorbance value is measured and recorded as Ac. As can be seen from fig. 1 and the data in table 1, the composition has a stronger DPPH radical scavenging ability than the dry dendrobium extract and the fresh dendrobium extract alone, and the scavenging effect is gradually enhanced as the sample concentration is increased within a certain range. The clearance rate of the composition sample is superior to that of the dendrobium used alone, and the clearance rate range is 47.76-68.89%; the experimental result shows that the antioxidant extract of the composition has certain advantages in the aspect of the capability of removing DPPH free radicals.
Example 3: OH free radical scavenging experiments
OH free radical is a free radical with high activity and strong toxicity generated by organisms and has great damage effect on biomacromolecules, cells and the like, and the removal capability of the OH free radical is one of the common important indexes for evaluating and measuring the strength of the antioxidant capability of antioxidant substances.
Precisely measuring 1.0mL7.5mmol/L FeSO4, 1.0mL7.5mmol/L phenanthroline, 2.0mLPH7.4 phosphate buffer solution and 1.0mL8.8mmol/L H2O2 for example 1, comparative example 1 and comparative example 2 respectively, immediately mixing, fully mixing, adding 1.0mL of each sample solution with different concentrations and 4.0mL of double distilled water, mixing uniformly, and reacting at 37 ℃ for 30 min; a control sample of L-ascorbic acid was prepared in the same manner, and the absorbance was measured and recorded using an ultraviolet spectrophotometer at 536 nm.

As shown in table 2 and fig. 2, the removing ability of each sample is gradually enhanced with the increase of the concentration, and the removing ability of the composition is better than that of the extracts of the dry dendrobium and the fresh dendrobium. Within the same concentration range, the removal rate of OH radicals from the composition samples ranged from 16.45 to 38.56%. Thus, the results of this test show that the compositions have certain advantages in their ability to scavenge the OH radicals.
As shown in the data of example 2 and example 3, the antioxidant capacity of the composition is still lower than that of L-ascorbic acid with antioxidant function, but the composition is a traditional Chinese medicine extract, the preparation process is simpler and is not chemically synthesized; when the extract is used in cosmetics, allergy is not easy to occur, and the traditional Chinese medicine extract is more compatible especially for users with sensitive skin.

Claims (5)
1. A fresh dendrobium antioxidant composition is characterized in that: comprises 20-40 parts of fresh dendrobium, 10-20 parts of kudzu root, 10-20 parts of coix seed, 2-5 parts of saffron, 5-10 parts of ginseng and 5-10 parts of liquorice.

A preparation method of an antioxidant is characterized by comprising the following steps: taking fresh dendrobium officinale, smashing, mixing with other components of the fresh dendrobium antioxidant composition in claim 1, decocting and extracting with water, taking filtrate, and concentrating to obtain antioxidant.

4. The method for producing an antioxidant according to claim 2 or 3, wherein: concentrating the filtrate to 1kg crude drug/L concentrated solution, freeze drying, and storing in lyophilized powder form.
5. Use of the antioxidant prepared by the process for the preparation of an antioxidant according to any one of claims 2 to 4 in cosmetics.
