Abstract

本发明公开了一株蜡样芽胞杆菌噬菌体DLn1及其应用。噬菌体DLn1保藏编号为:GDMCC No:61638‑B1。本发明提供的烈性噬菌体DLn1能特异性裂解蜡样芽胞杆菌,且噬菌体DLn1裂解量较大。裂解量大的噬菌体能在较短时间增殖,更好的用于抵抗蜡样芽胞杆菌的污染。同时,噬菌体DLn1对高温和酸碱具有耐受性,有较强的环境适应能力,不易在应用过程中失活,在牛奶中对目标菌有良好抑制效果。综上所述,噬菌体DLn1有望成为一种抑菌制剂用于防控食品中蜡样芽胞杆菌的污染。

Description

一株蜡样芽胞杆菌噬菌体DLn1及其应用
技术领域:
本发明属于生物技术领域,具体涉及一株蜡样芽胞杆菌噬菌体DLn1及其应用。
背景技术:
细菌是导致食源性疾病发生的主要病因,严重危害人们的健康,且抗生素的过度使用进一步加剧了这个情况。我国的主要食源性致病菌包括副溶血弧菌、沙门氏菌、金黄色葡萄球菌、蜡样芽胞杆菌、致泻大肠埃希菌等。其中,蜡样芽胞杆菌越来越受到人们的关注。据统计,从2003到2019年在我国由蜡样芽胞杆菌引起的食源性暴发事件有504起,死亡人数为6人,在食源性致病菌中排名第四。当人体摄入被蜡样芽胞杆菌污染的食物后,可引起呕吐或腹泻症状,严重时会导致死亡。芽胞的产生是蜡样芽胞杆菌容易污染食品的一个重要原因。具有耐受性的芽胞能够抵抗环境中不利因素的影响,并且在适宜的条件下重新转变成营养细胞,进一步生长繁殖。已有研究表明,蜡样芽胞杆菌对β-内酰胺、红霉素、四环素和碳青霉烯等抗生素具有耐药性。因此,利用抗生素防控蜡样芽胞杆菌不容乐观,开发新的防控手段势在必行。
噬菌体是一类侵染细菌的病毒,会干扰宿主菌正常的代谢过程,甚至导致宿主菌裂解死亡。因此,噬菌体及其衍生物在病原体生物控制中具有很大的潜力。在二十世纪,由于抗生素耐药问题日趋严重,噬菌体疗法重新进入人们的视野,被用于临床治疗并取得了一定的效果。多种噬菌体制剂也已被批准应用在食品生产中,如美国食品药品监督管理局在2006年批准List ShieldTM制剂作为食品添加剂,用于防控肉制品、水产品、乳制品中李斯特杆菌的污染。EcoShieldTM制剂及SalmoFreshTM制剂也被批准用于防控大肠杆菌和沙门氏菌的污染。为了增强治疗效果、扩大噬菌体制剂的裂解范围并预防耐受菌的出现,通常商品化的噬菌体制剂不是单一的噬菌体,而是由多种噬菌体组合而成。因此对新型噬菌体的分离和表征尤为重要。
虽然很多噬菌体制剂已被用于防控微生物的污染,但是目前用于防控蜡样芽胞杆菌污染的噬菌体仍十分有限,因此亟需分离出能够感染蜡样芽胞杆菌并可应用于食品加工和生产的噬菌体及其组合制剂。
发明内容:
本发明的第一个目的在于提供一株蜡样芽胞杆菌的新型烈性噬菌体DLn1,可将其应用于食品中抑制蜡样芽胞杆菌的生长。
为了实现上述目的,本发明采用以下技术方案:
本发明提供的烈性噬菌体DLn1(Bacillus cereus phage DLn1)是以蜡样芽胞杆菌为宿主分离纯化所得。所述噬菌体DLn1于2021年5月12日保藏于广东省微生物菌种保藏中心(GDMCC),地址:广东省广州市越秀区先烈中路100号大院59号楼5楼,邮编:510070,保藏编号为:GDMCC No:61638-B1。
本发明的噬菌体DLn1属于有尾噬菌体目Salasmaviridae科,通过全基因组测序与比对,表明该噬菌体与已有噬菌体相似性低,属于一株新型噬菌体。
本发明的噬菌体DLn1具有SEQ ID No.1所示的核苷酸序列。
本发明的噬菌体DLn1潜伏期短、裂解量大。潜伏期为15min,裂解量为634PFU/cell。
本发明的噬菌体DLn1具有良好的热稳定性和pH稳定性,在温度为4-55℃及pH为4-10之间可保持稳定效价。
本发明的噬菌体DLn1有相对较宽的宿主范围,可以裂解宿主克隆复合体ST-205的多个蜡样芽胞杆菌菌株。
本发明的噬菌体DLn1在牛奶中对蜡样芽胞杆菌具有良好抑菌效果。
因此,本发明的第二个目的是提供上述噬菌体DLn1在制备抑制蜡样芽胞杆菌制剂中的应用。
本发明的第三个目的是提供一种抑制蜡样芽胞杆菌的噬菌体制剂,其含有噬菌体DLn1作为活性成分。
优选,噬菌体DLn1在制备抑制食品中蜡样芽胞杆菌制剂中的应用。
进一步优选是噬菌体DLn1在制备抑制牛奶中蜡样芽胞杆菌制剂中的应用。
本发明的有益效果:

具体实施方式
为了具体阐明本发明所采取的手段及实施效果,以下结合实施例及附图对本发明作进一步说明。
实施例1、噬菌体DLn1的分离、纯化与增殖
1、样品的来源及处理
用于分离噬菌体的样本来自于广东省广州市污水,采集水样后进行10000×g离心20min使大部分杂质沉于底部,取上清液用0.45μm的滤膜过滤去除大部分环境细菌。加入50mM硫酸镁至上清液,静置30min后用0.22μm滤膜过滤,使噬菌体吸附至滤膜。随后用洗脱液(1%牛肉膏、3%Tween 80)浸泡滤膜并超声5min后,将洗脱液用0.22μm滤膜过滤除去细菌,滤液保存以备用。
2、噬菌体的分离、纯化
在3mLTSB培养基(含1mM CaCl2)中添加30μL培养至对数期的蜡样芽胞杆菌2177和100μL滤液,在温度为37℃、转速为200rpm的摇床过夜培养。随后用0.22μm滤膜过滤,将未纯化的噬菌体滤液于4℃保存。
采用双层琼脂平板法分离纯化噬菌体。将培养至对数期的蜡样芽胞杆菌2177与100μL噬菌体滤液加至TSB上层琼脂(含0.4%琼脂、1mM CaCl2)中,混匀后倒入TSB固体平板(含1.5%琼脂、1mM CaCl2)。待上层琼脂凝固后,于37℃倒置培养4h,当出现噬菌斑后,用接种针挑取单一噬菌斑并悬浮于1mLTSB培养基中。梯度稀释后取100μL与对数期的宿主菌蜡样芽胞杆菌2177混匀,再次采用双层平板法获得独立分散的噬菌斑。重复以上操作4-5次,直至出现大小形态均匀的噬菌斑(图1),即获得纯化的噬菌体DLn1。将纯化的噬菌体DLn1于4℃保存。
3、噬菌体DLn1的增殖
采用多次富集的方法可增殖噬菌体DLn1。取500μL过夜蜡样芽胞杆菌2177菌株接于含有1mM CaCl2的50mLTSB培养基中,培养1h后,加入1mL噬菌体DLn1,继续培养7h后,10000×g离心10min,离心之后过滤,重复以上操作3-5次,以提高噬菌体的效价,获得滴度为3×1010PFU/mL的噬菌体DLn1。
实施例2、噬菌体DLn1的形态观察
取10μL高滴度的噬菌体DLn1置于镀碳网的铜膜上,自然静置15min后,用干净的滤纸吸取多余的液体。晾干后用2%的磷酸钨染色10min,待自然干燥后,将制备好的染色片在透射电子显微镜下观察(图2)。噬菌体头长和宽分别为60.1±3.6nm,36.5±3.6nm,噬菌体尾长和宽分别为36.4±3.7nm和4.7±1.0nm。
实施例3、噬菌体DLn1基因组分析
提取噬菌体DLn1的基因组DNA进行分析。将上述所获得的噬菌体DLn1用聚乙二醇及氯化钠浓缩,获得更高滴度的噬菌体。随后采用苯酚氯仿法提取噬菌体DNA,通过Illumina Miseq对DNA进行测序并分析。经分析可知,噬菌体DLn1具有如SEQ ID NO:1的核苷酸序列。通过NCBI BLASTn确定噬菌体DLn1与已报道噬菌体的相似性。
通过测序及分析可知,噬菌体DLn1的基因组大小为25379bp,且为线性DNA。噬菌体DLn1与NCBI比对结果如表1所示,其中核酸一致性是Query Cover与Per.ident的乘积。通过表1可知,该噬菌体与已知的噬菌体相似性小于95%,表明该噬菌体属于新型噬菌体。结合电子显微镜观察及最新国际病毒分类学可知,噬菌体DLn1属于有尾噬菌体目Salasmaviridae科。

实施例4、噬菌体DLn1的保藏
上述分离的蜡样芽胞杆菌烈性噬菌体DLn1(Bacillus cereus phage DLn1)已送至广东省微生物菌种保藏中心(GDMCC)保藏,地址:广东省广州市越秀区先烈中路100号大院59号楼5楼,邮编:510070,保藏编号为:GDMCC No:61638-B1,保藏日期为:2021.05.12。
实施例5、噬菌体DLn1一步生长曲线的测定
预先将TSB(1mM CaCl2)培养基及噬菌体在37℃保温。取1mL生长于对数期的宿主菌蜡样芽胞杆菌2177,13000×g离心1min后重悬于1mLTSB(1mM CaCl2)培养基中。将噬菌体与菌液以感染复数(MOI)为0.1混合,吸附5min后13000×g离心1min,将上清过滤到新的离心管中以测定未吸附的噬菌体量。同时,将沉淀重悬于1mLTSB(1mM CaCl2)培养基中,取50μL到50mL的TSB培养基(1mM CaCl2)中,于37℃摇床震荡培养,每隔5min取样测定噬菌体效价,重复三次实验。以取样时间为横坐标,噬菌体效价与感染中心之比为纵坐标,绘制噬菌体DLn1一步生长曲线。通过一步生长曲线(图3)可知,噬菌体DLn1的潜伏期为15min,裂解量为634PFU/cell。
实施例6、噬菌体DLn1温度稳定性的测定
取1mL效价为108PFU/mL的噬菌体DLn1分别置于4℃、30℃、37℃、55℃、65℃、72℃、85℃中孵育1h,随后稀释适当倍数,采用双层琼脂法测定不同温度下噬菌体的效价。通过实验可知,噬菌体DLn1在温度为4-55℃稳定,65℃时活性下降,高于65℃后活性丧失(图4)。
实施例7、噬菌体DLn1酸碱稳定性的测定
用1M HCL和1M NaOH将TSB培养基调至不同pH(2~12)。取100μL噬菌体DLn1置于不同pH培养基中,使噬菌体DLn1最终浓度为108PFU/mL。30℃孵育1h后,稀释至适当倍数,采用双层琼脂法测定不同pH下噬菌体的效价。通过实验可知,噬菌体在pH为4-11之间稳定,在pH为3时活性下降,在pH为2和12时活性丧失(图5)。
实施例8、噬菌体DLn1宿主谱的测定
测定噬菌体DLn1潜在的宿主范围,将86株蜡样芽胞杆菌培养至对数期,随后采用双层琼脂点样法测定噬菌体DLn1对以上菌株的裂解能力。结果如表2所示。

一共测定了86株蜡样芽胞杆菌,其中除感染原始使用目标菌蜡样芽胞杆菌2177外,噬菌体DLn1还能感染其他17株蜡样芽胞杆菌,且这17株与宿主菌2177都属于ST-205克隆复合体。
实施例9、噬菌体DLn1在牛奶中抑菌能力的测定
挑取新鲜的蜡样芽胞杆菌2177单菌落悬浮于500μL磷酸盐缓冲溶液使OD600=1.2,取100μL宿主菌加入9.8mL牛奶中,菌液最终浓度为104CFU/mL,最后与100μL噬菌体混合,至MOI分别为100和1000。在25℃静置培养,分别于0h、3h、6h、12h和24h取样,稀释不同数量级铺板计数,测定在牛奶中噬菌体对蜡样芽胞杆菌2177生长的抑制效果。由图6可知,在培养3h后,与对照组相比,MOI为100和1000实验组中细菌数量显著降低,分别降低了0.88lg(CFU/mL)和2.21lg(CFU/mL),且在高MOI下,噬菌体对细菌抑制效果更明显。在6h,当MOI为100和1000时,实验组中细菌数量较对照组降低了4个数量级;同样,在12h时降低了3个数量级。培养至24h,实验组中细菌数量增至7.26lg(CFU/mL),与对照组无显著差异。从整体看,噬菌体DLn1在前12h抑菌效果明显,对防控食品中蜡样芽胞杆菌污染有较大的潜力。
