Abstract
多種の食中毒細菌に対して有効であって、効果的な新規の食中毒細菌のバクテリオファージ耐性化抑制剤及びその使用、食中毒細菌増殖抑制用組成物、並びに、食中毒細菌のバクテリオファージ耐性化抑制方法を提供する。食中毒細菌のバクテリオファージ耐性化抑制剤は、キレート剤又はナイシンを有効成分として含有する。食中毒細菌増殖抑制用組成物は、バクテリオファージと前記食中毒細菌のバクテリオファージ耐性化抑制剤とを含む。食中毒細菌のバクテリオファージ耐性化抑制剤の使用は、キレート剤又はナイシンを有効成分として含有する食中毒細菌のバクテリオファージ耐性化抑制剤の使用であって、食中毒細菌のバクテリオファージに対する耐性化を抑制するための使用である。食中毒細菌のバクテリオファージ耐性化抑制方法は、キレート剤又はナイシンを用いる方法である。
Description
本発明は、食中毒細菌のバクテリオファージ耐性化抑制剤及びその使用に関する。具体的には、本発明は、食中毒細菌のバクテリオファージ耐性化抑制剤、食中毒細菌のバクテリオファージ耐性化抑制剤の使用及び食中毒細菌のバクテリオファージ耐性化抑制方法に関する。
バクテリオファージは、細菌種に特異的に、細菌を溶菌するが、一部の細菌集団はバクテリオファージに対する耐性を有する。そのため、バクテリオファージ耐性菌への有効な対策が求められている。これを解決するために、複数のファージ株を混合したファージカクテルが食中毒細菌の制御のために用いられている。しかしながら、ファージカクテルを用いても、確実にバクテリオファージ耐性菌を制御できるわけではない。
これまで、確実に食中毒細菌を制御するために、ファージと抗菌物質との併用について検討されてきた。例えば、サルモネラ特異的溶菌ファージと、アルギン酸ラウリル(Lauric arginate:LAE)及び塩化セチルピリジニウム(Cetylpyridinium chloride:CPC)との併用によって鶏肉におけるサルモネラ菌を効果的に殺菌できたという報告がある(例えば、非特許文献1参照)。

しかしながら、サルモネラ菌に限定されない多種の食中毒細菌に対して有効であって、バクテリオファージに対する耐性化を抑制しながら、効果的に食中毒細菌の増殖を阻害できるバクテリオファージと抗菌物質との組み合わせについては未だ知られていなかった。
本発明は、上記事情に鑑みてなされたものであって、多種の食中毒細菌に対して有効であって、効果的な新規の食中毒細菌のバクテリオファージ耐性化抑制剤及びその使用、食中毒細菌増殖抑制用組成物、並びに、食中毒細菌のバクテリオファージ耐性化抑制方法を提供する。
すなわち、本発明は、以下の態様を含む。
本発明の第1態様に係る食中毒細菌のバクテリオファージ耐性化抑制剤は、キレート剤又はナイシンを有効成分として含有する。
上記第1態様に係る食中毒細菌のバクテリオファージ耐性化抑制剤において、前記キレート剤がエチレンジアミン四酢酸又はその塩であってもよい。
本発明の第1態様に係る食中毒細菌のバクテリオファージ耐性化抑制剤は、キレート剤又はナイシンを有効成分として含有する。
上記第1態様に係る食中毒細菌のバクテリオファージ耐性化抑制剤において、前記キレート剤がエチレンジアミン四酢酸又はその塩であってもよい。
本発明の第2態様に係る食中毒細菌増殖抑制用組成物は、バクテリオファージと、上記第1態様に係る食中毒細菌のバクテリオファージ耐性化抑制剤とを含む。
上記第2態様に係る食中毒細菌増殖抑制用組成物は、食品に対して用いられてもよい。
上記第2態様に係る食中毒細菌増殖抑制用組成物は、食品に対して用いられてもよい。
本発明の第3態様に係る食中毒細菌のバクテリオファージ耐性化抑制剤の使用は、キレート剤又はナイシンを有効成分として含有する食中毒細菌のバクテリオファージ耐性化抑制剤の使用であって、食中毒細菌のバクテリオファージに対する耐性化を抑制するための使用である。
本発明の第4態様に係る食中毒細菌のバクテリオファージ耐性化抑制方法は、キレート剤又はナイシンを用いる方法である。
上記態様の食中毒細菌のバクテリオファージ耐性化抑制剤及びその使用、並びに、食中毒細菌のバクテリオファージ耐性化抑制方法は、多種の食中毒細菌に有効であり、食中毒細菌のバクテリオファージに対する耐性化を効果的に抑制することができる。上記態様の食中毒細菌増殖抑制用組成物は、多種の食中毒細菌に有効であり、食中毒細菌の増殖を効果的に抑制することができる。
≪食中毒細菌のバクテリオファージ耐性化抑制剤≫
本実施形態の食中毒細菌のバクテリオファージ耐性化抑制剤は、キレート剤又はナイシンを有効成分として含有する。
本実施形態の食中毒細菌のバクテリオファージ耐性化抑制剤は、キレート剤又はナイシンを有効成分として含有する。
従来のバクテリオファージを用いた食中毒細菌の増殖抑制方法では、食中毒細菌のバクテリオファージに対する耐性化が問題となっていた。これに対し、本実施形態の食中毒細菌のバクテリオファージ耐性化抑制剤は、後述の実施例に示すように、バクテリオファージに、キレート剤又はナイシンを併用することで、キレート剤又はナイシンが食中毒細菌のバクテリオファージ耐性化を抑制し、バクテリオファージによる食中毒細菌の溶菌作用を高く維持することができる。そのため、食中毒細菌の増殖を効果的に抑制することができる。
よって、一実施形態において、本発明は、キレート剤又はナイシンを有効成分として含有する食中毒細菌のバクテリオファージ耐性化抑制剤の使用であって、食中毒細菌のバクテリオファージに対する耐性化を抑制するための、食中毒細菌のバクテリオファージ耐性化抑制剤の使用を提供する。
また、一実施形態において、本発明は、キレート剤又はナイシンを用いる食中毒細菌のバクテリオファージ耐性化抑制方法を提供する。
よって、一実施形態において、本発明は、キレート剤又はナイシンを有効成分として含有する食中毒細菌のバクテリオファージ耐性化抑制剤の使用であって、食中毒細菌のバクテリオファージに対する耐性化を抑制するための、食中毒細菌のバクテリオファージ耐性化抑制剤の使用を提供する。
また、一実施形態において、本発明は、キレート剤又はナイシンを用いる食中毒細菌のバクテリオファージ耐性化抑制方法を提供する。
また、本実施形態の食中毒細菌のバクテリオファージ耐性化抑制剤は、後述の実施例に示すように、グラム陽性菌及びグラム陰性菌の両方の食中毒細菌における、バクテリオファージに対する優れた耐性化抑制効果を有する。
なお、本明細書における「有効成分として含有する」とは、キレート剤又はナイシンをバクテリオファージに対する耐性化抑制効果が奏される程度に含有することを意味する。
食中毒を起こすグラム陽性菌としては、例えば、リステリア属菌(Listeria)、クロストリジウム属菌(Clostridium)、スタフィロコッカス属菌(Staphylococcus)、バチルス属菌(Bacillus)等が挙げられ、これらに限定されない。
リステリア属菌としては、例えば、リステリア・モノサイトゲネス(L. monocytogenes)等が挙げられる。
クロストリジウム属菌としては、例えば、ウェルシュ菌(クロストリジウム・パーフリンジェンス(C. perfringens))、ボツリヌス菌(クロストリジウム・ボツリナム(C. botulinum))等が挙げられる。
スタフィロコッカス属菌としては、例えば、スタフィロコッカス・オーレウス(S. aureus)、スタフィロコッカス・エピデルミス(S. epidermis)等が挙げられる。
バチルス属菌としては、例えば、バチルス・セレウス(B. cereus)、バチルス・ズブチリス(B. subtilis)等が挙げられる。
リステリア属菌としては、例えば、リステリア・モノサイトゲネス(L. monocytogenes)等が挙げられる。
クロストリジウム属菌としては、例えば、ウェルシュ菌(クロストリジウム・パーフリンジェンス(C. perfringens))、ボツリヌス菌(クロストリジウム・ボツリナム(C. botulinum))等が挙げられる。
スタフィロコッカス属菌としては、例えば、スタフィロコッカス・オーレウス(S. aureus)、スタフィロコッカス・エピデルミス(S. epidermis)等が挙げられる。
バチルス属菌としては、例えば、バチルス・セレウス(B. cereus)、バチルス・ズブチリス(B. subtilis)等が挙げられる。

カンピロバクター属菌としては、例えば、カンピロバクター・ジェジュニ (C. jejuni)、カンピロバクター・コリ (C. coli)、カンピロバクター・フェタス(C. fetus)等が挙げられる。

<キレート剤>
本実施形態の食中毒細菌のバクテリオファージ耐性化抑制剤に含まれるキレート剤としては、金属を捕捉するキレート作用を持つ化合物を適宜選択して用いることができる。また、本実施形態の食中毒細菌のバクテリオファージ耐性化抑制剤は、食品等を対象として用いることができることから、それぞれの国において食品添加物として認可されている化合物を用いることができる。このようなキレート剤としては、例えば、クエン酸類、リン酸類、エチレンジアミン四酢酸(Ethylenediaminetetraacetic acid:EDTA)、及び、これらの塩等が挙げられる。リン酸類としては、例えば、ヘキサメタリン酸等が挙げられる。これらキレート剤を、1種類単独で用いてもよく、2種類以上組み合わせて用いてもよい。
中でも、キレート剤としては、EDTA又はその塩を用いることができる。EDTAの塩として具体的には、例えば、エチレンジアミン四酢酸カルシウム二ナトリウム、エチレンジアミン四酢酸二ナトリウム等が挙げられる。
また、これらキレート剤は市販のものを用いてもよく、公知の方法を用いて合成してもよい。
本実施形態の食中毒細菌のバクテリオファージ耐性化抑制剤に含まれるキレート剤としては、金属を捕捉するキレート作用を持つ化合物を適宜選択して用いることができる。また、本実施形態の食中毒細菌のバクテリオファージ耐性化抑制剤は、食品等を対象として用いることができることから、それぞれの国において食品添加物として認可されている化合物を用いることができる。このようなキレート剤としては、例えば、クエン酸類、リン酸類、エチレンジアミン四酢酸(Ethylenediaminetetraacetic acid:EDTA)、及び、これらの塩等が挙げられる。リン酸類としては、例えば、ヘキサメタリン酸等が挙げられる。これらキレート剤を、1種類単独で用いてもよく、2種類以上組み合わせて用いてもよい。
中でも、キレート剤としては、EDTA又はその塩を用いることができる。EDTAの塩として具体的には、例えば、エチレンジアミン四酢酸カルシウム二ナトリウム、エチレンジアミン四酢酸二ナトリウム等が挙げられる。
また、これらキレート剤は市販のものを用いてもよく、公知の方法を用いて合成してもよい。
<ナイシン>
本実施形態の食中毒細菌のバクテリオファージ耐性化抑制剤に含まれるナイシンとしては、例えば、ナイシンA、ナイシンZ、ナイシンQ等が挙げられ、これらに限定されない。これらナイシンを、1種類単独で用いてもよく、2種類以上組み合わせて用いてもよい。
なお、ナイシンAは、乳酸菌Lactococcus lactisにより産生され、欧米等50カ国以上ですでに食品添加物として認可使用されている。ナイシンZは、ナイシンAに類似のタンパク質である。ナイシンAを構成する34個のアミノ酸残基のうち、N末端から27番目のアミノ酸残基が、ナイシンAではヒスチジン残基であるのに対し、ナイシンZではアスパラギン残基である点のみが異なる。
本実施形態の食中毒細菌のバクテリオファージ耐性化抑制剤に含まれるナイシンとしては、例えば、ナイシンA、ナイシンZ、ナイシンQ等が挙げられ、これらに限定されない。これらナイシンを、1種類単独で用いてもよく、2種類以上組み合わせて用いてもよい。
なお、ナイシンAは、乳酸菌Lactococcus lactisにより産生され、欧米等50カ国以上ですでに食品添加物として認可使用されている。ナイシンZは、ナイシンAに類似のタンパク質である。ナイシンAを構成する34個のアミノ酸残基のうち、N末端から27番目のアミノ酸残基が、ナイシンAではヒスチジン残基であるのに対し、ナイシンZではアスパラギン残基である点のみが異なる。
ナイシンは、公知の方法を用いて、乳酸菌を培養し、精製することによって得ることができる。具体的には、例えば、まず、MRS培地(Oxoid社製)を用いて、乳酸菌を培養する。次いで、培養上清をアンバーライト(Amberlite XAD-16、シグマ社製)処理してナイシンを吸着させる。次いで、アンバーライトを蒸留水及び40v/v%エタノールで洗浄する。次いで、0.1w/v%のトリフルオロ酢酸を含む70v/v%イソプロパノールでナイシンを溶出させる。次いで、溶出画分を、陽イオン交換カラム(SP-Sepharose FF、GEヘルスケアバイオサイエンス株式会社製)を用いて精製することで、ナイシンの精製品を得ることができる。また、陽イオン交換カラム後のナイシンを、必要に応じて、逆相クロマトグラフィーを用いて精製することで、さらにナイシンの精製度を高めることができる(参考文献1:Zendo T et al., “Identification of the Lantibiotic Nisin Q, a New Natural Nisin Variant Produced by Lactococcus lactis 61-14 Isolated from a River in Japan”, Biosci. Biotechnol. Biochem., vol.67, No.7, p1616-1619,2003)。
また、ナイシンの市販品を用いてもよく、具体的には、例えば、ナイシンA試薬(ナイシン2.5%含有、Sigma社製)等が挙げられ、これに限定されない。

<その他添加剤>
本実施形態の食中毒細菌のバクテリオファージ耐性化抑制剤は、食中毒細菌のバクテリオファージ耐性化抑制効果を妨げない範囲において、上記キレート剤又は上記ナイシン以外のその他の添加剤を含有していてもよい。
本実施形態の食中毒細菌のバクテリオファージ耐性化抑制剤は、食中毒細菌のバクテリオファージ耐性化抑制効果を妨げない範囲において、上記キレート剤又は上記ナイシン以外のその他の添加剤を含有していてもよい。
その他の添加剤としては、食品に使用可能である化合物を用いることができる。そのようなその他の添加剤として具体的には、例えば、塩類を含む緩衝剤、微量の金属、グリセロール、糖類、アミノ酸、酸化防止剤等が挙げられる。
塩類としては、例えば、ナトリウム、カリウム等が挙げられる。
微量の金属としては、例えば、Mg、Mn、Ca等が挙げられる。
糖類としては、例えば、マルトース、グルコース等が挙げられる。
アミノ酸としては、例えば、グリシン、アルギニン、リジン等が挙げられる。
酸化防止剤としては、例えば、L-アスコルビン酸、L-アスコルビン酸ナトリウム、エリソルビン酸、ジブチルヒドロキシトルエン、dl-α-トコフェロール、没食子酸プロピル、γ-オリザノール、カンゾウ油性抽出物、d-α-トコフェロール、フェルラ酸、ミックストコフェロール、ローズマリー抽出物等が挙げられる。

≪食中毒細菌増殖抑制用組成物≫
本実施形態の食中毒細菌増殖抑制用組成物は、バクテリオファージと、上記実施形態に係る食中毒細菌のバクテリオファージ耐性化抑制剤と、を含む。

<バクテリオファージ>
本実施形態の食中毒細菌増殖抑制用組成物に含まれるバクテリオファージとしては、特定の食中毒細菌に吸着して、特異的に該食中毒細菌を溶菌破壊できるものを適宜選択して用いることができる。バクテリオファージとして具体的には、例えば、大腸菌を宿主とするバクテリオファージとしては、T系ファージ、ΦX-174、λ、ΦX80、Qβ、P1等が挙げられる。T系ファージとしては、例えば、T1、T2、T3、T4、T5、T6、T7等が挙げられる。また、例えば、バチルス属菌を宿主とするバクテリオファージとしては、SZP01、SZP02等が挙げられる。
バクテリオファージとしてより具体的には、例えば、以下の(1)及び(2)に示すもの等が挙げられ、これらに限定されない。これらバクテリオファージを、1種類単独で用いてもよく、2種類以上組み合わせて用いてもよい。
(1)グラム陽性菌に対する特異的溶菌ファージ
L. monocytogenes No.193株特異的溶菌ファージ:193-S3株
(2)グラム陰性菌に対する特異的溶菌ファージ
C. jejuni特異的溶菌ファージ:P10株

本実施形態の食中毒細菌増殖抑制用組成物に含まれるバクテリオファージとしては、特定の食中毒細菌に吸着して、特異的に該食中毒細菌を溶菌破壊できるものを適宜選択して用いることができる。バクテリオファージとして具体的には、例えば、大腸菌を宿主とするバクテリオファージとしては、T系ファージ、ΦX-174、λ、ΦX80、Qβ、P1等が挙げられる。T系ファージとしては、例えば、T1、T2、T3、T4、T5、T6、T7等が挙げられる。また、例えば、バチルス属菌を宿主とするバクテリオファージとしては、SZP01、SZP02等が挙げられる。
バクテリオファージとしてより具体的には、例えば、以下の(1)及び(2)に示すもの等が挙げられ、これらに限定されない。これらバクテリオファージを、1種類単独で用いてもよく、2種類以上組み合わせて用いてもよい。
(1)グラム陽性菌に対する特異的溶菌ファージ

(2)グラム陰性菌に対する特異的溶菌ファージ

バクテリオファージは、その種類に応じて、公知慣用の方法を用いて、バクテリオファージを宿主となる菌体に感染させ、該菌体を培養することで得ることができる。
<キレート剤の含有量>
本実施形態の食中毒細菌増殖抑制用組成物中のキレート剤の含有量は、本実施形態の食中毒細菌増殖抑制用組成物の総容量に対して、0.01容量%より多くすることができ、0.02容量%以上0.5容量%以下とすることができ、0.025容量%以上0.1容量%以下とすることができる。キレート剤の含有量が上記下限値以上であることにより、食中毒細菌のバクテリオファージに対する耐性化をより効果的に抑制することができる。一方、キレート剤の含有量が上記上限値以下であることにより、食品に対して使用した場合に、食品中のキレート剤の含有量を食品添加物として適正な範囲とすることができる。
本実施形態の食中毒細菌増殖抑制用組成物中のキレート剤の含有量は、本実施形態の食中毒細菌増殖抑制用組成物の総容量に対して、0.01容量%より多くすることができ、0.02容量%以上0.5容量%以下とすることができ、0.025容量%以上0.1容量%以下とすることができる。キレート剤の含有量が上記下限値以上であることにより、食中毒細菌のバクテリオファージに対する耐性化をより効果的に抑制することができる。一方、キレート剤の含有量が上記上限値以下であることにより、食品に対して使用した場合に、食品中のキレート剤の含有量を食品添加物として適正な範囲とすることができる。
<ナイシンの含有量>
本実施形態の食中毒細菌増殖抑制用組成物中のナイシンの含有量は、本実施形態の食中毒細菌増殖抑制用組成物の総容量に対して、100IU/mL以上3000IU/mL以下とすることができ、500IU/mL以上2000IU/mL以下とすることができる。ナイシンの含有量が上記下限値以上であることにより、食中毒細菌のバクテリオファージに対する耐性化をより効果的に抑制することができる。一方、ナイシンの含有量が上記上限値以下であることにより、食品に対して使用した場合に、食品中のナイシンの含有量を食品添加物として適正な範囲とすることができる。

<形状及び形態>
本実施形態の食中毒細菌増殖抑制用組成物は、液状、半固形状又は固形状とすることができる。形状として具体的には、例えば、液剤、乳剤(エマルジョン)、ゲル剤、それらを含浸させた種々の剤形等が挙げられ、これらに限定されない。
本実施形態の食中毒細菌増殖抑制用組成物が液状である場合、その形態として具体的には、例えば、噴霧用容器入り、プッシュ式容器入り(例えば、液で出るタイプ等)、詰替用ボトル入り等が挙げられ、これらに限定されない。
本実施形態の食中毒細菌増殖抑制用組成物が半固形状又は固形状である場合、その形態として具体的には、例えば、噴霧用容器入り、プッシュ式容器入り(例えば、ジェルで出るタイプ等)、口腔内洗浄及びリンス剤、歯磨き剤、ディッピング剤、清拭綿、ペーパータオル、ウエットティッシュ等が挙げられ、これらに限定されない。
本実施形態の食中毒細菌増殖抑制用組成物は、液状、半固形状又は固形状とすることができる。形状として具体的には、例えば、液剤、乳剤(エマルジョン)、ゲル剤、それらを含浸させた種々の剤形等が挙げられ、これらに限定されない。

<使用方法>
本実施形態の食中毒細菌増殖抑制用組成物は、例えば、食品、食品が接触する道具、食品の加工調理器具、食品を加工調理する装置、食品を加工調理する施設、食品の加工調理に携わる人の手指等に存在する食中毒細菌のバクテリオファージ耐性化を抑制するために使用することができる。
本実施形態の食中毒細菌増殖抑制用組成物は、例えば、食品、食品が接触する道具、食品の加工調理器具、食品を加工調理する装置、食品を加工調理する施設、食品の加工調理に携わる人の手指等に存在する食中毒細菌のバクテリオファージ耐性化を抑制するために使用することができる。
食品としては、例えば果汁、野菜汁、農産物及びその加工品、魚介類及びその加工品、肉類(例えば鶏肉、牛肉、豚肉、羊肉等)及びその加工品、乳類(例えば牛乳等)及びその加工品等が挙げられる。中でも、食用の肉類、特に生食用の肉類に本実施形態の食中毒細菌増殖抑制用組成物を使用することができる。また、食品には、ヒト用の食品に限らず、動物用の飼料が含まれていてもよい。動物としては、家畜であってもよく、愛玩動物であってもよい。家畜としては、例えば、トリ、ブタ、ウシ、ヒツジ等が挙げられる。愛玩動物としては、例えば、ネコ、イヌ、ウサギ等が挙げられる。
また、例えば、ウシの飼料に本実施形態の食中毒細菌増殖抑制用組成物を混ぜて摂取させることにより、ウシ腸内に生息する食中毒の病原菌の増殖を効果的に抑制することができる。さらに、本実施形態の食中毒細菌増殖抑制用組成物を含む飼料を与えて飼育したウシは生食用の牛肉として加工することができる。
また、例えば、ウシの飼料に本実施形態の食中毒細菌増殖抑制用組成物を混ぜて摂取させることにより、ウシ腸内に生息する食中毒の病原菌の増殖を効果的に抑制することができる。さらに、本実施形態の食中毒細菌増殖抑制用組成物を含む飼料を与えて飼育したウシは生食用の牛肉として加工することができる。
本実施形態の食中毒細菌増殖抑制用組成物の使用方法について、詳細を以下に説明する。まず、食品等の対象物と、上記食中毒細菌増殖抑制用組成物とを接触させる。食品等の対象物と上記食中毒細菌増殖抑制用組成物とを接触させる時間としては、食品等の対象物に含まれる食中毒細菌の量に応じて適宜調整することができる。食品等の対象物と、上記食中毒細菌増殖抑制用組成物との接触方法としては、直接的な方法でもよく、間接的な方法でもよい。直接的な方法としては、例えば、噴霧、霧吹き、ディッピング、ソーキング等が挙げられる。間接的な方法としては、例えば、食品を加工、調理、保存、包装等する際に、該食品が接触する道具又は食品加工調理器具等の表面を、上記食中毒細菌増殖抑制用組成物で予め処理する方法等が挙げられる。
また、食品等の対象物と、上記食中毒細菌増殖抑制用組成物との接触は、例えば5℃以上45℃以下の温度で行うことができる。温度が上記範囲内であることにより、後述の実施例に示すように、上記食中毒細菌増殖抑制用組成物に含まれる上記キレート剤又は上記ナイシンにより、食中毒細菌のバクテリオファージに対する耐性化を抑制しながら、上記食中毒細菌増殖抑制用組成物に含まれるバクテリオファージが増殖し、食中毒細菌を効果的に溶菌することができる。
また、例えば、対象物が食品である場合、食品の表面積に対して、キレート剤の接触量を10μg/cm2以上程度となるようにすることができる。又は、食品の表面積に対して、ナイシンの接触量を5U/cm2以上程度となるようにすることができる。食品の表面積に対してキレート剤又はナイシンの接触量が上記下限値以上であることにより、バクテリオファージに対する耐性化抑制効果を充分に発揮することができる。
一方、食品の表面積に対してキレート剤又はナイシンの接触量の上限値としては、それぞれの国において食品添加物として食品に含まれるキレート剤又はナイシンの使用基準を上回らない程度の量とすることができる。
また、対象物が食品以外である場合においても、対象物の表面積に対してキレート剤又はナイシンの接触量が上記下限値以上であることにより、バクテリオファージに対する耐性化抑制効果を充分に発揮することができる。一方、対象物の表面積に対してキレート剤又はナイシンの接触量の上限値としては、特別な限定はなく、適切な製造コストとなる量であってもよい。
また、例えば、対象物が食品である場合、食品の表面積に対して、キレート剤の接触量を10μg/cm2以上程度となるようにすることができる。又は、食品の表面積に対して、ナイシンの接触量を5U/cm2以上程度となるようにすることができる。食品の表面積に対してキレート剤又はナイシンの接触量が上記下限値以上であることにより、バクテリオファージに対する耐性化抑制効果を充分に発揮することができる。
一方、食品の表面積に対してキレート剤又はナイシンの接触量の上限値としては、それぞれの国において食品添加物として食品に含まれるキレート剤又はナイシンの使用基準を上回らない程度の量とすることができる。
また、対象物が食品以外である場合においても、対象物の表面積に対してキレート剤又はナイシンの接触量が上記下限値以上であることにより、バクテリオファージに対する耐性化抑制効果を充分に発揮することができる。一方、対象物の表面積に対してキレート剤又はナイシンの接触量の上限値としては、特別な限定はなく、適切な製造コストとなる量であってもよい。
また、食品等の対象物と、上記食中毒細菌増殖抑制用組成物との接触の前に、食品等の対象物を、次亜塩素酸ナトリウム等を用いて殺菌処理してもよい。これにより、バクテリオファージの標的となる食中毒細菌以外の一般細菌の生菌数の増加を抑制しながら、上記食中毒細菌増殖抑制用組成物に含まれるバクテリオファージが増殖し、食中毒細菌を効果的に溶菌することができる。
以下、実施例及び比較例等を挙げて本発明をさらに詳述するが、本発明はこれらの実施例等に限定されるものではない。
[実施例1]カンピロバクター属菌に対するバクテリオファージと食品添加物との併用効果の検討
1.食品添加物単独での抗菌活性の確認
まず、日本国内で食品添加物として認可されており、抗菌活性を有することが知られている3種の食品添加物(ヘキサメタリン酸ナトリウム、エチレンジアミン四酢酸(Ethylenediaminetetraacetic acid:EDTA)、及び、酢酸ナトリウム)を用いて、各食品添加物単独での抗菌活性を確認した。
具体的には、96ウェルCell Culture Cluster に1mMのCaCl2及び10mMのMgSO4含有ブレインハートインフュージョン培地(brain heart infusion broth:BHI培地)を190μLずつ分注し、Campylobacter jejuni培養液の10倍希釈液を10μL(8.7×105CFU/mL)ずつ接種した。そこに各食品添加物を含有する溶液を20μLずつ、下記表1に示す濃度になるように添加し、42℃、微好気条件下で培養した。0時間後(すなわち、培養開始時)及び24時間後に、培養液をウマ血液寒天培地(Horse blood agar:HBA)にプレーティングし、42℃、48時間微好気培養した後、コロニー数から生菌数を測定した。また、食品添加物を添加しないものをコントロールとした。結果を以下の表1に示す。
なお、ウマ血液寒天培地(Horse blood agar:HBA)は、以下の手順で調製した。まず、ブルセラ基礎培地(BRUCELLA MEDIUM BASE、OXOID社製) を所定の濃度で調製し、121℃、15分間オートクレーブ滅菌した。次いで、滅菌したブルセラ基礎培地を55℃まで冷却し、45℃に加温したウマ溶血液(関東化学株式会社製)を終濃度5容量%となるように添加し、シャーレに分注した。使用するまで4℃で保存した。
1.食品添加物単独での抗菌活性の確認
まず、日本国内で食品添加物として認可されており、抗菌活性を有することが知られている3種の食品添加物(ヘキサメタリン酸ナトリウム、エチレンジアミン四酢酸(Ethylenediaminetetraacetic acid:EDTA)、及び、酢酸ナトリウム)を用いて、各食品添加物単独での抗菌活性を確認した。

なお、ウマ血液寒天培地(Horse blood agar:HBA)は、以下の手順で調製した。まず、ブルセラ基礎培地(BRUCELLA MEDIUM BASE、OXOID社製) を所定の濃度で調製し、121℃、15分間オートクレーブ滅菌した。次いで、滅菌したブルセラ基礎培地を55℃まで冷却し、45℃に加温したウマ溶血液(関東化学株式会社製)を終濃度5容量%となるように添加し、シャーレに分注した。使用するまで4℃で保存した。
表1から、0.1%EDTA及び2、3、4%酢酸ナトリウムを添加した場合、培養24時間後には初発菌数と比較して生菌数が低下したため、C. jejuniに対する抗菌活性が示された。
2.カンピロバクター属菌に対するバクテリオファージとEDTAとの併用効果の検討
次に、EDTA存在下で、液体培地中におけるC. jejuniに対するC. jejuni特異的溶菌ファージPC10の効果を調べた。
まず、1mMのCaCl2及び10mMのMgSO4含有BHI培地5mLに、C. jejuni培養液の10倍希釈液250μLを接種して、PC10(力価:5.3×108PFU/mL)100μL及びEDTA500μL(終濃度:0.001、0.01、0.025及び0.1容量%)をそれぞれ添加し、42℃、48時間微好気条件下で培養した。0時間後(すなわち、培養開始時)、4時間後、8時間後、12時間後及び24時間後に、培養液をHBAにプレーティングし、42℃、48時間微好気培養した後、コロニー数から生菌数を測定した。また、PC10及びEDTAのいずれも添加しないものをコントロールとした。結果を以下の図1に示す。なお、図1には、「コントロール」、「PC10単独」、「0.025容量%EDTA単独」、「0.1容量%EDTA単独」、「PC10と0.025容量%EDTAとの併用」、「PC10と0.1容量%EDTAとの併用」の結果を示した。
次に、EDTA存在下で、液体培地中におけるC. jejuniに対するC. jejuni特異的溶菌ファージPC10の効果を調べた。
まず、1mMのCaCl2及び10mMのMgSO4含有BHI培地5mLに、C. jejuni培養液の10倍希釈液250μLを接種して、PC10(力価:5.3×108PFU/mL)100μL及びEDTA500μL(終濃度:0.001、0.01、0.025及び0.1容量%)をそれぞれ添加し、42℃、48時間微好気条件下で培養した。0時間後(すなわち、培養開始時)、4時間後、8時間後、12時間後及び24時間後に、培養液をHBAにプレーティングし、42℃、48時間微好気培養した後、コロニー数から生菌数を測定した。また、PC10及びEDTAのいずれも添加しないものをコントロールとした。結果を以下の図1に示す。なお、図1には、「コントロール」、「PC10単独」、「0.025容量%EDTA単独」、「0.1容量%EDTA単独」、「PC10と0.025容量%EDTAとの併用」、「PC10と0.1容量%EDTAとの併用」の結果を示した。
図1から、PC10単独の場合は培養8時間後以降に生菌数が増加した。また、0.001容量%及び0.01容量%EDTAを併用した場合もPC10単独の場合と同様に培養8時間後以降に生菌数が増加した(図示せず)。一方、0.025容量%及び0.1容量%EDTAを併用した場合は、8時間後以降の菌の増殖が抑制され、生菌数をさらに低下させて、培養24時間後には生菌数が検出下限以下になった。
このことから、PC10と0.025容量%以上0.1容量%以下のEDTAとを併用した場合は、EDTAによりC. jejuniのPC10に対する耐性化が効果的に抑制され、これにより、効果的にC. jejuniの増殖が抑制された。

[実施例2]サルモネラ属菌に対するバクテリオファージとEDTAとの併用効果の検討
次に、Salmnella Enteritidisに対するS. Enteritidis特異的溶菌ファージSTG2とEDTAとの併用効果を調べた。
まず、1mMのCaCl2及び10mMのMgSO4含有BHI培地5mLに、S. Enteritidis培養液の10倍希釈液250μLを接種して、STG2(力価:1.0×1010PFU/mL)100μL及びEDTA500μL(終濃度:0.025及び0.1容量%)をそれぞれ添加し、42℃、48時間微好気条件下で培養した。0時間後(すなわち、培養開始時)、4時間後、8時間後、12時間後及び24時間後に、培養液をHBAにプレーティングし、42℃、48時間微好気培養した後、コロニー数から生菌数を測定した。また、STG2及びEDTAのいずれも添加しないものをコントロールとした。結果を以下の図2に示す。
次に、Salmnella Enteritidisに対するS. Enteritidis特異的溶菌ファージSTG2とEDTAとの併用効果を調べた。

図2から、STG2単独の場合は培養6時間後以降に生菌数が増加した。また、0.025容量%及び0.1容量%EDTA単独の場合、並びに、0.025容量%EDTAを併用した場合も、STG2単独の場合と同様に培養6時間後以降に生菌数が増加した。一方、0.1容量%EDTAを併用した場合は、6時間後以降の菌の増殖が抑制され、生菌数をさらに低下させて、培養24時間後には生菌数が検出下限以下になった。
このことから、STG2と0.1容量%のEDTAとを併用した場合は、EDTAによりS. EnteritidisのSTG2に対する耐性化が効果的に抑制され、これにより、効果的にS. Enteritidisの増殖が抑制された。

これまでに、EDTAは細菌に対するファージ感染の阻害剤として使用されるという報告がある(例えば、「参考文献2:Hungaro HM et al., “Use of bacteriophages to reduce Salmonella in chicken skin in comparison with a chemical agents.”, Food Research International, Vol. 52, Issue 1, p75-81, 2013.」参照)。
しかしながら、実施例1及び2で使用したEDTAの濃度は、上記報告で使用された濃度よりも低いことから、低濃度のEDTAであれば、C. jejuni及びS. Enteritidisの、ファージに対する耐性化抑制効果を示すことが明らかになった。
また、現在、EDTAは缶詰及び瓶詰食品に使用されており、その使用基準は0.25g/kgとなっている。そのため、より実用的な濃度である0.025%EDTAを以降の実施例で使用した。
しかしながら、実施例1及び2で使用したEDTAの濃度は、上記報告で使用された濃度よりも低いことから、低濃度のEDTAであれば、C. jejuni及びS. Enteritidisの、ファージに対する耐性化抑制効果を示すことが明らかになった。
また、現在、EDTAは缶詰及び瓶詰食品に使用されており、その使用基準は0.25g/kgとなっている。そのため、より実用的な濃度である0.025%EDTAを以降の実施例で使用した。
また、ファージは宿主菌の増殖能を利用して菌体内で増殖し、宿主菌を溶菌する。そのため、宿主菌が増殖できない環境下ではファージも増殖できずに宿主菌を溶菌できない。MOIを高く設定して1菌体に大量のファージが吸着できる環境にすれば、ファージが増殖しなくても宿主菌を溶菌するという報告があるが、実施例1及び2のMOIは10程度であった。
これに対し、ファージと低濃度のEDTAとの併用は、図1及び図2の結果からも明らかなように、相加効果ではなく、相乗効果であった。これは、低濃度のEDTAにより細菌のファージ耐性化が抑制されたことで、少量のファージであっても、溶菌作用が顕著に働いたためであると推察された。
これに対し、ファージと低濃度のEDTAとの併用は、図1及び図2の結果からも明らかなように、相加効果ではなく、相乗効果であった。これは、低濃度のEDTAにより細菌のファージ耐性化が抑制されたことで、少量のファージであっても、溶菌作用が顕著に働いたためであると推察された。
[実施例3]殺菌処理鶏皮におけるC. jejuniに対するPC10とEDTAとの併用効果の検討
次に、PC10とEDTAとの併用効果の実用性を調べるために、実際に鶏皮に接種したC. jejuniに対するPC10とEDTAとの併用効果を調べた。試料としては、市販の国産若鶏皮を使用した。実際に食鶏処理過程で使用される次亜塩素酸ナトリウムによる殺菌処理と、ファージ感染のための42℃での保持を加えた試料処理方法とについて検討した。
次に、PC10とEDTAとの併用効果の実用性を調べるために、実際に鶏皮に接種したC. jejuniに対するPC10とEDTAとの併用効果を調べた。試料としては、市販の国産若鶏皮を使用した。実際に食鶏処理過程で使用される次亜塩素酸ナトリウムによる殺菌処理と、ファージ感染のための42℃での保持を加えた試料処理方法とについて検討した。
まず、3cm×3cm(9cm2)にカットした市販鶏皮にC. jejuniの培養液の100倍希釈液(滅菌水で希釈)を20μL接種し、クリーンベンチ内で15分間静置した。次いで、市販鶏肉でも行われている0.02%次亜塩素酸ナトリウムによる殺菌を60秒間行い、純水で洗浄してキムタオルで水気を除去した。次いで、0.1%EDTAに60秒間浸漬し、水気を軽く除去したのち、PC10(力価:1.1×108PFU/mL)100μLを接種して、真空パックした。真空パックした鶏皮は42℃で1時間保持後、8℃で保存し、0時間後(すなわち、保存開始時)、1時間後、4時間後、8時間後、24時間後の生菌数を測定した。PC10及びEDTAを添加せずに次亜塩素酸ナトリウムのみで処理し、42℃で保持せず、8℃で保存したものをコントロールとした。
また、生菌数測定では、ストマッカー袋に採取した鶏皮検体に生理食塩水10mLを加えて60秒間ストマッカー処理し、生理食塩水で段階希釈した。バツラー培地と標準寒天培地とにそれぞれプレーティングし、バツラー培地では、37℃、48時間微好気培養後のコロニー数からC. jejuni生菌数を測定した。また、標準寒天培地では、37℃、48時間好気培養後のコロニー数から一般生菌数を測定した。結果を図3A(C. jejuni生菌数)及び図3B(一般生菌数)に示す。
なお、バツラー培地は、以下の手順で調製した。まず、コロンビア血液寒天基礎培地(関東化学株式会社製)を所定の濃度で調製し、121℃、15分間、オートクレーブ滅菌した。次いで、滅菌したコロンビア血液寒天基礎培地を55℃まで冷却し、45℃に加温したウマ溶血液(関東化学株式会社製)を終濃度5容量%となるように添加し、シャーレに分注して、寒天がゲル化後、使用した。
また、標準寒天培地は、以下の手順で調製した。まず、パールコア標準寒天培地(栄研化学株式会社製)を所定の濃度で調製し、121℃、15分間、オートクレーブ滅菌した。次いで、滅菌したパールコア標準寒天培地をシャーレに分注して、寒天がゲル化後、使用した。
また、生菌数測定では、ストマッカー袋に採取した鶏皮検体に生理食塩水10mLを加えて60秒間ストマッカー処理し、生理食塩水で段階希釈した。バツラー培地と標準寒天培地とにそれぞれプレーティングし、バツラー培地では、37℃、48時間微好気培養後のコロニー数からC. jejuni生菌数を測定した。また、標準寒天培地では、37℃、48時間好気培養後のコロニー数から一般生菌数を測定した。結果を図3A(C. jejuni生菌数)及び図3B(一般生菌数)に示す。
なお、バツラー培地は、以下の手順で調製した。まず、コロンビア血液寒天基礎培地(関東化学株式会社製)を所定の濃度で調製し、121℃、15分間、オートクレーブ滅菌した。次いで、滅菌したコロンビア血液寒天基礎培地を55℃まで冷却し、45℃に加温したウマ溶血液(関東化学株式会社製)を終濃度5容量%となるように添加し、シャーレに分注して、寒天がゲル化後、使用した。
また、標準寒天培地は、以下の手順で調製した。まず、パールコア標準寒天培地(栄研化学株式会社製)を所定の濃度で調製し、121℃、15分間、オートクレーブ滅菌した。次いで、滅菌したパールコア標準寒天培地をシャーレに分注して、寒天がゲル化後、使用した。
図3Aから、C. jejuni生菌数は42℃で1 時間保持後には1.2桁減少し、この後8℃で24時間保存しても生菌数の増加は認められなかった。この結果から、市販鶏皮上においても宿主菌の生育至適温度の42℃であれば、ファージが感染できることが示された。
また、図3Bから、42℃保持によって一般生菌が増殖することが懸念されたが、1時間程度であればほとんど増殖しないことが確かめられた。次亜塩素酸ナトリウムは水溶液中で生成した次亜塩素酸が細菌の細胞膜に作用してタンパク等を酸化することでその機能を阻害するため、グラム陽性菌及びグラム陰性菌に対して幅広く殺菌効果を示す。そのため、市販鶏皮上に存在する細菌に対して次亜塩素酸ナトリウムが作用したことで、42℃でも細菌の増殖が抑制されたと考えられた。
これらのことから、殺菌処理とEDTA処理とを組み合わせた前処理を行うことで、バクテリオファージを用いて食品のC. jejuni制御が可能であることが示された。
また、図3Bから、42℃保持によって一般生菌が増殖することが懸念されたが、1時間程度であればほとんど増殖しないことが確かめられた。次亜塩素酸ナトリウムは水溶液中で生成した次亜塩素酸が細菌の細胞膜に作用してタンパク等を酸化することでその機能を阻害するため、グラム陽性菌及びグラム陰性菌に対して幅広く殺菌効果を示す。そのため、市販鶏皮上に存在する細菌に対して次亜塩素酸ナトリウムが作用したことで、42℃でも細菌の増殖が抑制されたと考えられた。
これらのことから、殺菌処理とEDTA処理とを組み合わせた前処理を行うことで、バクテリオファージを用いて食品のC. jejuni制御が可能であることが示された。
このようにバクテリオファージとEDTAとの組み合わせにより、EDTAによりカンピロバクター及びサルモネラ等の食中毒細菌のバクテリオファージに対する耐性化を効果的に抑制でき、これにより、カンピロバクター及びサルモネラ等の食中毒細菌に対するバクテリオファージの溶菌効果が高められ、さらにその生育を長時間阻害できることが示された。
[実施例4]リステリア属菌に対するバクテリオファージとEDTAとの併用効果の検討
次に、グラム陽性菌であるListeria monocytogenes No.193株に対するL. monocytogenes No.193株特異的溶菌ファージ193-S3とEDTAとの併用効果を調べた。
まず、LB培地5mLに、Listeria monocytogenes No.193株の培養液(OD660=0.1)の10倍希釈液250μLを接種して、193-S3(力価:5.7×109PFU/mL)100μL及びEDTA500μL(終濃度:0.025容量%)をそれぞれ添加し、37℃、48時間好気条件下で振とう培養した。0時間後(すなわち、培養開始時)、3時間後、6時間後、24時間後及び48時間後に、培養液をトリプケースソイ寒天培地(Trypcase Soy Agar:TSA)にプレーティングし、37℃、48時間好気培養した後、コロニー数から生菌数を測定した。また、193-S3及びEDTAのいずれも添加しないものをコントロールとした。結果を以下の図4に示す。
次に、グラム陽性菌であるListeria monocytogenes No.193株に対するL. monocytogenes No.193株特異的溶菌ファージ193-S3とEDTAとの併用効果を調べた。
まず、LB培地5mLに、Listeria monocytogenes No.193株の培養液(OD660=0.1)の10倍希釈液250μLを接種して、193-S3(力価:5.7×109PFU/mL)100μL及びEDTA500μL(終濃度:0.025容量%)をそれぞれ添加し、37℃、48時間好気条件下で振とう培養した。0時間後(すなわち、培養開始時)、3時間後、6時間後、24時間後及び48時間後に、培養液をトリプケースソイ寒天培地(Trypcase Soy Agar:TSA)にプレーティングし、37℃、48時間好気培養した後、コロニー数から生菌数を測定した。また、193-S3及びEDTAのいずれも添加しないものをコントロールとした。結果を以下の図4に示す。
図4から、193-S3単独の場合は培養3時間後以降に生菌数が増加した。また、0.025容量%EDTA単独の場合、培養6時間後までゆるやかに生菌数が増加したが、培養6時間後以降は、生菌数はほぼ一定であった。一方、0.025容量%EDTAを併用した場合は、6時間後以降の菌の増殖が抑制され、生菌数をさらに低下させて、培養48時間後には生菌数が1×104CFU/mL以下になった。
このことから、193-S3と0.025容量%のEDTAとを併用した場合は、効果的にL. monocytogenesの増殖が抑制された。また、L. monocytogenesに対するファージと低濃度のEDTAとの併用は、図4の結果からも明らかであるが、上記図1及び上記図2の結果と同様に、相加効果ではなく、相乗効果であった。これは、低濃度のEDTAによりリステリア属菌のファージ耐性化が抑制されたことで、少量のファージであっても、溶菌作用が顕著に働いたためであると推察された。
このことから、193-S3と0.025容量%のEDTAとを併用した場合は、効果的にL. monocytogenesの増殖が抑制された。また、L. monocytogenesに対するファージと低濃度のEDTAとの併用は、図4の結果からも明らかであるが、上記図1及び上記図2の結果と同様に、相加効果ではなく、相乗効果であった。これは、低濃度のEDTAによりリステリア属菌のファージ耐性化が抑制されたことで、少量のファージであっても、溶菌作用が顕著に働いたためであると推察された。
以上の結果から、ファージと低濃度のEDTAとを併用することで、グラム陰性菌及びグラム陽性菌のいずれの食中毒細菌についても、増殖を効果的に抑制できることが明らかとなった。
[実施例5]食中毒細菌の混合培養液に対するバクテリオファージとナイシンとの併用効果の検討
次に、日本国内で食品添加物として認可されており、抗菌活性を有することが知られているナイシンを用いて、食中毒細菌の混合培養液に対する各細菌の特異的溶菌ファージのカクテルとナイシンとの併用効果を調べた。
まず、LB培地5mLに、S. Enteritidis、S. Typhimurium及びEscherichia coli O157:H7の培養液(OD660=0.1)の10倍希釈液250μLを接種して、S. Enteritidis特異的溶菌ファージSTG2(力価:0.3×1010PFU/mL)、S. Typhimurium特異的溶菌ファージSEG5(力価:0.3×1010PFU/mL)及びE. coli O157:H7特異的溶菌ファージPS5(力価:0.3×1010PFU/mL)を含むファージカクテル溶液100μL及びナイシン500μL(終濃度:500、1000及び2000IU/mL)(Sigma Aldrich社製)をそれぞれ添加し、37℃、24時間好気条件下で振とう培養した。0時間後(すなわち、培養開始時)、2時間後、4時間後、6時間後及び24時間後に、培養液をTSAにプレーティングし、37℃、48時間好気培養した後、コロニー数から生菌数を測定した。また、ファージカクテル溶液及びナイシンのいずれも添加しないものをコントロールとした。結果を以下の図5に示す。図5において、「Phage」とはファージカクテル溶液を意味する。
次に、日本国内で食品添加物として認可されており、抗菌活性を有することが知られているナイシンを用いて、食中毒細菌の混合培養液に対する各細菌の特異的溶菌ファージのカクテルとナイシンとの併用効果を調べた。
まず、LB培地5mLに、S. Enteritidis、S. Typhimurium及びEscherichia coli O157:H7の培養液(OD660=0.1)の10倍希釈液250μLを接種して、S. Enteritidis特異的溶菌ファージSTG2(力価:0.3×1010PFU/mL)、S. Typhimurium特異的溶菌ファージSEG5(力価:0.3×1010PFU/mL)及びE. coli O157:H7特異的溶菌ファージPS5(力価:0.3×1010PFU/mL)を含むファージカクテル溶液100μL及びナイシン500μL(終濃度:500、1000及び2000IU/mL)(Sigma Aldrich社製)をそれぞれ添加し、37℃、24時間好気条件下で振とう培養した。0時間後(すなわち、培養開始時)、2時間後、4時間後、6時間後及び24時間後に、培養液をTSAにプレーティングし、37℃、48時間好気培養した後、コロニー数から生菌数を測定した。また、ファージカクテル溶液及びナイシンのいずれも添加しないものをコントロールとした。結果を以下の図5に示す。図5において、「Phage」とはファージカクテル溶液を意味する。
図5から、ファージカクテル溶液単独の場合は培養4時間後以降に生菌数が増加した。また、500、1000及び2000IU/mLナイシン単独の場合、培養2時間後以降に生菌数が増加した。一方、500、1000及び2000IU/mLナイシンを併用した場合は、4時間後以降の菌の増殖が抑制され、生菌数をさらに低下させて、培養6時間後以降には生菌数が検出下限以下になった。
以上のことから、EDTAと同様に、ファージとナイシンとを併用することで、食中毒細菌の増殖を効果的に抑制できることが明らかとなった。
また、S. Enteritidis、S. Typhimurium及びE. coli O157:H7に対するファージカクテルとナイシンとの併用は、図5の結果からも明らかであるが、上記図1、上記図2及び上記図4の結果と同様に、相加効果ではなく、相乗効果であった。これは、ナイシンによりS. Enteritidis、S. Typhimurium及びE. coli O157:H7の各ファージに対する耐性化が抑制されたことで、少量のファージカクテルであっても、溶菌作用が顕著に働いたためであると推察された。
また、S. Enteritidis、S. Typhimurium及びE. coli O157:H7に対するファージカクテルとナイシンとの併用は、図5の結果からも明らかであるが、上記図1、上記図2及び上記図4の結果と同様に、相加効果ではなく、相乗効果であった。これは、ナイシンによりS. Enteritidis、S. Typhimurium及びE. coli O157:H7の各ファージに対する耐性化が抑制されたことで、少量のファージカクテルであっても、溶菌作用が顕著に働いたためであると推察された。
本実施形態の食中毒細菌のバクテリオファージ耐性化抑制剤は、多種の食中毒細菌に有効であり、食中毒細菌のバクテリオファージに対する耐性化を効果的に抑制することができる。本実施形態の食中毒細菌増殖抑制用組成物は、バクテリオファージと、前記食中毒細菌のバクテリオファージ耐性化抑制剤と、を含み、例えば、食品、調理器具等に対して、好適に用いられる。
Claims (6)
- キレート剤又はナイシンを有効成分として含有する食中毒細菌のバクテリオファージ耐性化抑制剤。
- 前記キレート剤がエチレンジアミン四酢酸又はその塩である請求項1に記載の食中毒細菌のバクテリオファージ耐性化抑制剤。
- バクテリオファージと、請求項1又は2に記載の食中毒細菌のバクテリオファージ耐性化抑制剤とを含む食中毒細菌増殖抑制用組成物。
- 食品に対して用いられる請求項3に記載の食中毒細菌増殖抑制用組成物。
- キレート剤又はナイシンを有効成分として含有する食中毒細菌のバクテリオファージ耐性化抑制剤の使用であって、食中毒細菌のバクテリオファージに対する耐性化を抑制するための、食中毒細菌のバクテリオファージ耐性化抑制剤の使用。
- キレート剤又はナイシンを用いる食中毒細菌のバクテリオファージ耐性化抑制方法。
