背景技术
大豆原产中国。中国各地均有栽培,大豆的营养成分非常丰富,其蛋白质含量高于谷类和薯类食物2.5~8倍,除糖类较低外,其他营养成分,如脂肪、钙、磷、铁和维生素等人体必需的营养物质,都高于谷类和薯类食物,它是一种理想的优质植物蛋白食物。
大豆中含有约40%的蛋白质,20%的脂肪,27%的碳水化合物,多种维生素和矿物质,豆浆中含有大豆异黄酮,具有良好的美白、舒缓以及抗氧化能力,如何让大豆中的营养成分更利于吸收,提高活性大豆异黄酮含量成为了亟待解决的问题。

本发明提供一种乳酸杆菌豆浆发酵产物滤液的制备方法及应用,所述的乳酸杆菌豆浆发酵产物滤液具有良好的美白、抗氧化作用。

一种乳酸杆菌豆浆发酵产物滤液的制备方法,包括以下步骤:
S1、将乳酸杆菌接种于固体培养基进行活化培养,活化后的乳酸杆菌再接种于液体培养基中培养,得到乳酸杆菌菌液;
S2、将3~6重量份豆浆粉、2~5重量份葡萄糖、1~3重量份蔗糖、0.4~1重量份酵母膏、0.02~0.1重量份磷酸二氢钾、90~99重量份去离子水加入发酵罐中,105℃~121℃灭菌20~40min,然后冷却至25~40℃,得到豆浆液;
S3、将乳酸菌菌液接种于豆浆液中,接种量为5~15wt%,在25~40℃下发酵18~40h,过滤,经巴氏灭菌,得到乳酸杆菌豆浆发酵产物滤液。
作为一种优选方案,所述乳酸杆菌为植物乳杆菌、德式乳杆菌、保加利亚乳杆菌、双歧杆菌、干酪乳杆菌、副干酪乳杆菌中的一种或多种。

S11、将乳酸杆菌接种于固体培养基进行活化培养,活化后的乳酸杆菌再接种于液体培养基中培养,得到乳酸杆菌菌液;
S12、将3~6重量份豆浆粉、2~5重量份葡萄糖、1~3重量份蔗糖、0.4~1重量份酵母膏、0.02~0.1%重量份磷酸二氢钾、90~99重量份去离子水加入发酵罐中,105℃~121℃灭菌20~40min,然后冷却至25~40℃,得到豆浆液;
S13、将乳酸菌菌液接种于豆浆液中,接种量为5~15wt%,在25~40℃下发酵18~40h,过滤,经巴氏灭菌,得到乳酸杆菌豆浆发酵产物滤液。
作为一种优选方案,所述乳酸杆菌为植物乳杆菌、德式乳杆菌、保加利亚乳杆菌、双歧杆菌、干酪乳杆菌、副干酪乳杆菌中的一种或多种。

S3、将水相调节pH至6~6.8,以1~1.8BV/h上NKA-2大孔吸附树脂柱,洗脱速度为2~3BV/h,去离子水洗2~3BV,70~90wt%乙醇溶液洗2~3BV,收集乙醇溶液洗脱液,干燥,即得白首乌提取物。
本发明通过对白首乌进行水蒸气蒸馏,水相大孔吸附树脂处理,具有美白作用的成分得到有效保留,不具有美白作用的成分被弃去、被洗脱,从而得到了具有优异美白作用的白首乌提取物。
本发明的有益效果:(1)所述的豆浆经特定菌种发酵后,大分子蛋白分解成为利于人体皮肤吸收的小分子多肽和多种氨基酸,活性大豆异黄酮含量增加,具有良好的美白、舒缓、抗氧化能力,能够广泛的应用于美白化妆品,且在美白方面与白首乌提取物具有协同增效作用。

为使本发明实施例的目的、技术方案和优点更加清楚,下面将对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有作出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
实施例1
一种乳酸杆菌豆浆发酵产物滤液的制备方法,包括以下步骤:
S1、将保加利亚乳杆菌接种于固体培养基在25℃下活化培养2d,活化后的乳酸杆菌再接种于液体培养基中,在28℃下培养2.5d,得到保加利亚乳杆菌菌液;
S2、将4重量份豆浆粉、3重量份葡萄糖、1.2重量份蔗糖、0.5重量份酵母膏、0.05重量份磷酸二氢钾、91.25重量份去离子水加入发酵罐中,110℃灭菌30min,然后冷却至28℃,得到豆浆液;
S3、将保加利亚乳杆菌菌液接种于豆浆液中,接种量为10wt%,在30℃下发酵24h,过滤,经巴氏灭菌,得到乳酸杆菌豆浆发酵产物滤液。
所述固体培养基按重量百分比计,包括:0.8%酪蛋白酶消化物、0.6%牛肉膏、0.3%酵母膏、0.15%柠檬酸三铵、0.4%乙酸钠、0.04%硫酸镁、0.2%硫酸锰、0.12%磷酸二氢钾、2%葡萄糖、0.08%吐温-80、1.5%琼脂、余量去离子水,总量100%。
所述液体培养基配方重量百分比计,包括:0.7%酪蛋白酶消化物、0.65%牛肉膏、0.28%酵母膏、0.15%柠檬酸三铵、0.35%乙酸钠、0.04%硫酸镁、0.2%硫酸锰、0.18%磷酸二氢钾、2%葡萄糖、0.08%吐温-80、余量去离子水,总量100%。
实施例2
一种乳酸杆菌豆浆发酵产物滤液的制备方法,包括以下步骤:
S1、将保加利亚乳杆菌接种于固体培养基在25℃下活化培养2d,活化后的乳酸杆菌再接种于液体培养基中,在28℃下培养2.5d,得到保加利亚乳杆菌菌液;
S2、将5重量份豆浆粉、2.5重量份葡萄糖、1.8重量份蔗糖、0.5重量份酵母膏、0.06重量份磷酸二氢钾、90.14重量份去离子水加入发酵罐中,115℃灭菌25min,然后冷却至32℃,得到豆浆液;
S3、将乳酸菌菌液接种于豆浆液中,接种量为8wt%,在28℃下发酵30h,过滤,经巴氏灭菌,得到乳酸杆菌豆浆发酵产物滤液。
所述固体培养基按重量百分比计,包括:0.6%酪蛋白酶消化物、0.8%牛肉膏、0.3%酵母膏、0.15%柠檬酸三铵、0.3%乙酸钠、0.04%硫酸镁、0.2%硫酸锰、0.15%磷酸二氢钾、1.8%葡萄糖、0.06%吐温-80、1.2%琼脂、余量去离子水,总量100%。
所述液体培养基配方重量百分比计,包括:0.6%酪蛋白酶消化物、0.8%牛肉膏、0.3%酵母膏、0.15%柠檬酸三铵、0.4%乙酸钠、0.04%硫酸镁、0.18%硫酸锰、0.15%磷酸二氢钾、2%葡萄糖、0.08%吐温-80、余量去离子水,总量100%。
实施例3
一种乳酸杆菌豆浆发酵产物滤液的制备方法,包括以下步骤:
S1、将保加利亚乳杆菌接种于固体培养基在25℃下活化培养2d,活化后的乳酸杆菌再接种于液体培养基中,在28℃下培养2.5d,得到保加利亚乳杆菌菌液;
S2、将4.2重量份豆浆粉、3.5重量份葡萄糖、1.2重量份蔗糖、0.5重量份酵母膏、0.08重量份磷酸二氢钾、90.52重量份去离子水加入发酵罐中,108℃灭菌30min,然后冷却至25℃,得到豆浆液;
S3、将乳酸菌菌液接种于豆浆液中,接种量为12wt%,在28℃下发酵24h,过滤,经巴氏灭菌,得到乳酸杆菌豆浆发酵产物滤液。
所述固体培养基按重量百分比计,包括:0.7%酪蛋白酶消化物、0.6%牛肉膏、0.25%酵母膏、0.12%柠檬酸三铵、0.3%乙酸钠、0.03%硫酸镁、0.2%硫酸锰、0.15%磷酸二氢钾、2%葡萄糖、0.06%吐温-80、1.8%琼脂、余量去离子水,总量100%。
所述液体培养基配方重量百分比计,包括:0.6%酪蛋白酶消化物、0.9%牛肉膏、0.26%酵母膏、0.14%柠檬酸三铵、0.3%乙酸钠、0.04%硫酸镁、0.19%硫酸锰、0.15%磷酸二氢钾、1.7%葡萄糖、0.07%吐温-80、余量去离子水,总量100%。
实施例4
一种美白组合物,由乳酸杆菌豆浆发酵产物滤液与白首乌提取物按照重量比1:0.4配制而成。
所述乳酸杆菌豆浆发酵产物滤液为实施例1制备得到的乳酸杆菌豆浆发酵产物滤液。
所述白首乌提取物的制备方法为:
S21、将白首乌药材洗净,干燥,粉碎至90目,得到白首乌粉末;
S2、将白首乌粉末加入到去离子水中,进行水蒸气蒸馏得到油水混合物,取水相;所述白首乌粉末与去离子水的料液比为1g:10ml;
S3、将水相调节pH至6.6,以1.5BV/h上NKA-2大孔吸附树脂柱,洗脱速度为2BV/h,去离子水洗2.5BV,90wt%乙醇溶液洗3BV,收集乙醇溶液洗脱液,干燥,即得白首乌提取物。
实施例5
一种美白组合物,由乳酸杆菌豆浆发酵产物滤液与白首乌提取物按照重量比1:0.3配制而成。
所述乳酸杆菌豆浆发酵产物滤液为实施例2制备得到的乳酸杆菌豆浆发酵产物滤液。
所述白首乌提取物的制备方法为:
S21、将白首乌药材洗净,干燥,粉碎至9目,得到白首乌粉末;
S2、将白首乌粉末加入到去离子水中,进行水蒸气蒸馏得到油水混合物,取水相;所述白首乌粉末与去离子水的料液比为1g:12ml;
S3、将水相调节pH至6.2,以1.2BV/h上NKA-2大孔吸附树脂柱,洗脱速度为2BV/h,去离子水洗2.5BV,90wt%乙醇溶液洗2BV,收集乙醇溶液洗脱液,干燥,即得白首乌提取物。
实施例6
一种美白组合物,由乳酸杆菌豆浆发酵产物滤液与白首乌提取物按照重量比1:0.25配制而成。
S21、将白首乌药材洗净,干燥,粉碎至80目,得到白首乌粉末;
S2、将白首乌粉末加入到去离子水中,进行水蒸气蒸馏得到油水混合物,取水相;所述白首乌粉末与去离子水的料液比为1g:12ml;
S3、将水相调节pH至6.4,以1.5BV/h上NKA-2大孔吸附树脂柱,洗脱速度为3BV/h,去离子水洗3BV,70wt%乙醇溶液洗3BV,收集乙醇溶液洗脱液,干燥,即得白首乌提取物。
对比例1
对比例1与实施例4不同之处在于,对比例1为单一的白首乌提取物,且所述白首乌提取物的制备方法与实施例4相同。
为了进一步证明本发明的效果,提供了以下测试方法:
1.美白实验:选择同一代对数生长期的B16细胞,用常规胰酶处理后,吹打成单细胞悬液,然后定量接种于96孔板。24小时后,弃去上清液,实验组分别加入含有测试的等量培养物(测试物分别为实施例1、4、5、6、对比例1所制备得到的乳酸杆菌、美白组合物、白首乌提取物),然后在37℃,5%的CO2饱和湿性条件下培养,对照孔加与实验组等量的RPMI1640培养液,培育三天后弃去上清液,用常规胰酶消化处理后,计数细胞密度,离心得到细胞沉淀,然后用PBS冲洗两次,离心弃去上清液,加入10%的DMSO的1N的NaOH液,80℃水浴50分钟,接着用酶标仪测定450nm处的吸光值。黑色素生成的抑制率(%)=(对照组的吸光度-试验组的吸光度/对照组的吸光度)×100%,具体结果见表1。

从表1中可看出,本发明所制备得到的乳酸杆菌豆浆发酵产物滤液、白首乌提取物具有良好的美白作用,且二者联用具有协同增效作用。

Claims (4)
1.一种美白组合物,其特征在于,由乳酸杆菌豆浆发酵产物滤液与白首乌提取物按照重量比1:0.2~0.5配制而成;
所述乳酸杆菌豆浆发酵产物滤液的制备方法为:
S11、将乳酸杆菌接种于固体培养基进行活化培养,活化后的乳酸杆菌再接种于液体培养基中培养,得到乳酸杆菌菌液;
S12、将3~6重量份豆浆粉、2~5重量份葡萄糖、1~3重量份蔗糖、0.4~1重量份酵母膏、0.02~0.1%重量份磷酸二氢钾、90~99重量份去离子水加入发酵罐中,105℃~121℃灭菌20~40min,然后冷却至25~40℃,得到豆浆液;
S13、将乳酸菌菌液接种于豆浆液中,接种量为5~15wt%,在25~40℃下发酵18~40h,过滤,经巴氏灭菌,得到乳酸杆菌豆浆发酵产物滤液;
所述白首乌提取物的制备方法为:
S21、将白首乌药材洗净,干燥,粉碎至60~100目,得到白首乌粉末;
S2、将白首乌粉末加入到去离子水中,进行水蒸气蒸馏得到油水混合物,取水相;所述白首乌粉末与去离子水的料液比为1g:8~15ml;
S3、将水相调节pH至6~6.8,以1~1.8BV/h上NKA-2大孔吸附树脂柱,洗脱速度为2~3BV/h,去离子水洗2~3BV,70~90wt%乙醇溶液洗2~3BV,收集乙醇溶液洗脱液,干燥,即得白首乌提取物。
2.根据权利要求1所述的美白组合物,其特征在于,所述乳酸杆菌为植物乳杆菌、德式乳杆菌、保加利亚乳杆菌、双歧杆菌、干酪乳杆菌、副干酪乳杆菌中的一种或多种。
3.根据权利要求1所述的美白组合物,其特征在于,所述固体培养基按重量百分比计,包括:0.5~1%酪蛋白酶消化物、0.5~1%牛肉膏、0.2~0.4%酵母膏、0.1~0.2%柠檬酸三铵、0.2~0.5%乙酸钠、0.02~0.05%硫酸镁、0.1~0.3%硫酸锰、0.1~0.2%磷酸二氢钾、1~3%葡萄糖、0.05~0.1%吐温-80、1~2%琼脂、余量去离子水。
4.根据权利要求1所述的美白组合物,其特征在于,所述液体培养基配方重量百分比计,包括:0.5~1%酪蛋白酶消化物、0.5~1%牛肉膏、0.2~0.4%酵母膏、0.1~0.2%柠檬酸三铵、0.2~0.5%乙酸钠、0.02~0.05%硫酸镁、0.1~0.3%硫酸锰、0.1~0.2%磷酸二氢钾、1~3%葡萄糖、0.05~0.1%吐温-80、余量去离子水。
