Abstract
本发明属于医用敷料的药物技术领域,具体涉及用于伤口皮肤修复的复方水凝胶贴片及其制备方法。本发明的制备方法包括如下步骤,(1)将明胶和单宁酸混合在去离子水中,明胶浓度为0.05~0.15g/ml,单宁酸浓度为5×10‑3g/ml,在45~50℃加热20~40分钟,产生最终明胶浓度为10%W/V的混合溶液;将pH值调节至10.0~11.0后加入熊果酸;(2)将获得的溶液在65~75℃下加热6小时,调节PH值到7.0~7.5,得到熊果酸水凝胶的酒红色透明前体溶液;(3)在45~50℃下剧烈搅拌下,向步骤(2)中加入10%W/V的NaIO4溶液,在45~50℃下搅拌混合物,发生溶胶‑凝胶转变,制得所述复方水凝胶贴片。本发明在外敷过程中发挥抗炎、抗氧化、促修复的作用,促进伤口加快恢复来缓解病人的痛苦。
Description
技术领域
本发明属于医用敷料的药物技术领域,具体涉及用于伤口皮肤修复的复方水凝胶贴片及其制备方法。
背景技术
据统计,我国超过2%的人经历着慢性创伤愈合的痛苦,全球每年用于伤口护理产品的费用高达150亿美元。传统伤口敷料如壳聚糖、透明质酸、藻酸盐和明胶等对慢性创面有一定的辅助治疗作用,但伤口的愈合程度仍主要依赖患者的自身修复能力,造成大量慢性难治性伤口因患者营养状态差、高龄、免疫力低下等原因而长期迁延不愈甚至导致死亡。因此,迫切需要研制出基于临床伤口护理需求的具有组织再生功能的新型敷料。
发明内容
根据现有技术上存在的缺陷,结合目前的研究前沿,本发明提供了涉及用于伤口皮肤修复的复方水凝胶贴片及其制备方法。
本发明是采用以下的技术方案实现的:
本发明提供了一种复方水凝胶贴片的制备方法,包括如下步骤,
(1)将明胶和单宁酸混合在去离子水中,明胶浓度为0.05~0.15g/ml,单宁酸浓度为5×10-3g/ml,在45~50℃加热20~40分钟,产生最终明胶浓度为10%W/V的混合溶液;将pH值调节至10.0~11.0后加入熊果酸;
(2)将获得的溶液在65~75℃下加热6小时,调节PH值到7.0~7.5,得到熊果酸水凝胶的酒红色透明前体溶液;
(3)在45~50℃下剧烈搅拌下,向步骤(2)中加入10%W/V的NaIO4溶液,在45~50℃下搅拌混合物,发生溶胶-凝胶转变,制得所述复方水凝胶贴片。
具体的,所述步骤(2)中熊果酸的终浓度为1×10-3~5×10-3g/ml。
其中,所述步骤(1)和步骤(2)中调节PH值的分别为氢氧化钠溶液和盐酸。
本发明还提供一种权上述方法制备的复方水凝胶贴片。
本发明还提供一种上述的复方水凝胶贴片的应用,所述复方水凝胶贴片应用于伤口皮肤修复。
与现有技术相比,本发明的有益效果:
糖尿病患者伤口难愈合的问题是临床上亟待解决的难题。本发明打破传统外敷材料的理念,将低成本、高治疗效果的中药加入外敷材料中,使其材料具有生物活性从而在外敷过程中发挥抗炎、抗氧化、促修复的作用,促进伤口加快恢复来缓解病人的痛苦。

具体实施方式
为了使本发明目的、技术方案更加清楚明白,下面结合附图,对本发明作进一步详细说明(如图1所示)。下述实施例中所述实验方法,如无特殊说明,均为常规方法;实施例中未注明具体技术或条件者,按照本领域内的文献所描述的技术或条件或者按照产品说明书进行;所述试剂和材料,如无特殊说明,均可从商业途径获得。
一、制备用于伤口皮肤修复的复方水凝胶贴片
实施例1
(1)将明胶(10克)和单宁酸(0.5克)混合在100毫升去离子水中,在50℃加热30分钟,产生最终明胶浓度为10%重量/体积的混合溶液。然后,使用5M氢氧化钠将pH值调节至11.0后加入0.5克熊果酸;
(2)将获得的溶液在70℃下加热6小时,用5M的盐酸调节PH值到7.5,得到熊果酸水凝胶的酒红色透明前体溶液;
(3)在50℃下剧烈搅拌下,将不同量的NaIO4溶液(10%,重量/体积)加入到前体溶液中。在50℃下进一步搅拌反应混合物,直到发生溶胶-凝胶转变。
二、效果检测
通过投射电子显微镜分别对实施例1制备的水凝胶进行形貌检测,从图2、图3可以看出,制备所得的水凝胶产生大量孔隙。
当然,以上仅为本发明的优选实施例而已,并不用于限制本发明,尽管参照前述实施例对本发明进行了详细的说明,对于本领域的技术人员来说,其依然可以对前述各实施例所记载的技术方案进行修改,或者对其中部分技术特征进行等同替换。凡在本发明的精神和原则之内,所作的任何修改、等同替换、改进等,均应包含在本发明的保护范围之内。
Claims (5)
1.一种复方水凝胶贴片的制备方法,其特征在于,包括如下步骤,
(1)将明胶和单宁酸混合在去离子水中,明胶浓度为0.05~0.15g/ml,单宁酸浓度为5×10-3g/ml,在45~50℃加热20~40分钟,产生最终明胶浓度为10%W/V的混合溶液;将pH值调节至10.0~11.0后加入熊果酸;
(2)将获得的溶液在65~75℃下加热6小时,调节PH值到7.0~7.5,得到熊果酸水凝胶的酒红色透明前体溶液;
(3)在45~50℃下剧烈搅拌下,向步骤(2)中加入10%W/V的NaIO4溶液,在45~50℃下搅拌混合物,发生溶胶-凝胶转变,制得所述复方水凝胶贴片。
2.根据权利要求1所述的制备方法,其特征在于,所述步骤(2)中熊果酸的终浓度为1×10-3~5×10-3g/ml。
3.根据权利要求2所述的制备方法,其特征在于,所述步骤(1)和步骤(2)中调节PH值的分别为氢氧化钠溶液和盐酸。
4.一种权利要求1-3中任一项所述方法制备的复方水凝胶贴片。
5.一种权利要求4所述的复方水凝胶贴片的应用,其特征在于,所述复方水凝胶贴片应用于伤口皮肤修复。
