本发明公开了一种含有干细胞提取液的面膜精华液及面膜。100mL的面膜精华液包括以下原料:干细胞提取液冻干粉50‑250mg,灯塔水母胶原蛋白冻干粉2‑7g,β烟酰胺单核苷酸200‑650mg,透明质酸2‑7g,辅酶Q1030‑120mg,褪黑素10‑100mg,新鲜酵母提取物100‑320mg,红花提取液4‑12mL,新鲜石斛提取液5‑20mL,植物精华素5‑20mL,表没食子儿茶素没食子酸酯1‑3g,其余为去离子水。本发明的面膜精华液具有良好的美容效果,制作成面膜具有良好的抗衰老性能,能够提高皮肤的水分和弹性,同时,改善皮肤的光泽。

本发明涉及一种含有干细胞提取液的面膜精华液及面膜,属于面膜技术领域。
背景技术
由于人面部表皮干细胞的减少,再生能力降低,表皮细胞的死亡脱落、功能减退等,随着日积月累皮肤出现斑点、皱纹增多等衰老的迹象。随着现在经济的发展,关于美容行业也是迎来了长足发展,各种抗衰产品受到广大爱美人士的青睐。
面膜是美容保养品的一种载体,面膜敷在脸上时,面膜里的水分和营养成分能把肌肤紧致包融,让水充分渗透入肌肤表皮的角质层,同时营养性或功效性物质会充分渗透进入皮肤的深层,软化皮肤,增强肌肤弹性,促进毛细血管扩张,清除毛孔的灰尘、化学污染物质、细菌和油脂污垢,加速皮肤深层的血液微循环,增强表皮各层细胞的活力,消除肌肤疲劳、衰老,维持年轻态。

为了克服现有技术的不足,本发明提供了一种含有干细胞提取液的面膜精华液及面膜。
本发明是通过以下技术方案来实现的:
一种含有干细胞提取液的面膜精华液,100mL的面膜精华液包括以下原料:干细胞提取液冻干粉50-250mg,灯塔水母胶原蛋白冻干粉2-7g,β烟酰胺单核苷酸200-650mg,透明质酸2-7g,辅酶Q10 30-120mg,褪黑素10-100mg,新鲜酵母提取物100-320mg,红花提取液4-12mL,新鲜石斛提取液5-20mL,植物精华素5-20mL,表没食子儿茶素没食子酸酯1-3g,其余为去离子水。

所述的一种含有干细胞提取液的面膜精华液,所述灯塔水母干细胞的提取液是通过以下步骤获得的:
(1)将活水螅清洁饲养24-48小时,用无菌生理盐水反复冲洗后,70%酒精消毒,在4℃无菌条件下去皮去触手,粉碎过滤,加细胞分离消化液和缓冲培养液后制成单细胞悬液,生理盐水离心洗剂3次,加适量培养基后,与带有选择标记的干细胞标志物抗体37℃二氧化碳孵箱中孵育45分钟,4℃冰浴过夜;
(2)用流式细胞仪对上述带有干细胞标记的细胞悬液进行干细胞分离,收集干细胞,4℃无菌条件下离心洗剂三次,去除标记物,加缓冲液制成细胞悬液;
(3)将所述细胞悬浮液加入预冷缓冲液中无菌条件下超声破碎细胞,4℃离心,收集上清液,测量蛋白质浓度,制成每ml含有1-2×108个细胞的干细胞提取物;
(4)将所得的干细胞提取物与白蛋白、生理盐水混合物、辅料混合均匀即得。
所述的一种含有干细胞提取液的面膜精华液,所述植物精华素为桑叶精华、玫瑰精华、金盏花精华和芦荟精华等量混合所得。
所述的一种含有干细胞提取液的面膜精华液,所述植物精华素是通过以下步骤获得的:将桑叶、玫瑰、金盏花和芦荟粉碎混合后采用70-80v/v%乙醇水溶液提取,得到提取液,将提取液浓缩后加入纯水,然后调节提取液的pH为6-7,调节温度为50-60℃,加入蛋白酶酶解45-60min;灭酶后经过膜过滤得到植物精华素。
所述的一种含有干细胞提取液的面膜精华液,所述新鲜酵母提取物是通过以下步骤获得的:对酵母细胞在温度为90-100℃条件下进行高温破壁处理,离心收集上清液,即为新鲜酵母提取物。
所述的一种含有干细胞提取液的面膜精华液,所述红花提取液是通过以下步骤获得的:将红花粉碎后采用70-80v/v%乙醇水溶液在温度为30-40℃条件下进行提取,得到乙醇提取液,将所述乙醇提取液浓缩后经过大孔树脂,采用80-90%的乙醇水溶液洗脱,浓缩后得到红花提取液。
所述的一种含有干细胞提取液的面膜精华液,所述新鲜石斛提取液是通过以下步骤获得的:将新鲜石斛破碎后置于水中在温度为90-100℃的条件下进行提取,得到水提液,将所述水提液浓缩后加入95v/v%乙醇水溶液溶解,经过HLB柱,采用乙醇洗脱,浓缩后得到新鲜石斛提取液。
一种含有干细胞提取液的面膜,采用权利要求1或2所述的含有干细胞提取液的面膜精华液,其特征是,用0.22μm无菌滤膜对面膜精华液进行除菌,超滤去色,加入消毒面膜纸后无菌封装,得到面膜。
干细胞提取液中的各种生长因子、基因调控因子等,能促进组织细胞重编程和逆分化及干细胞增生,能促进表皮细胞生长,毛囊细胞生发,真皮深层胶原蛋白表达,促进毛细血管增生,组织器官细胞和免疫细胞增生,全身细胞代谢活力增强,及抗衰老抗凋亡等作用。烟酰胺单核苷酸更有促进代谢和能源物质(ATP)生成等作用,具有促使组织活力旺盛之功效。

辅酶Q10除了具有改善代谢的作用外,具有强大的抗氧化作用,清除自由基,使细胞免受损害,保护细胞健康。皮肤的老化、皱纹的增加也与Q10含量有关,含量越低,皮肤越易老化,面部的皱纹也越多。辅酶Q10能增加皮肤中透明质酸的浓度,提高肌肤含水量,对改善暗沉肤色、减少皱纹、保持肌肤光滑、弹性和湿润性都有着良好的效果,从而增强角质细胞活力,减少细胞凋亡,让你更年轻。

本发明的面膜精华液采用干细胞提取液和β烟酰胺单核苷酸为主要原料,同时辅以灯塔水母胶原蛋白冻干粉、透明质酸,辅酶Q10,褪黑素,新鲜酵母提取物,红花提取液,新鲜石斛提取液和植物精华素,各原料协同作用,具有良好的美容效果,制作成面膜具有良好的抗衰老性能,能够提高皮肤的水分和弹性,同时,改善皮肤的光泽。

实施例1
一种含有干细胞提取液的面膜精华液,100mL的面膜精华液包括以下原料:干细胞提取液冻干粉50mg,灯塔水母胶原蛋白冻干粉2g,β烟酰胺单核苷酸200mg,透明质酸2g,辅酶Q10 30mg,褪黑素10mg,新鲜酵母提取物100mg,红花提取液4mL,新鲜石斛提取液5mL,植物精华素5mL,表没食子儿茶素没食子酸酯1g,其余为去离子水。
一种面膜,采用所述的面膜精华液,用0.22μm无菌滤膜对面膜精华液进行除菌,超滤去色,加入消毒面膜巾后无菌封装,得到面膜。
其中,所述灯塔水母干细胞的提取液是通过以下步骤获得的:

(4)将所得的干细胞提取物与白蛋白、生理盐水混合物、辅料混合均匀即得。

所述新鲜酵母提取物是通过以下步骤获得的:对酵母细胞在温度为90-100℃条件下进行高温破壁处理,离心收集上清液,即为新鲜酵母提取物。
所述红花提取液是通过以下步骤获得的:将红花粉碎后采用75v/v%乙醇水溶液在温度为35℃条件下进行提取,得到乙醇提取液,将所述乙醇提取液浓缩后经过大孔树脂,采用85%的乙醇水溶液洗脱,浓缩后得到红花提取液。其中,75v/v%乙醇水溶液的重量为红花的1-2倍。
所述新鲜石斛提取液是通过以下步骤获得的:将新鲜石斛破碎后置于水中在温度为95℃的条件下进行提取,得到水提液,将所述水提液浓缩后加入95v/v%乙醇水溶液溶解,经过HLB柱,采用乙醇洗脱,浓缩后得到新鲜石斛提取液。其中,新鲜石斛和水的重量比为1:2。
实施例2
所述的一种含有干细胞提取液的面膜精华液,100mL的面膜精华液包括以下原料:干细胞提取液冻干粉150mg,灯塔水母胶原蛋白冻干粉4.5g,β烟酰胺单核苷酸425mg,透明质酸4.5g,辅酶Q10 60mg,褪黑素50mg,新鲜酵母提取物160mg,红花提取液10mL,新鲜石斛提取液8mL,植物精华素10mL,表没食子儿茶素没食子酸酯2g,其余为去离子水。
一种面膜,采用所述的面膜精华液,用0.22μm无菌滤膜对面膜精华液进行除菌,超滤去色,加入消毒面膜巾后无菌封装,得到面膜。
所述的一种含有干细胞提取液的面膜精华液,所述干细胞提取液指灯塔水母干细胞的提取液。
干细胞是一种具有再生和自我更新的细胞,即具有增殖、自我修复、大量制造及分化成后代能力的细胞。对于人体坏死或病变组织的修复,对老化凋亡细胞的替换都具有重要意义。但人体干细胞培养技术复杂,费用昂贵,做一次治疗动辄几十万元,但动植物的干细胞,特别是灯塔水母干细胞海水即可养殖,费用低廉,可大量养殖,广泛使用,真正使干细胞的好处进到寻常百姓家。
本实施例所用的灯塔水母(Turritopsis nutricula)干细胞是从其幼年阶段生命形式水螅(Hydra)中提取干细胞(见申请人专利:CN105920044B,一种水螅干细胞制剂及其制备方法与应用)。
灯塔水母水螅属腔肠动物,其身体主要由干细胞构成。目前研究发现,灯塔水母天生具有“返老还童”的能力,是现今世界上唯一不死的生物。灯塔水母“返老还童”是通过细胞转分化过程实现的,即从老年转化为幼年类型。所以说,灯塔水母的生命是没有终结的。灯塔水母在死前回到了生命的开端,重新演绎了自己的生长发育。
灯塔水母之所以永生,是因为它有再生基因。如果将这种能够使衰老细胞逆分化成为干细胞的基因及其调节分化的细胞因子,通过面膜精华液,从皮肤毛囊吸收进入皮下组织细胞,就可调节人体细胞基因逆分化,使人类细胞年轻化成为干细胞,就可以使面部皮肤返老还童,使人脸如同回到少年时代,使人更年轻漂亮。
灯塔水母是极富含胶原蛋白的生物。灯塔水母胶原蛋白的价值,更是难以比拟。让我们听听科学家的研究结论。世界著名的意大利莱切萨伦托大学的海洋生物学家皮莱诺教授(Prof.Piraino,a biologist at the University of Salento in Lecce,Italy)说,已经证明水母对我们很有用(从药物到营养药品甚至健康的美味都可利用)。以胶原蛋白为例,它是人类结缔组织的主要结构蛋白,也可以用作骨移植和整形手术(医美)的支架来植入人体。但并不是所有类型的动物胶原蛋白都与所有类型的人类细胞相容。这是因为水母的胶原蛋白是原始的,与多种人类细胞类型兼容(jellies'collagen is primitive andcompatible with a wide variety of human cell types)。
他说,如果你把胶原蛋白分解成小块,它就会起到抗氧化剂的作用。因为它有助于吸收一种叫做自由基的化学物质,而自由基会破坏细胞(he says that if you breakcollagen into smaller pieces it acts as an antioxidant,a molecule that isdeemed healthy because it helps soak up chemicals called radicals that candamage cells.)。
他还从水母中分离出一种化合物似乎有抗癌活性。他现在正在研究这种化合物是否对一系列不同的癌症有效(that appears to have anticancer activity.He is nowworking to find out whether the compound is effective against a range ofdifferent cancers.)。

实施例3
一种含有干细胞提取液的面膜精华液,100mL的面膜精华液包括以下原料:干细胞提取液冻干粉250mg,灯塔水母胶原蛋白冻干粉7g,β烟酰胺单核苷酸650mg,透明质酸7g,辅酶Q10 120mg,褪黑素100mg,新鲜酵母提取物320mg,红花提取液12mL,新鲜石斛提取液20mL,植物精华素20mL,表没食子儿茶素没食子酸酯3g,其余为去离子水。
一种含有干细胞提取液的面膜精华液,100mL的面膜精华液包括以下原料:干细胞提取液冻干粉200mg,灯塔水母胶原蛋白冻干粉6g,β烟酰胺单核苷酸600mg,透明质酸6g,辅酶Q10 100mg,褪黑素200mg,新鲜酵母提取物300mg,红花提取液10mL,新鲜石斛提取液10-20mL,植物精华素10-20mL,表没食子儿茶素没食子酸酯2g,其余为去离子水。
一种面膜,采用所述的面膜精华液,用0.22μm无菌滤膜对面膜精华液进行除菌,超滤去色,加入消毒面膜巾后无菌封装,得到面膜。
其中,所述灯塔水母干细胞的提取液是通过以下步骤获得的:
(1)将活水螅清洁饲养24-48小时,用无菌生理盐水反复冲洗后,70%酒精消毒,在4℃无菌条件下去皮去触手,粉碎过滤,加细胞分离消化液和缓冲培养液后制成单细胞悬液,生理盐水离心洗剂3次,加适量培养基后,与带有选择标记的干细胞标志物抗体37℃二氧化碳孵箱中孵育45分钟,4℃冰浴过夜;
(2)用流式细胞仪对上述带有干细胞标记的细胞悬液进行干细胞分离,收集干细胞,4℃无菌条件下离心洗剂三次,去除标记物,加缓冲液制成细胞悬液;
(3)将所述细胞悬浮液加入预冷缓冲液中无菌条件下超声破碎细胞,4℃离心,收集上清液,测量蛋白质浓度,制成每ml含有1-2×108个细胞的干细胞提取物;
(4)将所得的干细胞提取物与白蛋白、生理盐水混合物、辅料混合均匀即得。
所述植物精华素是通过以下步骤获得的:将桑叶、玫瑰花、金盏花及芦荟等量混合粉碎后采用乙醇提取,得到提取液,将提取液浓缩后加入纯水,然后调节提取液的pH和温度,加入蛋白酶酶解45-60min;灭酶后经过膜过滤得到植物精华素。
其中,乙醇的重量为桑叶、玫瑰花、金盏花及芦荟重量的1-2倍,所述提取液的pH为6,酶解温度为50-60℃,加入木瓜蛋白酶和胰蛋白酶。

所述新鲜石斛提取液是通过以下步骤获得的:将新鲜石斛破碎后置于水中在温度为95℃的条件下进行提取,得到水提液,将所述水提液浓缩后加入95v/v%乙醇水溶液溶解,经过HLB柱,采用乙醇洗脱,浓缩后得到新鲜石斛提取液。其中,新鲜石斛和水的重量比为1:2。
实验方案及结果:
随机选取80名健康女志愿者,志愿者年龄控制为35-55岁,并随机将80名志愿者对应分成4组,每组20人,其中三组志愿者对应采用以上实施例的面膜,第四组志愿者采用5v/v%甘油水溶液敷脸,且四组志愿者均在每周一、周三、周五晚上八点敷脸15min,坚持使用28天。
对志愿者做以下检测:
使用专门的皮肤弹性(含水量)测试仪来测量受试者面部皮肤的弹性和含水量。同时对肤质进行检测。根据皮肤在短时间内恢复的状况来评定弹性的大小,弹性值1为最大,即皮肤100%恢复原状。结果见表1。
表1各组志愿者使用前和使用28天后的皮肤状态
由表1结果可知,与对照组相比,本发明的面膜在抗皱与保湿水平已经达到显著效果,在使用本发明实施例1-3的面膜后,增加了皮肤的水分含量,有效减缓了皱纹的产生,而且肌肤新生有活力,肤色光亮透晰,皮肤弹性增强,此外修复效果随时间增长而越发明显。
以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明技术原理的前提下,还可以做出若干改进和变形,这些改进和变形也应视为本发明的保护范围。
Claims (10)
1.一种含有干细胞提取液的面膜精华液,其特征是,100mL的面膜精华液包括以下原料:干细胞提取液冻干粉50-250mg,灯塔水母胶原蛋白冻干粉2-7g,β烟酰胺单核苷酸200-650mg,透明质酸2-7g,辅酶Q1030-120mg,褪黑素10-100mg,新鲜酵母提取物100-320mg,红花提取液4-12mL,新鲜石斛提取液5-20mL,植物精华素5-20mL,表没食子儿茶素没食子酸酯1-3g,其余为去离子水。
2.根据权利要求1所述的一种含有干细胞提取液的面膜精华液,其特征是,100mL的面膜精华液包括以下原料:干细胞提取液冻干粉200mg,灯塔水母胶原蛋白冻干粉6g,β烟酰胺单核苷酸600mg,透明质酸6g,辅酶Q10100mg,褪黑素200mg,新鲜酵母提取物300mg,红花提取液10mL,新鲜石斛提取液10-20mL,植物精华素10-20mL,表没食子儿茶素没食子酸酯2g,其余为去离子水。
3.根据权利要求1或2所述的一种含有干细胞提取液的面膜精华液,其特征是,所述干细胞提取液指灯塔水母干细胞的提取液。
4.根据权利要求3所述的一种含有干细胞提取液的面膜精华液,其特征是,所述灯塔水母干细胞的提取液是通过以下步骤获得的:
(1)将活水螅清洁饲养24-48小时,用无菌生理盐水反复冲洗后,70%酒精消毒,在4℃无菌条件下去皮去触手,粉碎过滤,加细胞分离消化液和缓冲培养液后制成单细胞悬液,生理盐水离心洗剂3次,加适量培养基后,与带有选择标记的干细胞标志物抗体37℃二氧化碳孵箱中孵育45分钟,4℃冰浴过夜;
(2)用流式细胞仪对上述带有干细胞标记的细胞悬液进行干细胞分离,收集干细胞,4℃无菌条件下离心洗剂三次,去除标记物,加缓冲液制成细胞悬液;
(3)将所述细胞悬浮液加入预冷缓冲液中无菌条件下超声破碎细胞,4℃离心,收集上清液,测量蛋白质浓度,制成每ml含有1-2×108个细胞的干细胞提取物;
(4)将所得的干细胞提取物与白蛋白、生理盐水混合物、辅料混合均匀即得。
5.根据权利要求1或2所述的一种含有干细胞提取液的面膜精华液,其特征是,所述植物精华素为桑叶精华、玫瑰精华、金盏花精华和芦荟精华等量混合所得。
6.根据权利要求4所述的一种含有干细胞提取液的面膜精华液,其特征是,所述植物精华素是通过以下步骤获得的:将桑叶、玫瑰、金盏花和芦荟粉碎混合后采用70-80v/v%乙醇水溶液提取,得到提取液,将提取液浓缩后加入纯水,然后调节提取液的pH为6-7,调节温度为50-60℃,加入蛋白酶酶解45-60min;灭酶后经过膜过滤得到植物精华素。
7.根据权利要求1或2所述的一种含有干细胞提取液的面膜精华液,其特征是,所述新鲜酵母提取物是通过以下步骤获得的:对酵母细胞在温度为90-100℃下进行高温破壁处理,离心收集上清液,即为新鲜酵母提取物。
8.根据权利要求1或2所述的一种含有干细胞提取液的面膜精华液,其特征是,所述红花提取液是通过以下步骤获得的:将红花粉碎后采用70-80v/v%乙醇水溶液在温度为30-40℃条件下进行提取,得到乙醇提取液,将所述乙醇提取液浓缩后经过大孔树脂,采用80-90%的乙醇水溶液洗脱,浓缩后得到红花提取液。
9.根据权利要求1或2所述的一种含有干细胞提取液的面膜精华液,其特征是,所述新鲜石斛提取液是通过以下步骤获得的:将新鲜石斛破碎后置于水中在温度为90-100℃的条件下进行提取,得到水提液,将所述水提液浓缩后加入95v/v%乙醇水溶液溶解,经过HLB柱,采用乙醇洗脱,浓缩后得到新鲜石斛提取液。
10.一种含有干细胞提取液的面膜,采用权利要求1-9任一权利要求所述的面膜精华液,其特征是,用0.22μm无菌滤膜对面膜精华液进行除菌,超滤去色,加入消毒面膜纸后无菌封装,得到面膜。
