技术领域
本发明属于化学合成物技术领域,涉及一种texaphyrin-叶酸螯合物及其制备方法与应用。
背景技术
当人们试图选择性地靶向治疗癌性病变以实现更高的疗效时,肿瘤治疗剂和显像剂面临相当大的障碍。迄今为止,人们已在癌症肿瘤上鉴定出多种过度表达受体,为设计具有更高选择性和特异性的肿瘤药物提供了宝贵的靶标。
叶酸受体(folate receptor)是一个显著的靶标,已被证明在多种癌细胞系中过度表达。[1]以此为基础,已开发出多种显像剂和治疗剂,针对单一化疗或显像剂中的多种癌症类型。[2]尽管这是个有价值的目标,但FDA尚未批准使用叶酸为靶向癌症治疗剂或显像剂。
特沙弗林(Texaphyrin)能够在“扩大型” 5配位袋位中结合金属。它可与金属阳离子形成24种不同的稳定的1:1配合物的能力已被证明,在癌症治疗和成像中的应用中也有应用证明。Texaphyrins在近红外范围(700-900 nm)的射线有强烈吸收,因此它可以独特地提供体内激发。另外,Texaphyrins在人类受试者中表现出对肿瘤的选择性和人体毒性可耐受的特征。此外,改变Texaphyrin核心中的中心配位金属可以调整Texaphyrin的功能。人们已经进行了将Texaphyrins掺入纳米颗粒的努力,例如专利公开号为CN107735402B的中国发明专利于2016年4月14日公开的一种德克萨卟啉-磷脂缀合物,包含与磷脂的脂质侧链共价连接的德克萨卟啉、德克萨卟啉衍生物或德克萨卟啉类似物;又例如将for-texaphyrin(GdTx)包裹在用于体内MRI的纳米颗粒以及热疗剂中。迄今为止,尚无Texaphyrin-叶酸螯合物(Texaphyrin-folate conjugate)的报道。
发明内容
本发明一方面,提供了一种Texaphyrin-叶酸盐螯合物(Texaphyrin-folateconjugate),其包含Texaphyrin,Texaphyrin衍生物或Texaphyrin同系物以共价键与叶酸或叶酸衍生物的O-(烷基氨基)二氨基苯酚衍生物结合的螯合物。
另一方面,提供了一种纳米颗粒,其包含上述的Texaphyrin-叶酸盐螯合物。
进一步地,本发明特别提供了制备Texaphyrin-叶酸盐螯合物的方法,尤其是制备邻苯二酚叶酸和1:1稳定金属络合物的方法。这些金属包括:锰、铁、钴、锌、钇、锝、镉、铟、铋、钐、铕、钆、铽、镝、钬、铒、铥、镱和镥。
再一方面,提供了本发明所述的Texaphyrin-叶酸盐螯合物或纳米颗粒用于治疗的应用,包括放射增敏,放射疗法,光热疗法。还可用于医学成像,包括MRI,双光子,SPECT,PET,CT,荧光和超声。
本发明在此首次提出了该化合物在癌症成像和治疗中的设计与合成。鉴于叶酸受体在多种癌细胞系中的高度过度表达,再加上Texaphyrin在肿瘤治疗和影像学领域的出色表现,相比其他叶酸螯合体尔言,texaphyrin-叶酸螯合物提供了一种肿瘤特异性药物。

本发明描述了新的靶向肿瘤的分子体,Texaphyrin-叶酸同系物(Texaphyrin-folate analogue)的制备。尽管先前已经使用许多分子来装饰Texaphyrin,但是本发明利用合理的合成设计展示了一类新型的化合物。
在现有技术中人们已经注意到,二氨基苯酚提供了得到功能化Texaphyrin结构的有效方法。

Texaphyrin-叶酸结合物的合成路线如附图1所示,具体如下:
将4-(4-溴丁氧基)-1,2-二硝基苯(3):1,4-二溴丁烷(30 mmol)添加到1(2.07mmol),无水K2CO3(16.2 mmol)和KI(2.07 mmol)的混合物中,在干燥的DMF(110毫升)中。在80℃下搅拌2小时后,加入水(150mL),并用乙酸乙酯(3×35mL)萃取。 合并有机层,用水(35mL),饱和碳酸氢钠溶液(2×25mL),盐水(25mL)洗涤,经MgSO4干燥,过滤,并真空浓缩,得到3。

将4-(3,4-二硝基苯氧基)丁-1-胺(5):4(0.054mmol)加入到THF(10mL)和水合肼(0.5mL)的溶液中。将混合物在75℃下搅拌6h,然后减压除去溶剂。将残留物溶于最少量的二氯甲烷中,然后通过柱色谱纯化,得到5。
N2-(4-((((2-氨基-4-氧代-3,4-二氢蝶呤-6-基)甲基)氨基)苯甲酰基)-N5-(4-(3,4-二硝基苯氧基)丁基)谷氨酰胺(6 ):将叶酸(0.189mmol),DCC(0.192mmol)和HONb(0.192mmol)加入到DMF(8mL)的溶液中。将混合物在室温下在黑暗中搅拌16小时。将固体过滤后,将5(0.189mmol)加入到滤液中。在环境温度下搅拌24小时后,在剧烈搅拌下将混合物逐渐倒入无水Et 2 O(20mL)中,并冷却至0℃。通过过滤收集沉淀物,用Et 2 O洗涤,在高真空下干燥,得到化合物6。
N2-(4-((((2-氨基-4-氧代-3,4-二氢蝶呤-6-基)甲基)氨基)苯甲酰基)-N5-(4-(3,4-二氨基苯氧基)丁基)谷氨酰胺(7 ):向RBF中添加6(0.859 mmol),并将钯碳(91.5mg,85.9μmol)溶于10 mL无水甲醇中,然后添加浓HCl(213.8μL,2.58 mmol)。然后将反应容器密封,并用氢气气氛吹扫几次,以活化钯催化剂。一旦完成,将反应容器在室温下在氢气氛下放置3小时。完成后,通过LCMS分析确认,将混合物过滤,并真空浓缩,得到7。
N2-(4-((((2-amino-4-oxo-3,4-dihydropteridin-6-yl)methyl)氨基)苯甲酰基)-N5-(4-((((6E,9E)-13,33, 34,53-四乙基-14,54-二甲基-31H-11l2,51l2-1l2,51l2,7,9-四氮杂-1,3,5( 2,5)-三吡咯烷-8(1,2)-苯并环环癸烷-6,9-二烯-84-基)氧基)丁基)谷氨酰胺(9):向反应容器中添加7(0.538 mmol)和三吡喃二醛( 0.538 mmol)溶于6 mL无水甲醇中,然后加入浓HCl(89.2 µL,1.08 mmol)。然后在避光的同时将反应容器加热至50°C 30分钟。完成后,将溶剂减压完全除去。然后将残余物用己烷(5 x 10 mL)洗涤,除去疏水性彩色杂质,得到8,为深红色固体。
代表性的螯合合成,钆(III)络合物10:向反应容器中加入8(34.02 02mol),水合乙酸g(III)(35.73μmol)和三乙胺(47.43μL,340.25μmol),在4 mL中加入甲醇,并在60°C下在空气中搅拌2小时。完成后,将溶剂完全除去,并将己烷(3 x 10 mL)添加到残留物中,除去浅粉红色杂质,得到9(75%,25.5μmol),为深绿色固体。
镥(III)络合物:向反应容器中加入8 mL(23μmol),水合乙酸镥(III)(24.14μmol)和三乙胺(229μmol)的5 mL甲醇溶液,搅拌至在52℃下通风1.5小时。完成后,将溶剂完全除去,并将己烷(3 x 10 mL)添加到残留物中,除去浅粉红色杂质,得到呈绿色固体的镥(II)络合的Texaphyrin-叶酸酯:14.5μmol,收率63%。
铁(II)络合物:在5 mL甲醇中,向反应容器中添加8(15.63μmol),乙酸铁(II)(16.41μmol)和三乙胺(156.33μmol),并搅拌至空气中在52℃下放置1.25小时。完成后,将溶剂完全除去,并将己烷(3 x 10 mL)添加到残留物中,除去浅粉红色杂质,得到铁(II)络合的草绿色Texaphyrin-叶酸盐,为深绿色固体:10.8μmol,收率69%。
钴(II)配合物:向反应容器中加入在5 mL甲醇中的8(13.79 μmol),乙酸钴(II)(14.48 μmol)和三乙胺(137.94μ mol),并搅拌至空气中在52℃下放置45分钟。完成后,将溶剂完全除去,并将己烷(3 x 10 mL)添加到残留物中,除去浅粉红色杂质,得到深绿色固体形式的钴(II)络合的Texaphyrin-叶酸盐:11.2μmol,81%收率。
锰(II)配合物:向反应容器中加入在5 mL甲醇中的8(15.81μmol),乙酸锰(II)(16.60μmol)和三乙胺(158.13μmol),并在空气中搅拌开放在52℃下1.5小时。完成后,将溶剂完全除去,并将己烷(3 x 10 mL)添加到残余物中,除去浅粉红色杂质,得到钴(II)络合的Texaphyrin-叶酸,为深绿色固体:12.1μmol,收率76%。
镓(III)配合物:在5 mL甲醇中向反应容器中添加8(15.81μmol),乙酸镓(III)(16.60μmol)和三乙胺(158.13μmol),并搅拌至空气中在52 ℃下1.5小时。完成后,将溶剂完全除去,并将己烷(3 x 10 mL)添加至残留物中,除去浅粉红色杂质,得到镓(III)络合的草绿色Texaphyrin-叶酸酯,为深绿色固体:11.4μmol,收率72%。
锰(II)络合物,钇(III)络合物,镉(III)络合物,铟(III)络合物,铋(III)络合物,钐(III)络合物,铕(III)络合物,钬(III)络合物,铒( III)络合物,钍(III)络合物,镱(III)络合物和铼(II)络合物,镓(III)络合物,锆(II)络合物和铷(I)络合物。可以注意到未复合的碳11,氮13,氧15和氟18衍生物可以进行双重PET分析。
可能的Texaphyrin-叶酸共轭物螯合库如图2所示。 Gd-texaphyrin的MRI能力
Texaphyrins具有独特而强大的螯合能力,可与多种金属离子形成稳定的1:1络合物。有了这种能力,Texaphyrin金属螯合物在磁共振成像(MRI)中的能力已被评估。首先被检查的具有MRI活性的Texaphyrin是Motexafin(Gd-texaphyrin),其在临床前和临床阶段试验中显示MRI对比增强。1-2这些评估在临床癌症模型中利用Gd-texaphyrin作为放射增敏剂,并且同时通过MRI检测肿瘤剂量反应。 Gd-texaphyrin独特且有利的内在特性已导致其进入针对于非小细胞肺癌衍生脑转移患者的III期临床试验。尽管与单独放疗相比,接受Gd-texaphyrin和全脑放射治疗的患者神经功能的进展有所延迟,但FDA并未批准Gd-texaphyrin。3另一种用于MRI对比增强评估的顺磁性Texaphyrin是Mn-nanotexaphyrin。新型的自组装两亲性Texaphyrin-磷脂缀合物,产生出了一个基于有机物的多功能纳米颗粒。4作者选择研究一种基于顺磁性的Mn-texaphyrin的试剂,因为它具有同时降低T1和T2的能力和组织中的T2松弛常数。 然后在带有颈淋巴结转移的VX-2头颈肿瘤兔体内评估Mn–nanotexaphyrin。为评估其在淋巴造影术中增强造影剂的能力,在肿瘤近端部位皮下注射了Mn–nanotexaphyrin,经过2小时T1和T2加权成像,旨在增强从肿瘤部位转移至转移灶的淋巴引流的可视化疾病淋巴结。 成像结果显示,从肿瘤部位到相邻的转移性淋巴结的淋巴引流的可视性大大增强。 概念验证工作表明,Mn-nanotexaphyrin可作为前哨淋巴结(SLNs)活检程序的MRI造影剂。 这些报告证明了Texaphyrins用作顺磁性金属离子的热力学和动力学稳定大环螯合剂的能力,以增强MRI的对比度。
体内MRI:所有动物均按照组织动物保护委员会,安大略省研究用动物法和加拿大动物保护委员会制定的政策接受了人类护理。所有动物研究均已通过ORGANIZAION动物保健委员会的协议批准。将重量为2.0至3.5 kg的雄性新西兰白兔(Charles River,美国马萨诸塞州威尔明顿)注射300μLVX-2肿瘤细胞悬液(5 x 106 cfu / mL),以诱导肿瘤进入鼓泡肌。接种后两周,在VX2细胞注射部位形成了肿瘤,所有兔子均出现了至少一个颈淋巴结转移。向一只通过CT成像确认的淋巴结转移的兔子皮下注射1.5 mL 8 mg / mL的Ga-texaphyrin-叶酸,皮下围绕肿瘤区域(类似于使用tech 99的前哨淋巴结活检(SLNB)程序)。注射后2小时后,在7特斯拉临床前MR成像系统(Biospec,公司,德国埃特林根)上进行MRI。对于兔子图像,成像使用B-GA20S梯度线圈和Bruker制造的内径为15.5 cm的正交圆柱RF体积线圈。将兔子定向在有机玻璃支架内的腹卧位置,并首先将其首先插入孔中。使用MR兼容系统将麻醉直接传送到鼻锥。在基线进行冠状和轴向T2加权成像,在皮下注射造影剂后2小时进行冠状和轴向T2加权和T1加权成像。所有采集都共享1x1x1.5 mm的空间分辨率。冠状动脉采集具有110x80毫米的视野,110x80的矩阵尺寸和36个切片覆盖范围。轴向采集具有96x72 mm的视野,96x72 mm的矩阵尺寸和55个切片覆盖范围。所有T2加权图像集均以66.5毫秒的有效回波时间(回波时间9.5毫秒;稀有因子14)和重复时间6000毫秒获得。所有T1加权图像集均以9.5毫秒的有效回波时间(回波时间9.5毫秒;稀有因子2)和1000毫秒的重复时间获得。采集时间从冠状72加权成像的4分24秒到轴向r2加权成像的7分30秒不等。

将Mn-texaphyrin-叶酸盐(10 mg / kg)静脉注射到原位PC3前列腺癌小鼠模型(n= 6)中,并评估T1和T2成像的MRI对比增强。 Mn-texaphyrin叶酸使能的T1和T2加权成像的MRI显示,造影剂给药后信号明显增强。与预扫描图像相比,注射后30分钟的T1和T2加权图像显示信号改善,边界清晰,可以可视化肿瘤组织结构。为了评估Mn-texaphyrin-叶酸盐作为MRI造影剂,还获取了T1和T2图。该图表明在45分钟时在肿瘤的外围区域中T1和T2值降低,并且保留了造影剂。这些数据共同表明,Mn-texaphyrin-叶酸盐有必要进一步发展为MRI造影剂。
MR成像使用B-GA12梯度线圈和Bruker制造的内径为35 mm的正交圆柱射频(RF)体积线圈。将小鼠俯卧在塑料支架内,头先插入孔中。麻醉(1.8%异氟醚)直接递送至鼻锥。成像包括基线,注射后15、45、90和120分钟的疗程。每个会话提供轴向T2加权和T1加权图像集以及轴向定量T2和T1映射采集,分别需要4分钟,4分钟36 s,14分钟57 s和16分钟40 s。常用参数包括0.25 mm x 0.25 mm×1.0 mm空间分辨率,120×92矩阵尺寸和30 mm×23 mm视场。分别以48和10毫秒的有效回波时间(RARE因子分别为12和2,在8毫秒的公共回波时间)以及4000和1000毫秒的重复时间获取T2和T1加权图像集。定量T2映射使用了多重自旋回波技术(从12–384 ms进行32次回波; 12 ms重新聚焦间隔)。定量T1映射使用可变重复时间方法(RARE因子2;七个重复时间400、800、1200、1600、2000、3500、5000 ms)。 T2和T1地图生成使用了在线布鲁克处理。
实施例2
原位PC3前列腺肿瘤模型:无胸腺裸雄鼠购自Harlan实验室。 在腹部切入腹膜5毫米长; 从腹腔部分切除膀胱和精囊,以暴露前列腺背叶。 注射5×105个PC-3M-luc-C6细胞(表达Lucifer的细胞系,Caliper)
使用28号针头从原位到前列腺背叶。 将器官放回腹腔,并缝合肌肉壁和皮肤。 皮下注射0.05 mg kg-1进行镇痛。 通过MRI(Biospec 70/30 USR,Bruker)监测原位前列腺肿瘤的生长。

5.根据权利要求1所述的一种texaphyrin-叶酸螯合物,其特征在于,与选自锰、铁、钴、锌、钇、锝、镉、铟、铋、钐 、铕、钆、铽、镝、钬、铒、铥、镱和镥的金属络合。
6.一种纳米颗粒,其特征在于,包含权利要求1-5任一项所述texaphyrin-叶酸螯合物。
8.如权利要求1-6任一项所述的一种texaphyrin-叶酸螯合物或者纳米颗粒的应用,用于放射增敏、放射疗法或光热疗法中。
