Abstract
本发明涉及一种具有增强男性性功能功能的食品组合物及其制备方法,包括以下组分及其质量份数:牡蛎低聚肽1~6份、甲鱼低聚肽1~5份、黑番茄提取物2~5份、青钱柳提取物0.5~4份、可乐果提取物0.1~0.6份以及番红花提取物0.01~0.3份。本发明的具有增强男性性功能的食物组合物采用的上述六种天然来源的药食同源的食品级原料,安全无毒副作用,不会产生依耐性以及抗药性,也不会在体内蓄积,能从细胞、组织和代谢水平上协同作用和增效,显著提高男性性功能。并且,分别采用闪式提取协同膜分离法以及减压提取法对不同组分进行提取制备,能有效减少对活性成分的破坏,提高提取物得率,最大限度地保留各主要活性成分,从而能在减少单份服用量的同时,精确定量摄入。

技术领域
本发明涉及功能食品技术领域,尤其涉及一种具有增强男性性功能的食品组合物及其制备方法。
背景技术
目前,随着生活节奏的加快以及生活压力的增大,我国男性的性功能状况不容乐观,男性性功能低下的发生率越来越高,有报道显示,我国40岁以上人群中,高达52.5%的男性忍受着性功能低下的折磨,尤其是近几年男性的阳痿、早泄、不育等疾病发生率明显增加,严重影响患者身心健康。
性功能是生活质量必要且重要的组成部分,男性性功能低下的发生率升高可由多种因素引起,如社会、环境、疾病、心理、年龄等各方面的原因,临床表现为性欲低下,血中雄性激素(主要为睾酮)水平明显下降,精液中精子减少及活力低下。其中,因年龄增长导致的雄性激素水平降低几乎是不可避免的:男性在40岁之后其血中睾酮水平每年约下降1%~2%,到45~75岁时血中睾酮水平可比年轻时下降60%,而当血中睾酮水平低于3μg/L时,即可诊断为男性性功能低下。
随着社会的发展和医疗水平的进步,性功能低下问题日益受到人们的重视,虽然九成以上的男性普遍认为性功能低下影响他们的男性气概,然而到医院就诊的不足一成,更多的患者则是自行选用一些延时性产品,如他非类或中药类产品。他非类产品能够有效抑制5型磷酸二酯酶的活性,从而使一氧化氮能充分扩张阴茎海绵体的血管平滑肌,进而带来长达半小时的勃起,这种勃起也不仅限于阴茎,比如鼻粘膜也会因此充血,给服用者带来闭塞的症状,消化道充血也会带来消化不良,此外还会带来头痛(最常见)、面部潮红(常见)、心衰、低血压、皮疹等一系列副作用,严重危害男性健康。
另外,也有很大一部分性功能低下人群倾向于服用中药类产品,目前该类中药类产品主要以鹿鞭、海狗鞭、牛鞭、鹿茸等动物器官和/或蛤蚧、海马、蚕蛹等动物本身和/或淫羊藿、肉苁蓉、玛咖、锁阳、蛇床子、阳起石、巴戟天、东革阿里、冬虫夏草、人参、菟丝子、黄精、枸杞子等药材之间的配伍组合。例如:申请号为CN202011159692.6(公开号为CN112043775A)的中国发明专利《种益肾壮阳用的中药组合物及其制备方法》、申请号为CN202010612940.1(公开号为CN111821377A)的中国发明专利《一种能改善男性性功能的金龟三鞭酒及其制备方法》、申请号为CN201911404514.2为(公开号CN111012839A)的中国发明专利《一种健身补肾药酒及其制备方法》、申请号为CN201510557121.0(公开号为CN105079183A)的中国发明专利《改善性功能障碍的玛咖组合物》等专利均公开了该类产品。上述专利虽然均宣称具有改善男性性功能的作用,但是所用的部分药物具有一定的毒性作用,长期服用可能会带来肝肾功能的损伤以及外源性激素摄入引起的内分泌紊乱,对男性健康造成影响。
发明内容
本发明所要解决的第一个技术问题是针对现有技术而提供一种具有增强男性性功能的食品组合物,该食品组合物安全有效、无毒副作用,并且不用产生依赖性且不会在人体体内蓄积。
本发明所要解决的第二个技术问题是针对现有技术而提供一种上述食品组合物的制备方法,该制备方法提取物得率高,并能最大程度地保留各主要活性成分。
本发明解决上述第一个技术问题所采用的技术方案为:一种具有增强男性性功能功能的食品组合物,其特征在于,包括以下组分及其质量份数:
牡蛎低聚肽1~6份、甲鱼低聚肽1~5份、黑番茄提取物2~5份、青钱柳提取物0.5~4份、可乐果提取物0.1~0.6份以及番红花提取物0.01~0.3份。
进一步,优选地,上述具有增强男性性功能功能的食品组合物包括以下组分及其质量份数:
牡蛎低聚肽4份、甲鱼低聚肽3份、黑番茄提取物3.5份、青钱柳提取物2.8份、可乐果提取物0.3份以及番红花提取物0.1份。
进一步,所述牡蛎低聚肽及甲鱼低聚肽中分子量小于1000D的低聚肽成分分别占95%以上,而分子量处于180~500D的低聚肽成分分别占65%以上,从而能进一步抵御胃肠道对上述牡蛎低聚肽及甲鱼低聚肽的消化。
进一步,所述番红花提取物中番红花总苷含量高于30%,上述青钱柳提取物中多糖含量高于35%,上述可乐提取物中天然咖啡因含量高于10%。
进一步,所述食品组合物包括可接受的食用辅料。从而有利于采用常规技术手段制备成任何一种适合食用的形式,例如固体饮料、饮料、糖果、饼干、果冻等。
进一步,所述食用辅料为食品级辅料,且包括稳定剂、增味剂、赋形剂或护色剂中的至少一种,从而便于制备食用的各种剂型或满足不同口味喜好的消费者的需求。
进一步,所述牡蛎低聚肽以及甲鱼低聚肽均通过生物酶解协同膜分离法再经后处理而制得;所述黑番茄提取物及番红花提取物均通过闪式提取协同膜分离法再经冷冻干燥法处理而制得;所述青钱柳提取物及可乐果提取物均通过闪式提取协同减压提取法再经后处理而制得。采用闪式提取协同膜分离法以及减压提取法对不同类别成分进行提取,能有效减少制备工艺中对活性成分的破坏,提高提取物得率,最大限度地保留各主要活性成分,同时能有效解决冷冻干燥工艺中普遍存在的微生物超标的问题,提高产品的安全性,此外,还能有效减少提取时间,降低能耗。
进一步,所述牡蛎低聚肽以及甲鱼低聚肽的制备方法分别如下:
(1)牡蛎肉/甲鱼肉的前处理:将新鲜的牡蛎肉/甲鱼肉在1~2.5%食用白醋中浸泡10~30min后用清水漂洗,取出沥净,接着将沥净的原料用70~80℃热水处理3~10min(热水处理的目的是对原料进行灭菌,同时也有利于后续的酶解反应),取出沥干后捣碎,于pH7.0~9.0、料液比1:3~1:8、温度30~37℃条件下加入30~60kU/L碱性脂肪酶脱脂20~60min,再次清水漂洗沥干后得到脱脂牡蛎肉糜/脱脂甲鱼肉糜;
(2)调浆:将制得的脱脂牡蛎肉糜/脱脂甲鱼肉糜按料液比1:3~1:8加入纯净水后经高速匀浆拌匀分散,得到牡蛎肉分散液/甲鱼肉分散液;
(3)酶解:将制得的牡蛎肉分散液/甲鱼肉分散液加热并使其温度保持在40~50℃之间,接着在分散液中加入动物蛋白酶、木瓜蛋白酶和中性蛋白酶中的至少一种,搅拌均匀后酶解3h~6h,酶解结束后加热煮沸7min灭酶,得到牡蛎酶解液/甲鱼酶解液;其中,动物蛋白酶的加入量是分散液中底物含量的0~0.5%,木瓜蛋白酶的加入量是分散液中底物含量的0~0.3%,中性蛋白酶加入的量为分散液中底物含量的0~0.5%;优选地,在上述分解液中加入混合酶液进行酶解,该混合酶液包括质量比为2:3~5:1的动物蛋白酶和木瓜蛋白酶。
(4)膜分离:对灭酶后的酶解液进行离心去杂,收集清液,清液依次经微孔滤膜过滤及活性炭柱处理后得酶解精制液,再将酶解精制液调节pH至8.0~8.2并使酶解液通过分子截留量为1000D的超滤膜,得到低聚肽酶解液;
(5)浓缩干燥:将低聚肽酶解液依次进行真空浓缩和喷雾干燥,得到所述牡蛎低聚肽/甲鱼低聚肽。
进一步,所述黑番茄提取物以及番红花提取物的制备方法分别如下:
(1)预处理与闪式提取:选择新鲜成熟的黑番茄果实,经榨汁处理后,汁液另存备用,残渣中仍有部分功效成分,需加入3~8倍体积的水作为提取溶剂,于室温下闪式提取)5~120s后进行离心分离后得到提取液,与上述汁液混合,得到黑番茄澄清液;
选择番红花干品,加入3~8倍体积的水作为提取溶剂,于室温下提取5~120s后进行离心分离后得到番红花澄清液;
(2)微滤与反渗透浓缩:将黑番茄澄清液/番红花澄清液通过微滤去除细菌,并通过反渗透将提取液浓缩至相对密度1.05~1.15;
(3)冷冻干燥:将上述浓缩液经冷冻干燥后得黑番茄提取物、番红花提取物,其中,黑番茄提取物得率高于6%,番红花提取物得率高于40%。
进一步,所述青钱柳提取物及可乐果提取物的制备方法分别如下:
(1)闪式提取:将粗粉碎的青钱柳/可乐果加入提取罐中,加入10~25倍量水作为提取溶剂,于室温下提取5~120s;
(2)减压提取:开启闪式提取器与减压提取器间的连通阀门,料液在压差作用下进入减压提取器进行提取,提取温度为30~60℃,提取压力为0.05~0.1MPa,提取时间为10~40min,直至获得提取液;
(3)离心、浓缩:将所得提取液进行离心分离,得澄清液,并进行减压浓缩,浓缩温度为50℃,真空度控制在-0.05~-0.08Mpa,所得浸膏相对密度控制在1.10~1.20;
(4)喷雾干燥:将浸膏进行喷雾干燥,得到上述青钱柳提取物/可乐果提取物,其中,青钱柳提取物得率高于40%,可乐果提取物得率高于20%。
为进一步解决上述第二个技术问题所采用的技术方案为:一种如上所述的具有增强男性性功能功能的食品组合物的制备方法,其特征在于包括以下步骤:
(1)分别制备牡蛎低聚肽和甲鱼低聚肽;
(2)分别制备黑番茄提取物和番红花提取物;
(3)分别制备青钱柳提取物和可乐果提取物;
(4)按上述质量份数取上述步骤(1)、(2)、(3)中制备的牡蛎低聚肽、甲鱼低聚肽、黑番茄提取物、番红花提取物、青钱柳提取物以及可乐果提取物,按照重量占比小的部分先逐级递增、重量比接近的组分先混合的原则混合至均匀,即得到所述食品组合物。
与现有技术相比,本发明的优点在于:本发明将牡蛎低聚肽、甲鱼低聚肽、黑番茄提取物、青钱柳提取物、可乐果提取物以及番红花提取物进行科学组配,其中,牡蛎低聚肽和甲鱼低聚肽能有效释放牡蛎与甲鱼中的小分子活性低聚肽,而小分子活性低聚肽能有效抵御胃肠道的消化,最大限度的保留了其原有活性,通过旁路途径、胞饮及主动运输等多种方式穿过小肠上皮细胞进入血液,随血液循环而靶向性地运输至细胞中发挥作用,牡蛎低聚肽能有效提高血清睾酮含量,甲鱼低聚肽能促进身体循环的改善,搭配以扩张血管的青钱柳提取物和促进血液循环的番红花提取物,可有效加速活性物质在体内的快速吸收和代谢利用,并能促进阴茎海绵体内小动脉及血管窦的平滑肌细胞舒张,改善阴茎勃起状态,提高勃起质量,而可乐果提取物中含有天然咖啡因,能起到抗疲劳作用,进一步巩固和提高性能力,另外,组合物中的黑番茄提取物除能有效掩盖牡蛎低聚肽及甲鱼低聚肽的鲜腥味,解决部分人对口感的不适应问题,此外,因其还含有番茄红素,对前列腺具有很好的保护作用,其抗氧化功能能有效减轻自由基对生殖系统的损伤,进而增强膀胱组织的抗氧化能力,减少脂质过氧化,并促进受损组织器官的恢复。可见,本发明的具有增强男性性功能的食物组合物采用的上述六种天然来源的药食同源的食品级原料,安全无毒副作用,不会产生依耐性以及抗药性,也不会在体内蓄积,能从细胞、组织和代谢水平上协同作用和增效,显著提高男性性功能。
本发明食品组合物的制备方法中分别采用闪式提取协同膜分离法以及减压提取法对不同组分进行提取制备,能有效减少制备过程中对活性成分的破坏,提高提取物得率,有效减少提取时间,降低能耗,最大限度地保留各主要活性成分,从而能在减少单份服用量的同时,精确定量摄入,此外,能有效解决传统冷冻干燥工艺中普遍存在的微生物超标的问题,提高产品的安全性。

具体实施方式
以下结合附图实施例对本发明作进一步详细描述。
实施例1:
按质量份数计,本实施例中具有增强男性性功能功能的食品组合物包括以下组分:牡蛎低聚肽2份、甲鱼低聚肽1.6份、黑番茄提取物5份、青钱柳提取物4份、可乐果提取物0.6份以及番红花提取物0.5份。
上述食品组合物的制备方法如下:
(1)制备牡蛎低聚肽/甲鱼低聚肽;
将新鲜的牡蛎肉/甲鱼肉浸泡在1.5%的食用白醋中30min后用清水漂洗,做脱腥处理,再将沥净后的原料用75℃热水处理5min,取出沥干后再用组织捣碎机捣碎,于pH8.0、料液比1:5、温度30℃条件下加入45kU/L碱性脂肪酶脱脂40min,再次清水漂洗沥干后得到脱脂牡蛎肉糜/脱脂甲鱼肉糜。
将脱脂牡蛎肉糜/脱脂甲鱼肉糜按料液比1:5分别加入纯净水后经高速匀浆拌匀分散,得到牡蛎肉分散液/甲鱼肉分散液。将上述牡蛎肉分散液/甲鱼肉分散液加热并使其温度保持在50℃,加入肉糜含量0.2%的动物蛋白酶和0.3%的木瓜蛋白酶,搅拌均匀后酶解4h。酶解结束后加热煮沸7min灭酶,将灭酶后的各酶解液分别进行离心去杂,收集清液,启动膜过滤设备,清液经过微孔滤膜过滤后继续过活性炭柱,得到酶解精制液,再将酶解精制液调节pH至8.0,使酶解液通过分子截留量为1000D的超滤膜,得到低聚肽酶解液。
将各低聚肽酶解液分别依次进行真空浓缩和喷雾干燥,得到上述的牡蛎低聚肽/甲鱼低聚肽。经检测,本实施例制备的牡蛎低聚肽和甲鱼低聚肽中分子量小于1000D的低聚肽成分分别占97%、96%,分子量处于180~500D的低聚肽成分分别占69%、66%。
(2)制备黑番茄提取物/番红花提取物;
选取新鲜成熟的黑番茄果实,经榨汁处理后,汁液另存备用,残渣中仍有部分功效成分,需加入5倍量水作为提取溶剂,于室温下闪式提取60s,后进行离心分离后得到提取液,与上述汁液混合,得到黑番茄澄清液。选择番红花干品,加入8倍量水作为提取溶剂,于室温下提取40s,后进行离心分离后得到番红花澄清液。将黑番茄澄清液/番红花澄清液通过微滤去除细菌,并通过反渗透将提取液浓缩至相对密度1.05~1.15;浓缩液经冷冻干燥后得黑番茄提取物/番红花提取物。本实施例制备的黑番茄提取物得率为7.15%;番红花提取物得率为44.42%,提取物中番红花总苷含量32.11%。
(3)制备青钱柳提取物/可乐果提取物;
将粗粉碎的青钱柳/可乐果加入提取罐中,加入12倍量水作为提取溶剂,于室温下提取40s。开启闪式提取器与减压提取器间的连通阀门,料液在压差作用下进入减压提取器进行提取,提取温度为45℃,提取压力为0.07MPa,提取时间为10min,得提取液。将所得提取液进行离心分离,得澄清液,并进行减压浓缩,浓缩温度为50℃,真空度控制在-0.05~-0.08Mpa,所得浸膏相对密度控制在1.10~1.20。将浸膏进行喷雾干燥,得到青钱柳提取物/可乐果提取物。
本实施例制备的青钱柳提取物得率为32.21%,提取物中多糖含量为11.25%,总黄酮含量为8.11%;可乐果提取物得率为20.62%,提取物中天然咖啡因含量为11.01%。
(4)将上述步骤(1)、(2)、(3)中制备的牡蛎低聚肽、甲鱼低聚肽、黑番茄提取物、番红花提取物、青钱柳提取物、可乐果提取物,按照重量占比小的部分先逐级递增、重量比接近的组分先混合的原则混合至均匀,即得到上述功能食品。
实施例2:
按质量份数计,本实施例中具有增强男性性功能功能的食品组合物包括以下组分:牡蛎低聚肽6份、甲鱼低聚肽5份、黑番茄提取物2份、青钱柳提取物0.5份、可乐果提取物0.2份、番红花提取物0.01份。
上述食品组合物的制备方法如下:
(1)制备牡蛎低聚肽/甲鱼低聚肽;
将新鲜的牡蛎肉/甲鱼肉浸泡在2.5%的食用白醋中10min后用清水漂洗,做脱腥处理,再将沥净后的原料用80℃热水处理3min,取出沥干后再用组织捣碎机捣碎,于pH8.0、料液比1:4、温度37℃条件下加入60kU/L碱性脂肪酶脱脂30min,再次清水漂洗沥干后得到脱脂牡蛎肉糜/脱脂甲鱼肉糜。
将脱脂牡蛎肉糜/脱脂甲鱼肉糜按料液比1:3分别加入纯净水后经高速匀浆拌匀分散,得到牡蛎肉分散液/甲鱼肉分散液。将上述牡蛎肉分散液/甲鱼肉分散液加热并使其温度保持在40℃,加入肉糜含量0.5%的动物蛋白酶和0.1%的木瓜蛋白酶,搅拌均匀后酶解4h。酶解结束后加热煮沸7min灭酶,将灭酶后的各酶解液分别进行离心去杂,收集清液,启动膜过滤设备,清液经过微孔滤膜过滤后继续过活性炭柱,得到酶解精制液,再将酶解精制液调节pH至8.0,使酶解液通过分子截留量为1000D的超滤膜,得到低聚肽酶解液。
将各低聚肽酶解液分别依次进行真空浓缩和喷雾干燥,得到上述的牡蛎低聚肽/甲鱼低聚肽。经检测,本实施例制备的牡蛎低聚肽和甲鱼低聚肽中分子量小于1000D的低聚肽成分分别占98%、97%,分子量处于180~500D的低聚肽成分分别占73%、69%。
(2)制备黑番茄提取物/番红花提取物;
选取新鲜成熟的黑番茄果实,经榨汁处理后,汁液另存备用,残渣中仍有部分功效成分,需加入8倍量水作为提取溶剂,于室温下闪式提取90s,后进行离心分离后得到提取液,与上述汁液混合,得到黑番茄澄清液。选择番红花干品,加入3倍量水作为提取溶剂,于室温下提取90s,后进行离心分离后得到番红花澄清液。将黑番茄澄清液/番红花澄清液通过微滤去除细菌,并通过反渗透将提取液浓缩至相对密度1.05~1.15;浓缩液经冷冻干燥后得黑番茄提取物/番红花提取物。本实施例制备的黑番茄提取物得率为7.27%;番红花提取物得率为43.09%,提取物中番红花总苷含量31.87%。
(3)制备青钱柳提取物/可乐果提取物;
将粗粉碎的青钱柳/可乐果加入提取罐中,加入20倍量水作为提取溶剂,于室温下提取100s。开启闪式提取器与减压提取器间的连通阀门,料液在压差作用下进入减压提取器进行提取,提取温度为55℃,提取压力为0.06MPa,提取时间为30min,得提取液。将所得提取液进行离心分离,得澄清液,并进行减压浓缩,浓缩温度为50℃,真空度控制在-0.05~-0.08Mpa,所得浸膏相对密度控制在1.10~1.20。将浸膏进行喷雾干燥,得到青钱柳提取物/可乐果提取物。
本实施例制备的青钱柳提取物得率为35.32%,提取物中多糖含量为12.38%,总黄酮含量为8.87%;可乐果提取物得率为22.77%,提取物中天然咖啡因含量为12.37%。
(4)将上述步骤(1)、(2)、(3)中制备的牡蛎低聚肽、甲鱼低聚肽、黑番茄提取物、番红花提取物、青钱柳提取物、可乐果提取物,按照重量占比小的部分先逐级递增、重量比接近的组分先混合的原则混合至均匀,即得到上述功能食品。
实施例3:
按质量份数计,本实施例中具有增强男性性功能功能的食品组合物包括以下组分:牡蛎低聚肽4份、甲鱼低聚肽3份、黑番茄提取物3.5份、青钱柳提取物2.8份、可乐果提取物0.3份、番红花提取物0.1份。
上述功能食品的制备方法为:
(1)制备牡蛎低聚肽/甲鱼低聚肽;
将新鲜的牡蛎肉/甲鱼肉浸泡在1.5%的食用白醋中15min后用清水漂洗,做脱腥处理,再将沥净后的原料用75℃热水处理5min,取出沥干后再用组织捣碎机捣碎,于pH8.0、料液比1:5、温度35℃条件下加入45kU/L碱性脂肪酶脱脂30min,再次清水漂洗沥干后得到脱脂牡蛎肉糜/脱脂甲鱼肉糜。
将脱脂牡蛎肉糜/脱脂甲鱼肉糜按料液比1:3分别加入纯净水后经高速匀浆拌匀分散,得到牡蛎肉分散液/甲鱼肉分散液。将上述牡蛎肉分散液/甲鱼肉分散液加热并使其温度保持在45℃,加入肉糜含量0.3%的动物蛋白酶和0.2%的木瓜蛋白酶,搅拌均匀后酶解4h。酶解结束后加热煮沸7min灭酶,将灭酶后的各酶解液分别进行离心去杂,收集清液,启动膜过滤设备,清液经过微孔滤膜过滤后继续过活性炭柱,得到酶解精制液,再将酶解精制液调节pH至8.0,使酶解液通过分子截留量为1000D的超滤膜,得到低聚肽酶解液。
将各低聚肽酶解液分别依次进行真空浓缩和喷雾干燥,得到上述的牡蛎低聚肽/甲鱼低聚肽。经检测,本实施例制备的牡蛎低聚肽和甲鱼低聚肽中分子量小于1000D的低聚肽成分分别占98%、97%,分子量处于180~500D的低聚肽成分分别占71%、68%。
(2)制备黑番茄提取物/番红花提取物;
选取新鲜成熟的黑番茄果实,经榨汁处理后,汁液另存备用,残渣中仍有部分功效成分,需加入3倍量水作为提取溶剂,于室温下闪式提取40s,后进行离心分离后得到提取液,与上述汁液混合,得到黑番茄澄清液。选择番红花干品,加入8倍量水作为提取溶剂,于室温下提取60s,后进行离心分离后得到番红花澄清液;将黑番茄澄清液/番红花澄清液通过微滤去除细菌,并通过反渗透将提取液浓缩至相对密度1.05~1.15;浓缩液经冷冻干燥后得黑番茄提取物/番红花提取物。本实施例制备的黑番茄提取物得率为7.75%;番红花提取物得率为45.62%,提取物中番红花总苷含量32.87%。
(3)制备青钱柳提取物/可乐果提取物;
将粗粉碎的青钱柳/可乐果加入提取罐中,加入15倍量水作为提取溶剂,于室温下提取60s.开启闪式提取器与减压提取器间的连通阀门,料液在压差作用下进入减压提取器进行提取,提取温度为50℃,提取压力为0.05MPa,提取时间为10min,得提取液。将所得提取液进行离心分离,得澄清液,并进行减压浓缩,浓缩温度为50℃,真空度控制在-0.05~-0.08Mpa,所得浸膏相对密度控制在1.10~1.20。将浸膏进行喷雾干燥,得到青钱柳提取物/可乐果提取物。
本实施例制备的上述青钱柳提取物得率为39.76%,提取物中多糖含量为15.57%,总黄酮含量为9.84%;可乐果提取物得率为25.54%,提取物中天然咖啡因含量为13.86%。
(4)将上述步骤(1)、(2)、(3)中制备的牡蛎低聚肽、甲鱼低聚肽、黑番茄提取物、番红花提取物、青钱柳提取物、可乐果提取物,按照重量占比小的部分先逐级递增、重量比接近的组分先混合的原则混合至均匀,即得到上述功能食品。
对比例1:
按重量份数计,本对比例中的食品组合物包括以下组分:牡蛎低聚肽4份、甲鱼低聚肽3份、复合提取物6.7份。
上述原料的来源/制备方法为:
(1)牡蛎低聚肽、甲鱼低聚肽均为市面在售原料,且为低聚肽行业主流厂家生产:牡蛎低聚肽由武汉天天好生物制品有限公司生产,甲鱼低聚肽由浙江中得农业集团有限公司生产。经检测,所选市售牡蛎低聚肽、甲鱼低聚肽中分子量小于1000D的低聚肽成分分别占81%、73%,分子量处于180~500D的低聚肽成分分别占45%、32%。
(2)复合提取物的制备;
选择新鲜成熟的黑番茄果实,经榨汁处理后,汁液另存备用,残渣与番红花干品、粗粉碎的青钱柳和可乐果一同加入提取罐中,以实施例3中各提取物得率推算各原料添加比例,加入20倍量水,浸泡30min后采用传统提取法进行提取,自煮沸后开始计时,提取60min后得提取液;将所得提取液进行离心分离,得澄清液。将上述澄清液进行减压浓缩和喷雾干燥,得到复合提取物。
(3)将步骤(1)中市售牡蛎低聚肽、甲鱼低聚肽与(2)中复合提取物混合均匀即得本比较例上述的食品组合物。
对比例2:
按重量份数计,本对比例中的食品组合物包括以下组分:牡蛎低聚肽4份、甲鱼低聚肽3份。
上述原料的制备方法与实施例3中牡蛎低聚肽、甲鱼低聚肽的制备方法相同,将二者混合均匀即得本比较例上述的食品组合物。
对比例3:
本比较例上述的食品组合物。:黑番茄提取物3.5份、青钱柳提取物2.8份、可乐果提取物0.3份、番红花提取物0.1份。
上述原料的制备方法与实施例3中黑番茄提取物、青钱柳提取物、可乐果提取物、番红花提取物的制备方法相同,将四者混合均匀即得。
本发明对各实施例和对比例制备的主要活性物质混合物进行了动物模型效果试验,具体为基于小鼠交配实验模型的功效评价试验,该实验委托浙江大学生物系统工程与食品科学学院完成。
试验方法:ICR雄性小鼠,体重20±2g,自由进食标准颗粒饲料及饮水,保持环境温度25±2℃,光照周期12h:12h条件下适应性饲养一周后开始正式实验。另需相应数量ICR雌性小鼠用于性功能评价实验。
随机将雄性小鼠分成10组,每组10只,分别为:实施例1组、实施例2组以及实施例3低剂量组(1.0g/kg);实施例3中剂量组(2.0g/kg);实施例3高剂量组(4.0g/kg);对比例1组;对比例2组;对比例3组;阳性对照组(肾宝片)以及正常对照组。其中,实施例1组、实施例2组、实施例3中剂量组、对比例1组、对比例2组、对比例3组均以2.0g/kg的给药剂量用生理盐水溶解后灌胃,实施例3低剂量组、实施例3高剂量组分别以1.0g/kg、4.0g/kg的给药剂量用生理盐水溶解后灌胃,阳性对照组根据肾宝片的推荐服用剂量折算,按0.96g/kg的给药剂量用生理盐水溶解后灌胃,正常对照组以等容量的生理盐水灌胃,每日根据体重状况给予小鼠相应剂量的受试物,每日一次,连续灌胃6周,于固定日期测定小鼠体重和称量饲料重量,并记录。末次给药后,进行性功能测试,在每个测试开始时,将雄性小鼠单独放在笼子里,使其适应15min。雌性小鼠于实验前48h皮下注射5μg苯甲酸雌二醇,并于实验前5h皮下注射5μg孕酮诱发动情。实验在昏暗的房间进行(19:00-23:00),雌性和雄性小鼠放在同一个笼子里,观察并摄录25min内小鼠交配行为。统计各组小鼠的性行为参数,包括:(1)捕捉次数:雄性小鼠捕捉雌性小鼠的次数;(2)骑跨潜伏期:引入雌性小鼠后出现第一次骑跨行为的时间间隔;(3)骑跨次数:雄性小鼠在雌性小鼠身上骑跨行为的次数;(4)插入潜伏期:从测试开始到雄鼠有第一次插入行为的时间;(5)插入次数:25分钟内插入行为的次数;(6)射精潜伏期:第1次插入至射精所需要的时间;(7)射精次数:雄性小鼠25分钟内射精的次数;(8)射精后的间隔:雄鼠射精后到再次插入之间的时间间隔。待交配实验结束后次日,将眼球的血液样本收集到管中,在3500rpm离心5min后,获得血清,放置-20℃冰箱待用,测定生化指标。采血后,处死小鼠,取睾丸并在冰浴上用生理盐水洗去浮血,放入福尔马林中保存做病理切片(HE染色),取阴茎海绵体、附睾、精囊腺、附睾、肝脏用锡箔纸包好后放入-80℃保存备用。所有雌性小鼠在性功能实验后实行二氧化碳安乐死。
试验结果如下:
由表1可知,与正常对照组比较,阳性对照组小鼠的捕捉次数、骑跨次数、插入次数、射精潜伏期、射精次数等指标均显著上升(P<0.01),而骑跨潜伏期、插入潜伏期、射精后间隔期均明显下降(P<0.05,P<0.01),表明阳性对照具有提高小鼠性功能的作用。与阳性对照组比较,实施例3中、高剂量组小鼠的捕捉次数、骑跨次数、插入次数、射精潜伏期、射精次数等指标均明显上升(P<0.05,P<0.01),而骑跨潜伏期、插入潜伏期、射精后间隔期均明显下降(P<0.05,P<0.01),表明实施例3的中、高剂量组在提高小鼠性功能方面的作用要强于市售的肾宝片。进一步将实施例3中剂量组与对比例1组、对比例2组、对比例3组的数据进行比较,提示将两种低聚肽与四种提取物经科学配伍,具有良好的协同增效作用,且采用闪式提取协同膜分离法再经冷冻干燥法以及闪式提取协同减压提取法所制备提取物的功效优于传统混合提取法。
由表2可知,与正常对照组比较,阳性对照组及实施例3低、中、高剂量组小鼠的血清睾酮水平、促卵泡激素水平、促黄体生成素水平、血清NO水平以及海绵体NO水平均明显上升(P<0.05,P<0.01),而海绵体PDE5水平明显下降(P<0.05,P<0.01)。与阳性对照组比较,实施例3中、高剂量组小鼠的血清睾酮水平、促卵泡激素水平、促黄体生成素水平、血清NO水平、海绵体NO水平均明显上升(P<0.05,P<0.01),海绵体PDE5水平呈下降趋势,但差异不显著(P>0.05),这与小鼠表观性行为学指标结果一致,表明实施例3的中、高剂量组的作用要强于市售的肾宝片,提示本发明的食品组合物可通过提高小鼠血清睾酮、促卵泡激素、促黄体生成素、血清NO及海绵体NO水平来提高性功能。进一步将实施例3中剂量组与各对比例组进行比较,可得出上述小鼠行为学指标中相类似结果。
通过伊红对小鼠附睾中的精子进行染色,结果如图1所示,表面与正常对照组相比,各处理组对精子形态没有显著影响。
从分子机制角度分析,睾酮是在睾丸间质细胞的线粒体和内质网中由胆固醇经过一系列酶促反应而合成。其间,胆固醇由类固醇急性调节蛋白(StAR)介导而从线粒体膜外转移至膜内,而细胞色素P450侧链裂解酶(P450scc)是负责该过程的主导酶,负责将胆固醇转化为孕烯醇酮,随后光滑内质网内的3β-羟固醇脱氢酶(3β-HSD)再将其转化为孕酮,最后孕酮在17β-HSD3的作用下转化为睾酮,其中起限速作用的是StAR。基于小鼠睾酮水平的上升,推测合成睾酮的相关蛋白酶水平提高,因此进一步通过免疫组化测定睾丸组织StAR与HSD17B3的表达。由表3和图2可知,与正常对照组比较,阳性对照组及实施例2组、实施例3低、中、高剂量组和对比例1组小鼠睾丸组织StAR与HSD17B3均明显上升(P<0.05,P<0.01)。与阳性对照组比较,实施例3低、中、高剂量组小鼠的睾丸组织StAR与HSD17B3均明显上升(P<0.05,P<0.01),与前面推测的结果一致。
本发明对制备的主要活性物质混合物还进行了人群食用效果验证,组织56位不同年龄段的志愿者进行为期10天的试用,其中40岁以上人群占94.64%,10天后,认为效果显著的有40人,认为有一定效果的有11人,认为无效的有5人,整体有效率高达91.07%。
随机选取两名志愿者症状予以说明。
志愿者1:马××,46岁,试用前,性欲差,难勃起,勃起后硬度差,不持久。试用第3天,晨勃现象明显;试用10天后,勃起硬度及性生活时间明显延长,并多次表达要继续试用的愿望。
志愿者2:李×,56岁,试用前,精力差、时常身体乏力,腰膝酸软,性欲望极低,几乎无性生活,勃起困难。试用第4天,晨勃现象明显,精力明显感觉充沛,勃起时间及勃起硬度明显延长,有性生活欲望;试用10天后,性生活欲望强烈,性生活时间明显延长,质量明显提高。

Claims (6)
1.一种具有增强男性性功能功能的食品组合物,其特征在于,由以下组分及其质量份数组成:
牡蛎低聚肽1~6份、甲鱼低聚肽1~5份、黑番茄提取物2~5份、青钱柳提取物0.5~4份、可乐果提取物0.1~0.6份以及番红花提取物0.01~0.3份,
所述牡蛎低聚肽以及甲鱼低聚肽的制备方法分别如下:
(1) 牡蛎肉/甲鱼肉的前处理:将新鲜的牡蛎肉/甲鱼肉在1~2.5%食用白醋中浸泡10~30min后用清水漂洗,取出沥净,接着将沥净的原料用70~80℃热水处理3~10min,取出沥干后捣碎,于pH7.0~9.0、料液比1:3~1:8、温度30~37℃条件下加入30~60kU/L碱性脂肪酶脱脂20~60min,再次清水漂洗沥干后得到脱脂牡蛎肉糜/脱脂甲鱼肉糜;
(2)调浆:将制得的脱脂牡蛎肉糜/脱脂甲鱼肉糜按料液比1:3~1:8加入纯净水后经高速匀浆拌匀分散,得到牡蛎肉分散液/甲鱼肉分散液;
(3)酶解:将制得的牡蛎肉分散液/甲鱼肉分散液加热并使其温度保持在40~50℃之间,接着在分散液中加入动物蛋白酶、木瓜蛋白酶和中性蛋白酶中的至少一种,搅拌均匀后酶解3h~6h,酶解结束后加热煮沸7min灭酶,得到牡蛎酶解液/甲鱼酶解液;其中,动物蛋白酶的加入量是分散液中底物含量的0~0.5%,木瓜蛋白酶的加入量是分散液中底物含量的0~0.3%,中性蛋白酶加入的量为分散液中底物含量的0~0.5%;
(4)膜分离:对灭酶后的酶解液进行离心去杂,收集清液,清液依次经微孔滤膜过滤及活性炭柱处理后得酶解精制液,再将酶解精制液调节pH至8.0~8.2并使酶解液通过分子截留量为1000D的超滤膜,得到低聚肽酶解液;
(5)浓缩干燥:将低聚肽酶解液依次进行真空浓缩和喷雾干燥,得到所述牡蛎低聚肽/甲鱼低聚肽;
所述青钱柳提取物及可乐果提取物的制备方法分别如下:
(1)闪式提取:将粗粉碎的青钱柳/可乐果加入提取罐中,加入10~25倍量水作为提取溶剂,于室温下提取5~120s;
(2)减压提取:开启闪式提取器与减压提取器间的连通阀门,料液在压差作用下进入减压提取器进行提取,提取温度为30~60℃,提取压力为0.05~0.1MPa,提取时间为10~40min,直至获得提取液;
(3)离心、浓缩:将所得提取液进行离心分离,得澄清液,并进行减压浓缩,浓缩温度为50℃,真空度控制在-0.05~-0.08Mpa,所得浸膏相对密度控制在1.10~1.20;
(4)喷雾干燥:将浸膏进行喷雾干燥,得到上述青钱柳提取物/可乐果提取物,其中,青钱柳提取物得率高于40%,可乐果提取物得率高于20%,
所述牡蛎低聚肽及甲鱼低聚肽中分子量小于1000D的低聚肽成分分别占95%以上,而分子量处于180~500D的低聚肽成分分别占65%以上,
所述番红花提取物中番红花总苷含量高于30%,上述青钱柳提取物中多糖含量高于35%,上述可乐果提取物中天然咖啡因含量高于10%。
2.如权利要求1所述的具有增强男性性功能功能的食品组合物,其特征在于,所述食品组合物包括可接受的食用辅料。
3.如权利要求2所述的具有增强男性性功能功能的食品组合物,其特征在于,所述食用辅料为食品级辅料,包括稳定剂、增味剂、赋形剂或护色剂中的至少一种。
4.如权利要求1~3任一项所述的具有增强男性性功能功能的食品组合物,其特征在于,所述黑番茄提取物及番红花提取物均通过闪式提取协同膜分离法再经冷冻干燥法处理而制得。
5.如权利要求4所述的具有增强男性性功能功能的食品组合物,其特征在于,所述黑番茄提取物以及番红花提取物的制备方法分别如下:
(1)预处理与闪式提取:选择新鲜成熟的黑番茄果实,经榨汁处理后,汁液另存备用,残渣中仍有部分功效成分,需加入3~8倍体积的水作为提取溶剂,于室温下闪式提取5~120s后进行离心分离后得到提取液,与上述汁液混合,得到黑番茄澄清液;
选择番红花干品,加入3~8倍体积的水作为提取溶剂,于室温下提取5~120s后进行离心分离后得到番红花澄清液;
(2)微滤与反渗透浓缩:将黑番茄澄清液/番红花澄清液通过微滤去除细菌,并通过反渗透将提取液浓缩至相对密度1. 05~1.15;
(3)冷冻干燥:将上述浓缩液经冷冻干燥后得黑番茄提取物/番红花提取物,其中,黑番茄提取物得率高于6%,番红花提取物得率高于40%。
6.一种如权利要求1~5任一项所述的具有增强男性性功能功能的食品组合物的制备方法,其特征在于包括以下步骤:
(1)分别制备牡蛎低聚肽和甲鱼低聚肽;
(2)分别制备黑番茄提取物和番红花提取物;
(3)分别制备青钱柳提取物和可乐果提取物;
(4)按上述质量份数取上述步骤(1)、(2)、(3)中制备的牡蛎低聚肽、甲鱼低聚肽、黑番茄提取物、番红花提取物、青钱柳提取物以及可乐果提取物,按照重量占比小的部分先逐级递增、重量比接近的组分先混合的原则混合至均匀,即得到所述食品组合物。
