Abstract
本发明公开了一种低灰分淫羊藿提取物及其制备方法,其采用酒精水溶液或水为溶媒对淫羊藿药材进行提取,提取液经固液分离后进行浓缩,浓缩液醇沉,固液分离,清液干燥得低灰分淫羊藿提取物。本发明利用无机物金属离子在酒精溶液与水溶液中的溶解度不同,降低所得提取物中的灰分。

本发明涉及物质提取技术领域,具体涉及一种低灰分淫羊藿提取物及其制备方法。
背景技术
淫羊藿提取物具有增强免疫、补肾壮阳、强健筋骨、祛风除湿之功效,可用于治疗骨质疏松、更年期综合征、乳房肿块、高血压、冠心病等疾病,广泛应用于保健食品、膳食补充剂行业。
目前淫羊藿提取物的生产主要是通过大孔吸附树脂分离纯化工艺降低提取物产品中的灰分,但对于大孔吸附树脂工艺进行纯化要求的条件较高,步骤繁琐,而未经过大孔吸附树脂分离纯化的提取物灰分通常在15%以上。
发明内容
本发明旨在克服大孔吸附树脂分离纯化工艺存在的弊端,提供一种低灰分淫羊藿提取物及其制备方法,其以酒精水溶液或水为溶媒对淫羊藿药材进行提取,提取液经固液分离后进行浓缩,浓缩液醇沉,固液分离,取清液,利用无机物金属离子在酒精溶液与水溶液中的溶解度不同,降低所得提取物中的灰分。
为了实现上述发明目的,本发明提供的技术方案如下:
一种低灰分淫羊藿提取物的制备方法,包括以下步骤:
(1)提取:将淫羊藿药材投入提取设备中,加入酒精水溶液或水进行提取,收集提取液;
(2)固液分离:将步骤(1)得到的提取液经过过滤机或离心机处理,收集澄清溶液;
(3)浓缩:将步骤(2)得到的澄清溶液经真空浓缩设备进行浓缩,得浓缩液A;
(4)醇沉:向步骤(3)所得浓缩液A中逐步加入酒精进行醇沉;
(5)固液分离:对醇沉液进行固液分离,收集上清。
(6)浓缩:将步骤(5)所得上清液进行浓缩,得浓缩液B。
(7)干燥:将步骤(6)所得浓缩液B进行干燥,得低灰分淫羊藿提取物。
优选的,步骤(1)中,所述酒精水溶液的可为任意酒精浓度,提取温度为60℃~100℃。
优选的,步骤(3)中,所述浓缩液的生药比为1:1~3:1。
优选的,步骤(4)中,所述醇沉操作,将95%乙醇或无水乙醇加入快速搅拌中的浓缩液中,醇沉液的酒精浓度为60%~90%。
本发明还提供了采用上述低灰分淫羊藿提取物的制备方法制备得到的淫羊藿提取物。该淫羊藿提取物中灰分小于10%,淫羊藿苷含量大于11%。
本发明与现有技术相比,本发明具有以下优点:
1、仅使用最常规、最简单的设备即可将提取物中灰分降至10%以下,对设备的要求低。
2、相对于大孔吸附树脂提纯工艺,操作方便、简单,提纯效果相当。

实施例1
结合图1所示,本实施例的一种低灰分淫羊藿提取物及其制备方法,包括以下步骤:
(1)提取:将250kg淫羊藿投入5吨提取罐中,加入20倍水升温进行提取,提取温度95℃,提取3次,每次2h,提取完成后将提取液转入离心机原料罐。
(2)固液分离:将提取液经过离心处理,收集清液。
(3)浓缩:将经过离心处理的提取液用双效浓缩器进行浓缩,浓缩至约150L转入醇沉罐。
(4)醇沉:开启搅拌,将800L 95%乙醇以60L/h的流速加入醇沉罐中,继续搅拌0.5h,然后转入离心机原料罐。
(5)固液分离:将醇沉液用离心机进行分离,分开收集固体及清液。
(6)浓缩:将清液转入双效浓缩器进行浓缩,60℃,浓缩至约60L。
(7)带式真空干燥:用带式真空干燥将浓缩液进行干燥,所得淫羊藿提取物指标参见表1。
实施例2
本实施例的一种低灰分淫羊藿提取物及其制备方法,包括以下步骤:
(1)提取:将250kg淫羊藿投入5吨提取罐中,加入18倍50%乙醇升温进行提取,提取温度78℃,提取3次,每次2h,提取完成后将提取液转入离心机原料罐。
(2)蒸馏、出渣:三次提取完成后,罐底部通蒸汽进行蒸馏,至馏出液醇度10%以下,回收乙醇用于调配提取液。提取罐中残液排放后,出渣,清洗提取罐。
(3)固液分离:将提取液经过离心处理,收集清液。
(4)浓缩:将经过离心处理的提取液用刮膜浓缩器进行浓缩,浓缩至约150L转入醇沉罐。
(5)醇沉:开启搅拌,将800L 95%乙醇以60L/h的流速加入醇沉罐中,继续搅拌0.5h,然后转入离心机原料罐。
(6)固液分离:将醇沉液用离心机进行分离,分开收集固体及清液。
(7)浓缩:将清液转入刮膜浓缩器进行浓缩,60℃,浓缩至约50L。
(8)带式真空干燥:用带式真空干燥将浓缩液进行干燥,所得淫羊藿提取物指标参见表1。
实施例3
本实施例的一种低灰分淫羊藿提取物及其制备方法,包括以下步骤:
(1)提取:将250kg淫羊藿投入5吨提取罐中,加入18倍60%乙醇升温进行提取,提取温度75℃,提取3次,每次2h,提取完成后将提取液转入离心机原料罐。
(2)蒸馏、出渣:三次提取完成后,罐底部通蒸汽进行蒸馏,至馏出液醇度10%以下,回收乙醇用于调配提取液。提取罐中残液排放后,出渣,清洗提取罐。
(3)固液分离:将提取液经过离心处理,收集清液。
(4)浓缩:将经过离心处理的提取液用刮膜浓缩器进行浓缩,浓缩至约200L转入醇沉罐。
(5)醇沉:开启搅拌,将600L 95%乙醇以60L/h的流速加入醇沉罐中,继续搅拌0.5h,然后转入离心机原料罐。

(7)浓缩:将清液转入刮膜浓缩器进行浓缩,60℃,浓缩至约50L。

(1)提取:将淫羊藿药材投入提取设备中,加入酒精水溶液或水进行提取,收集提取液;
(2)固液分离:将步骤(1)得到的提取液经过过滤机或离心机处理,收集澄清溶液;
(3)浓缩:将步骤(2)得到的澄清溶液经真空浓缩设备进行浓缩,得浓缩液A;
(4)醇沉:向步骤(3)所得浓缩液A中逐步加入酒精进行醇沉;
(5)固液分离:对醇沉液进行固液分离,收集上清。
(6)浓缩:将步骤(5)所得上清液进行浓缩,得浓缩液B。
(7)干燥:将步骤(6)所得浓缩液B进行干燥,得低灰分淫羊藿提取物。
2.根据权利要求1所述的制备方法,其特征在于,步骤(1)中:所述酒精水溶液的可为任意酒精浓度,提取温度为60℃~100℃。
