Abstract
本发明公开了一种宁心助眠组合物及其制备方法与应用。所述宁心助眠组合物包括生牡蛎壳、酸枣仁、百合、茯苓、红枣、玫瑰花、炒麦芽、山楂、甘草、陈皮、新鲜牡蛎肉、贻贝、γ‑氨基丁酸。所述宁心助眠组合物成分天然,配比科学,无毒副作用,并经动物体内实验证实,其可明显提高戊巴比妥钠阈下剂量睡眠发生率并缩短巴比妥钠睡眠潜伏期,具有改善睡眠的作用,进而能够用来制备改善睡眠的食品或保健品。
Description
技术领域
本发明属于食品领域,具体涉及一种宁心助眠组合物及其制备方法与应用。
背景技术
随着社会环境的变迁,人们工作和生活压力日益增加,失眠人群逐渐增多。失眠作为目前社会的常见病、多发病,对人们的健康水平和生活质量产生了严重的影响。失眠主要表现为:入睡困难、浅睡易醒、醒后难以入睡、多梦,常伴有白天萎糜不振、四肢无力、反应迟钝、工作效率低下、头痛、记忆力减退等症状。据调查研究,我国1/3的成年人受到失眠的困扰,而其中1/3的人已达到重度失眠的程度。
目前,西医针对失眠的治疗主要采用苯二氮卓类、唑吡坦、阿米替林等药物,上述药物均存在副作用大、易出现耐药性、成瘾性等问题。中成药中针对失眠的药物包括复方手参丸、三宝片、安眠补脑口服液、参芝安神口服液等,但其也存在起效慢、不宜长期服用等缺点。
据统计,失眠患者中仅21%的人就诊,而且大部分患者对安神功效的保健食品、功能性食品具有迫切需求,因此该类产品市场需求量巨大。目前市面上改善睡眠功能保健食品同质化严重,大部分以褪黑素作为主要原料,然而褪黑素改善睡眠疗效还需进一步验证,同时由于褪黑素是人体正常分泌物,人为地补充褪黑素,会抑制人体自身的分泌功能影响机体平衡。因此安全有效、特色鲜明的安神功能产品的开发极具价值。
发明内容
本发明的目的在于提供的是一种宁心助眠组合物及其制备方法与应用。所述宁心助眠组合物成分天然,安全可靠,具有明显的改善睡眠的作用。
为实现上述发明目的,本发明采用以下技术方案予以实现:
本发明提供了一种宁心助眠组合物,按重量份计,所述宁心助眠组合物包括生牡蛎壳10份-50份、酸枣仁5份-50份、百合2份-50份、茯苓2份-50份、红枣2份-50份、玫瑰花2份-50份、炒麦芽2份-50份、山楂2份-50份、甘草2份-50份、陈皮2份-50份、牡蛎肉5份-50份、贻贝5份-50份。
进一步的,按重量份计,所述宁心助眠组合物包括生牡蛎壳15份-25份、酸枣仁10份-20份、百合2份-10份、茯苓2份-10份、红枣2份-10份、玫瑰花2份-10份、炒麦芽2份-10份、山楂2份-10份、甘草2份-10份、陈皮2份-5份、牡蛎肉5份-15份、贻贝5份-15份。
优选的,按重量份计,所述宁心助眠组合物包括生牡蛎壳20份、酸枣仁15份、百合5份、茯苓5份、红枣5份、玫瑰花5份、炒麦芽5份、山楂5份、甘草5份、陈皮3份、牡蛎肉10份、贻贝10份。
进一步的,按重量份计,所述宁心助眠组合物还包括γ-氨基丁酸0.1份-10份,矫味剂1份-100份。
优选的,按重量份计,所述宁心助眠组合物还包括γ-氨基丁酸0.1份。
进一步的,所述宁心助眠组合物的制备方法包括以下步骤:
(1)分别称取原材料为生牡蛎壳、酸枣仁、百合、茯苓、红枣、玫瑰花、炒麦芽、山楂、甘草、陈皮,粉碎至20目-80目,加入原材料总重量的8-12倍的水,过滤后合并所有提取液,60-80℃减压浓缩,喷雾干燥,得到固体粉末1;
(2)称取新鲜牡蛎肉和贻贝,匀浆后,加入新鲜牡蛎肉和贻贝总重量1-3倍的水混匀,调节pH至2.5-3,加入酶制剂,保温酶解1-6h,酶解后将酶解液煮沸,离心,上清液60-80℃减压浓缩,喷雾干燥,得到固体粉末2;
(3)将固体粉末1和固体粉末2混匀,加入γ-氨基丁酸混匀,得到宁心助眠组合物。
进一步的,所述步骤(2)中酶制剂包括胃蛋白酶、胰蛋白酶、菠萝蛋白酶、木瓜蛋白酶、中性蛋白酶、碱性蛋白酶中的至少一种。
本发明还提供了所述的宁心助眠组合物在用于制备改善睡眠的食品或保健品中的应用。
进一步的,由所述宁心助眠组合物改善小鼠睡眠的有效剂量推算至临床每日成人用量为1-50g。
进一步的,所述宁心助眠组合物通过提高模型动物的睡眠发生率,缩短睡眠潜伏期来达到改善睡眠的作用。
进一步的,所述食品或保健品为膏滋、口服液、固体饮料剂型或咀嚼片。
与现有技术相比,本发明具有的优点及有益效果为:
本发明在中医药理论指导下,结合现代化学和药理学研究,精选酸枣仁、牡蛎、茯苓、红枣、百合等多种食用历史悠久的药食两用中药材,并依据上述中药的特性及配伍关系,凭借长期临床经验和大量的实验数据,得出了原材料间的最佳配比,并加入了具有补益功效的海洋贝类活性肽及具有镇静、健脑、促生长作用的γ-氨基丁酸,得到了具有宁心助眠组合物。所述宁心助眠组合物制备方法简单、效果显著,取材天然,适口性好,且无任何毒副作用;并经动物体内实验证实,所述宁心助眠组合物能够提高睡眠发生率,缩短睡眠潜伏期,进而改善睡眠,因此其适用于产业化生产,对失眠、虚烦等虚阳上浮之症有显著的效果。
具体实施方式
以下结合具体实施例对本发明的技术方案做进一步详细的说明。
实施例1
宁心助眠组合物的制备,包括以下步骤:
(1)称取牡蛎(生)20g,酸枣仁15g,百合5g,茯苓5g,红枣5g,玫瑰花5g,麦芽(炒)5g,山楂5g,甘草5g,陈皮3g,将各种原材料粉碎成20-80目的粗粉,用10倍重量的水加热回流提取3次,过滤,合并三次所得提取液,70℃减压浓缩,喷雾干燥,得固体粉末1;其中,所述牡蛎(生)为未经过煅烧的生牡蛎壳。
(2)称取新鲜牡蛎肉10g和贻贝10g,匀浆,加入1倍重量的水,调pH至2.5,加入0.5%(v/v)的胃蛋白酶,37℃酶解1h后调pH为10,再加入0.5%(v/v)的碱性蛋白酶,酶解1.5h,于100℃煮沸10min,冷却后离心,上清液70℃减压干燥,喷雾干燥,得固体粉末2;
(3)将固体粉末1和固体粉末2混匀,再加入0.1gγ-氨基丁酸,即得宁心助眠组合物。
(4)所述宁心助眠组合物还可以与1-100g的矫味剂(蜂蜜、木糖醇中的至少一种)混合后制成膏滋、口服液、固体饮料剂型、咀嚼片等形式的宁心助眠食品。
实施例2
为观察实施例1制备的宁心助眠组合物对小鼠睡眠的影响,本实验通过直接睡眠试验、延长戊巴比妥钠睡眠时间试验、戊巴比妥钠阈下剂量催眠试验、巴比妥钠睡眠潜伏期试验,观察其改善睡眠的功效。延长戊巴比妥钠睡眠时间实验、戊巴比妥钠阈下剂量催眠试验和巴比妥钠睡眠潜伏期试验三项试验中二项阳性,且无明显直接睡眠作用,可判定宁心助眠组合物具有改善睡眠功能作用。
1、实验样品及实验方法
动物:昆明小鼠,雄性。PVC盒饲养,每盒5只,室温20~26℃,日温差≤4℃,相对湿度40~70%,昼夜明暗交替时间为12h/12h。
实验设计:
实验动物接收后检疫3-5天,剔除不合格动物。随机分为对照组、低剂量组(2.5g/kg宁心助眠组合物)、高剂量组(5g/kg宁心助眠组合物)、地西泮组(3mg/kg),每组12只。连续灌胃受试物30天。试验期间每周称量一次动物体重。根据体重调整灌胃的受试物剂量。正常对照组给予等容量的纯净水。阳性对照组给予3mg/kg剂量的地西泮。
(1)直接睡眠试验
动物末次给不同剂量受试样品后,观察动物是否出现睡眠现象。睡眠以翻正反射消失为指标。当小鼠置于背卧位时,能立即翻正身位。如超过30-60s不能翻正者,即认为翻正反射消失,进入睡眠。翻正反射恢复即为动物觉醒,翻正反射消失至恢复这段时间为动物睡眠时间,记录各组入睡动物数及睡眠时间。
(2)延长戊巴比妥钠睡眠时间试验
做正式实验前先进行预实验,确定使动物100%入睡、但又不使睡眠时间过长的戊巴比妥钠剂量为30-60mg/kg,用此剂量做正式实验。动物末次给不同剂量受试样品后30min,给各组动物腹腔注射戊巴比妥钠,注射量为0.2mL/20g,以翻正反射消失为指标,观察受试样品能否延长戊巴比妥钠睡眠时间。
(3)戊巴比妥钠阈下剂量催眠试验
正式实验前先进行预实验,确定戊巴比妥钠阈下催眠剂量为16-30mg/kg,即80-90%小鼠翻正反射不消失的戊巴比妥钠最大阈下剂量。动物末次给不同剂量受试样品30min后,各组动物腹腔注射戊巴比妥钠最大阈下催眠剂量,记录30min内入睡动物数(翻正反射消失达1min以上者)。
(4)巴比妥钠睡眠潜伏期试验
做正式实验前先进行预实验,确定使动物100%入睡、但又不使睡眠时间过长的巴比妥钠的剂量为200-300mg/kg,用此剂量做正式实验。动物末次给不同剂量受试样品30min后,给各组动物腹腔注射巴比妥钠,注射量为0.2mL/20g,以翻正反射消失为指标,观察受试样品对巴比妥钠睡眠潜伏期的影响。
2、实验结果
(1)直接睡眠试验
参见表1,动物末次给予纯净水及不同剂量样品后,各受试物组和对照组小鼠均未出现睡眠现象且活动正常,表明本发明的宁心助眠组合物对小鼠无直接催眠作用。

(2)延长戊巴比妥钠睡眠时间试验
动物末次给予纯净水及不同剂量样品后,腹腔注射戊巴比妥钠(40mg/kg)。结果见表2,与对照组相比,高剂量组和低剂量组小鼠睡眠时间无显著性差异。阳性药地西泮组小鼠睡眠时间与对照组比较显著延长(P<0.05)。

(3)戊巴比妥钠阈下剂量催眠试验
动物末次给予纯净水及不同剂量样品后,腹腔注射戊巴比妥钠最大阈下催眠剂量(24mg/kg)。结果见表3,与对照组相比,高剂量组与低剂量组小鼠睡眠发生率均显著增加(P<0.01)。阳性药地西泮组小鼠睡眠发生率与对照组比较显著增加(P<0.01)。

(4)巴比妥钠睡眠潜伏期试验
动物末次给予纯净水及不同剂量样品后,腹腔注射巴比妥钠(260mg/kg)。结果见表4,与对照组相比,高剂量组和低剂量组小鼠睡眠潜伏期均显著缩短(P<0.05)。阳性药地西泮组小鼠睡眠潜伏期显著缩短(P<0.01)。

(5)对动物体重的影响
试验期间,各组动物一般状况良好。各样品对动物体重的影响见表5,高剂量和低剂量连续灌胃30天对动物体重与对照组比较无明显差异,说明宁心助眠组合物对动物不产生影响。

3、实验结论
宁心助眠组合物连续灌胃给予小鼠30天后,“戊巴比妥钠阈下剂量催眠试验”和“巴比妥钠睡眠潜伏期试验”两项试验结果均为阳性,并且未发现宁心助眠组合物有直接睡眠作用,因此说明宁心助眠组合物对小鼠具有改善睡眠功效。
以上实施例仅用于说明本发明的技术方案,而非对其进行限制;尽管参照前述实施例对本发明进行了详细的说明,对于本领域的普通技术人员来说,依然可以对前述实施例所记载的技术方案进行修改,或者对其中部分技术特征进行等同替换;而这些修改或替换,并不使相应技术方案的本质脱离本发明所要求保护的技术方案的精神和范围。
Claims (10)
1.一种宁心助眠组合物,其特征在于,按重量份计,所述宁心助眠组合物包括生牡蛎壳10份-50份、酸枣仁5份-50份、百合2份-50份、茯苓2份-50份、红枣2份-50份、玫瑰花2份-50份、炒麦芽2份-50份、山楂2份-50份、甘草2份-50份、陈皮2份-50份、牡蛎肉5份-50份、贻贝5份-50份。
2.根据权利要求1所述的宁心助眠组合物,其特征在于,按重量份计,所述宁心助眠组合物包括生牡蛎15份-25份、酸枣仁10份-20份、百合2份-10份、茯苓2份-10份、红枣2份-10份、玫瑰花2份-10份、炒麦芽2份-10份、山楂2份-10份、甘草2份-10份、陈皮2份-5份、牡蛎肉5份-15份、贻贝5份-15份。
3.根据权利要求1或2所述的宁心助眠组合物,其特征在于,按重量份计,所述宁心助眠组合物还包括γ-氨基丁酸0.1份-10份,矫味剂1份-100份。
4.根据权利要求2所述的宁心助眠组合物,其特征在于,所述宁心助眠组合物的制备方法包括以下步骤:
(1)分别称取原材料为生牡蛎壳、酸枣仁、百合、茯苓、红枣、玫瑰花、炒麦芽、山楂、甘草、陈皮,粉碎后加水热回流提取2-4次,过滤后合并所有提取液,减压浓缩,喷雾干燥,得到固体粉末1;
(2)称取新鲜牡蛎肉和贻贝,匀浆后加水混匀,调节pH,加入酶制剂,保温酶解1-6 h,酶解后将酶解液煮沸,离心,上清液减压干燥,喷雾干燥,得到固体粉末2;
(3)将固体粉末1和固体粉末2混匀,加入γ-氨基丁酸混匀,得到宁心助眠组合物。
5.根据权利要求4所述的宁心助眠组合物,其特征在于,所述步骤(1)中水的重量为原材料总重量的8-12倍;所述步骤(2)中水的重量为新鲜蛏肉和贻贝总重量的1-3倍。
6.根据权利要求4所述的宁心助眠组合物,其特征在于,所述步骤(2)中酶制剂包括胃蛋白酶、胰蛋白酶、菠萝蛋白酶、木瓜蛋白酶、中性蛋白酶、碱性蛋白酶中的至少一种。
7.权利要求1-3任一项所述的宁心助眠组合物在用于制备改善睡眠的食品或保健品中的应用。
8.根据权利要求7所述的宁心助眠组合物在用于制备改善睡眠的食品或保健品中的应用,其特征在于,所述宁心助眠组合物的每日成人用量为1-50g。
9.根据权利要求7所述的宁心助眠组合物在用于制备改善睡眠的食品或保健品中的应用,其特征在于,所述宁心助眠组合物通过提高模型动物的睡眠发生率,缩短睡眠潜伏期来达到改善睡眠的作用。
10.根据权利要求7所述的宁心助眠组合物在用于制备改善睡眠的食品或保健品中的应用,其特征在于,所述食品或保健品为膏滋、口服液、固体饮料剂型或咀嚼片。
