Abstract
本发明提供一种MOF微针贴片的制备方法及应用,该方法通过聚二甲基硅氧烷PDMS模具复制和紫外光固化,以甲基丙烯化透明质酸(MeHA)或甲基丙烯化明胶(GelMA)为主要成分制备得到针体柔软、多孔并负载MOF材料的微针贴片;本发明制备的MOF微针贴片可贴附在皮肤创面处,其柔软的微针针体可以增大有效成分与创面之间的表面积并减轻治疗过程的疼痛感。针体成分可诱导血管生成、促进上皮细胞分化;针体内负载的MOF材料有抗菌作用,从而有助于加速创面愈合,应用范围广泛。本发明MOF微针贴片制备方法成本低、操作简易,实用性强,适宜推广。
Description
技术领域
本发明属于生物医用材料技术领域,具体涉及一种MOF微针贴片的制备方法及应用。
背景技术
外伤造成的急性创面常伴有皮肤损伤以及一部分软组织的缺损,这常常给患者带来巨大的痛苦。目前创面修复主要以清创止血、镇静止痛、伤口缝合等方式为主,而这些方法容易使患者受到二次痛苦并产生皮肤瘢痕,创面修复效果也有待提高。
微针疗法是一种微创、无痛和高效的透皮疗法,可以在皮肤内形成大量的微孔道从而有利于药物的渗透,因此广泛应用于透皮给药。由于微米级的微针针体不会刺激皮下神经末梢,故不会造成患者额外的痛苦。在创面修复中,微针疗法通过提高与创面的接触面积以及透皮给药效率,从而能够更好地发挥创面修复效果。但尖锐的针体容易对创面造成二次损伤,在非无菌条件下甚至会引起严重的伤口感染。因此,迫切需要开发出无痛、抗菌的微针贴片用于创面的修复。基于此,本发明提出一种MOF微针贴片的制备方法及应用,制备针体柔软、多孔并负载MOF材料的微针贴片,对于减轻创面细菌污染、促进创面修复过程具有积极作用。
发明内容
本发明的目的是针对现有技术的不足,提供一种MOF微针贴片的制备方法及应用,本发明基于MOF材料优异的抗菌性能,以甲基丙烯化透明质酸 (MeHA) 或甲基丙烯化明胶(GelMA)为针体主要成分,制备得到针体柔软、多孔并负载MOF材料的微针贴片,可加速创面的修复过程。
为实现上述技术目的,本发明采取的技术方案为
一种MOF微针贴片的制备方法,包括以下步骤:
S1、MOF微针贴片原材料溶液的制备
将MOF材料微颗粒与磷酸缓冲盐溶液(PBS)混合,置于超声波清洗机中超声3小时以均匀分散,得到含有MOF材料微颗粒的磷酸缓冲盐溶液(PBS)溶剂,然后将上述溶剂与微针原材料充分混匀,并向其中加入光引发剂,获得待固化MOF微针贴片原材料溶液,最后避光保存在-4℃冰箱备用;
S2、MOF微针贴片的制备
在PDMS微针模具上滴加待固化MOF微针贴片原材料溶液,随后通过高速离心法使MOF微针贴片原材料溶液填充PDMS微针模具的针体,离心后除去模具表面多余的液体,通过紫外光固化的方法使模具内部的MOF微针贴片原材料溶液固化成型;待材料充分固化后从模具上剥离,得到MOF微针贴片。
进一步的,S1中,所述微针原材料选自甲基丙烯化透明质酸(MeHA)、甲基丙烯化明胶(GelMA)及聚乙二醇二丙烯酸酯(PEGDA)中的一种或几种。
进一步的, S1中,所述MOF材料选自ZIF-8、HKUST-1、Zn-Vitamin MOF及 Cu-Vitamin MOF 中的一种或多种。
进一步的,S1中,所述光引发剂为2-羟基-2-甲基苯丙酮。
进一步的,S2中,固化过程中将模具置于冰上,以防止固化过程中材料的变形。
进一步的,S2中,先用 0.1% (w/v) 的MOF微针贴片原材料溶液作为灌洗液对PDMS微针模具进行灌洗,用紫外光固化仪在模具上照射1分钟,随后清除灌洗液再进行MOF微针贴片的制作。
进一步的,MOF微针贴片原材料溶液在固化之前避光放置。
进一步的,所述PDMS微针模具由ETPTA微针模板翻模制得。
进一步的,所述MOF微针贴片呈圆形,针体为圆锥形,针体的底部直径为310μm,高度为700μm。
本发明还提供了一种通过上述制备方法制得的MOF微针贴片,包括MOF微针贴片的基底部和针体部,制成的MOF微针贴片质地柔软、富有弹性,并且负载的MOF材料能够有效的抗菌,从而有助于创面的愈合过程。
本发明还提供了以上所述的MOF微针贴片在创面修复领域中的应用。
本发明的有益效果:
(1)本发明提供的用于创面修复的MOF微针贴片的制备方法,通过PDMS微针模具填充待固化MOF微针贴片原材料溶液和紫外光固化的方法,制备得到了针体柔软、多孔并负载MOF材料的微针贴片,制备方法简单、可靠;
(2)本发明提供的MOF微针贴片针体柔软,有助于保护创面,防止创面受到二次损伤,从而有利于创面愈合;
(3)本发明采用甲基丙烯化透明质酸(MeHA)或甲基丙烯化明胶(GelMA)以及MOF材料作为MOF微针贴片原材料溶液主要成分,有利于发挥MOF微针贴片在创面愈合中的抗感染、减轻炎症以及促血管生成的功能,最终有助于创面的快速愈合。

本发明提供一种MOF微针贴片的制备方法,包括以下步骤:
S1、MOF微针贴片原材料溶液的制备
用研钵将MOF材料粗颗粒研磨成MOF材料微颗粒,随后将MOF材料微颗粒与磷酸缓冲盐溶液(PBS)混合,置于超声波清洗机中超声3小时以均匀分散,得到含有MOF材料微颗粒的磷酸缓冲盐溶液(PBS)溶剂,然后将上述溶剂与微针原材料充分混匀,并向其中加入光引发剂(1% (v/v) 的2-羟基-2-甲基苯丙酮),获得待固化MOF微针贴片原材料溶液,之后避光保存在-4℃冰箱备用;
S2、MOF微针贴片的制备
在PDMS微针模具(由ETPTA微针模板翻模制得)上滴加待固化MOF微针贴片原材料溶液,随后通过高速离心法使MOF微针贴片原材料溶液填充PDMS微针模具的针体,离心后除去模具表面多余的液体,通过紫外光固化的方法(100W紫外光下照射1分钟)使模具内部的MOF微针贴片原材料溶液固化成型,固化期间需将模具置于冰上防止固化过程中材料的变形;待材料充分固化后从模具上剥离,得到MOF微针贴片,所述MOF微针贴片呈圆形,针体为圆锥形,针体的底部直径为310μm,高度为700μm。
本发明中,所述微针原材料选自甲基丙烯化透明质酸(MeHA)、甲基丙烯化明胶(GelMA)及聚乙二醇二丙烯酸酯(PEGDA)中的一种或几种;所述MOF材料选自ZIF-8、HKUST-1、Zn-Vitamin MOF及 Cu-Vitamin MOF 中的一种或多种;所述光引发剂为2-羟基-2-甲基苯丙酮;MOF微针贴片原材料溶液在固化之前需要避光放置。
本发明S2中,先用 0.1% (w/v) 的MOF微针贴片原材料溶液作为灌洗液对PDMS微针模具进行灌洗,用紫外光固化仪在模具上照射1分钟,随后清除灌洗液后再进行MOF微针贴片的制作。
基于上述制备方法制备的用于创面修复的MOF微针贴片,包括由甲基丙烯化透明质酸(MeHA)或甲基丙烯化明胶(GelMA)制成的MOF微针贴片,包括基底部和针体部,针尖柔软,内部多孔结构明显;MOF材料微颗粒负载在针尖内部,通过释放金属离子发挥抗菌性能。这种MOF微针贴片在创面愈合中可以发挥抗感染、减轻炎症以及促血管生成的功能,最终有助于创面的快速愈合。
本发明基于MOF材料优异的抗菌性能,通过PDMS模具复制和紫外光固化的方法,将MOF微针贴片原材料溶液固化成微针的形态,从而制得MOF微针贴片,制备过程简单、有效。
实施例1
一种以甲基丙烯化透明质酸(MeHA)和ZIF-8微颗粒作为主成分的ZIF-8微针贴片,按照如下方法制备:
S1、配置待固化ZIF-8微针贴片原材料溶液作为浇注材料
用研钵将ZIF-8粗颗粒研磨成微颗粒,然后使用pH=6的磷酸缓冲盐溶液 (PBS) 与ZIF-8微颗粒混合并置于超声波清洗机中超声3小时以均匀分散,从而获得含有ZIF-8的PBS溶剂;再将该溶剂与3% (w/v)的 MeHA 、1% (v/v)的2-羟基-2-甲基苯丙酮和0.068% (w/v)的N-N-二甲基丙烯酰胺充分混匀以获得待固化的ZIF-8微针贴片原材料溶液,该待固化ZIF-8微针贴片原材料溶液在紫外光照射下1分钟可使其固化成型,避光保存在-4℃冰箱备用;
S2、ZIF-8微针贴片的制备
首先,用0.1% (w/v)的 MeHA、1% (v/v) 的2-羟基-2-甲基苯丙酮以及磷酸盐缓冲液(PBS)作为灌洗液滴加在PDMS微针模具(385个针体)的表面,在100W紫外固化仪下照射1分钟以灌洗模具,然后用移液枪吸出灌洗液;将待固化ZIF-8微针贴片原材料溶液滴加在PDMS微针模具(385个针体)的表面,用水平高速离心机充分离心5分钟使待固化ZIF-8微针贴片原材料溶液充分填充到PDMS微针模具的针状孔2-2内;然后在 100W 紫外光固化仪下照射1分钟使ZIF-8微针贴片原材料溶液固化,期间需将模具置于冰上防止固化过程中材料的变形;最后可分离出ZIF-8微针贴片与PDMS微针模具,如图3所示。
实施例2
一种以甲基丙烯化明胶(GelMA)作为主成分的ZIF-8微针贴片,按照如下方法制备:
(1)配置待固化ZIF-8微针贴片原材料溶液作为浇注材料
用研钵将ZIF-8粗颗粒研磨成微颗粒,然后用pH=6的磷酸缓冲盐溶液 (PBS) 与ZIF-8微颗粒混合并置于超声波清洗机中超声3小时以均匀分散,从而获得含有ZIF-8微颗粒的PBS溶剂;将该溶剂与10% (w/v) GelMA 、10% (v/v) PEGDA以及1%(v/v) 2-羟基-2-甲基苯丙酮混合作为待固化微针贴片原材料溶液并避光放置;该待固化微针原材料溶液在紫外光照射下 1 分钟可使微针原材料溶液固化;
(2)ZIF-8微针贴片的制备
首先,将0.1% (w/v) 的GelMA 溶液作为灌洗液加在PDMS微针模具(400个针体)的表面,在100W紫外固化仪下照射1分钟以灌洗模具,然后用移液枪吸出灌洗液;将ZIF-8微针贴片原材料溶液滴加在PDMS微针模具(400个针体)的表面,通过高速离心法使待固化微针原材料溶液充分填充到PDMS微针模具的针状孔内。然后在 100W 紫外光固化仪下照射 1分钟,使待固化微针原材料溶液固化;最后与PDMS微针模具分离得到ZIF-8微针贴片。
试验例
用于创面修复的ZIF-8微针贴片作用于大鼠背部圆形创面:
以实施例1制备的ZIF-8微针贴片为例,用10% (w/v)水合氯醛腹腔注射6只重量150-200g 的雌性大鼠,背部人为制造直径为1cm 的圆形创面;
将上述大鼠模型随机分配为对照组和实验组,每组3只,对照组:不接受任何治疗;实验组:把甲基丙烯化透明质酸(MeHA)和ZIF-8微颗粒作为主成分的ZIF-8微针贴片敷在创面表面,然后用1624W透明敷贴固定ZIF-8微针贴片防止微针脱落,通过甲基丙烯化透明质酸缓慢降解作用以及ZIF-8微颗粒释放的锌离子的抗菌作用进行创面修复;治疗9天后,从大鼠背部创面取样进行 HE 染色。
如图5所示的HE 染色结果可知,对照组皮肤的创面面积更大,几乎未见上皮化;而实验组创面面积小,可见大量上皮再生,皮肤恢复较快,表明ZIF-8微针贴片有利于创面的愈合。
因此,本发明通过聚二甲基硅氧烷(PDMS)模具复制和紫外光固化的方法,以甲基丙烯化透明质酸 (MeHA) 或甲基丙烯化明胶(GelMA)为针体主要成分,制备得到针体柔软、多孔并负载MOF材料的微针贴片,对于减轻创面细菌污染、促进创面修复过程具有积极作用。
以上仅是本发明的优选实施方式,本发明的保护范围并不仅局限于上述实施例,凡属于本发明思路下的技术方案均属于本发明的保护范围,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理前提下的若干改进和润饰,应视为本发明的保护范围。
Claims (10)
1.一种MOF微针贴片的制备方法,其特征在于,包括以下步骤:
S1、MOF微针贴片原材料溶液的制备
将MOF材料微颗粒与磷酸缓冲盐溶液混合,置于超声波清洗机中超声3小时以均匀分散,得到含有MOF材料微颗粒的磷酸缓冲盐溶液溶剂,然后将上述溶剂与微针原材料充分混匀,并向其中加入光引发剂,获得待固化MOF微针贴片原材料溶液,之后避光保存在-4℃冰箱备用;
S2、MOF微针贴片的制备
在PDMS微针模具上涂抹待固化MOF微针贴片原材料溶液,随后通过高速离心法使MOF微针贴片原材料溶液充分填充到PDMS微针模具的针体,离心后除去模具表面多余的液体,通过紫外光固化的方法使模具内部的MOF微针贴片原材料溶液固化成型;待材料充分固化后从模具上剥离,得到MOF微针贴片。
2.根据权利要求1所述的MOF微针贴片的制备方法,其特征在于,S1中,所述微针原材料选自甲基丙烯化透明质酸、甲基丙烯化明胶及聚乙二醇二丙烯酸酯中的一种或几种。
3.根据权利要求1所述的MOF微针贴片的制备方法,其特征在于,S1中,所述MOF材料选自ZIF-8、HKUST-1、Zn-Vitamin MOF及 Cu-Vitamin MOF 中的一种或多种。
4.根据权利要求1所述的MOF微针贴片的制备方法,其特征在于,S1中,所述光引发剂为2-羟基-2-甲基苯丙酮。
5.根据权利要求1所述的MOF微针贴片的制备方法,其特征在于,S2中,固化过程中将模具置于冰上,以防止固化过程中材料的变形。
6.根据权利要求1所述的MOF微针贴片的制备方法,其特征在于,S2中,先用MOF微针贴片原材料溶液作为灌洗液对PDMS微针模具进行灌洗,用紫外光固化仪在模具上照射1分钟,随后清除灌洗液再进行MOF微针贴片的制作。
7.根据权利要求1所述的MOF微针贴片的制备方法,其特征在于,MOF微针贴片原材料溶液在固化之前需低温避光存放。
8.根据权利要求1所述的MOF微针贴片的制备方法,其特征在于,所述MOF微针贴片呈圆形,针体为圆锥形,针体的底部直径为310μm,高度为700μm。
9.一种采用权利要求1-8任一项所述的制备方法制得的MOF微针贴片,其特征在于,包括柔软多孔的基底部、针体部以及针体多孔材料内部负载的MOF微颗粒。
10.权利要求1~9任意一项所述的MOF微针贴片在创面修复领域中的应用。
