技术领域
本发明涉及一株青贮菌株及其应用。
背景技术
黑麦草是一种优质牧草,叶多质嫩,适口性好,可直接喂养草食动物,也可以与精料混合饲喂猪、兔、家禽和鱼等养殖动物。黑麦草青贮可解决供求上出现的季节不平衡和地域不平衡问题,也可解决盛产期雨季不宜调制干草的困难,并获得较青刈玉米品质更为优良的青贮料。发酵良好的青贮黑麦草,具有浓厚的醇甜水果香味,是最佳的冬季饲料。黑麦草营养价值高,富含蛋白质、矿物质和维生素,粗蛋白4.93%,粗脂肪1.06%,无氮浸出物4.57%,钙0.075%,磷0.07%,其中粗蛋白、粗脂肪比本地杂草含量高出3倍。
黑麦草青贮过程一直采用自然青贮或商品通用青贮接种剂,由于青草中乳酸菌菌体丰度低、外源乳酸菌存在适应性弱与竞争性低等问题,黑麦草青贮过程中有时会出现比较大的物料损失甚至失败。
发明内容
本发明提供一株黑麦草青贮菌株,该菌株具有产酸多、生长速度快等优点。
一株黑麦草青贮菌株,其为植物乳杆菌(Lactobacillus plantarum)1707,保藏在中国微生物菌种保藏管理委员会普通微生物中心,保藏号为CGMCC No.20962。
上述黑麦草青贮菌株在黑麦草青贮中的应用。
进一步地,在黑麦草中加入稻糠,再接种植物乳杆菌(Lactobacillus plantarum)1707进行青贮。
青贮过程的基本环境适宜乳酸菌的生长,但前期物料中(特别是表面)会残存空气,因此一些好氧的微生物与乳酸菌一起生长,竞争利用物料中的养分,造成物料腐败损失及后期pH的升高。因此,选用本发明黑麦草青贮菌株可提高青贮饲料中乳酸菌的起始丰度,而且生长速度快,能够加速青贮过程中起始阶段乳酸菌的生长产酸,对青贮有重要意义。
黑麦草中水分含量比较大,直接接种青贮,致使浸出水较多,易造成丁酸菌繁殖产生丁酸,使得青贮品质下降甚至失败,因此本发明加入稻糠以调整物料水分。
本发明植物乳杆菌(Lactobacillus plantarum)1707于2017年7月分离自黑龙江省安达市,黑龙江蓬勃牧草有限公司的黑麦草青贮物料。
植物乳杆菌(Lactobacillus plantarum)1707为植物乳杆菌,属于乳杆菌属(Lactobacillus);保藏在中国微生物菌种保藏管理委员会普通微生物中心(CGMCC),保藏地址是北京市朝阳区北辰西路1号院3号,中国科学院微生物研究所,保藏编号为CGMCCNo.20962,保藏日期为2020年10月29日。
具体实施方式
实施例1
4%葡萄糖MRS液体培养基在108℃下灭菌处理15min,接种植物乳杆菌(Lactobacillus plantarum)1707后静止保温28℃培养48hr后检测总酸,总酸采用酸碱滴定法。
植物乳杆菌(Lactobacillus plantarum)1707的产酸量为3.24±0.24%。
实施例2
黑麦草青草在组织捣碎机中加3倍水捣碎,双层纱布过滤,过滤液中补加0.5%葡萄糖,接种植物乳杆菌(Lactobacillus plantarum)1707,静止保温28℃培养,在规定时间检测有效活菌数。
植物乳杆菌(Lactobacillus plantarum)1707培养12hr有效活菌数(0.92±0.17)×108cfu/ml,培养24hr有效活菌数(1.51±0.17)×108cfu/ml,培养36hr有效活菌数(1.49±0.17)×108cfu/ml。
实施例3
黑麦草鲜草切碎至3~5cm,在108℃下灭菌处理15min,冷却后分别接种2.5%的植物乳杆菌(Lactobacillus plantarum)1707菌液和市售青贮接种剂商品(植物乳杆菌)菌液,搅拌混合均匀,分装小袋每袋500g后,抽真空后封口,置28℃下恒温培养一定时间后,检测总酸。
植物乳杆菌(Lactobacillus plantarum)1707培养12hr总酸(以乳酸计%干基)为1.41±0.13%,培养24hr总酸(以乳酸计%干基)为5.50±0.28%,培养36hr总酸(以乳酸计%干基)为5.67±0.13%,培养48hr总酸(以乳酸计%干基)为5.64±0.13%,培养72hr总酸(以乳酸计%干基)为5.66±0.12%。
市售青贮接种剂商品(植物乳杆菌)培养12hr总酸(以乳酸计%干基)为1.00±0.15%,培养24hr总酸(以乳酸计%干基)为4.25±0.13%,培养36hr总酸(以乳酸计%干基)为5.40±0.25%,培养48hr总酸(以乳酸计%干基)为5.50±0.12%,培养72hr总酸(以乳酸计%干基)为5.52±0.20%。
植物乳杆菌(Lactobacillus plantarum)1707产酸在24hr后趋于平稳,而市售青贮接种剂商品(植物乳杆菌)产酸明显滞后,在36hr后趋于平稳,达到峰值后二者差异不显著。表明植物乳杆菌(Lactobacillus plantarum)1707作为黑麦草青贮菌株比市售青贮接种剂商品更具优势。
实施例4
黑麦草鲜草切碎至3~5cm加入10%稻糠,在混合机内接种2.5%的植物乳杆菌(Lactobacillus plantarum)1707菌液和市售青贮接种剂商品(植物乳杆菌)菌液,搅拌均匀,通过打包机塑料膜密封打包,置室内自然青贮1个月,开包检查青贮效果。单生产批次鲜草加工量7吨,鲜草包重量约500公斤。
植物乳杆菌(Lactobacillus plantarum)1707和市售青贮接种剂商品(植物乳杆菌)对黑麦草进行青贮,在1个月青贮饲料的感官指标基本一致,无明显的腐败与大面积霉菌斑点,呈现出正常的青贮物料状态。表1的结果表明,使用本发明植物乳杆菌(Lactobacillus plantarum)1707对黑麦草+稻糠进行打包青贮,与市售青贮接种剂商品(植物乳杆菌)相比在pH、氨肽氮/总氮、中性洗涤纤维、酸性洗涤纤维等四项指标差异显著,呈正向变化;对有机酸乳酸、乙酸、丁酸指标差异不显著,乳酸与乙酸有小幅度提高。

2.如权利要求1所述的黑麦草青贮菌株在黑麦草青贮中的应用。
3.根据权利要求2所述的黑麦草青贮菌株在黑麦草青贮中的应用,其特征在于黑麦草中加入稻糠青贮。
4.根据权利要求3所述的黑麦草青贮菌株在黑麦草青贮中的应用,其特征在于稻糠的加入量为黑麦草重量的10±1%。
