本发明提供一种基于玫瑰花用于缓解癫痫的医用配方食品及其制备方法,每100mL所述医用配方食品中含有:蛋白质3~12 g,油脂5~20 g,碳水化合物2~8 g,玫瑰花水提液5~20 g,GABA 0.1~0.5 g,维生素预混料0.05~0.5 g,矿物质预混料1~10 g,大豆磷脂1~10 g,余量为水;将营养素和大豆磷脂分别在水和油脂中溶解得到水相和油相,将水相和油相混合在一起,经过剪切、均质、灭菌,得到所述的医用配方食品。本发明通过各种成分的协同作用,利用玫瑰花的抗氧化作用,辅以GABA对癫痫的缓解作用,延长癫痫的发作周期。

本发明涉及一种健康食品领域,尤其涉及一种基于玫瑰花用于缓解癫痫的医用配方食品及其制备方法,属于生物技术领域。
背景技术
癫痫是一种慢性神经系统疾病,目前我国约600万以上的癫痫患者,每年新发癫痫患者65万-70万。虽然目前癫痫的诊疗取得了很大的进展,但难治性癫痫患者的数量却在日益增多。
癫痫的发病是由中枢神经系统兴奋与抑制之间不平衡导致的,其主要与离子通道、神经递质及神经胶质细胞的改变有关。基因突变会导致离子通道功能异常,即有缺陷的基因编码有缺陷的离子通道蛋白而发病,其中钠离子、钾离子、钙离子通道与癫痫相关性的研究较为明确;当兴奋性神经递质过多或抑制性递质过少,都能使兴奋与抑制间失衡,使神经元过度放电,引起癫痫;当星形胶质细胞对谷氨酸或γ氨基丁酸的摄取能力发生改变时可导致癫痫发作。中枢神经系统中重要的抑制性神经性神经递质有γ-氨基丁酸(GABA)和甘氨酸,兴奋性神经递质包括谷氨酸(Glu)和天冬氨酸。

2009年,Freitas等(Freitas R M D, Nascimento K G D, Ferreira P M P, etal. Neurochemical changes on oxidative stress in rat hippocampus duringacutephase ofpilocarpine-induced seizures[J]. Pharmacology Biochemistry andBehavior, 2010, 94: 341-345.)通过大鼠实验证实,氧化应激在癫痫发病中起重要作用,且癫痫的持续时间与氧化损伤呈正相关。
玫瑰(Rosa rugosa Thunb.)为多年生落叶性灌木,属蔷薇科(Rosaceae)蔷薇属(Rosa)植物,主产于法国、俄罗斯、保加利亚等国,我国主要种植区为山东、甘肃、云南等地。在我国,玫瑰花还是一种具有悠久历史的药食同源植物。2010年,我国卫生部已批准重瓣红玫瑰(Rose rugosa cv. plena)作为普通食物资源用于食品制作和加工。据中国药典记载,玫瑰花蕾性温味甘,具有活血、调经、镇静安神、健脾以及治疗跌打损伤等功效。
玫瑰花的提取方式主要分为水提取法、有机溶剂提取法。张莲莉等(张莲莉, 戴伟锋,覃豪, 等. 三种玫瑰花茶中黄酮类成分的分析及其抗氧化活性研究[J]. 昆明理工大学学报(自然科学版), 2019, 44(05): 84-90.)三种玫瑰花茶汤中黄酮类成分含量均分别高于其甲醇提取液;金边和平阴玫瑰花茶汤的抗氧化活性均高于其甲醇提取液,苦水玫瑰茶汤抗氧化活性低于其甲醇提取液。利用有机溶剂提取法得到的玫瑰花提取液不适用添加到食品中。
玫瑰花中主要的活性成分包括挥发油、多酚类、黄酮类、多糖类等多种化合物,具有减少和消除自由基、抗氧化和抗菌等作用。

玫瑰花具有消除自由基和抗氧化的作用,可以缓解氧化应激,从而缓解癫痫,基于这一原理,配制一种基于玫瑰花用于缓解癫痫的医用配方食品。
发明内容
本发明提供一种基于玫瑰花用于缓解癫痫的医用配方食品及其制备方法,目的在于利用玫瑰花的抗氧化作用对氧化应激的影响,辅以GABA作为脑中抑制性神经递质对癫痫的缓解,延长患者的癫痫发作周期。
本发明提供一种基于玫瑰花用于缓解癫痫的医用配方食品为乳剂,制备步骤如下:
①医用配方食品复配
所述医用配方食品每100 mL含有:
蛋白质3~12 g,
油脂5~20 g,
碳水化合物2~8 g,
玫瑰花水提液5~20 g,
GABA 0.1~0.5 g,
维生素预混料0.05~0.5 g,
矿物质预混料1~10 g,
大豆磷脂1~10 g,
余量为水。
所述蛋白质为浓缩乳清蛋白、水解乳清蛋白、大豆分离蛋白以及酪蛋白酸钠的一种或几种的混合物。
所述油脂为大豆油、花生油、亚麻籽油、紫苏籽油以及中链甘油三酯中的一种或几种混合物。
所述玫瑰花水提液为玫瑰花按照料液比1:10~1:25(g:mL),温度70~90℃,时间10~20 min浸泡后过滤得到的滤液,利用旋转蒸发仪浓缩,浓缩后滤液中固含物40~60%,多糖类0.1~0.5 g/L,黄酮类0.5~1.5 g/L,多酚类150~200 g/L。
所述矿物质预混料1 g中包含碳酸钙30~80 mg、氯化镁15~20 mg、食用盐50~100mg、氯化钾80~150 mg、磷酸二氢钾20~60 mg、一水合硫酸亚铁0.5~1.0 mg、一水合硫酸锌0.2~0.6 mg、亚硒酸钠0.001~0.01 mg、无水硫酸铜0.03~0.06 mg、柠檬酸锰0.01~0.05 mg、碘化钾0.005~0.01 mg,余量为矿物质预混料载体。
所述矿物质预混料载体为麦芽糊精。
所述维生素预混料1 g中包含维生素A 2~5 mg、维生素D 0.03~0.06 mg、抗坏血酸250~500 mg、维生素E 30~60 mg、维生素K1 0.1~0.5 mg、硫胺素3~6 mg、核黄素2~6 mg、烟酰胺8~15 mg、泛酸15~20 mg、盐酸吡哆醇3~6 mg、叶酸0.5~2.5 mg、维生素B12 0.003~0.020 mg、生物素0.05~0.25 mg,余量为维生素预混料载体。
所述维生素预混料载体为麦芽糊精。
②医用配方食品制备
水相:将蛋白质、碳水化合物、矿物质预混料、维生素预混料、GABA和玫瑰花提取液加入到30~60℃的蒸馏水中,充分搅拌至溶解;
油相:将乳化剂加入到油脂中,充分搅拌至溶解;
将水相与油相混合均匀,利用高速剪切机混合均匀,转速为1500~3000 r/min,剪切时间为5~20 min,得到初乳;
利用高压均质机对初乳进行均质处理,均质条件为:200~800 bar,均质2~8次;
均质完成后,灌装灭菌,灭菌条件为:110~125℃,10~30 min;取出并冷却至室温,即得到医用配方食品。
本发明提供的医用配方食品含有大豆分离蛋白和浓缩乳清蛋白等蛋白质,具有丰富的氮源,更好的补充所需要的蛋白质,富含中链脂肪酸、DHA等不饱和脂肪酸,对癫痫的氧化应激有重要的抑制作用。
本发明提供的医用配方食品特别添加GABA和玫瑰花提取液,GABA可以增加脑中GABA的含量,维持脑中的兴奋性神经递质与抑制性神经递质之间的平衡,玫瑰花提取液具有减少和消除自由基、抗氧化等作用,缓解氧化应激,达到缓解癫痫的效果。
本发明提供的医用配方食品采用麦芽糊精等低血糖生成指数(GI)的碳水化合物,有助于病人血糖的控制。
本发明提供的医用配方食品是一种复合食品,其结合了大豆分离蛋白、浓缩乳清蛋白等蛋白质,补充所需要的蛋白质,结合色氨酸缓解癫痫患者的氧化应激,结合麦芽糊精等低GI的碳水化合物,控制病人血糖,通过各种成分的协同作用,缓解癫痫患者的氧化应激,提高患者的免疫力,从而缓解癫痫的发病。该癫痫患者特殊医学用途配方食品可以作为营养食品为难治性癫痫患者提供营养支持,也可以作为普通癫痫患者的营养支持。

水相:将大豆分离蛋白、麦芽糊精、矿物质预混料、维生素预混料、GABA和玫瑰花提取液加入到40℃的蒸馏水中,充分搅拌至溶解;
油相:将大豆磷脂加入到亚麻籽油中,充分搅拌至溶解;
将水相与油相混合均匀,利用高速剪切机混合均匀,转速为2500 r/min,剪切时间为15 min,得到初乳;
利用高压均质机对初乳进行均质处理,均质条件为:600 bar,均质4次;
均质完成后,灌装灭菌,灭菌条件为:115℃,20 min;取出并冷却至室温,即得到医用配方食品。
实施例2:
①医用配方食品复配
所述医用配方食品每100 mL含有:
浓缩乳清蛋白3 g,
紫苏籽油5 g,
低聚果糖2 g,
玫瑰花水提液5 g,
GABA 0.1 g,
维生素预混料0.1 g,
矿物质预混料2 g,
大豆磷脂2 g,
余量为水;
所述玫瑰花水提液为玫瑰花按照料液比1:10(g:mL),温度70℃,时间20 min浸泡后过滤得到的滤液,利用旋转蒸发仪浓缩,浓缩后滤液中固含物60%,多糖类0.1 g/L,黄酮类0.5 g/L,多酚类150 g/L;
所述矿物质预混料1g包含碳酸钙56 mg、氯化镁18.3 mg、食用盐83 mg、氯化钾111mg、磷酸二氢钾40 mg、一水合硫酸亚铁0.83 mg、一水合硫酸锌0.4 mg、亚硒酸钠0.003 mg、无水硫酸铜0.044 mg、柠檬酸锰0.025 mg、碘化钾0.007 mg,余量为麦芽糊精;
所述维生素预混料1 g包含维生素A 3.6 mg、维生素D 0.048 mg、抗坏血酸336mg、维生素E 48 mg、维生素K1 0.264 mg、硫胺素4.2 mg、核黄素4.2 mg、烟酰胺12 mg、泛酸17.4 mg、盐酸吡哆醇4.2 mg、叶酸1.332 mg、维生素B12 0.0078 mg、生物素0.132 mg,余量为麦芽糊精。所述维生素预混料载体为麦芽糊精;
②医用配方食品制备
水相:将浓缩乳清蛋白、低聚果糖、矿物质预混料、维生素预混料、GABA和玫瑰花提取液加入到30℃的蒸馏水中,充分搅拌至溶解;
油相:将大豆磷脂加入到紫苏籽油中,充分搅拌至溶解;
将水相与油相混合均匀,利用高速剪切机混合均匀,转速为1500 r/min,剪切时间为20 min,得到初乳;
利用高压均质机对初乳进行均质处理,均质条件为:200 bar,均质8次;
均质完成后,灌装灭菌,灭菌条件为:110℃,30 min;取出并冷却至室温,即得到医用配方食品。
实施例3:
①医用配方食品复配
所述医用配方食品每100 mL含有:
酪蛋白酸钠12 g,
中链甘油三酯20 g,
麦芽糊精8 g,
玫瑰花水提液20 g,
GABA 0.5 g,
维生素预混料0.5 g,
矿物质预混料10 g,
大豆磷脂8 g,
余量为水;
所述玫瑰花水提液为玫瑰花按照料液比1:30(g:mL),温度90℃,时间10 min浸泡后过滤得到的滤液,利用旋转蒸发仪浓缩,浓缩后滤液中固含物40%,多糖类0.5 g/L,黄酮类1.5 g/L,多酚类200 g/L;
所述矿物质预混料1g包含碳酸钙56 mg、氯化镁18.3 mg、食用盐83 mg、氯化钾111mg、磷酸二氢钾40 mg、一水合硫酸亚铁0.83 mg、一水合硫酸锌0.4 mg、亚硒酸钠0.003 mg、无水硫酸铜0.044 mg、柠檬酸锰0.025 mg、碘化钾0.007 mg,余量为麦芽糊精;

②医用配方食品制备
水相:将酪蛋白酸钠、麦芽糊精、矿物质预混料、维生素预混料、GABA和玫瑰花提取液加入到50℃的蒸馏水中,充分搅拌至溶解;
油相:将大豆磷脂加入到中链甘油三酯中,充分搅拌至溶解;
将水相与油相混合均匀,利用高速剪切机混合均匀,转速为3000 r/min,剪切时间为10 min,得到初乳;
利用高压均质机对初乳进行均质处理,均质条件为:800 bar,均质2次;
均质完成后,灌装灭菌,灭菌条件为:121℃,10 min;取出并冷却至室温,即得到医用配方食品。
本发明提供的医用配方食品对小鼠癫痫发病的影响:
选用昆明小鼠,雄性,体重18-22 g,分为4组,每组10只,首先构建癫痫小鼠模型。1-3组为实验组分别用实施例1-3医用配方食品进行灌胃;4组为对照组,灌胃生理盐水,正常饲养。按照2.5 mL/10 g剂量每天灌胃小鼠,连续7天。每天称1次体重,并观察发病情况,最后一次灌胃1 h后眼球取血,利用丙二醛检测试剂盒、过氧化物歧化酶检测试剂盒、谷胱甘肽过氧化物酶检测试剂盒、SOD商业检测试剂盒检测丙二醛的含量和过氧化物歧化酶、谷胱甘肽过氧化物酶、SOD的酶活。医用配方食品对小鼠体重的影响结果见表1,对癫痫的发病影响结果见表2,对丙二醛含量和酶活影响见表3。

由表1可知,与阴性对照组比较,实施例1-3组小鼠体重增加无显著性差异(P>0.05)。可见,该制剂对小鼠体重增长未见明显影响,可以维持小鼠的正常生理功能。

由表2可知,本发明提供的医用配方食品可以显著延长小鼠癫痫发作周期,有利于缓解癫痫。

由表3可知,本发明提供的医用配方食品相对于阴性对照组可以提高过氧化物歧化酶、谷胱甘肽过氧化物酶、SOD的酶活和降低丙二醛的含量,可以缓解小鼠癫痫。

Claims (8)

1.①医用配方食品复配
所述医用配方食品每100 mL含有:
蛋白质3~12 g,
油脂5~20 g,
碳水化合物2~8 g,
玫瑰花水提液5~20 g,
GABA 0.1~0.5 g,
维生素预混料0.05~0.5 g,
矿物质预混料1~10 g,
大豆磷脂1~10 g,
余量为水;
②医用配方食品制备
水相:将蛋白质、碳水化合物、矿物质预混料、维生素预混料、GABA和玫瑰花提取液加入到30~50℃的蒸馏水中,充分搅拌至溶解;
油相:将乳化剂加入到油脂中,充分搅拌至溶解;
将水相与油相混合均匀,利用高速剪切机混合均匀,转速为1500~3000 r/min,剪切时间为10~20 min,得到初乳;
利用高压均质机对初乳进行均质处理,均质条件为:200~800 bar,均质2~8次;
均质完成后,灌装灭菌,灭菌条件为:110~125℃,10~30 min;取出并冷却至室温,即得到医用配方食品。
2.根据权利要求1所述的医用配方食品,其特征在于,所述蛋白质为浓缩乳清蛋白、水解乳清蛋白、大豆分离蛋白以及酪蛋白酸钠的一种或几种的混合物。
3.根据权利要求1所述的医用配方食品,其特征在于,所述油脂为大豆油、花生油、亚麻籽油、紫苏籽油以及中链甘油三酯中的一种或几种混合物。
4.根据权利要求1所述的医用配方食品,其特征在于,所述碳水化合物为麦芽糊精和低聚果糖中的一种或两种的混合物。
5.根据权利要求1所述的医用配方食品,其特征在于,所述玫瑰花水提液为玫瑰花按照料液比1:10~1:25(g:mL),温度70~90℃,时间10~20 min浸泡后过滤得到的滤液,利用旋转蒸发仪浓缩,浓缩后滤液中固含物40~60%,多糖类0.1~0.5 g/L,黄酮类0.5~1.5 g/L,多酚类150~200 g/L。
6.根据权利要求1所述的医用配方食品,其特征在于,所述矿物质预混料1 g中包含碳酸钙30~80 mg、氯化镁15~20 mg、食用盐50~100 mg、氯化钾80~150 mg、磷酸二氢钾20~60mg、一水合硫酸亚铁0.5~1.0 mg、一水合硫酸锌0.2~0.6 mg、亚硒酸钠0.001~0.01 mg、无水硫酸铜0.03~0.06 mg、柠檬酸锰0.01~0.05 mg、碘化钾0.005~0.01 mg,余量为矿物质预混料载体。
7.根据权利要求1所述的医用配方食品,其特征在于,所述维生素预混料1 g中包含维生素A 2~5 mg、维生素D 0.03~0.06 mg、抗坏血酸250~500 mg、维生素E 30~60 mg、维生素K1 0.1~0.5 mg、硫胺素3~6 mg、核黄素2~6 mg、烟酰胺8~15 mg、泛酸15~20 mg、盐酸吡哆醇3~6 mg、叶酸0.5~2.5 mg、维生素B12 0.003~0.020 mg、生物素0.05~0.25 mg,余量为维生素预混料载体。
8.根据权利要求1所述的医用配方食品,其特征在于,剪切转速为1500~3000 r/min,剪切时间为10~20 min;均质条件为:200~800 bar,均质2~8次;灭菌条件为:110~125℃,10~30min。
