Abstract
The invention provides a piglet diarrhea resistant probiotic preparation which comprises the following components in parts by weight: 10-15 parts of freeze-dried powder of bacillus subtilis, 4-6 parts of freeze-dried powder of bacillus licheniformis and 1-3 parts of folium artemisiae argyi fermentation freeze-dried powder; the preservation number of the bacillus subtilis is CGMCC No.9660, and the content of viable bacteria in the freeze-dried powder of the bacillus subtilis is more than or equal to 40 hundred million/g; the preservation number of the bacillus licheniformis is CGMCC No.5686, and the viable bacteria content in the freeze-dried powder of the bacillus licheniformis is more than or equal to 50 hundred million/g. The invention also provides a method for preparing the preparation and application of the preparation in preparing medicaments for preventing piglet diarrhea. The piglet diarrhea resistant probiotic preparation can obviously reduce the incidence rate of piglet diarrhea and has good practical application and market prospect.

In the following examples, the folium artemisiae argyi fermentation freeze-dried powder is obtained by the following preparation method:
mixing the ground folium artemisiae argyi powder and the boiled potatoes according to the weight ratio of 3:1, adding sterile water to enable the weight ratio of solid to liquid to be 2.5:1, inoculating 2% of Aspergillus niger YM33182 to ferment, and fermenting at 28-32 ℃ for 36 hours; draining the fermented product, and freeze-drying to obtain grape seed fermented dry powder; the Aspergillus niger YM33182 is deposited as follows: CCTCC No. M206H 3.
Example 1
1. Experimental animals, namely, weaning Jiangxiang pigs and piglets from a Jiangxiang local farm.
2. The feed additive comprises the following components: lyophilized powder of Bacillus subtilis (with preservation number of CGMCC No.9660) with viable bacteria content of 40 hundred million/g, lyophilized powder of Bacillus licheniformis (with preservation number of CGMCC No.5686) with viable bacteria content of 50 hundred million/g, and folium Artemisiae Argyi fermented lyophilized powder.
3. Diarrhea incidence factor pretest
Basal feed formulation (w/w): 48% of corn, 10% of broken rice, 10% of bran, 10% of bean cake, 8% of fish meal, 2.5% of green material, 0.5% of calcium carbonate, 0.5% of calcium phosphate and 0.5% of salt.
The addition amount of the freeze-dried powder is 0.5 percent (w/w), the freeze-dried powder (the viable bacteria content is more than or equal to 40 hundred million/g) of bacillus subtilis (the preservation number is CGMCC No.9660), the freeze-dried powder (the viable bacteria content is more than or equal to 50 hundred million/g) of bacillus licheniformis (the preservation number is CGMCC No.5686) and the folium artemisiae argyi fermentation freeze-dried powder are respectively added into the basic feed and are respectively marked as an experimental group 1, an experimental group 2 and an experimental group 3.
80 piglets of the Jiangxiang pigs of the experimental animals are taken, the weaning day of the batch of piglets is 12-14 days after birth and breast milk feeding, the batch of piglets is divided into 4 groups, and 20 piglets in each group are respectively fed with basic feed (used as a control group), feed of the experimental group 1, feed of the experimental group 2 and feed of the experimental group 3. The feeding time was 50 days, the feeding was performed twice daily, water was freely drunk, and the incidence of diarrhea was examined, and the results are shown in Table 1. 4. Preliminary optimization experiment
The method comprises the steps of combining bacillus subtilis (with the preservation number of CGMCC No.9660) freeze-dried powder, bacillus licheniformis (with the preservation number of CGMCC No.5686) freeze-dried powder and folium artemisiae argyi fermentation freeze-dried powder in different modes to obtain a feed additive, adding the feed additive into a basic feed according to the addition of 0.5% (w/w), inspecting the influence of a mixture on the diarrhea incidence rate of piglets, feeding 20 experimental animals of each experimental group according to a feeding method of diarrhea incidence factor pre-test. The group settings and experimental results are shown in Table 2, Table 2
5. Optimization experiment
According to the pre-optimization experiment, the mixing of the bacillus subtilis (with the preservation number of CGMCC No.9660), the bacillus licheniformis (with the preservation number of CGMCC No.5686) and the folium artemisiae argyi fermentation freeze-dried powder has obvious influence on the incidence rate of piglet diarrhea. After a large number of groping experiments, the feed additive is prepared by adopting the following formula and is added into a basic feed (the addition amount is 0.5 percent (w/w)), and when the number of the experimental animals weaned from the fragrant pig in Yangtze river is 100, the number of diarrhea occurrence is only 1, namely the diarrhea occurrence rate is 1%.
The feed additive amount formula is as follows:
according to the weight portion, 12 portions of freeze-dried powder of bacillus subtilis, 5 portions of freeze-dried powder of bacillus licheniformis and 2 portions of folium artemisiae argyi fermentation freeze-dried powder.
6. Experiment on feed suitability
On the basis of the optimization experiment, the applicability of the feed additive screened by the optimization experiment to different feeds is tested, and the feed formula adopted by the experiment is as follows:
the formula of the feed 1 is as follows: 60 w/w% of rice bran, 30 w/w% of corn flour and 10 w/w% of current season vegetables; the formula of the feed 2 is as follows: 74.86% of corn, 5.5% of wheat bran, 17.5% of soybean meal, 0.8% of stone powder, 0.5% of calcium hydrophosphate, 0.3% of salt, 0.15% of sodium bicarbonate, 0.04% of lysine hydrochloride, 0.08% of choline, and,0.03% of sweetening agent and 0.24% of compound vitamin (consisting of vitamin E, vitamin B2, vitamin K, folic acid, pantothenic acid and nicotinic acid according to the weight ratio of 4:2:1:2:3: 3); the formula of the feed 3 is as follows: 65% of corn, 1.8% of wheat middling, 12.60% of fermented soybean meal, 14.17% of soybean meal, 2.8% of fish meal, 0.28% of oil extraction, 0.91% of stone powder, 1% of calcium hydrophosphate, 0.32% of salt, 0.06% of choline chloride, 0.05% of flavor element, composite premix (containing 100ml/kg of manganese, 1332mg/kg of zinc, 2664mg/kg of iron, 167mg/kg of copper, 5mg/kg of iodine, 5mg/kg of selenium, 57000IU/kg of vitamin A, vitamin D36600IU/kg, vitamin E366 IU/kg, vitamin B2 96mg/kg)1%。
240 weaned Jiangxiang piglets are selected as experimental animals and divided into 6 groups, 40 piglets in each group are fed according to an optimization experiment method, and the diarrhea incidence rate is inspected. Wherein, each feed is provided with a control group (not added with feed additives) and an experimental group (added with feed additives according to 0.5% (w/w)). The experimental results are shown in table 3, and the results show that the additive obtained by the invention can be applied to various feeds, and remarkably reduce the diarrhea incidence rate of piglets.

The formulation of the probiotic formulation (i.e. the feed additive described in example 1) for the adjustment of the anti-piglet diarrhea was as follows:
10 parts of freeze-dried powder of bacillus subtilis, 6 parts of freeze-dried powder of bacillus licheniformis and 1 part of freeze-dried fermented folium artemisiae argyi powder.
100 weaned Jiangxiang piglets are selected as experimental animals and are bred according to the method in the optimization experiment in the example 1, and the result shows that the diarrhea incidence rate is 2%.
Example 3
The formulation of the probiotic formulation (i.e. the feed additive described in example 1) for the adjustment of the anti-piglet diarrhea was as follows:
15 parts of freeze-dried powder of bacillus subtilis, 4 parts of freeze-dried powder of bacillus licheniformis and 3 parts of folium artemisiae argyi fermentation freeze-dried powder.

10-15 parts of freeze-dried powder of bacillus subtilis, 4-6 parts of freeze-dried powder of bacillus licheniformis and 1-3 parts of folium artemisiae argyi fermentation freeze-dried powder;
the preservation number of the bacillus subtilis is CGMCC No.9660, and the content of viable bacteria in the freeze-dried powder of the bacillus subtilis is more than or equal to 40 hundred million/g; the preservation number of the bacillus licheniformis is CGMCC No.5686, and the content of viable bacteria in the freeze-dried powder of the bacillus licheniformis is more than or equal to 50 hundred million/g;
the folium artemisiae argyi fermentation freeze-dried powder is prepared by the following preparation method:
