Abstract
本发明公开了一种酪氨酸酶抑制剂,其采用下述方法制备而成:(1)酶解:将牦牛的副产物进行脱脂和除杂处理,之后加入复合酶酶解液酶解,酶解结束后灭酶,冷却至常温后离心,收集上清液冷冻干燥得到酶解液冻干粉;(2)纯化:将酶解液冻干粉用超纯水溶解,然后用截留分子量为3000Da的超滤膜进行分离纯化,将超滤获得的3000Da以下的组分冷冻干燥得牦牛胶原蛋白肽,即为本发明酪氨酸酶抑制剂。本发明酪氨酸酶抑制剂具有较佳的酪氨酸酶抑制作用,同时绿色环保,无毒,活性稳定,在美容产品和护色剂领域具有良好的应用前景。

技术领域
本发明涉及一种酶抑制剂,具体涉及一种酪氨酸酶抑制剂及应用。
背景技术
酪氨酸酶是一种氧化酶,是机体内调节黑色素生成的限速酶,其活性的控制即可改变黑色素的合成与生成量。在美容行业中,酪氨酸酶抑制剂以被制备成各类的预防皮肤晒黑、增白、祛斑的美容产品和保健食品,在美白功效上得到广泛运用。然而,在保鲜行业中酪氨酸酶同样扮演着重要角色,是引起食品加工与储藏过程中酶促褐变的主要酶,尤其在一些蔬菜与水果的微加工中,过多的黑色素分泌,引起色泽与外观的褐变,不仅降低营养价值和食品品质更是造成经济损失。因此近年来通过降低酪氨酸酶活性,改善酶促褐变的情况得到越来越多的关注。
目前在防止酶促褐变的主要措施仍是酪氨酸酶抑制剂的运用,如半胱氨酸、抗坏血酸、柠檬酸、铁取代磷酸盐等已应用于食品的保鲜中,都取得了一定抗褐变成效。但随着生活品质的提高,人们对于绿色天然的理念追求越来越明显,因此开发一种大众能接受的绿色环保天然的抗酶促褐变的食品护色剂势在必行。
本发明从高原牦牛提取的胶原蛋白肽,安全性高,吸收性强,具有直接抑制酪氨酸酶的效果,更是一种近年流行的纯天然多肽抑制剂,可用于制备各类具有酪氨酸酶抑制作用的产品。
发明内容
针对上述问题,本发明的目的在于提供一种酪氨酸酶抑制剂及应用,本发明酪氨酸酶抑制剂具有较佳的酪氨酸酶抑制作用,同时绿色环保,无毒,活性稳定,在美容产品和护色剂领域具有良好的应用前景。
本发明通过下述技术方案实现。
一种酪氨酸酶抑制剂,其采用下述方法制备而成:
(1)酶解
将牦牛的副产物进行脱脂和除杂处理,之后加入复合酶酶解液酶解,酶解结束后灭酶,冷却至常温后离心,收集上清液冷冻干燥得到酶解液冻干粉;
(2)纯化
将酶解液冻干粉用超纯水溶解,然后用截留分子量为3000Da的超滤膜进行分离纯化,将超滤获得的3000Da以下的组分冷冻干燥得牦牛胶原蛋白肽,即为本发明酪氨酸酶抑制剂。
作为具体技术方案,所述步骤(1)中,牦牛的副产物为牦牛皮、牛骨中的一种或两者的混合物。
作为具体技术方案,所述步骤(1)中,酶解的具体条件为:于85~95℃水溶液中保温预变性2~4h,冷却至45~50℃后调节pH 8~9,之后加入复合酶酶解液酶解4~6h。
作为具体技术方案,所述步骤(1)中,复合酶解液为碱性蛋白酶和丝氨酸蛋白酶的复合物。
作为具体技术方案,所述酪氨酸酶抑制剂抑制酪氨酸酶的IC50为3.15mg/mL,且具有浓度依赖性。
一种如上述的酪氨酸酶抑制剂在食品天然护色剂中的应用。
作为具体技术方案,所述食品天然护色剂可抑制鲜切果蔬食品褐变。
作为具体技术方案,所述食品天然护色剂中含有质量分数为0.1%~1%的上述酪氨酸酶抑制剂,含有质量分数为0.05%~0.5%的维生素C。

本发明的有益效果为:
本发明发现高原牦牛提取的胶原蛋白肽纯化后具有直接抑制酪氨酸酶的效果,可用于制备护色剂剂防止鲜切果蔬的褐变,也可用于制备美白皮肤的化妆品。由于其生物法天然提取和可食性,作为护色剂具备较高的安全性,其无色无味和本身的营养功效,作为护色剂不影响食品的营养价值与口味,其良好的抑制酪氨酸酶活性和抗氧化作用,作为护色剂具有一定防褐变的协同作用,同时适合普通人群家用和企业生产加工使用,符合近年来人们追求的绿色环保安全理念。

具体实施方式
下面结合具体实施方式对本发明做进一步的说明,需要指出的是以下实施方式仅是以例举的形式对本发明所做的解释性说明,但本发明的保护范围并不仅限于此,所有本领域的技术人员以本发明的精神对本发明所做的等效的替换均落入本发明的保护范围。
实施例1
本发明酪氨酸酶抑制剂的制备方法如下:
(1)酶解
称取牦牛皮100g,加入质量浓度为5%的Na2CO3和5%的NaCl溶液进行脱脂和除杂处理,纯水洗净后,于85~95℃水溶液中保温预变性2~4h,冷却至45~50℃后,调节pH 8~9,加入复合酶酶解液酶解4~6h。酶解结束后,高温灭酶活,冷却至常温。冷却后于10000rpm离心20min,收集上清液,冷冻干燥得酶解液冻干粉。
(2)纯化
将酶解液冻干粉用超纯水溶解,然后用截留分子量为3000Da的超滤膜进行分离纯化,将超滤获得的3000Da以下的组分冷冻干燥,最终获得目的牦牛胶原蛋白肽9.8g,即为本发明酪氨酸酶抑制剂。
实施例2
本发明酪氨酸酶抑制剂牦对抑制酪氨酸酶的活性检测。
试剂:蘑菇酪氨酸酶(T3824-25KU)及左旋多巴(D9628-5G)均为sigma产品,其它试剂磷酸二氢钠和磷酸氢二钠均为国药集团上海化学试剂公司产品。
酪氨酸酶用0.2M磷酸缓冲液(pH=6.8)溶解配制成100U/mL,左旋多巴用0.2M磷酸缓冲液溶解配置成0.003M,将本发明酪氨酸酶抑制剂用0.2M磷酸缓冲液溶解配置成1、2、4、8、16mg/mL的不同浓度溶液,按表一的反应体系进行酶促反应检测其对酪氨酸酶的抑制作用。将PBS、酪氨酸酶和本发明酪氨酸酶抑制剂分别按表一体积加入A、B、C、D管,37℃烘箱孵育10min后加入底物左旋多巴,37℃再孵育10min停止反应,依次点入96孔板,酶标仪492nm下检测各管吸光值,结果见图1。本发明酪氨酸酶抑制剂的酪氨酸酶抑制率按以下公式计算:
酶抑制率(%)=[(B-A)-(D-C)]/(B-A)*100
由图1可知,不同浓度的本发明酪氨酸酶抑制剂(1、2、4、8、16mg/mL),对酪氨酸酶的抑制作用分别为3.59,32.7,52.9,64.4,77.8%,具有一定的浓度依赖性,IC50为3.15mg/mL。

本发明酪氨酸酶抑制剂在食品天然护色剂中应用时,抑制鲜切果蔬食品褐变的效果。
分别取本发明酪氨酸酶抑制剂1g和维生素C 0.5g,用1000ml的纯净水溶解配制成护色液。取新鲜、无机械损伤和无病害的马铃薯,水洗后去皮,切成薯条迅速吹干,将薯条完全侵泡在护色液中10min后捞出吹干,与不浸泡的相比,室温下放置24h可防止褐变,密封-20℃保存,可保护薯条不发生褐变。
实施例4
本发明酪氨酸酶抑制剂在食品天然护色剂中应用时,抑制鲜切果蔬食品褐变的效果。
分别取本发明酪氨酸酶抑制2g和维生素C1g,用1000ml的纯净水溶解配制成护色液。取新鲜、无机械损伤和无病害的苹果,洗干净后去皮切片,将苹果块完全浸泡在护色液中10min后捞出吹干,与不浸泡的相比,室温下放置24h可防止褐变,密封-20℃保存,可保护苹果块不发生褐变。
Claims (10)
1.一种酪氨酸酶抑制剂,其特征在于采用下述方法制备而成:
(1)酶解
将牦牛的副产物进行脱脂和除杂处理,之后加入复合酶酶解液酶解,酶解结束后灭酶,冷却至常温后离心,收集上清液冷冻干燥得到酶解液冻干粉;
(2)纯化
将酶解液冻干粉用超纯水溶解,然后用截留分子量为3000Da的超滤膜进行分离纯化,将超滤获得的3000Da以下的组分冷冻干燥得牦牛胶原蛋白肽,即为本发明酪氨酸酶抑制剂。
2.如权利要求1所述的一种酪氨酸酶抑制剂,其特征在于所述步骤(1)中,牦牛的副产物为牦牛皮、牛骨中的一种或两者的混合物。
3.如权利要求1所述的一种酪氨酸酶抑制剂,其特征在于所述步骤(1)中,酶解的具体条件为:于85~95℃水溶液中保温预变性2~4h,冷却至45~50℃后调节pH 8~9,之后加入复合酶酶解液酶解4~6h。
4.如权利要求1所述的一种酪氨酸酶抑制剂,其特征在于所述步骤(1)中,复合酶解液为碱性蛋白酶和丝氨酸蛋白酶的复合物。
5.如权利要求1所述的一种酪氨酸酶抑制剂,其特征在于所述酪氨酸酶抑制剂抑制酪氨酸酶的IC50为3.15mg/mL,且具有浓度依赖性。
6.一种如权利要求1所述的酪氨酸酶抑制剂在食品天然护色剂中的应用。
7.如权利要求6所述的酪氨酸酶抑制剂在食品天然护色剂中的应用,其特征在于所述食品天然护色剂可抑制鲜切果蔬食品褐变。
8.如权利要求6所述的酪氨酸酶抑制剂在食品天然护色剂中的应用,其特征在于所述食品天然护色剂中含有质量分数为0.1%~1%的权利要求1所述酪氨酸酶抑制剂,含有质量分数为0.05%~0.5%的维生素C。
9.一种如权利要求1所述的酪氨酸酶抑制剂在美白化妆品中的应用。
10.如权利要求9所述的酪氨酸酶抑制剂在美白化妆品中的应用,其特征在于所述美白化妆品包括美白洗面奶、美白面膜、美白面霜。
