Description
The technical field is as follows:
the invention relates to the technical field of plant beverages, and particularly relates to an anti-fatigue beverage prepared from Ampelopsis grossedentata leaves and a preparation method thereof.
Background art:
ampelopsis grossedentata is a Vitaceae ampelopsis plant, and the leaves of Ampelopsis grossedentata are approved by the former national health committee of 12 months and 24 days in 2013 to be a new resource food. The Ampelopsis grossedentata leaves and the extract thereof are rich in flavone, polysaccharide, various trace elements and amino acid, and have the effects of clearing away heat and toxic materials, enhancing resistance, resisting inflammation, inhibiting bacteria, resisting oxidation, enhancing immunity and the like. GABA, globally known as gamma-aminobutyric acid, is a non-proteinogenic amino acid, mainly found in vertebrates, plants and microorganisms. GABA is used as an inhibitory neurotransmitter in human body and has various effects of improving sleep, promoting relaxation, lowering blood pressure, relieving fatigue, improving emotion and the like. Sugar substitutes have a relatively high sweetness and low self-caloric content and can be used in products to reduce caloric intake. In addition, part of sugar substitutes have little influence on blood sugar and are very friendly to diabetes patients.
Although the beverage in the market is various, the demand of consumers for the anti-fatigue type beverage is high. The natural and healthy components are selected to create the product with the functions of eliminating fatigue, relieving pressure, resisting oxidation, improving immunity and reducing blood pressure for consumers, and the product has great significance.
The invention content is as follows:
the invention aims to overcome the defects of the prior art and provide the ampelopsis grossedentata leaf anti-fatigue beverage and the preparation method thereof.
The invention aims to provide an anti-fatigue beverage containing Ampelopsis grossedentata leaves, which comprises the following components: ampelopsis grossedentata leaf extract, sodium citrate, water, sodium carboxymethylcellulose, xanthan gum, gamma-aminobutyric acid (GABA) and a sweetener.
The invention provides an anti-fatigue beverage of Ampelopsis grossedentata leaves, which takes Ampelopsis grossedentata leaves and GABA as main functional raw materials. The Ampelopsis grossedentata leaf extract has the effects of enhancing immunity, resisting oxidation, clearing away heat and toxic materials, relieving sore throat, resisting inflammation and the like. GABA has effects of improving sleep, promoting relaxation, lowering blood pressure, relieving fatigue, improving emotion, etc., and has synergistic effect of Ampelopsis grossedentata leaf extract and GABA to greatly improve antifatigue effect. In addition, the product selects sugar substitute as sweetener, which can reduce calorie intake and control blood sugar level well.
Preferably, the ampelopsis grossedentata leaf anti-fatigue beverage comprises the following components in parts by mass: 0.1-1 part of ampelopsis grossedentata leaf extract, 0.05-0.3 part of sodium citrate, 400-700 parts of water, 0.01-0.5 part of sodium carboxymethylcellulose, 0.01-0.5 part of xanthan gum, 0.01-0.1 part of gamma-aminobutyric acid and 1-4 parts of sweetener. After the sodium carboxymethylcellulose and the xanthan gum are compounded and added into the anti-fatigue beverage of the Ampelopsis grossedentata leaves, the stability of a solution system is improved, so that the anti-fatigue beverage of the Ampelopsis grossedentata leaves is clearer and more transparent. The anti-fatigue beverage of ampelopsis grossedentata leaves is prepared by selecting pure water as the component.
Further preferably, the coating comprises the following components in parts by mass: 0.15-0.8 part of ampelopsis grossedentata leaf extract, 0.05-0.3 part of sodium citrate, 500-600 parts of water, 0.01-0.2 part of sodium carboxymethylcellulose, 0.01-0.1 part of xanthan gum, 0.02-0.07 part of gamma-aminobutyric acid and 1.7-2.6 parts of sweetener.
Preferably, the sweetener is selected from more than one of mogroside, stevioside, xylitol and erythritol.
The invention also protects a preparation method of the ampelopsis grossedentata leaf anti-fatigue beverage, which comprises the following steps:
(1) dissolving Ampelopsis grossedentata leaf extract in a part of water, and collecting supernatant to obtain Ampelopsis grossedentata leaf extract solution;
(2) adding the sodium carboxymethylcellulose and the xanthan gum into the rest water to be completely dissolved to obtain a solution of the sodium carboxymethylcellulose and the xanthan gum;
(3) and (3) uniformly mixing and dissolving the ampelopsis grossedentata leaf extract solution obtained in the step (1) and the solution of sodium carboxymethylcellulose and xanthan gum obtained in the step (2) with sodium citrate, gamma-aminobutyric acid and a sweetening agent, homogenizing, degassing, carrying out instantaneous sterilization at an ultra-high temperature (the sterilization temperature is 135 ℃ and the sterilization time is 10s), cooling and filling to obtain the ampelopsis grossedentata leaf anti-fatigue beverage.
Preferably, the temperature of the water in the step (1) is 80-100 ℃.
Preferably, the ampelopsis grossedentata leaf extract in the step (1) is prepared by the following steps: cleaning Ampelopsis grossedentata leaves, removing impurities, extracting with a solvent, filtering, concentrating, drying, and pulverizing or granulating to obtain Ampelopsis grossedentata leaf extract.

Experimental example 1: animal fatigue test of fatigue-resistant Ampelopsis grossedentata leaf beverage obtained in example 4
1. Laboratory animal
40 healthy male KM mice, weighing 18-22g, were bred adaptively for 5 days at room temperature of 23-25 ℃ and relative humidity of 55-60% before the experiment. Alternate light and shade for 12h, freely drink water, freely eat standard feed, and the treatment of the animal in the experimental process accords with the animal ethical standard.
2. Experimental groups and methods of administration
40 mice were randomly divided into 4 groups, which were a normal group, a GABA group, a Ampelopsis grossedentata aqueous extract group, and an anti-fatigue drink group, 10 mice per group. The normal group does not do anything. The administration dose of the mice was obtained by converting the equivalent dose according to the body surface area, and the administration was carried out in a 10-fold manner as compared with the equivalent dose. The GABA group and the Ampelopsis grossedentata water extract group are respectively and independently administered to mice, and the independent administration amount is the same as the GABA or the extract in the anti-fatigue beverage. The administration method comprises the following steps: the administration is carried out by gavage for 1 time at 9 am every day for 30 days, and normal group is gavage with normal saline with corresponding volume.
3. Observation indexes are as follows:
3.1 judge the fatigue degree of the mice by the time of exhaustion of swimming. The mice were given a load of 6% and were forced to swim to exhaustion, and were judged to be exhausted when they were submerged in water for 10 seconds and could not emerge from the water surface, and the exhaustion swimming time was recorded.
3.2 serum urea, lactate, myoglycogen assay: collecting blood by an eyeball-picking method 1h after the mouse exhaustive swimming is finished, collecting the blood into a centrifuge tube, standing for 1h, centrifuging for 10min at 4 ℃ 4000r/min, taking supernatant, and storing in a refrigerator at-80 ℃ for later use. The left hind leg skeletal muscle was collected from dissected mice and stored in a freezer at-80 ℃ for future use. Detection was performed according to the kit instructions.
Data were processed using software GaphPad Prism 8.0.2, Kolmogorov-Smirnov test for normal test, and Brown-Forsythe test for homogeneity of variance. One-way ANOVA is adopted for data which are in accordance with normal distribution and uniform in variance, and Tukey is adopted for pairwise comparison; normal distribution but variable data were obtained using Brown-Forythe and Welch ANOVA, and Dunnett's T3 for pairwise comparisons. Data are expressed as mean ± standard deviation.
The experimental result shows that the beverage of the example 4 has obvious anti-fatigue effect, the anti-fatigue effect is stronger than that of singly taking the ampelopsis grossedentata aqueous extract or GABA, and the ampelopsis grossedentata aqueous extract and GABA are compounded to show a certain synergistic effect.
Table 1: experiment of exhaustive swimming time and Biochemical index level of mice in each group
Note: p < 0.05, P < 0.01, compared to normal group.
