背景技术
大多动物细胞分泌具有各种尺寸和成分的源自细胞内的细胞外囊泡。已知原核生物和真核生物均分泌细胞外囊泡。
细胞外囊泡是具有小至约20nm且大至约5μm的直径的膜结构囊泡。细胞外囊泡在其尺寸和组成具有非均质性,并且具有外来体(exosomes,约30nm至约100nm)、核外粒体(ectosomes)、微囊泡(microvesicles,约100nm至约1000nm)、微粒(microparticles),外膜囊泡(outer membrane vesicles)等多个不同的种。细胞外囊泡的特性受到作为其起源的细胞的特性的影响。
细胞内物质(例如,脱氧核糖核酸(DNA;Deoxyribo Nucleic Acid)、核糖核酸(RNA;Ribonucleic Acid)、蛋白质等)可以被自然地承载于细胞外囊泡中且分泌到细胞外。细胞外囊泡具有与生物膜成分相同的成分,其生物相容性(biocompability)较高,其尺寸也属于纳米级,因此,物质送递效率很优秀。因此,目前进行使用细胞外囊泡代替现有的脂质体等送递系统来送递药物的研究。然而,在靶蛋白承载于细胞外囊泡中时,靶蛋白到细胞外囊泡中的承载效率较低。因此,存在对能够将靶蛋白以优秀的效率稳定地承载于细胞外囊泡中的技术的要求。
发明内容
技术问题

一方面的实施例提供源自包括编码一个或更多靶蛋白的一个或更多多核苷酸的重组微生物的细胞外囊泡(EV),其中,所述微生物为乳酸菌或酵母。
所述靶蛋白可以被连接至信号肽。换言之,所述靶蛋白可以是信号肽和靶蛋白的融合蛋白。所述重组微生物可以将所述靶蛋白以增加的量承载于所述细胞外囊泡中。在这种情况下,与包括编码一个或更多没有信号肽的靶蛋白的一个或更多多核苷酸的重组微生物相比,所述重组微生物可以具有提高的细胞外囊泡(extracellular vesicle;EV)承载能力。所述细胞外囊泡承载能力是指靶蛋白包括在细胞外囊泡(EV)中的程度或靶蛋白在细胞外囊泡(EV)中的表达程度。所述细胞外囊泡承载能力可以是指相比于不包括编码靶蛋白的多核苷酸的母体微生物的承载能力。
所述信号肽可以由SEQ ID NO:4的核苷酸序列编码,或SEQ ID NOS:21至60的氨基酸序列中的任何一个、或包括其中任何一个的序列或类似于其中任何一个的序列编码。
对所述重组微生物而言,所述乳酸菌可以是属于选自乳杆菌(Lactobacillus)、乳球菌(Lactococcus),以及双歧杆菌(Bifidobacterium)属之属。所述乳酸菌可以是副干酪乳杆菌(Lactobacillus paracasei)、断乳杆菌(Lactobacillus brevis),或植物乳杆菌(Lactobacillus plantarum)。
对所述重组微生物而言,所述酵母可以属于选自酵母属(Saccharomyces)、毕赤酵母属(Pichia),以及汉逊酵母(Hansenula)属之属。Saccharomyces可以是酿酒酵母(S.cerevisiae)。Pichia可以是巴斯德毕赤酵母(Pichia pastoris),Hansenula可以是多型汉逊酵母(Hansenula polymorpha)。
在所述重组微生物中,所述靶蛋白可以是生长因子、细胞因子、抗体、酶、抑制性蛋白,或其片段。所述生长因子可以是成纤维细胞生长因子。所述靶蛋白可以被选自成纤维细胞生长因子(fibroblast growth factor:FGF)、表皮生长因子(epidermal growthfactor:EGF)、肝细胞生长因子(hepatocyte growth factor:HGF)、胰岛素样生长因子(insulin-like growth factor:IGF)、胎盘生长因子(placenta growth factor:PGF)、血小板衍生生长因子(platelet-derived growth factor:PDGF)、转化生长因子(transforming growth factor:TGF)、血管内皮生长因子(vascular endothelial growthfactor:VEGF)、硫氧还蛋白(thioredoxin:TRX)、白细胞介素-1(interleukin-1:IL-1)、白细胞介素-10、白细胞介素-22、白细胞介素-13,以及肿瘤坏死因子(tumor necrosisfactor:TNF)。所述靶蛋白可以被选自,例如,IL-22、EGF、IGF1、FGF1(以下也称为成纤维细胞生长因子(acidic fibroblast growth factor:aFGF))、FGF2(以下也称为碱性成纤维细胞生长因子(basic fibroblast growth factor:bFGF))、FGF7(以下也称角化细胞生长因子(keratinocyte growth factor:KGF))、TGFa,以及TRX。

另一方面的实施例提供一种用于将一个或更多靶蛋白递送到个体的组合物,其包括作为活性成分的源自所述重组微生物的细胞外囊泡和载体。
在关于所述重组微生物和所述组合物的实施例中,所述细胞外囊泡可以是从所述微生物的培养基中分离的。换言之,所述细胞外囊泡可以被分泌到细胞外。所述细胞外囊泡可以具有约20nm至约500nm,例如,约20nm至约200nm,或约100nm至约200nm的平均直径。所述细胞外囊泡可以包括所述靶蛋白。所述靶蛋白可以位于所述EV的膜上或其内部。
所述细胞外囊泡可以是通过能够从培养液中分离所述细胞外囊泡的任何方法来分离的。例如,所述细胞外囊泡可以通过离心分离、超离心分离、通过过滤器的过滤、超滤法、凝胶过滤色谱法、离子交换色谱法、沉淀、免疫沉淀反应、预流电泳、毛细管电泳,或其组合分离。所述分离方法可以包括用于除去杂质的洗涤,以及浓缩等的过程。所述细胞外囊泡可以是通过下文中的分离所述细胞外囊泡的方法来产生的。所述细胞外囊泡可以是通过使用具有10kD以上,例如,50kD以上、100kD以上、300kD以上,或500kD以上的截止值的超细过滤器对所述微生物培养液进行超滤的方法产生的。所述细胞外囊泡是通过以100,000xg以上超离心分离所述微生物培养液来沉淀的。所述分离可以是通过根据下文中的第七实施例的细胞外囊泡的生产方法产生生产的。
在关于所述组合物的实施例中,所述载体可以是在生理学上可接受的,例如可以在药学上或化妆品制备学上可接受的。所述载体可以包括通常施用的盐水、无菌水、任氏液、缓冲液、环糊精、葡萄糖溶液、麦芽糊精溶液、甘油、乙醇、脂质体,或其组合。并且,所述载体可以包括抗氧化剂、稀释剂、分散剂、表面活性剂、结合剂、润滑剂,或其组合。

施用量可以通过患者的体重、年龄、性别、健康状态、日常饮食、施用时间、施用方法、排泄率以及疾病的严重程度等变更。日剂量是指足以通过被使用于需要治疗的个体来治疗减轻的疾病状态的有效成分的量。基于体重为70kg的成年个体,施用量为约0.01mg/日至1000mg/日,或约0.01mg/日至约500mg/日,并且可以按照预定的时间间隔进行每天一次至几次的分割施用。
所述组合物可以是化妆品组合物。所述化妆品组合物可以包括通常使用于化妆品组合物的成分。所述化妆品组合物可以包括诸如抗氧化剂、稳定剂、增溶剂、维生素、颜料、香料的常规的辅助剂和载体。
所述化妆品组合物可以是溶液、悬浮液、乳浊液、面团、凝胶、护肤霜、护肤液、面粉、油、粉状粉底、乳液粉底、蜡状粉底、或喷雾剂。所述化妆品组合物可以具有诸如营养霜、收敛化妆水、柔肤化妆水、护肤液、精华素、营养凝胶或按摩霜的剂型。
所述组合物可以被用于在个体内促进成纤维细胞或角质形成细胞的生长或胶原蛋白的合成。所述组合物可以被用于防止皮肤老化或减轻皱纹。在这种情况下,所述靶蛋白可以是生长因子。
所述组合物可以是用于向所述个体局部地送递所述靶蛋白。所述组合物可以是用于通过经皮(transdermally)、皮肤内(intradermally)、口部,或粘膜(transmucosally)以及粘膜内送递。
对所述组合物而言,所述个体可以是哺乳动物。所述哺乳动物可以是人、狗、猫、马,或猪。

所述培养可以是在有用于所述微生物的生长的培养基中进行孵育。所述培养可以在已知适合乳酸菌或酵母的条件,例如温度和搅拌条件下进行。
从所述培养物中分离所述细胞外囊泡的步骤可以包括从培养物中分离细胞外囊泡的任何方法。
所述分离可以包括:离心分离所述培养物以获得上清;过滤所述上清;以及通过超离心分离所述过滤物来获得沉淀物。
在所述分离中,可以以约1,000至约20,000xg进行离心分离。在所述过滤中,所述过滤可以是使用超微过滤器的过滤。所述过滤可以是通过使用10kD以上,例如,50kD以上、100kD以上、300kD以上,或500kD以上的截止值的超细过滤器对所述上清进行超滤的。在超离心分离所述过滤物来获得沉淀物的步骤中,所述超离心分离可以是以100,000xg以上,例如,以约100,000xg至约200,000xg进行的。
所述方法可以进一步包括悬浮沉淀物。

最佳实施模式
在下文中,通过实施例进一步详细地描述本公开。然而,这些实施例仅用于示例性地说明本公开,并不用于限制本公开的范围。
实施例1:源自酵母细胞的细胞外囊泡(Extracellular Vesicle;EV)
制备用于表达靶蛋白的重组酵母,并从该酵母分离细胞外囊泡。具体过程如下。使用酿酒酵母(Saccharomyces cerevisiae)作为酵母细胞。
1.表达载体的制备
图1示出用于在酵母细胞中表达靶蛋白的表达载体。该表达载体是使用质粒pRS416 GPD(SEQ ID:NO.1)的序列来制备的,所述靶蛋白为hEGF1、hIGF1、hFGF1、hFGF2、hTGF alpha,以及hTRX。hEGF1、hIGF1、hFGF1、hFGF2、hTGF alpha,以及hTRX分别具有SEQID:NOS 14、15、12、13、17,和18的氨基酸序列,其分别由SEQ ID:NOS 5、6、7、8、10和11的核苷酸序列来编码。FGF7可以具有SEQ ID NO:9的核苷酸序列,其氨基酸序列可以是被SEQ IDNO:9的核苷酸序列编码的氨基酸序列。
根据所述靶蛋白,图1中的载体被命名为p416G-MF-hEFG1(IGF1、FGF1、FGF2、TGFalpha,以及TRX)。
首先,根据要求,根据S.cerevisiae的密码子使用频率由MicroGene公司合成对密码子优化的靶蛋白基因,即人EGF1、IGF1、FGF1、FGF2、TGF alpha,以及TRX基因。通过使用p416GPD载体(ATCC87360)(SEQ ID NO:1)将各基因制成图1中的表达载体。图1的表达载体包括在靶蛋白基因的上游连接编码S.cerevisiae的交配因子(mating factor)alpha-1信号肽(MF)的多核苷酸(SEQ ID NO:4)的序列。使用没有连接编码信号肽(MF)的多核苷酸(SEQ ID NO:4)的基因作为对照组。并且,使用p426GPD载体(ATCC87361)(SEQ ID NO:2)代替p416GPD(ATCC87360)载体通过相同的方式制备载体。所述p416GPD载体为以低拷贝(lowcopy)存在于细胞内的载体,所述p426GPD为以高拷贝(high copy)存在于细胞内的载体。在p416GPD和p426GPD中,GPD是指启动子GPD的核苷酸序列(SEQ ID NO:3)。
在图1中,所述载体包括作为S.cerevisiae的复制起点的CEN/Ars序列、氨苄西林抗性基因(Ampicillin resistance gene:Ampr)序列、作为大肠杆菌(Escherichia coli;E.coli)的复制起点序列的ColE1 ori序列、作为S.cerevisiae的启动子序列的启动子GPD序列、作为S.cerevisiae的CYCterminator序列的ScCYCterm序列、作为噬菌体的复制起点的F1 ori序列、S.cerevisiae的启动子、ORF、终止子序列(ScURA3p-URA3)。
2.在酵母中靶蛋白的表达
根据LiCl法将p416G-MF-hEGF1(IGF1、FGF1、FGF2、TGF alpha,以及TRX)载体转化到S.cerevisiae CEN.PK2-1菌株中。将所获得的转化的菌株在2mL的minimal ura-dropout培养基(无氨基酵母氮源(Yeast nitrogen base without amino acids)(Sigma-Aldrich:Cat.No.Y0626)6.7g/L)),以及Yeast synthetic drop-out without uracil(Sigma-Aldrich:Cat.No.Y1501)1.92g/L、葡萄糖2(w/v)%一次培养一天,并将所培养的菌株接种于包括1%的酸水解酪素(Casamino acids)的15ml的minimal ura-drop out培养基以使原始OD600为0.5,然后进行本培养。以220rpm搅拌的同时在30℃下进行所述本培养两天,并且准备直接使用除去菌体的上清的试料组。并且,使用100kDa截止值薄膜(AmiconUltra-15 Centrifugal Filter Unit with Ultracel-10membrane(100K)millipore:Cat.No.UFC910024)过滤所述上清来获得浓缩的滤液,并将该滤液以150,000xg超离心分离(ultracentrifugation)2个小时来分离细胞外囊泡(EV)并将其悬浮在1ml的磷酸缓冲盐溶液(phosphate buffer saline;PBS)中。在此,通过对所述上清液以及所获得的细胞外囊泡试料进行蛋白免疫印迹法(western blot)来确认蛋白的表达程度。
图2示出在从用p416G-MF-hEGF1(IGF1、FGF1、FGF2、TGF alpha,以及TRX)转化的酿酒酵母(S.cerevisiae)分离的上清液和细胞外囊泡中靶蛋白的表达程度。分道1和2为示出各靶蛋白连接到信号肽MF的融合蛋白的表达程度的蛋白免疫印迹照片。分道1包括在酵母细胞内表达且从培养液中分离的所有蛋白,即承载或没承载于细胞外囊泡中的所有靶蛋白。分道2仅表示承载于细胞外囊泡中的靶蛋白。如图2中所示,在所有六个实验组中,靶蛋白均以显著地增加的量承载于细胞外囊泡中。
图3示出在从用p416G-hFGF1、p416G-MF-hFGF1、p416G-hTRX,以及p416G-MF-hTRX转化的S.cerevisiae分离的上清液和细胞外囊泡中靶蛋白的表达程度。换言之,图3示出根据是否有信号肽而被细胞外囊泡捕获的程度。
分道1表示从在没有信号肽的情况下表达的菌株的培养液中获得的细胞外囊泡内的靶蛋白,分道2表示从与信号肽序列一起表达且分泌的菌株的培养液中获得的细胞外囊泡内的靶蛋白的表达。如图3所示,与在细胞内表达的靶蛋白即在每一亿的EV中0.667ng的FGF1和0.047ng的TRX相比,在表达并分泌到细胞外时表达在细胞外囊泡中的靶蛋白的量即在每一亿的EV中2.648ng的FGF1和35.518ng的TRX显著地多。
3.确认含有生长因子的细胞外囊泡对细胞增殖的效能
测量根据第二节记载的方法分离的细胞外囊泡内的各靶蛋白的浓度,然后,将各蛋白用PBS稀释10倍,并从20μL的初始浓度开始通过4个步骤稀释。将20μL的各稀释液添加于各孔中包括NIH3T3细胞株或5,000个HaCat细胞的96孔板,然后在37℃下培养48小时。然后,将10μL的CCK-8试剂盒(Cell Counting Kit-8(Dojindo))溶液添加于各孔中。在两个小时后,在450nm处测量吸光度。对于FGF1、FGF2,以及IGF使用了NIH3T3细胞,对于TGFa和EGF使用了所述HaCat细胞。
图4示出从酵母中分离的含有生长因子的细胞外囊泡对细胞增殖的效果。在图4中,SC是指酿酒酵母(Saccharomyces cerevisiae)。
结果,所述包括靶蛋白的细胞外囊泡剂量依赖性地增加细胞数。在图4中,除了使用用FGF1转化的巴斯德毕赤酵母(Pichia pastoris)或多型汉逊酵母(Hansenulapolymorpha)之外,Pichia EV-FGF1以及Hansenula EV-FGF1是经过与使用S.cerevisiae相同的过程获得的。在图4中,横轴表示培养基内的细胞外囊泡内含有的靶蛋白的浓度(w/v、ng/ml)。纵轴表示对所使用的含有细胞外囊泡的溶液中细胞增殖的程度与对照组(100%)进行比较并使用百分比表示其结果。
4.确认IL-22的表达
通过使用IL-22作为靶蛋白根据第一节的记载制作表达载体p426G-MF-IL-22,并根据第二节的记载将所述表达载体转化到S.cerevisiae CEN.PK2-1菌株中。使用了相同而不包括IL-22的p426G-MF载体作为对照组。
具体地,将Colo205细胞株在96孔板中的RPMI培养基中并在37℃下培养48个小时后,使用p426G-MF-IL22载体和p426G-MF载体转化来纯化分别源自表达IL-22的酵母或不表达IL-22的酵母的细胞外囊泡。将所述细胞外囊泡以0.5mg/mL的浓度悬浮在PBS中,将其添加于96孔板,每孔20μL,并进一步在37℃下培养6个小时。然后,通过从细胞株中提取蛋白来通过IL-10的表达程度间接地比较IL-22的表达程度。IL-22具有SEQ ID:NO 19的氨基酸序列。已知IL-22在促进IL-10的生产。
图5示出分别使用包括IL-22的细胞外囊泡或不包括IL-22的细胞外囊泡处理的Colo205细胞株中提取的蛋白来确认的IL-10蛋白生产量。如图5中所示,与不包括IL-22的细胞外囊泡相比,在使包括IL-22的细胞外囊泡接触细胞来进行培养的情况下,IL-10蛋白的生产量显著地增加。在图5中,分道1表示使用不包括IL-22的细胞外囊泡处理的Colo205细胞株的IL-10蛋白的生产量程度,分道2表示使用包括IL-22的细胞外囊泡处理的Colo205细胞株的IL-10蛋白的产量程度。
5.源自酵母的细胞外囊泡和细胞的融合
从未转化的S.cerevisiae CEN.PK2-1菌株如上所述分离了细胞外囊泡。将1ml的所分离的细胞外囊泡(0.5mg/ml的PBS)在5μM的CFSE(5-Carboxyfluorescein N-hydroxysuccinimidyl ester)溶液中在常温下放置30分钟。接着,使用PD-10去盐柱(desalting column)(GE)从溶液中除去残留的CFSE来获得CFSE标记的细胞外囊泡。将NIH3T3细胞株在96孔板的每孔中在RPMI培养基0.2mL中在37℃下培养48个小时后,添加在PBS中的10μL(赤色)或20μL(绿色)的CFSE标记的细胞外囊泡并在37℃下进一步培养24个小时。此后,使用PBS洗涤细胞。通过流式细胞仪(flow cytometer)通过残留的细胞,并测量其荧光程度。作为对照组,使用相同的CFSE标记0.5μg/ml的BSA并使用20μl的其产物。
图6示出通过细胞流分析观察CFSE标记的细胞外囊泡与细胞融合的程度的结果。在图6中,对照组(negative control)(左侧的图表)使和CFSE标记的BSA接触的细胞,实验组(右侧的图表)表示使CFSE标记的细胞外囊泡10μL(赤色)和20μL(绿色)和细胞接触后观察的结果。结果,如在图6的右侧图表表示,细胞被CFSE染色,由此确认细胞外囊泡与细胞融合且引起细胞外囊泡的成分被导入细胞中。NIH3T3细胞为标准成纤维细胞(fibroblast)细胞株。
6.确认源自酵母的细胞外囊泡对皮肤的毒性
根据OECD指南通过对人工皮肤的毒性试验来测量源自酵母的细胞外囊泡对皮肤的毒性。使用NeodermTM-ED(Taigo Science公司产品)作为人工皮肤。
分离源自酿酒酵母(S.cerevisiae)、巴斯德毕赤酵母(Pichia pastoris),或多型汉逊酵母(Hansenula polymorpha)的细胞外囊泡。根据第二节的记载分离了源自酿酒酵母(S.cerevisiae)的细胞外囊泡。除了使用巴斯德毕赤酵母(Pichia pastoris)以及多型汉逊酵母(Hansenula polymorpha)之外,根据第二节的记载分离了源自巴斯德毕赤酵母(Pichia pastoris)或多型汉逊酵母(Hansenula polymorpha)的细胞外囊泡。
将所分离的细胞外囊泡、作为阴性对照组的PBS,以及作为阳性对照组的5%SDS分别以30μL的量涂覆在NeodermTM-ED人工皮肤上并孵育在37℃下15分钟。然后,使用PBS洗涤所述人工皮肤并将其沉淀在12孔板中的2ml分析培养基(assay medium)(Taigo Science公司)并在37℃额外孵育42小时。
捞取所孵育的人工皮肤并将其移到0.3%的MTT溶液(0.3mg/ml)后在37℃下孵育三个小时。然后,再次捞取所述人工皮肤并使用8mm的活检穿孔器(biopsy punch)来分离各组织,且将所述组织投入于500μL的0.04N HCL-异丙醇并进行脱色四个小时。在570nm处测量吸光度后,对其与对照组中的吸光度进行比较来计算存活率(%)。结果,当所测量的存活率为阳性对照组和阴性对照组值的中间值或更高时,确定没有毒性。根据以下的式计算了存活率。
存活率=试验物质吸光度/阴性对照组吸光度×100
图7示出测量源自酵母的细胞外囊泡对皮肤的毒性的。在图7中,1表示阴性对照组(PBS);2表示阳性对照组(5%SDS);3表示源自S.cerevisiae的细胞外囊泡;4表示源自P.pastoris的细胞外囊泡;5表示源自H.polymorpha的细胞外囊泡。
实施例2:源自乳酸菌(lactic acid bacteria:LAB)细胞的细胞外囊泡
制备表达靶蛋白的重组乳酸菌,并从该乳酸菌分离细胞外囊泡。具体过程如下。使用副干酪乳杆菌(Lactobacillus paracasei)LMT1-21(KCTC13422BP)、断乳杆菌(Lactobacillus brevis)LMT1-46(KCTC13423BP)和/或植物乳杆菌(Lactobacillusplantarum)LMT1-9(KCTC13421BP)作为乳酸菌细胞。
1.制备基因表达载体
为了靶基因,从蛋白的氨基酸序列使用密码子优化工具(http://sq.idtdna.com/ CodonOpt)获得为所使用的乳酸菌优化的密码子的核苷酸序列,设计在该序列的两个末端具有对BamHI和XhoI限制酶的识别序列的序列,并合成具有该序列的DNA(Macrogen公司,韩国)。使用BamHI和Xhol限制酶来切断合成的基因。并且,对于母载体pMT182-PR4(SEQ IDNO:20)而言,使用相同的限制酶来切断,使用凝胶纯化试剂盒(Gel purification kit)来纯化,并使用碱性磷酸酶(AP;alkaline phosphatase)来脱磷。该母载体包括用于表达所述靶蛋白的启动子PR4和用于将所述靶蛋白分泌到细胞外的信号肽SP4(SEQ ID NO:21)。
将通过这种方法准备的1μL的载体DNA、3μL的插入物(insert)DNA、0.5μL的T4DNA连接酶(Takara公司,日本),以及缓冲溶液1μL添加到蒸馏水5.5μL中来使总体积为10μL。将该反应溶液在16℃下孵育12个小时来进行连接反应,并将所获得的连接产物通过萨姆布鲁克等的方法(Sambrook et al.,Molecular Cloning:A laboratory manual,2nd ed.1989)转化到大肠杆菌十大菌株中。分析并确认了从各群组获得的质粒的序列。使用FGF1、FGF2、EGF、IGF、KGF、TGFa、TRX,以及IL-22作为靶蛋白。其分别具有SEQ ID:NOS1、2、3、4、5、6、7,以及8的氨基酸序列。
2.乳酸菌转化
将所获得的克隆的DNA转化到三种乳酸菌。将各菌株在50mL的MRS中培养直至OD600为0.5后在4℃下以7,000rpm离心分离10分钟,并用25mL的冰冷(ice-cold)EPS(含有1mM的K2HPO4KH2PO4,pH7.4,1mM的MgCl2以及0.5M的蔗糖)洗涤两次。通过将该细胞悬浮在1mL的冰冷EPS中来制造可用于电穿孔法(electroporation)的反应潜能细胞(competent cell),并保存在-80℃的冷藏箱(deep freezer)中。将40μL的反应潜能细胞和1μg/μL的载体DNA转移到试管并放置在冰上五分钟。在25μF、8kV/cm,以及400ohms条件下提供搏动后,立即添加1mL的MRS液体培养基并在37℃下培养一个小时左右。将该细胞涂抹在包括10μg/ml的氯霉素的MRS培养基后在37℃下培养49小时来获得转化的细胞。
3.分离细胞外囊泡
从所获得的所述转化的乳酸菌(lactic acid bacteria;LAB)菌株中,将KCTC13422BP菌株在MRS液体培养基中在37℃下静置培养16个小时,然后将其以2%
(w/v)体积接种于MRS液体培养基并静置培养16个小时。通过将所获得的培养物以5,000xg离心分离15分钟来获得除去乳酸菌的上清液,然后通过使用100kDa的截留分子量(molecular weight cut-off;MWCO)超滤膜将该上清超滤并浓缩为20倍。通过将该浓缩液以150,000xg超离心分离3个小时来获得沉淀的团粒,并将该团粒用PBS重悬来获得细胞外囊泡溶液。通过使用Nanosight NS300(马尔文)来测量所获得的细胞外囊泡的尺寸和数量。将其结果表示在图8中。
图8示出从转化的乳酸菌中分离的细胞外囊泡的尺寸和浓度分布的。在图8中,横轴表示直径,纵轴表示浓度(粒子/ml)。在图8中,所使用的乳酸菌为KCTC13422BP菌株,靶蛋白为FGF1。
如图8所示,细胞外囊泡中90%的粒子以80nm至250nm的粒子尺寸分布。
4.确认在细胞外囊泡中是否存在靶蛋白
对在第三节中获得的EV溶液进行蛋白免疫印迹法以确认在EV中是否存在于靶蛋白中。所述细胞外囊泡是从以通过使用编码FGF1或TRX的基因克隆pMT182-PR4来而获得的载体转化的KCTC13422BP菌株(以下也称LMT1-21)中分离的。此时,所述基因使用与信号肽即SP4序列融合的或没有与其融合的序列。
通过以下的方法进行蛋白免疫印迹。将4x的上样缓冲液(thermo)、10x的还原剂(reducing agent)(thermo)添加于5μL的EV溶液中,然后在用于十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE;sodium dodecyl sulfate-polyacrylamide gelelectrophoresis)的凝胶上进行电泳。将该凝胶的蛋白移到硝酸纤维膜,然后,将该膜在包括5%的脱脂乳作为封闭液的含吐温20的生理盐水缓冲液(TBST;Tris-buffered salinewith tween 20)中孵育两个小时来封闭。然后,使用TBST洗涤三次,各次五分钟,并将膜和第一抗体添加到封闭液并孵育两个小时来引起抗原-抗体结合。在TBST洗涤后,添加二次抗体。放置一个小时后,通过使用增强电化学(ECL;enhanced electrochemical)系统来确认靶蛋白的分量和位置。
图9示出对EV溶液进行蛋白免疫印迹法的结果。如图9所示,FGF1和TRX表达在从通过使用分别与信号肽融合的FGF1和TRX克隆pMT182-PR4来获得的载体转化的LMT1-21中分离出来的细胞外囊泡中,由此可见,FGF1和TRX存在于细胞外囊泡中。
图10示出对从通过使用包括编码与信号肽基因融合或没有与信号肽基因融合的FGF1的基因的重组载体即pMT182-PR4-FGF1或pMT182-PR4-SP4-FGF1载体转化的LMT1-21中分离的细胞外囊泡进行蛋白免疫印迹的结果。在图10中,分道1表示在不使用信号肽而表达FGF1基因的情况下的细胞外囊泡,分道2表示在表达编码信号肽和FGF1的融合蛋白的基因的情况下的细胞外囊泡。
5.确认源自乳酸菌的含有生长因子的细胞外囊泡对细胞增殖的效能
根据第三节所记载的方法,将从1L的乳酸菌培养液中分离的细胞外囊泡悬浮在1ml的PBS中。将DMEM培养基中的NIH3T3细胞株(或HaCat细胞)以5,000细胞/孔接种(seeding)于96孔板的每孔,并在37℃下培养48个小时。然后,将20μL的含有表达生长因子的细胞外囊泡的所述溶液或作为对照组的PBS添加于每孔中。在相同的条件下培养48个小时后,将10uL的CCK-8试剂盒(Dojindo)溶液添加于每孔。在两个小时后,在450nm处测量吸光度。对FGF1、FGF2,以及IGF使用了NIH3T3细胞,并对KGF、TGFa以及EGF使用了HaCat细胞。
图11示出从乳酸菌中分离的含有生长因子的细胞外囊泡对细胞增殖的效果。在图11中,LAB是指乳酸菌(lactic acid bacterium)(KCTC13422BP菌株)。结果,所述含靶蛋白的细胞外囊泡剂量依赖性地增加细胞浓度。在图11中,横轴表示包括在细胞外囊泡中的靶蛋白的浓度(w/v)。纵轴是对所使用的含有细胞外囊泡的溶液中的细胞增殖的程度与对照组进行比较并使用百分比表示其结果。
6.包括生长因子的细胞外囊泡的效能:确认IL-10的表达
根据第一节和第二节制备表达IL-22的载体,并将该载体转化到LMT1-21中。根据第三节,从使用表达IL-22的载体转化的LMT1-21中分离EV。为了确认该EV在细胞中是否促进IL-10的表达,间接地推测是否存在IL-22。
具体地,将Colo205细胞株在96孔板上RPMI培养基中在37℃下培养48个小时,从其分离源自表达IL-22的乳酸菌或不表达IL-22的乳酸菌的细胞外囊泡,将该细胞外囊泡以0.5mg/mL的浓度悬浮在PBS中,将其以20μL添加于每孔中并进一步培养在37℃下6个小时。然后,在从细胞中提取蛋白即进行细胞溶解(lysis)来获得溶解产物(lysate),并在所述蛋白中IL-10的表达程度进行了比较。
图12为对与源自LMT1-21的EV接触的源自细胞的蛋白进行蛋白免疫印迹时的照片。在图12中,分道1表示PBS,分道2表示源自LMT1-21的EV,分道3表示源自表达IL-22的LMT1-21的EV。
7.源自乳酸菌的细胞外囊泡和细胞的融合
从未转化的乳酸菌菌株(KCTC13422BP)中,如上所述分离了细胞外囊泡。将所分离的1ml的细胞外囊泡(0.5mg/ml)在5μM的CFSE溶液中在常温下放置30分钟。接着,使用PD-10去盐柱(desalting column)(GE)从溶液中除去残留的CFSE来获得CFSE标记的细胞外囊泡。将NIH3T3细胞株在96孔板的各孔中在0.2mL的RPMI培养基中培养48个小时后,在每个孔中添加PBS中的CFSE标记的10μL(赤色)或20uL(绿色)细胞外囊泡并进一步培养24个小时。此后,使用PBS洗涤细胞。通过流式细胞仪(flow cytometer)通过残留的细胞,并测量其荧光程度。作为对照组,使用相同的CFSE标记0.5μg/ml的BSA并使用20μl的其产物。
图13示出通过细胞流分析观察CFSE标记的细胞外囊泡与细胞融合的程度的结果。在图13中,对照组(negative control)(左侧图表)表示使CFSE标记的BSA和细胞互相接触,实验组(右侧图表)表示使CFSE标记的10μL(赤色)的细胞外囊泡、20μL的细胞外囊泡(绿色)和细胞互相接触后观察到的结果。结果,如在图13的右侧图表表示,细胞由CFSE染色,由此可以确认细胞外囊泡与细胞融合且细胞外囊泡的成分导入细胞中。NIH3T3细胞为标准成纤维细胞(fibroblast)细胞株。
8.确认源自乳酸菌的细胞外囊泡对皮肤的毒性
根据OECD指南通过对人工皮肤的毒性试验来测量源自乳酸菌的细胞外囊泡对皮肤的毒性。使用NeodermTM-ED(Taigo Science公司产品)作为人工皮肤。
将分离源自LMT1-21、LMT1-9,或LMT1-46的细胞外囊泡。根据第二节的记载分离这些细胞外囊泡。所分离的细胞外囊泡、作为对照组的PBS,以及作为阳性对照组的5%SDS以各30μL的分量涂抹在NeodermTM-ED人工皮肤上,接着孵育在37℃下15分钟。接着,将所述人工皮肤用PBS洗涤并沉浸在12孔板中的2ml的分析培养基(assay medium)(Taigo Sicence公司产品)中并在37℃下进一步孵育42个小时。
捞取所孵育的人工皮肤并将其移到0.3%的MTT溶液(0.3mg/ml)中来孵育3个小时。然后,再次捞取人工皮肤并使用8mm的活检穿孔器(biopsy punch)来分离组织并投入500μl的0.04N HCl-异丙醇中并脱色4个小时。在570nm处测量吸光度后与对照组的吸光度进行比较来计算存活率(%)。结果,当所测量的存活率为阳性对照组和阴性对照组值的中间值或更高时,确定没有毒性。根据以下的式计算了存活率。
存活率=试验物质吸光度/阴性对照组吸光度×100
图14示出测量源自乳酸菌的细胞外囊泡对皮肤的毒性。在图14中,1表示阴性对照组(PBS)、2表示阳性对照组(5%SDS)、3表示源自LMT1-46的细胞外囊泡、4表示源自LMT1-9的细胞外囊泡,5表示源自LMT1-21的细胞外囊泡。
实施例3:对包含生长因子的细胞外囊泡的细胞增殖效率和裸生长因子的细胞增殖效率的比较
1.包括细胞外囊泡的生长因子的制备
分别通过在实施例1的第三项相同的方式从使用p416G-MF-EGF、p416G-MF-FGF1以及p416G-MF-FGF2转化的Pichia pastoris分离了包括生长因子的细胞外囊泡。将其中每一个悬浮在PBS中来调节EGF的浓度至10ug/ml且将FGF1或FGF2的浓度调节至1ug/ml。作为对照组,从AbCam购买了裸EGF、FGF1,以及FGF2蛋白,并将其悬浮在PBS直至成为如上所述的浓度。
2.对包括生长因子的细胞外囊泡的细胞增殖和裸生长因子的细胞增殖的效果的比较
从TaiGo Science公司购买人工皮肤即NeodermTM-ED。用PBS洗涤所述人工皮肤,并将2mL的PBS、2mL的如上所述制备的包括细胞外囊泡生长因子的溶液,以及2mL的如上所述制备的包括裸EGF、FGF1,或FGF2蛋白的对照组溶液添加到12孔板的孔中,接着在37℃下进一步培养24个小时。在用PBS洗涤三次后,人工皮肤在37℃下被固定于4%的多聚甲醛溶液(Sigma,美国)18个小时且被用Leica Biosystems制成冷冻切片。通过使用用于EGF-EV的抗Ki-67抗体和对照组且使用用于FGF1-EV和FGF2-EV的抗胶原质抗体(AbCam)和对照组来进行免疫组织化学法(Immunohistochemistry;IHC),然后投入3,3'二氨基联苯胺(3,3'Diaminobenzidine;DAB)。其结果在显微镜下照相。整体上,丰富的Ki-67或胶原质被观察为褐色。Ki-67是表皮细胞增殖的生物标志物。
如图15中所示,与使用所述PBS和所述对照组的处理(A行)相比,从EGF-EV处理观察到更加良好的表皮细胞增殖。并且,与使用PBS或对照蛋白(B行和C行)相比,使用FGF1-EV和FGF2-EV时观察到更加良好的胶原质合成。根据这些结果,与裸生长因子相比,包括在细胞外囊泡中的生长因子对细胞增殖更有效,与生长因子类型无关。并且,与包括在细胞外囊泡中或不包括在细胞外囊泡中的任何其它生长因子相比,FGF2-EV最有效。
实施例4:对生长因子稳定性的比较
1.相比于裸EGF稳定性的EGF-EV稳定性
通过实施例1的第二项相同的方法准备了源自Pichia pastoris的EGF-EV和所述对照蛋白,即不包括在EV中的EGF蛋白。简略地,所述EGF-EV或所述EGF蛋白被悬浮在1ml的PBS中直至其浓度为10ug/ml,并且在40℃下培养8周。两周一次,为了细胞增殖活性分析,所述样品被等分并用PBS稀释直至100ng/ml的浓度。
DMEM培养基中的HaCat细胞以5,000细胞/孔的密度被接种于96孔板的每孔中并在37℃下培养48个小时。然后,向其添加了20μL的EGF-EV、对照蛋白,PBS的各种样品。所述细胞在相同的条件下被培养48个小时,然后将10uL的cell counting kit-8(Dojindo)溶液添加于每孔中。在两个小时后,在450nm处测量吸光度。
如图16所示,包括在EV中的EGF比裸EGF更稳定。
2.相比于裸FGF2的稳定性的FGF2-EV的稳定性
通过实施例1的第二项相同的方法准备了源自Pichia pastoris的EGF2-EV和所述对照蛋白,即不包括在EV中的FGF-2蛋白。简略地,所述FGF2-EV或所述FGF2蛋白被悬浮在1ml的PBS中直至其浓度为10ug/ml,并且在常温下培养4周。为了细胞增殖活性分析,各样品被定期地等分并用PBS稀释直至100ng/ml的浓度。
DMEM培养基中的NIH3T3细胞以5,000细胞/孔的密度被接种于96孔板的每孔中并在37℃下培养48个小时。然后,向其添加了20uL的FGF2-EV、对照蛋白,PBS的各种样品。所述细胞在相同的条件下被培养48个小时,然后将10μL的cell counting kit-8(Dojindo)溶液添加于每孔中。在两个小时后,在450nm处测量吸光度。
如图17所示,包括在EV的FGF2比裸FGF2更稳定。
工业实用性
根据一实施例的重组微生物可以被用于从所述细胞外囊泡中有效地分离细胞外囊泡或靶蛋白。
根据另一实施例,用于将所述细胞外囊泡和靶蛋白送递到个体的组合物可以被用于将所述靶蛋白有效地送递到个体。
根据另一实施例,治疗个体的疾病的方法可以被用于有效地治疗所述疾病。
根据另一实施例,将化妆品应用于个体地方法可以被用于将化妆品有效地应用于个体。

Claims (11)
1.一种源自包括编码一个以上的靶蛋白的一个以上的多核苷酸的重组微生物的细胞外囊泡,其中,所述微生物为酵母,其中所述酵母属于选自酵母属、毕赤酵母属和汉逊酵母属中的属,并且其中所述靶蛋白连接有信号肽,因此与所述靶蛋白没有连接到所述信号肽相比,所述微生物将所述靶蛋白以增加的量承载于所述细胞外囊泡中,其中所述信号肽由SEQ ID NO:4的核苷酸序列编码,并且所述细胞外囊泡包含所述靶蛋白,
其中,所述靶蛋白选自由成纤维细胞生长因子1(FGF1)、成纤维细胞生长因子2(FGF2)、胰岛素样生长因子1(IGF1)、KGF、TGFα、TRX和IL-22组成的组中的一种或多种。
2.根据权利要求1所述的细胞外囊泡,其中,所述细胞外囊泡是从所述重组微生物的培养液中分离的。
3.根据权利要求2所述的细胞外囊泡,其中,所述细胞外囊泡是通过将所述微生物培养液以100,000xg以上超离心分离来沉淀分离的。

6.一种用于向个体送递靶蛋白的组合物,其包括作为活性成分的根据权利要求1至5中任何一项的细胞外囊泡和载体,其中所述个体是哺乳动物,其中所述组合物用于减轻皮肤的皱纹、美白皮肤、阻断紫外线或减轻炎症,其中所述靶蛋白选自由成纤维细胞生长因子1(FGF1)、成纤维细胞生长因子2(FGF2)、胰岛素样生长因子1(IGF1)、KGF、TGFα、TRX和IL-22组成的组中的一种或多种。
7.根据权利要求6所述的组合物,其用于经皮地、在皮肤内、口部,经粘膜地,或粘膜内地送递靶蛋白。
8.根据权利要求7所述的组合物,其具有药学或化妆学上的用途。
9.一种用于向个体送递靶蛋白的组合物,所述组合物具有药学上的用途,其包括作为活性成分的根据权利要求1至5中任何一项的细胞外囊泡和载体,其中所述个体是哺乳动物,其中所述组合物用于减轻皮肤的皱纹、美白皮肤、阻断紫外线或减轻炎症,其中所述靶蛋白是表皮生长因子(EGF)。
10.根据权利要求1至5中任何一项的细胞外囊泡在制备用于减轻皮肤的皱纹、美白皮肤、阻断紫外线或减轻炎症的药物中的用途,其中所述靶蛋白选自由成纤维细胞生长因子1(FGF1)、成纤维细胞生长因子2(FGF2)、表皮生长因子(EGF)、胰岛素样生长因子1(IGF1)、KGF、TGFα、TRX和IL-22组成的组中的一种或多种。
11.根据权利要求1至4中任何一项的细胞外囊泡在制备用于减轻皮肤的皱纹、美白皮肤、阻断紫外线或减轻炎症的化妆品中的用途,其中所述靶蛋白选自由成纤维细胞生长因子1(FGF1)、成纤维细胞生长因子2(FGF2)、胰岛素样生长因子1(IGF1)、KGF、TGFα、TRX和IL-22组成的组中的一种或多种。
