Abstract
本发明属于牡丹加工技术领域,具体涉及一种含有牡丹甾醇和α亚麻酸的组合物,以及该组合物在制备用于辅助治疗类风湿炎症药物中的应用、在制作食品中的应用。本发明所提供的组合物中,牡丹甾醇和α亚麻酸的重量比例为0.5~0.8:8~11。本发明的有益效果在于:(1)本发明以组合物的方式,将牡丹甾醇和α亚麻酸有机结合起来,共同协同作用,使它们应用于制备辅助治疗类风湿炎症药物中之后,其效果显著;(2)本发明的组合物来源于天然的植物,相对于对人体有副作用的西药来说,本发明的产品具有无副作用、安全性高的特点;(3)本发明的组合物中,原料丰富、成本低廉,易于推广应用及大规模生产。

技术领域
本发明属于牡丹加工技术领域,具体涉及一种含有牡丹甾醇和α亚麻酸的组合物,以及该组合物在制备用于辅助治疗类风湿炎症药物中的应用、在制作食品中的应用。
背景技术
类风湿关节炎(RA)的发病可能与遗传、感染、性激素等有关。RA关节炎的病理主要有滑膜衬里细胞增生、间质大量炎性细胞浸润,以及微血管的新生及软骨和骨组织的破坏等。一般治疗强调休息及关节制动,功能锻炼,理疗、外用药等辅助治疗缓解关节症状。
关于上述的类风湿疾病,目前并没有特效药,常采用的主要有非甾类抗炎药、慢作用抗风湿免疫抑制化学制剂、及植物药等临床应用。对治疗类风湿关节炎具有缓解疼痛的作用,但作用机制与疗效均有待提高。
《三组类风湿关节炎治疗方案的药物经济学分析》一文中披露了,方案1:柳氮磺氨吡啶+甲氨蝶呤+美洛昔康;方案2:羟氯喹+甲氨蝶呤+美洛昔康;方案3:来氟米特+甲氨蝶呤+美洛昔康,这三组药物的治疗效果。
以上的方案中,柳氮磺氨吡啶可能会引起会引起过敏反应、肝功能受损、肾脏受损、恶心、呕吐,腹泻等副作用;
甲氨蝶呤可能会导致不良反应包括1.胃肠道反应:咽喉炎、恶心、呕吐、腹痛、腹泻、消化道出血,食欲减退常见,偶见假膜性或出血性肠炎等。除此之外,还有可能致使肝功能损害,包括黄疸、丙氨酸氨基转移酶、碱性磷酸酶,γ-谷氨酰转肽酶等增高等副作用。
美洛昔康可能会导致胃肠道不适:常见消化不良、腹痛、恶心、腹泻等不良反应,偶有食管炎、结肠炎及消化道溃疡、出血等症状出现;此外,对于神经系统也会产生如下的不良影响:头痛、眩晕、耳鸣、嗜睡等。上述的药物或多或少的都会对人体产生一定的不良影响。
因此,需要开发一种有效的来源于植物的安全性高、无副作用、能用于辅助治疗类风湿炎症药物中的营养素。
发明内容
为了解决上述的技术问题,本发明提供了一种以牡丹甾醇和α亚麻酸为主要原料且无副作用、安全性高、应用效果好且成本低廉的组合物;
将上述的组合物应用于药物及食品的制备中,尤其是应用于能辅助预防或治疗风湿或者是类风湿类的药物或食品中,是本发明所要重点保护的范围。
本发明所提供的一种组合物,其中有效成分为牡丹甾醇和α亚麻酸;牡丹甾醇和α亚麻酸的重量比例为0.5~0.8:8~11。
本发明所采用的牡丹甾醇满足:其中的异岩藻甾醇占比为50~60%,燕麦甾烯醇占比为15~21%,β-谷甾醇占比为5~9%,角鲨烯3~5%。由于不同采摘时期的牡丹原料中所含的甾醇会呈现出一定变化,且不同批次的牡丹甾醇中各成分的含量也有轻微的波动,但是牡丹甾醇中各成分的含量大体上是在如上的范围内浮动。本发明人通过实验,发现满足上述含量范围原料的牡丹甾醇均能达到本发明的效果。
优选的,牡丹甾醇满足:
其中的异岩藻甾醇占比为51~59%,燕麦甾烯醇占比为16~20%,β-谷甾醇占比为5~8%,角鲨烯4~5%。

α亚麻酸是以牡丹籽油或者亚麻籽油、紫苏籽油为原料制备获得的。
每天8.5~11.8mg/kg;其中,牡丹甾醇和α亚麻酸的重量为0.5~0.8mg/kg:8~11mg/kg;
优选的,组合物在药物中的剂量为每天10mg/kg;牡丹甾醇和α亚麻酸的重量为0.6mg/kg:9.4mg/kg。
上述的药物还包括药学上可接受的辅料;
上述的药物为注射剂、胶囊剂、片剂、颗粒剂、粉剂、气雾剂中的任一种;本发明的药物中,辅料选自稀释剂、崩解剂、粘合剂、润滑剂、表面活性剂、抗氧化剂、防腐剂和稳定剂中的赋形剂中的任一种;
崩解剂可以是低取代的羟丙基纤维素;
稀释剂选自淀粉、糊精、羧甲基淀粉、羟丙基淀粉、改良淀粉、改性预胶化淀粉、乳糖、糖粉、微晶纤维素、磷酸钙、磷酸氢钙、碳酸钙中的至少一种;
润滑剂选自硬脂酸镁、硬脂酸、分子量4000~8000的聚乙二醇、硬脂酸钙、硬脂酸锌中的至少一种;
助流剂选自滑石粉、二氧化硅中的至少一种;
以牡丹甾醇和α亚麻酸为主要有效成分的组合物在制备具有防治类风湿炎症的食品中的应用,也是本发明所要保护的范围。
上述的食品还包括食品添加剂;
优选的为甜味剂、酸味剂、食用香精、稳定剂、抗氧化剂、微量元素、维生素类中的至少一种,但并不仅限于以上的种类;
上述的食品可以是固态食品、液体食品、膏装食品、冻状食品、风干食品;
优选的,食品为面条、面包、饼干、乳制品、罐头、膨化食品、糖果、饮料、烤鱼片中的任一种,或者是其它的休闲小食品;不仅限于以上的种类;
若是以该组合物为原料制备的食品是饮料、面包、蛋糕、饼干或其它糕点等,可以添加以下的原料作为甜味剂:比如蔗糖、乳糖、半乳糖、麦芽糖、D-甘露糖、木糖、D-山梨糖醇、葡萄糖、麦芽糖醇、木糖醇、甜蜜素、甘草、天门冬酰苯丙氨酸甲酯、1,4,6-三氯蔗糖、柚甘、新橙皮等;
具体的,比如:甜蜜素、安赛蜜、甜菊糖、甘草、甘草酸一钾、甘草酸二钠、甘草酸三钾和三钠、甘草酸铵、乙酰磺胺酸钾、蔗糖、乳糖、山梨糖醇、异麦芽三糖醇、乳糖醇、葡萄糖、果葡糖浆、赤藓糖醇、AK糖、糖精、木糖醇、山梨糖醇、木糖醇、低聚糖(水苏糖、棉籽糖、麦芽低聚糖、龙胆低聚糖、异麦芽低聚糖、环糊精、木糖低聚糖)、麦芽糖醇、新橙皮苷二氢查耳酮、糖精、糖精钠、环己基氨基磺酸钠、天门冬酰苯丙氨酸甲酯阿力甜、环己基氨基磺酸钠、索马甜、寡糖醇、潘糖醇、果寡糖、半乳糖、安赛蜜、三氯蔗糖、果糖、阿斯巴甜、蛋白糖、罗汉果甜苷、蜂蜜、甘草甜素、甘草酸铵、多聚果糖、多聚葡萄糖、低聚木糖、低聚甘露糖、异麦芽酮糖醇、低聚甘露糖、固体聚乙二醇中的任一种;
同样的,在制备糕点类食品时,也可以采用上述的甜味剂,比如烘烤类的糕点,蒸煮类的糕点、油炸类的糕点等等,例如绿豆糕、豆沙糕,或者是月饼等食品;
若是制备饮料类食品,包括固体饮料、液体饮料、口服液等;饮料的种类可以是蛋白饮料、乳制品饮料、果汁饮料、蔬菜饮料、乳酸菌饮料、咖啡饮料(包括牡丹咖啡)等;可以适当的加入酸味剂、食用香精、稳定剂,所述的酸味剂包括富马酸、琥珀酸、蚁酸、马来酸、甘氨酸、乙酸、酒石酸、DL-苹果酸、L-抗坏血酸、乳酸、柠檬酸、苹果酸、柠檬酸、柠檬酸钠、柠檬酸一钠、柠檬酸钾、酒石酸、果醋中的任一种;
食用香精可选自液态香精、油溶性香精、乳化性香精;可选自山楂香精、抹茶香精、玉米香精、板栗香精、红薯香精、桂花香精、桔子香精、薄荷香精、柠檬香精、橙子香精等各种果蔬或植物香精中的任一种;
稳定剂可选自食用胶、酯类,比如结冷胶、β-环糊精、淀粉、硬脂酸盐、单甘酯、蔗糖酯、双甘油甘油硬脂酸一酯、四甘油硬脂酸一酯、琥珀酰甘油一酯、双乙酰琥珀酰甘油一酯等等;
抗氧化剂可选自VE、VC、异维生素C钠、茶多酚、植酸、生姜提取物、叔丁基羟基茴香醚(BHA)、二丁基羟基甲苯(BHT)、叔丁基对苯二酚(TBHQ)、没食子酸丙酯(PG)、抗坏血酸棕榈酸酯(AP)、硫代二丙酸二月桂酸酯(DLTP)、4-己基间苯二酚(4-HR)中的任一种;
还可以包括其它辅料:抗坏血酸、防腐剂、着色剂、护色剂、膨松剂、抗性糊精、乳粉、碳酸氢钠、碳酸氢铵、蛋白粉、苯甲酸钠、山梨酸钾、乳酸;聚甘油脂肪酸酯、蔗糖脂肪酸酯及酶分解卵磷脂、功能性成分、微量元素比如维生素A、维生素B、维生素K中的任一种。
α亚麻酸是一种多不饱和脂肪酸,也是一种必需的脂肪酸。关于α亚麻酸对风湿或者是类风湿性关节炎的防治机理,分析如下:
细胞因子IL-1β和TNF-α作用于炎症细胞,包括刺激胶原蛋白酶的产生和使白细胞渗透所必须的粘附分子表达增加。实验证明,兔关节内的IL-1β可引起关节炎症,临床关节炎病人静脉注射抗TNF-α单克隆抗体可以抑制关节炎症。类风湿关节炎的成功治疗和抑制滑膜细胞因子和细胞粘附分子表达是密切相关的。而α亚麻酸中所含有的类花生酸物质,比如花生四烯酸可以作为关节炎症的介导,对于关节炎症的表现形式如疼痛、肿胀、白细胞浸润等产生作用,防治关节中的软骨丢失、骨吸收和关节障碍,因此在体内增加α亚麻酸在一定程度是可以抑制IL-1β和TNF-α的产生的,从这个角度来看,在体内增加α亚麻酸能够抑制IL-1β和TNF-α产生、或者是说增加α亚麻酸能降低正常人和关节炎病人体内的IL-1β和TNF-α单核细胞的合成从而达到预防关节疾病比如风湿或类风湿的发生;有文献披露,通过补充亚麻籽油可使人体内的IL-1β和TNF-α的产生被抑制大约30%。
但是仅仅通过α亚麻酸来辅助预防或治疗关节疾病如风湿或类风湿等关节炎症,其作用还是比较有限的。
目前关于牡丹甾醇的研究,非常的有限,比如张睿在《响应面优化牡丹籽甾醇的提取及其抗氧化活性研究》一文中对牡丹甾醇中对牡丹籽甾醇的提取及其抗氧化活性进行了研究,发现牡丹甾醇具有较好的抗氧化活性;陈刚等人通过超声波辅助提取牡丹果皮中的甾醇,对其进行了优化。关于牡丹甾醇的进一步研究,鲜有文献披露。而且目前所集中的研究主要是侧重于牡丹籽油中的牡丹甾醇,对于牡丹花粉中的牡丹甾醇的研究,更是少之又少。
本发明人结合此生对于植物油尤其是花生油、牡丹籽油的研究及实践生产经验(包括参与牡丹籽油标准的制定),通过自身数十年如一日的沉浸于牡丹中相关活性功能成分的研究,惊喜的发现,将从牡丹(或亚麻籽、或紫苏)中获得的α亚麻酸与牡丹甾醇相结合,其对于关节炎症的防治效果优异,其防治效果远远高于仅仅使用α亚麻酸或者是仅仅使用牡丹甾醇来辅助防治。
本发明的有益效果在于:
(1)本发明以组合物的方式,将牡丹甾醇和α亚麻酸有机结合起来,共同协同作用,使其应用于制备辅助治疗治疗类风湿炎症药物中之后,其效果显著;
(2)本发明所提供的组合物来源于天然的植物,相对于对人体有副作用的西药来说,本发明的产品具有无副作用、安全性高的特点;
(3)本发明的组合物原料来源丰富、成本低廉,易于推广应用及大规模生产。

本发明的牡丹甾醇、α亚麻酸微囊粉,由天宝牡丹科技有限公司提供;该公司所提供的牡丹甾醇异岩藻甾醇占比约为50~60%,燕麦甾烯醇占比约为15~21%,β-谷甾醇占比约为5~9%,角鲨烯约3~5%。
其它常见原料来源于市售产品。
实施例1
一种组合物,该组合物中有效成分为牡丹甾醇和α亚麻酸,牡丹甾醇和α亚麻酸的重量比例约为0.6:9.4;牡丹甾醇满足:其中的异岩藻甾醇占比为55.31%,燕麦甾烯醇占比为18.67%,β-谷甾醇占比为6.65%,角鲨烯4.43%,其余为其它的甾醇类或者是其它的物质。
将牡丹甾醇和α亚麻酸按上述的重量配比进行物理混合,获得组合物。
实施例2
一种组合物,该组合物中有效成分为牡丹甾醇和α亚麻酸,牡丹甾醇和α亚麻酸的重量比例约为0.75:9;牡丹甾醇满足:其中的异岩藻甾醇占比为53.46%,燕麦甾烯醇占比为17.38%,β-谷甾醇占比为5.72%,角鲨烯3.89%,其余为其它的甾醇类或者是其它的物质。
实施例3
一种组合物,该组合物中有效成分为牡丹甾醇和α亚麻酸,牡丹甾醇和α亚麻酸的重量比例为0.5:8;牡丹甾醇满足:其中的异岩藻甾醇占比为54.25%,燕麦甾烯醇占比为19.63%,β-谷甾醇占比为6.17%,角鲨烯4.65%,其余为其它的甾醇类或者是其它的物质。
实施例4
一种组合物,该组合物中有效成分为牡丹甾醇和α亚麻酸,牡丹甾醇和α亚麻酸的重量比例约为0.7:9;牡丹甾醇满足:其中的异岩藻甾醇占比为54.05%,燕麦甾烯醇占比为16.28%,β-谷甾醇占比为5.94%,角鲨烯4.37%,其余为其它的甾醇类或者是其它的物质。
实施例5
将实施例1中的产品制作为药片,具体步骤如下:
(1)将实施例1中的组合物与甘露醇混合,然后加入低取代的羟丙基纤维素、淀粉、环状糊精、乳糖,在掺混器中混合15分钟,获得最终混合物,将最终混合物加入到辊压机中干法制粒,获得颗粒;
(2)将(1)中所获得的物料使用直径为2mm的MUST冲头,通过压片机制备具有1~2kp的硬度和每10片约75mg重量的片状药物。
实施例6
将实施例1中的产品制作为颗粒剂药物,具体步骤如下:
取实施例1中的组合物,甘露醇、糊精、淀粉、糖份,将上述的原料混合均匀,加入蒸馏水,制成软材,过20目筛,烘干,过18目筛,分装,获得颗粒剂药物。
实施例7
将实施例1中的产品制作为胶囊型药物,具体步骤如下:
(1)将实施例1中的组合物与微晶纤维素混合均匀,在高速混合状态下向混合粉末中喷入浓度为95%的甘露醇,将所得的混合物用挤压机挤压破碎成颗粒物,干燥,再将颗粒物粉碎过80目筛;
(2)将淀粉、糊精与(1)中的物料混合,获得混合粉;
(3)将助硫剂与润滑剂与(2)中的混合粉在混合机中混合5~10分钟,获得最终混合物,作为胶囊内的填充物;
(4)将(3)中的填充物灌装到胶囊壳中,密封,获得胶囊剂的药物。
实施例8
利用实施例1中的组合物来制备粉末状的固体饮料,包括以下的步骤:
(1)将菊花浸膏置于粉碎机内粉碎,过40目筛,获得菊花粉末;
(2)将红糖放入粉碎机内粉碎,过80目筛;
(3)将菊花粉末和红糖、实施例1中的组合物输送至槽形混合机中,混合均匀,制作成软材;
(4)将软材输送至颗粒机,过10目筛,制作成湿颗粒,放入沸腾干燥机内通风干燥,得到干颗粒,过10目筛整粒备用。
实施例9
利用实施例1中的组合物来制备粉末状的液体饮料,包括以下的步骤:
(1)取实施例1中的组合物,再取甜味剂木糖醇,置于配液罐中,补充水,使其完全溶解,进行调配,混合并搅拌均匀,过滤,获得混合调配液;
(2)均质、灌装:将(1)中获得的调配液置于高压均质机中,在45~80℃、20~50MPa的条件下进行均质,均质后的液体进行灌装、封口;
(3)灭菌、检验:将封口后的液体在120℃的条件下灭菌15分钟,冷却,包装,获得液体饮料。
上述的饮料中,所加的甜味剂及水,可以视情况根据市场调查后的大众所喜口感进行调配,适当的时候可加入酸味剂、稳定剂、抗氧化剂。本发明所提供的仅仅是实施例1中组合物的应用总体的方法,并不侧重于保护某种具体的产品的生产工艺,而无论是上述的片剂、还是颗粒剂、胶囊剂的药物以及各种形态的食品,其工艺均为常规的工艺,因此,关于各物料的配比,在此并不详述。
实施例10
10.1临床实验:风湿或类风湿患者口服本发明实施例1中的组合物,每组共50例,其中男性15例,女性35例;年龄为50~70周岁;病程为1~15年;给药标准为:
对照组:市售的治疗风湿药物:布洛芬(按说明书中的剂量服用);
组别1:牡丹甾醇与α亚麻酸微囊粉在药物中的剂量均为每天10mg/kg;再与对照组中的药物结合服用;其中,牡丹甾醇和α亚麻酸的重量分别为0.6mg/kg:9.4mg/kg;
组别2:牡丹甾醇与α亚麻酸微囊粉在药物中的剂量均为每天3mg/kg;其中,牡丹甾醇和α亚麻酸的重量分别为0.1mg/kg:2.9mg/kg;再与对照组中的药物结合服用;
组别3:将牡丹甾醇按10mg/kg的量与对照组中的药物结合服用;
组别4:将α亚麻酸微囊粉按10mg/kg的量与对照组中的药物结合服用。
治疗方案:
疗效判断标准:根据2002年中化医学会风湿病学分会《类风湿关节炎诊治指南》中缓解标准:
(1)晨僵时间低于15分钟;
(2)无疲劳感;
(3)无关节痛;
(4)活动时无关节痛或关节压痛;
(5)无关节或腱鞘肿胀;
(6)血沉(魏氏法),女性小于30mm/h;男性小于20mm/h。符合5条或5条以上,并至少连续2个月诊断为临床缓解,有活动性血管炎,心包炎,胸膜炎,肌炎和近期无原因的体重下降或发热,则不能诊断为缓解。
疗效结果如下表所示:

注:治疗12周后,χ2=0.216(P>0.05);治疗24周后,χ2=0.431(P>0.05),差异无统计学意义。
从表1中的结果可以看出,组别1中的有效率最达,当经过24周的治疗之后,总的有效率达到了88%;
组别2中的牡丹甾醇与α亚麻酸微囊粉的用量仅为组别1中的1/3左右,结果表明其有效率较组别1要低;说明牡丹甾醇与α亚麻酸微囊粉的用量达到一定比例之后,对于风湿性疾病才具有显著的效果;
组别3中仅采用牡丹甾醇和布洛芬,组别4中仅采用α亚麻酸微囊粉和布洛芬,结果表明其有效率也远远的低于组别1。
对照组中,仅采用布洛芬,其有效率最低。这说明牡丹甾醇与α亚麻酸微囊粉对于预防和辅助治疗类风湿性关节炎中发挥了较大的作用。
10.2关于本发明的组合物应用后病患的血沉和类风湿因子等检查数据,如下:本发明人从各个组别中分别选择了一些病患的检测数据(50~70岁之间),具体如下:

注:正常人的血沉(ESR)参考值为:男0~15mm/h;女0~20mm/h;类风湿因子RF:阴性:(0~20IU/ml)
从以上表2~6中的血沉及类风湿因子检测数据可以看出,组别1中,患者经过12周、24周的药物治疗之后,血沉和类风湿因子均达到了正常值的范围;
而组别2~4中的效果较组别1要差。
对照组中的效果不太理想,这说明了将普通的治疗风湿或类风湿的药物与本发明的牡丹甾醇与α亚麻酸微囊粉相结合之后,其治疗效果优异。
类风湿性是一种自身免疫病,属于免疫失调引起的疾病。专家们认为,ω-3脂肪酸具有免疫调控作用,可抑制T细胞、B细胞等免疫活性细胞的活化、增殖,抑制细胞因子的产生,调节免疫应答,因此,ω-3多不饱和脂肪酸具有避免或减轻炎症作用。从以上的组4中可以看出,病患在服用α亚麻酸微囊粉之后,风湿类疾病的有效率相对于对照组显著提高。
植物甾醇是通过减弱炎症模型下的细胞活力,降低吞噬能力以及NO、TNF-α的生成从而减轻炎症的。TNF-α习称肿瘤坏死因子,它可以促进T细胞产生各种炎症因子,进而促进炎症反应的发生。它是一种主要的免疫调节和前炎性因子,因其在类风湿关节炎(RA)中的表现上调而被认为在其病理机制中发挥重要作用。在RA中TNF-α主要产生于RA滑膜中的滑膜巨噬细胞并刺激纤维母细胞增殖及淋巴细胞的活性。
本发明的组别1、2中有效率高于组别3、4,这说明牡丹甾醇与α亚麻酸微囊粉相结合,起到了协同增效的作用。分析原因,可能是牡丹甾醇能显著的降低炎症细胞的活力,而此时α亚麻酸中的ω-3脂肪酸抑制T细胞、B细胞等免疫活性细胞的活化、增殖,抑制细胞因子的产生。两者互相配合从而达到了协同增效的作用。
Claims (10)
1.一种组合物,其特征在于,所述的组合物中有效成分为牡丹甾醇和α亚麻酸;牡丹甾醇和α亚麻酸的重量比例为0.5~0.8:8~11。
2.如权利要求1所述的组合物在制备治疗关节炎症药物中的应用。
3.如权利要求1所述的关节炎症为风湿炎症或者是类风湿炎症。
4.如权利要求2~3中任一项所述的应用,其特征在于,所述的牡丹甾醇是以牡丹花粉为原料提取获得的;所述的牡丹甾醇满足:其中的异岩藻甾醇占比为50~60%,燕麦甾烯醇占比为15~21%,β-谷甾醇占比为5~9%,角鲨烯3~5%。
5.如权利要求4所述的应用,其特征在于,α亚麻酸是以牡丹籽油或者亚麻籽油、紫苏籽油中的任一种为原料制备获得的。
6.如权利要求4所述的应用,其特征在于,所述组合物在药物中的剂量为每天8.5~11.8mg/kg;其中,牡丹甾醇和α亚麻酸的重量为0.5~0.8 mg/kg:8~11 mg/kg;
优选的,组合物在药物中的剂量为每天10mg/kg;牡丹甾醇和α亚麻酸的重量分别为0.6mg/kg、9.4mg/kg。
7.如权利要求4所述的应用,其特征在于,所述的药物还包括药学上可接受的辅料。
8.所述的药物为注射剂、胶囊剂、片剂、颗粒剂、粉剂、气雾剂中的任一种;
如权利要求4所述的组合物在在制备具有预防风湿或类风湿炎症的食品中的应用。
9.如权利要求8所述的应用,其特征在于,所述的食品还包括食品添加剂;优选的为甜味剂、酸味剂、食用香精、稳定剂、抗氧化剂、微量元素、维生素类中的至少一种。
10.如权利要求9所述的应用,其特征在于,所述的食品为固态食品、液体食品、膏装食品、冻状食品、风干食品;
优选的,所述的食品为面条、面包、饼干、乳制品、罐头、膨化食品、糖果、饮料、烤鱼片中的任一种。
