Abstract
本发明涉及医药用品技术领域,具体的讲涉及一种牡丹细胞活性液及其制备方法,由下述成份重量百分比的牡丹籽油配置得到:牡丹皂苷25~30%、牡丹多糖10~30%、岩藻甾醇15~25%、@亚麻酸2~8%、亚油酸5~15%、维生素6~10%;本发明中牡丹籽提取物、牡丹花提取物、牡丹叶提取物、牡丹根提取物具有良好的生物容性,柔韧性,成膜性,并具有一定的抗菌、消炎、止血的作用,可促进伤口组织再生,修复和愈合;隔离伤口与细菌的混合,保护创面,缓解疼痛;本发明不含防腐剂,因为它是一个低水分含量的自然系统,是亲水性的,可以和和水以任意比例混溶,含有生理安全的磷脂和植物来源的多元醇,天然无刺激。

技术领域
本发明涉及医药用品技术领域,具体的讲涉及一种牡丹细胞活性液及其制备方法。
背景技术
随着生活水平的提高,人们越来越重视口腔的管理,并且已经开始认识到只有使用天然安全的原料才不会对口腔造成伤害,真正起到保护和修复的功效,围绕着天然、安全以及更有效的口腔原料的研究与开发奖能满足越来越高的要求。
但现有的各种保健牙膏、口腔保护液大多是以膏体或需清水复漱的液体形式存在,不仅使用场合和时间有限,使用不方便,而且在清水复漱口后,对牙齿及口腔的保健功能将会减低或消失,影响保健的效果,同时,功效单一,未能真正做到防止口腔细菌的产生,而仅是注重杀菌,预防和保健作用效果差。
口腔溃疡是常见的一类口腔粘膜病,据报道发病率在10%-30%。临床主要表现为口腔内散布的圆形或椭圆形的小糜烂面,周围充血水肿,患者常常自觉疼痛,影响进食、说话等日常生活行为。目前认为口腔溃疡的主要发病因素与机体免疫功能下降有关,另外内分泌因素、精神因素也会影响发病,糖尿病以及免疫功能低下者会厌恶溃疡愈合时间。遗憾的是,迄今为止,世界上还未有任何药物或方法能彻底根治口腔溃疡,人们只能通过不同的药物促进溃疡,人们只能通过不同的药物促进溃疡愈合、缩短病程、降低溃疡的发作频率或延长溃疡间歇期。
发明内容
因此本发明提出一种牡丹细胞活性液及其制备方法,填补现有技术的空白,提供一种具有细胞修复口腔溃疡及胃粘膜修复的细胞活性液,根据中医药理论,清除口腔疾病(牙龈炎、冠周炎、口腔黏膜炎)、睡眠不好、精神紧张、消化不良等造成的口臭,喷到炎症部位,减轻患者疼痛,缓解红肿充血症状仰制或杀灭致病菌。
本发明的技术方案是这样实现的:一种牡丹细胞活性液,由下述成份重量百分比的牡丹籽油配置得到:牡丹皂苷25~30%、牡丹多糖10~30%、岩藻甾醇15~25%、@亚麻酸2~8%、亚油酸5~15%、维生素6~10%。
进一步地,所述成份重量百分比的牡丹籽油配置得到:牡丹皂苷26~30%、牡丹多糖20~30%、岩藻甾醇26~23%、@亚麻酸4~8%、亚油酸5~10%、维生素7~9%。
进一步地,所述成份重量百分比的牡丹籽油配置得到:牡丹皂苷28%、牡丹多糖30%、岩藻甾醇22%、@亚麻酸6%、亚油酸6%、维生素8%。
一种牡丹细胞活性液的制备方法,取牡丹籽提取物、牡丹花提取物、牡丹根提取物,加乙醇回流提取2次,每次1h,提取浓缩液,牡丹籽油混合冰草、薄荷混合用纱布包好,放入萃取釜中,使用亚临界萃取方式;所述萃取温度恒定60℃,选取萃取压力为25MPa,乙醇浓度为95%,固定比6:1,萃取时间为4h,流体流速35kg/h,萃取结束后收集提取液。
通过上述公开内容,本发明的有益效果为:本发明中牡丹籽提取物、牡丹花提取物、牡丹叶提取物、牡丹根提取物具有良好的生物容性,柔韧性,成膜性,并具有一定的抗菌、消炎、止血的作用,可促进伤口组织再生,修复和愈合;隔离伤口与细菌的混合,保护创面,缓解疼痛;本发明不含防腐剂,因为它是一个低水分含量的自然系统,是亲水性的,可以和水以任意比例混溶,含有生理安全的磷脂和植物来源的多元醇,天然无刺激。

实施例1

实施例2

实施例3
一种牡丹细胞活性液,由下述成份重量百分比的牡丹籽油配置得到:牡丹皂苷30%、牡丹多糖12%、岩藻甾醇25%、@亚麻酸8%、亚油酸15%、维生素10%。
取牡丹籽提取物、牡丹花提取物、牡丹根提取物,加乙醇回流提取2次,每次1h,提取浓缩液,牡丹籽油混合冰草、薄荷混合用纱布包好,放入萃取釜中,使用亚临界萃取方式;所述萃取温度恒定60℃,选取萃取压力为25MPa,乙醇浓度为95%,固定比6:1,萃取时间为4h,流体流速35kg/h,萃取结束后收集提取液。
实施例4
本发明的各组成分及其重量配比是,牡丹籽提取物、牡丹花提取物、牡丹根提取物总提取物25~30%;牡丹籽油50~60%。
主要成分制备口腔细胞活性液,质量控制标准,并做观察,口腔修复液不被口腔中生产的细菌发酵利用,仰制链接菌生产及酸的产生;其次他能促进唾液分泌,减缓PH值下降,减少牙齿的酸蚀。
结果:制备工艺可行,质量控制方法简便、可靠,平均回收率99.96%。
结论:应用总有率97.67%,疗效确切,无不良反应。
牡丹籽提取物中含有油酸、亚油酸、亚麻酸等不饱和脂肪酸,在抗氧化能力和清除自由基方面有重要作用。皮肤衰老与自由基有密切关系,牡丹籽油能在一定程度上阻止自由基引起的过氧化反应,从而预防衰老和淡化色素斑渍;a一亚麻酸能促进皮肤吸收,从而增加皮肤弹性和光泽,可广泛应用于沐浴露、防晒霜、面膜等化妆品中,发挥其清除自由基、抗炎抗感染、抗辐射以及增强免疫力等作用。
牡丹花提取物含有人体必需脂肪酸、蛋白质、氨基酸、糖类、维生素C VC,VB1,VB2、类胡萝卜素、矿物质元素及微量元素(Cu,Fe,Zn,Mn,Co,Ni)等。它具有提高机体免疫力、预防动脉硬化、通经活血、滋养活肤等独特功效。
牡丹叶提取物活性成分有酸、酚、糖、蛋白质和黄酮等。牡丹叶中总黄酮的含量在1.47%~1.57%,其提取液对猪油和芝麻油均能起到抗氧化的作用,对猪油的影响略逊于芝麻油。
牡丹根提取物为菇类及贰类、酚及其贰类、生物碱、挥发油、植物甾醇、有机酸等,其中酚及酚贰类的含量相对略高。丹皮的主要活性成分为芍药普、丹皮多糖和丹皮酚等。芍药普具有抗心肌缺血、生血、抗肿瘤、抗过敏、抗炎等作用;丹皮多糖能够降血糖、具有预防及改善糖尿病及其并发症的作用。丹皮酚具有抗血栓、抗动脉粥样硬化、抗氧化、抗病毒等作用,菊花、蒲公英具有清热解毒之功效。
1、仪器
亚临界萃取流体萃取;高速万能粉碎机;紫外可见分光光度计、电动搅拌器、温度计、电子天平、电炉、水浴锅、旋转蒸发器。
2、方法及结果
制备工艺流程
根据药物的性质设计口腔细胞活性液的制备工艺,取牡丹籽提取物50g、牡丹花提取物50g、牡丹根提取物50g,加乙醇回流提取2次,每次1h,提取浓缩液,牡丹籽油混合冰草、薄荷混合用纱布包好,放入萃取釜中,开启亚临界萃取方式。设定实验条件:萃取温度恒定60℃,选取萃取压力为25MPa,乙醇浓度为95%,固定比6:1,萃取时间为4h,流体流速35kg/h,萃取结束后收集提取液。
2.1质量控制:形状本品为金黄色粘稠液体。
2.2无菌检测:取本品,按无菌检查法(中国药典1995年版二部)检查,应符合规定。
2.3本发明制品中可添加各种口感香液,诸如水果香型口感液等,以增加本发明制品的口感。
2.4本发明的使用方法是:每日早饭前,喷洒在口腔内,不用清水复漱,即可起到口腔消炎的作用。
3、质量检查:本制剂为金黄色,色泽均匀,味道清凉爽口,在口腔熔化时间为20min。
4、具体实施时,在一个利用随机取样、双盲并配合安慰剂控制的研究中,通过反复的清洗试验来观察牡丹籽油口腔修复液的产品效果,本实验采用三种配方,通过口腔的湿度、炎症的反应、分析不同配方对受损失的口腔的作用,得出结论:只要包含牡丹提取物的牡丹籽油口腔修复液的产品对受伤的口腔起到消炎、止血、修复、愈合的温度功效。
本发明中组合配合科学,天然安全,无副作用,各组成分之间协同作用,功效相符想成,且其制备方法有效保护了组方中有效成分的活性,使本发明具有细胞修复口腔溃疡及伤口创面的细胞活性液,喷到炎症部位,减轻患者疼痛,缓解红肿充血症状抑制杀灭致病菌。
最后说明的是,以上实施例仅用以说明本发明的技术方案而非限制,尽管参照较佳实施例对本发明进行了详细说明,本领域的普通技术人员应当理解,可以对本发明的技术方案进行修改或者等同替换,而不脱离本发明技术方案的宗旨和范围,其均应涵盖在本发明的权利要求范围当中。

1.一种牡丹细胞活性液,其特征在于:由下述成份重量百分比的牡丹籽油配置得到:牡丹皂苷25~30%、牡丹多糖10~30%、岩藻甾醇15~25%、@亚麻酸2~8%、亚油酸5~15%、维生素6~10%。
2.根据权利要求1所述的一种牡丹细胞活性液,其特征在于:所述成份重量百分比的牡丹籽油配置得到:牡丹皂苷26~30%、牡丹多糖20~30%、岩藻甾醇26~23%、@亚麻酸4~8%、亚油酸5~10%、维生素7~9%。
3.根据权利要求2所述的一种牡丹细胞活性液,其特征在于:所述成份重量百分比的牡丹籽油配置得到:牡丹皂苷28%、牡丹多糖30%、岩藻甾醇22%、@亚麻酸6%、亚油酸6%、维生素8%。
4.如权利要求1或2或3所述的一种牡丹细胞活性液的制备方法,其特征在于:取牡丹籽提取物、牡丹花提取物、牡丹根提取物,加乙醇回流提取2次,每次1h,提取浓缩液,牡丹籽油混合冰草、薄荷混合用纱布包好,放入萃取釜中,使用亚临界萃取方式。
5.根据权利要求4所述的一种牡丹细胞活性液的制备方法,其特征在于:所述萃取温度恒定60℃,选取萃取压力为25MPa,乙醇浓度为95%,固定比6:1,萃取时间为4h,流体流速35kg/h,萃取结束后收集提取液。
