The alcohol liposome contains ethanol in liposome, phospholipid is the main chemical component in the bilayer structure of the alcohol liposome, and the alcohol liposome contains relatively high volume fraction of ethanol, i.e. high concentration of low molecular weight alcohol is added on the basis of the liposome. The existence of ethanol enhances the flexibility of the liposome, and after the low molecular weight alcohol is added, the ethanol replaces the moisture near the head group of the lipid bilayer, thereby increasing and improving the mobility of the bilayer, and the ethanol molecule also changes the close arrangement of phospholipid molecules between skin cutin layers, promotes the drug to permeate skin barriers, and improves the transdermal absorption effect.
The ethosome can be fused with the cell membrane of the organism, and the wrapped content is transferred into the cell membrane; ethosomes are easily deformed to penetrate the various pores of the skin barrier, such as the phospholipid layer between the cuticles and the pilosebaceous glands, etc.; the ethosome is a zwitterionic surfactant, has oleophilic and hydrophilic properties, and can effectively encapsulate various compounds (fat-soluble and water-soluble molecules including protein polypeptide molecules) to penetrate to reach the deep layer of the skin. Therefore, it can be said that the flexible liposome and the ethosome have increased membrane flexibility, have increased deformability when crossing the skin barrier, are more suitable for crossing the keratinocyte gap, and are ideal carriers for carrying macromolecules.
3. The microemulsion has the characteristics of low viscosity, isotropy and thermodynamic stability, and is a transparent or semitransparent oil-water mixed system. The microemulsion can increase the solubility of the drug, increase the transdermal rate of the drug, improve the stability of the drug, prolong the action time of the drug, maintain constant blood concentration, and has simple preparation process and the like.
4. According to the scheme, two methods of the ethosome and the microemulsion for promoting the skin to absorb the drug are adopted, so that the absorption efficiency of the drug is further improved, the effect is fast, the pertinence is strong, and the treatment effect is good.
5. The coupling agent is creatively added into the gel heating agent, has the functions of coupling, conduction and permeation, so that the product is more closely contacted with the skin, the energy is efficiently and uniformly absorbed by the skin and tissues, the synergistic effects of coupling, conduction and permeation are achieved, the treatment speed is accelerated, the effect is quick, and the defects that the patch on the market generally adopts the traditional process and the simple skin permeation and absorption principle are overcome.
6. The heating bag can locally heat the affected part, dilate capillary vessels, and accelerate blood circulation and tissue metabolism; the couplant is applied, so that energy is efficiently and uniformly absorbed by skin and tissues, and the couplant has the effects of coupling, conduction and permeation; acting on skin and subcutaneous tissue, promoting molecular movement in cells, accelerating metabolism, promoting drug absorption, rapidly transporting to affected part, and improving rapid therapeutic effect.
