本发明属于水产养殖技术领域,尤其涉及一种防治马苏大麻哈鱼传染性造血器官坏死病的复方药物和方法。
背景技术
马苏大麻哈鱼(Oncorhynchus masou),鲑形目、鲑料、大麻哈鱼属,是重要的经济鱼类,在中国仅分布于绥芬河和图们江,目前已濒危。采用人工繁育鱼苗,进行增殖放流。马苏大麻哈鱼苗培育过程中,传染性造血器官坏死病的危害比较大,病原是一种弹状病毒(Rhabdovirus),该病是一种急性传染病,死亡率高达90%~100%,主要危害马苏大麻哈鱼稚鱼及幼鱼。近几年,马苏大麻哈鱼苗种培育规模逐渐增大,发病率也有增加的趋势。
现有方法对马苏大麻哈传染性造血器官坏死病的防治是用化学消毒剂在养殖马苏大麻哈水体中进行喷洒,不能有效杀灭鱼体内的病毒,还容易对马苏大麻哈鱼苗造成二次伤害,防治效率很低。有报道称,对于虹鳟种鱼的防治方法是注射传染性造血器官坏死病免疫灭活疫苗,对于虹鳟鱼有一定的效果,还没有应用于马苏大麻哈鱼,而且注射的方式操作是在每一条鱼的身上,鱼苗较小要做到精准注射,操作难度大、劳动量大而且繁琐,不易应用推广。
发明内容
本发明针对上述问题,提供一种马苏大麻哈鱼传染性造血器官坏死病的防治方法。操作简便、易行,经过一定规模的养殖实验,证明能有效提高马苏大麻哈鱼苗患传染性造血器官坏死病的存活率,使用本方法后的存活率经统计学方法检验效果显著。

一种防治马苏大麻哈鱼传染性造血器官坏死病的复方药物,由如下重量份的原料配制而成:金银花17%~23%、玄参15%~25%、野菊花12%~18%、苦楝叶14%~16%、白术7%~13%、干姜5%~15%、鱼腥草3%~7%、甘草2%~8%。
本发明还提供了一种马苏大麻哈鱼传染性造血器官坏死病的防治方法包括以下步骤:
步骤S1、在养殖马苏大麻哈鱼的池塘底部铺设U形纳米管,纳米管直径为15~30mm,用无油润滑空气压缩机充气增氧,无油润滑空气压缩机功率为1.5~3.5KW,管内空气流量0.1~0.6m3/hm,使养殖水曝气,保持水中溶解氧大于7mg/L。
步骤S2、蛋氨酸碘浸浴:将池塘水排出至水深0.2~0.5m,用蛋氨酸碘浸洗马苏大麻哈鱼,用量是1~2g/m3,浸洗时间是15~40min,浸洗后再将水注入池塘至水位不小于1~1.5m,发现病死鱼苗,及时捞出,深埋处理,防止病原传播。
步骤S3、养殖水消毒:将癸甲溴铵碘溶液稀释后全池泼洒,浓度是0.05~0.12g/m3(以癸甲溴铵计),每隔一天一次,连续2~5次。
步骤S4、在实施步骤S1~S3时,所投喂日粮饲料中添加维生素C和维生素E,维生素C日粮添加量是400~550mg/kg;维生素E日粮添加量是200~350mg/kg,日粮中连续添加这2种维生素投喂15~30d。
步骤S5、在实施步骤S1-S4时将金银花17%~23%、玄参15%~25%、野菊花12%~18%、苦楝叶14%~16%、白术7%~13%、干姜5%~15%、鱼腥草3%~7%、甘草2%~8%粉碎,均匀混合后制成复方药物,每千克鱼体重用上述复方药物5~10g添加日粮饲料中,连续投喂5~7d,作为一个疗程,间隔10~15d投喂一个疗程,连续3个疗程。
步骤S6、在实施步骤S5后,将五倍子、大黄粉碎后,用水煮沸20~30min,全池塘喷洒,五倍子、大黄在池塘水中浓度分别是5~8mg/L、1~2mg/L。
本发明通过S1-S6个步骤给马苏大麻哈鱼传染性造血器官坏死病养殖水体曝气增氧、蛋氨酸碘浸浴、养殖水用癸甲溴铵碘溶液消毒、饲料中投喂较高用量的维生素C和维生素E、复方药物以及全池喷洒五倍子、大黄的方法,实验结果说明上述用量和方法对马苏大麻哈鱼传染性造血器官坏死病有显著的防治效果。
本发明的马苏大麻哈鱼传染性造血器官坏死病的防治方法具有以下优点:改良了养殖水质,通过纳米管较大空气流通量水体的曝气和增氧的双重作用,降低了病鱼产生的有害产物,如氨、硫化氢、亚硝酸盐等有害物的浓度;癸甲溴铵碘溶液稀释后全池泼洒,消除了养殖水体中病原,对病患鱼体作了消毒处理;
日粮饲料中添加高剂量的维生素C和维生素E,增强了病鱼的机体免疫力;日粮饲料中添加金银花17%~23%、玄参15%~25%、野菊花12%~18%、苦楝叶14%~16%、白术7%~13%、干姜5%~15%、鱼腥草3%~7%、甘草2%~8%的复方药物,对病鱼病情有明显改善,患病鱼摄食量明显增大,日摄食率由0.16%提高到2.71%;存活率有显著提高,由10.2%提高到93.8%。

为了更好地了解本发明的目的、结构及功能,下面结合附图,对本发明一种马苏大麻哈鱼传染性造血器官坏死病的防治方法做进一步详细的描述。以下结合附图和具体实施例,对本发明作进一步描述:
实施例1:
1.在池塘底部设置纳米管1,呈U形铺设,纳米管直径为15mm,池塘外部设有1.5kw无油润滑空气压缩机2与池塘底部纳米管相连。池塘设有进水口4和排水口3。根据水中溶解氧量,开关无油润滑空气压缩机,纳米管内空气流量是0.1m3/hm,保持水中溶解氧量大于7mg/L。
2.蛋氨酸碘浸浴:将池塘水排出至水深0.2m,用蛋氨酸碘浸洗马苏大麻哈鱼,用量是1g/m3,浸洗时间是15min,浸洗后再将水注入池塘至水位1.0m,发现病死鱼,及时捞出,深埋处理,防止病原传播。
3.养殖水消毒:将癸甲溴铵碘溶液稀释后全池泼洒,使池塘水浓度为0.05g/m3(以癸甲溴铵计),每隔一天一次,连续2次。
4.在实施步骤1-3时,投喂日粮饲料中添加维生素C和维生素E,维生素C日粮添加量是400mg/kg;维生素E日粮添加量是200mg/kg,日粮中连续添加这2种维生素投喂15d。

实施例2:
1.在池塘底部设置纳米管1,呈U形铺设,纳米管直径为20mm,池塘外部设有3.0kw无油润滑空气压缩机2与池塘底部纳米管相连。池塘设有进水口4和排水口3。根据水中溶解氧量,开关无油润滑空气压缩机,纳米管内空气流量是0.3m3/hm,保持水中溶解氧量大于7mg/L。
2.蛋氨酸碘浸浴:将池塘水排出至水深0.4m,用蛋氨酸碘浸洗马苏大麻哈鱼,用量是1.5g/m3,浸洗时间是30min,浸洗后再将水注入池塘至水位1.3m,发现病死鱼,及时捞出,深埋处理,防止病原传播。
3.养殖水消毒:将癸甲溴铵碘溶液稀释后全池泼洒,使池塘水浓度为0.1g/m3(以癸甲溴铵计),每隔一天一次,连续3次。
4.在实施步骤1-3时,投喂日粮饲料中添加维生素C和维生素E。维生素C日粮添加量是500mg/kg;维生素E日粮添加量是300mg/kg,日粮中连续添加这2种维生素投喂20d。
5.在实施步骤1-4时将金银花20%、玄参20%、野菊花15%、苦楝叶15%、白术10%、干姜10%、鱼腥草5%、甘草5%粉碎,均匀混合后制成复方药物,每千克鱼体重用上述复方药物7g添加日粮饲料中,连续投喂6d,作为一个疗程,间隔12d投喂一个疗程,连续3个疗程。
6.在实施步骤5后,将五倍子、大黄粉碎后,用水煮沸25min,全池塘喷洒,五倍子、大黄在池塘水中浓度分别是7mg/L、1.5mg/L。
实施例3:
1.在池塘底部设置纳米管1,呈U形铺设,纳米管直径为30mm,池塘外部设有3.5kw无油润滑空气压缩机2与池塘底部纳米管相连。池塘设有进水口4和排水口3。根据水中溶解氧量,开关无油润滑空气压缩机,纳米管内空气流量是0.6m3/hm,保持水中溶解氧量大于7mg/L。
2.蛋氨酸碘浸浴:将池塘水排出至水深0.5m,用蛋氨酸碘浸洗马苏大麻哈鱼,用量是2g/m3,浸洗时间是40min,浸洗后再将水注入池塘至水位1.5m。发现病死鱼,及时捞出,深埋处理,防止病原传播。
3.养殖水消毒:将癸甲溴铵碘溶液稀释后全池泼洒,使池塘水浓度为0.12g/m3(以癸甲溴铵计),每隔一天一次,连续5次。
4.在实施步骤1-3时,投喂日粮饲料中添加维生素C和维生素E。维生素C日粮添加量是550mg/kg;维生素E日粮添加量是350mg/kg。日粮中连续添加这2种维生素投喂30d。
5.在实施步骤1-4时将金银花23%、玄参17%、野菊花16%、苦楝叶16%、白术8%、干姜6%、鱼腥草6%、甘草8%粉碎,均匀混合后制成复方药物。每千克鱼体重用上述复方药物10g添加日粮饲料中。连续投喂7d,作为一个疗程。间隔15d投喂一个疗程,连续3个疗程。
6.在实施步骤5后,将五倍子、大黄粉碎后,用水煮沸30min,全池塘喷洒,五倍子、大黄在池塘水中浓度分别是8mg/L、2mg/L。
验证效果:以本发明3个实施例作为实验组,以现有防治方法的2个池塘作为对照组。对照组和实验组选取池塘面积相同为长40m,宽20m的长方形池塘,马苏大麻哈鱼大小规格、患病数比例实验组和对照组无显著差异,放养密度相同条件下。
患病鱼具有传染性造血器官坏死病的典型特征:病变细胞核膜增厚,核染色质处于边缘位置,颗粒状,核内染色质减少,纵长形,一个细胞有两个或多个细胞核,细胞变圆形呈葡萄状堆聚。病鱼体黑色,眼球突出,腹腔积水膨大,鳍条基部充血;鳃及内脏无血色,口腔、骨骼肌、脂肪组织、腹膜、脑膜、鳔及心包膜有出血点,肠出血。治疗2个月后各组防治方法日摄食率见表1、各组防治方法存活率见表2,日摄食率由下式计算:
日摄食率=每日饲料摄入量/[(初始重量+终末重量)/2]
数据统计分析采用SPSS23.0软件处理,用单因素方差分析对数据进行显著性检验。所有分析取P<0.05为差异显著。

由表1说明,2个月的治疗,对照组日摄食率由0.27%降低到平均值的0.16%;三个实施例的实验组由治疗前的0.27%提高到平均值的2.71%。实验组日摄食率显著高于对照组日摄食率,表明实施例的方法有效恢复了鱼体生理代谢机能。

由表2可知,对照组和实验组在治疗前发病率基本相近的情况下,2个对照组平均存活率是10.2%;3个实验组平均存活率是93.8%,均显著高于对照组。
验证了本发明的防治方法对马苏大麻哈鱼传染性造血器官坏死病有较好的防治效果,并控制了病原体在患病鱼体到健康鱼的传播。
可以理解,本发明是通过一些实施例进行描述的,本领域技术人员知悉的,在不脱离本发明的精神和范围的情况下,可以对这些特征和实施例进行各种改变或等效替换。另外,在本发明的教导下,可以对这些特征和实施例进行修改以适应具体的情况及材料而不会脱离本发明的精神和范围。因此,本发明不受此处所公开的具体实施例的限制,所有落入本申请的权利要求范围内的实施例都属于本发明所保护的范围内。
Claims (9)
1.一种防治马苏大麻哈鱼传染性造血器官坏死病的复方药物,其特征在于,由如下重量份的原料配制而成:金银花17%~23%、玄参15%~25%、野菊花12%~18%、苦楝叶14%~16%、白术7%~13%、干姜5%~15%、鱼腥草3%~7%、甘草2%~8%。
2.一种防治马苏大麻哈鱼传染性造血器官坏死病的方法,其特征在于,包括以下步骤,且以下步骤顺次进行;
步骤S1、养殖水曝气,在养殖马苏大麻哈鱼的池塘底部铺设U形纳米管,用无油润滑空气压缩机充气增氧,使养殖水曝气,保持水中溶解氧大于7mg/L;
步骤S2、蛋氨酸碘浸浴:将池塘水排出至水深0.2~0.5m,用蛋氨酸碘浸洗马苏大麻哈鱼,浸洗后再将水注入池塘至水位不小于1~1.5m;
步骤S3、养殖水消毒:将癸甲溴铵碘溶液稀释后全池泼洒;
步骤S4、在实施步骤S1~S3时,所投喂日粮饲料中添加维生素C和维生素E;
步骤S5、在实施步骤S1-S4时将金银花17%~23%、玄参15%~25%、野菊花12%~18%、苦楝叶14%~16%、白术7%~13%、干姜5%~15%、鱼腥草3%~7%、甘草2%~8%粉碎,均匀混合后制成复方药物,并将其添加至日粮饲料中。
3.根据权利要求2所述的防治马苏大麻哈鱼传染性造血器官坏死病的方法,其特征在于,还包括步骤S6、在实施步骤S5后,将五倍子、大黄粉碎后,用水煮沸,全池塘喷洒。
4.根据权利要求2所述的防治马苏大麻哈鱼传染性造血器官坏死病的方法,其特征在于,所述步骤S1中,纳米管直径为15~30mm,无油润滑空气压缩机功率为1.5~3.5KW,管内空气流量0.1~0.6m3/hm。
5.根据权利要求2所述的防治马苏大麻哈鱼传染性造血器官坏死病的方法,其特征在于,所述步骤S2中,蛋氨酸碘浸浴:蛋氨酸碘浸洗马苏大麻哈鱼,用量是1~2g/m3,浸洗时间是15~40min,发现病死鱼苗,及时捞出,深埋处理,防止病原传播。
6.根据权利要求2所述的防治马苏大麻哈鱼传染性造血器官坏死病的方法,其特征在于,所述步骤S3中,养殖水消毒:以癸甲溴铵计,癸甲溴铵碘溶液浓度是0.05~0.12g/m3,每隔一天一次,连续2~5次。
7.根据权利要求2所述的防治马苏大麻哈鱼传染性造血器官坏死病的方法,其特征在于,所述步骤S4中,维生素C日粮添加量是400~550mg/kg;维生素E日粮添加量是200~350mg/kg,日粮中连续添加这2种维生素投喂15~30d。
8.根据权利要求2所述的防治马苏大麻哈鱼传染性造血器官坏死病的方法,其特征在于,所述步骤S5中,所述复方药物的添加量为每千克鱼体重使用复方药物5~10g,连续投喂5~7d,作为一个疗程,间隔10~15d投喂一个疗程,连续3个疗程。
9.根据权利要求2所述的防治马苏大麻哈鱼传染性造血器官坏死病的方法,其特征在于,在实施所述步骤S5后增加步骤S6,将五倍子、大黄粉碎后,用水煮沸20~30min,全池塘喷洒,五倍子、大黄在池塘水中浓度分别是5~8mg/L、1~2mg/L。
