原花青素(OPC)是一种有着特殊分子结构的生物类黄酮,由儿茶素、表儿茶素及表儿茶素没食子酸酯通过C4-C6或C4-C8连接而成的不同聚合度的混合物。原花青素水溶性好,极易被机体吸收,生物利用度高达90%以上,它是目前自然界植物来源中发现的抗氧化、清除自由基能力最强的物质之一。原花青素是主要存在于黑枸杞、葡萄籽、银杏叶、柏树、松树皮、黑米种皮、玫瑰花等多种植物中的天然成分。申请公布号为CN 108619423 A的专利公开了一种原花青素抗衰老组合物及其制备工艺及其制备工艺和应用。
紫檀芪(Pterostilbene)是白藜芦醇的甲基化衍生物。现代药理研究表明,紫檀芪具有多种药理活性,比如抗氧化、抗炎等。由于紫檀芪存在两个甲氧基,因此紫檀芪具有亲脂性和吸收好、生物利用度高的优点。紫檀芪是主要存于蓝莓和紫檀心材中的天然成分。
但目前并没有将原花青素、紫檀芪组合作为美白抗皱剂应用于化妆品和保健品的报道。
发明内容
本发明的目的是提供一种能够高效美白、抗皱的组合物,能够有效美白、抗皱,同时安全不致敏。
本发明将原花青素B2、紫檀芪复配,结果发现二者复配后具有抗皱,美白协同作用,同时安全不致敏,减少了单一成分的使用量,从而实现了本发明的目的。
本发明的复配型天然美白抗皱组合物,其含有原花青素B2和紫檀芪作为活性成分。
优选,所述的原花青素B2和紫檀芪按物质的量比30-70:70-30组合,优选为1:1。
进一步优选,所述的原花青素B2和紫檀芪的浓度都为15~30μM/L。
本发明的第二个目的是提供上述复配型天然美白抗皱组合物在制备食品、保健品、化妆品中的应用,特别是美白淡斑、抗衰老的护肤品中的应用。
本发明的全部原料均来源于天然植物,2种提取物在一定的比例下进行配伍,协同增强美白及抗氧化效果。通过组分中各天然提取物间存在的相互促进作用,协调增强其美白功效,使得该组合物具有美白保湿、抗衰老、抗皱纹、淡化色斑等功效。同时能快速改善肤质、提高皮肤弹性、平缓皱纹等。
本发明方法得到的天然植物源美白抗皱组合物性质稳定,应用于护肤产品中无刺激、抗过敏、美白淡斑、抗衰老。本发明制备方法简单,功效显著,具有很好的应用前景。

具体实施方式:
下面将通过实施例对本发明作进一步的描述,这些描述并不是对本发明内容作进一步的限定。本领域的技术人员应理解,对本发明内容所作的等同替换,或相应的改进,仍属于本发明的保护范围之内。
原花青素B2购自天津尖峰天然产物研究开发有限公司。紫檀芪为中国科学院华南植物园天然产物化学生物学实验室制备。
实施例1美白性能测定实验
酪氨酸酶是黑色素生成的关键酶,抑制酪氨酸酶的活性对延缓褐变和抑制黑色素形成具有重大意义,是美白皮肤的重要方法之一。在一定条件下,L-多巴能在酪氨酸酶的作用下生成多巴醌,该物质在475nm处有最大吸收峰,而底物L-多巴和酪氨酸酶吸收较小。若将待测样品加入其中,如果能够抑制酪氨酸酶的活性,势必会影响多巴醌的生成。多巴醌生成减少越多,说明待测物对酪氨酸酶的抑制作用越强,对酪氨酸酶抑制作用强就可作为美白的关键。
1、采用酶标仪来测定L-DOPA经催化生成的多巴醌的吸光度。按照表1所示,用移液枪向96孔酶标板中从左到右加入试剂(PBS和样品-原花青素B2、紫檀芪、阳性对照曲酸,其中样品的终浓度分别为如图1所示,用0.1%的DMSO溶液溶解),加入50uL酪氨酸酶溶液(200U/mL,用0.1mmol/LpH 6.8磷酸缓冲溶液溶解)后将酶标板置于37℃人工气候箱孵育10min后,再加入50uL底物1mmol/L的L-DOPA溶液(用0.1mmol/LpH 6.8磷酸缓冲溶液溶解),反应10min后,测定其在475nm处的吸光度。底物为L-DOPA。酪氨酸酶抑制率按下式计算。

试验结果如图1所示,图1是原花青素B2和紫檀芪抑制酪氨酸酶活性的曲线,原花青素B2和紫檀芪对酪氨酸酶活性抑制率的IC50分别为59.2±3.3μM/L、48.7±3.1μM/L,所以两者抑制酪氨酸酶活性基本相当,原花青素B2的活性略强于紫檀芪。阳性对照曲酸对酪氨酸酶抑制率的IC50为21.2μM/L,这说明原花青素B2和紫檀芪为很好的天然酪氨酸酶抑制剂。
2、协同增效作用的试验
参照1的酪氨酸酶抑制实验,样品为原花青素B2、紫檀芪、原花青素B2和紫檀芪的混合物,具体如表2所示,
原花青素B2和紫檀芪单独或联合应用的酪氨酸酶抑制率如表2所示。通过CompuSyn软件分析,得到CI。由表2可见,原花青素B2和紫檀芪联合应用后的抑制率均比单独使用的抑制率高,且CI均<1,这说明原花青素B2和紫檀芪在酪氨酸酶抑制能力上联合应用具有协同增效的作用。

实施例2抗皱效果检测和分析
皮肤作为人体最大的器官正是因为细胞外基质的丢失而产生衰老现象,据研究,皮肤厚度每十年大约减少6%。在皮肤的基质中,其中一种称为弹性蛋白酶的大分子纤维状蛋白,最为重要。皮肤出现松弛、皱纹、失去弹性等一系列老化现象大部分是由于弹性蛋白被降解或者变性,所以,可通过抑制弹性蛋白酶的活性来降低弹性蛋白的降解率,恢复皮肤的弹性、减少皱纹。
1、采用酶标仪用公知方法检测样品对弹性蛋白酶的抑制率。按照表3所示,在96孔板中加入样品溶液(原花青素B2或紫檀芪,终浓度如图2所示,溶剂为0.1%的DMSO溶液)和去离子水、130μL含有1.015mmol/L反应底物N-(甲氧基琥珀酰基)-L-丙氨酰-L-丙氨酰-L-脯氨酰-L-缬氨酸-4-硝基苯胺标准溶液的0.1mol/L Tris-Cl缓冲溶液(pH 8.0),在25℃下温育5min,加入15μL弹性蛋白酶溶液(0.5U/mL),继续在25℃下温育30min,然后测定410nm波长的吸光度(A410)。用去离子水替换样品水溶液,作为参比溶液同样测定吸光度(表3),按下式计算抑制率。

图2是原花青素B2和紫檀芪抑制弹性蛋白酶活性的曲线。原花青素B2和紫檀芪对弹性蛋白酶活性抑制率的IC50分别为75.7±3.8μM/L、73.2±3.1μM/L,所以两者都具有一定的抑制弹性蛋白酶的活性,是天然的弹性蛋白酶抑制剂。
2、协同增效作用
参照1的实验方法进行测试,只是样品如表4所示;
原花青素B2和紫檀芪单独或联合应用的弹性蛋白酶抑制率如表4所示。通过CompuSyn软件分析,得到CI。由表4可见,两种天然成分联合应用后的抑制率均比单独使用的抑制率高,且CI均<1,这说明原花青素B2和紫檀芪在弹性蛋白酶抑制能力上联合应用具有协同增效的作用。

实施例3:天然美白抗皱组合物的制备和应用
在一容器中,取0.5g亚麻籽胶、0.2g洋槐胶溶于25g去离子水中,加热到60℃得到粘稠的液体一;取另一容器,加入0.5g原花青素B2、0.25g紫檀芪、混匀,再加入10g丁二醇、2g戊二醇与3g己二醇,溶解,添加去离子水58.55g,混匀得到液体二;将液体二加入到液体一中,搅拌25min后降温,得到乳液,即为具有美白、修复效果的天然乳液。
上述制备的天然乳液,用于具有美白及抗皱功效的护肤品中,含有该天然乳液的美白抗皱精华液如表5所示:

将上述成分按其含量混合均匀,制得天然植物源美白抗皱组合物。
皮肤刺激性实验:以25名女性和25名男性为对象,50名志愿者早晚两次,涂抹0.05g天然植物源美白抗皱组合物于左前臂的正常皮肤上,连续测试七天,期间出现红肿、瘙痒、浮肿、丘疹均为产生过敏现象。
试验结果如表6所示:

通过本试验结果,可了解到实施例3的天然植物源美白抗皱组合物对皮肤无刺激,说明采用本发明的天然植物源美白抗皱组合物刺激性小,温和,安全性高。
为验证本发明效果,征集40名志愿者进行皮肤测试
测试对象:实验组为40名女性,年龄为25~45岁。所有志愿者每天使用所在组的实施例组合物,试验时间为一个月,在第30天时测试皮肤的弹性和美白程度。实验过程中不得使用其它任何护肤产品,而且受试者全部完成了验证过程,实验结果见表7。

从上表结果可以看出,本发明的天然植物源美白抗皱组合物的试用均产生较好的效果,由此可见本发明的天然植物源美白抗皱组合物可以有效的提高皮肤弹性与皮肤光泽度,能使使用者皮肤明显变白。本实验组的志愿者均无过敏或其它毒副反应。
上述实施例为本发明较佳的实施方式,但本发明的实施方式并不受上述实施例的限制,其他的任何未背离本发明的精神实质与原理下所作的修改、等同替换、改进等均为等效的置换方式,都包含在本发明的保护范围之内。
Claims (2)
1.一种复配型天然美白抗皱组合物,其特征在于,由原花青素B2和紫檀芪组成并作为活性成分,所述的原花青素B2和紫檀芪按物质的量比1:1组合,所述的原花青素B2和紫檀芪的浓度都为15~30μmol/L。
2.权利要求1所述的复配型天然美白抗皱组合物在制备美白淡斑、抗衰老的护肤品中的应用。
